Žmogaus virusų paviršiaus glikoproteinų ekspresijos tyrimas mielėse Pichia pastoris / Expression of human virus surface glycoproteins in yeast pichia pastoris

Vienas pagrindinių biomedicininės paskirties baltymų gamybos iššūkių yra pigių ir saugių ekspresijos sistemų, tinkamų glikoproteinų sintezei, paieška bei esamų sistemų tobulinimas. Vaistai, sukurti baltymų pagrindu, sudaro apie ketvirtadalį naujai patvirtinamų vaistų rinkos, o apie 60% jų sudaryti iš glikoproteinų. Dabar glikoproteinų sintezei naudojamos žinduolių kultūros turi keletą trūkumų. Jose gaunamų rekombinantinių baltymų kaina yra didelė, ribotas tūrinis našumas, ląstelės lėtai dauginasi ir auga, būna užkrėstos retrovirusais, gaunamas heterogeniškas produktas ir užima daug laiko sukurti stabilią ląstelių liniją. Itin intensyviai vykstantis tinkamų ekspresijos sistemų kūrimas kol kas nedavė norimų rezultatų. Mielių, kaip ir augalų bei vabzdžių, ekspresijos sistemos, dėl keletos priežasčių yra įvardijamos kaip vienos pagrindinių kandidatų užimti šią vietą. Visų pirma, mielės yra pripažintos kaip saugus organzimas, jų gentika, biochemija ir fiziologija yra gerai ištirta, be to, sėkmingai pradėti kurti rekombinantiniai mielių kamienai su sudėtingu žinduolių tipo N-glikozilinimu. Vis tik mielėse susintetintų glikoproteinų, tinkamų farmacijos pramonei, skaičius yra labai nedidelis, dažniausiai glikoproteinai nebūna tinkamai suvynioti ir modifikuoti, o esmininės priežastys, paaiškinančios mielių trūkumus sintetinant tokio tipo baltymus, nėra išaiškintos. Eukariotų genų inžinerijos laboratorijoje jau yra sukaupta nemaža patirties sintetinant mielėse virusinius glikoproteinus... [toliau žr. visą tekstą] / Growing market of the glycoprotein based drugs increases demands of safe, cheap and effective expression systems for production of glycoproteins. Mammalian cell cultures, which are being used for this purpose, are very expensive and ineffective. Yeasts are rising as one of the best alternatives. Well known genetics, biochemistry and physiology are only few advantages. Yeasts are also considered to be safe and easy to manipulate organism. Still, despite introducing humanized glycosylation pathways, yeast based expression systems are not able to produce glycoproteins for pharmaceutics, with a very few exceptions. So, further researches in adapting yeast for glycoproteins synthesis must be made. This work is directed for this purpose. In this work mumps, measles and influenza virus surface glycoproteins were expressed in yeast Pichia pastoris. Results show, that mumps virus hemagliutinin-neuraminidase is not synthesized in P.pastoris. Synthesis of measles virus (MeV) hemagliutinin (H) glycoprotein was not effective, with recombinant protein not possessing characteristics of native analogue. MeV-H was found in two forms: unglycosylated polypeptide precursor and glycosylated form, both aggregated and insoluble in non-ionic detergent. Increase in MeV-H expression level resulted in extensive accumulation of unglycosylated MeV-H protein precursors in the cytoplasm of yeast cells. Darbas susideda iš dviejų dalių: teorinio aprašo ir praktinio kūrybinio darbo. Teorinėje dalyje apžvelgiama darbo eigą, kūrybinio darbo idėjos paieška , priemonės ir atlikimo techniką, dailės terminai, abstrakcionizmas, abstrakcijos. Įsigilinama į tapybos esme. Tapomos abstrakcijos lyginamos su pirmojo abstrakcionizmo pradininko Vasilijaus Kandinskio darbais. The theoretical part also inquires into the heart of the matter.The painted abstarctions are compared with the works of Vasilij Kandinskij, the initiator of abstractionizm. Mehanizmi delovanja atrazina na steroidogenu aktivnost Leydig-ovih ćelija peripubertalnih pacova / The mechanism of atrazine action on steroidogenesis in peripubertal rat Leydig cells

<p>Rezultati prikazani u ovom radu opisuju efekte in vivo primene atrazina (2-hloro-4-<br />etilamino-6-izopropilamino-s-triazin) na ex vivo steroidogenezu u Leydig-ovim ćelijama<br />peripubertalnih pacova (tretiranih sa 50 mg/kg i 200 mg/kg telesne mase od 23. do 50.<br />dana starosti). Dobijeni rezultati jasno ukazuju da 28-dnevna in vivo primena atrazina<br />snažno inhibira testikularnu steroidogenezu, smanjujući ekspresiju gena za steroidogene<br />enzime i druge regulatorne proteine uključene u kontrolu testikularne steroidogeneze u<br />Leydig-ovim ćelijama peripubertalnih pacova. Rezultati in vivo primene atrazina<br />pokazuju da atrazin snažno inhibira ekspresiju gena za luteinizirajući hormon receptor<br />(LHR), skevendžer receptor-B1 (SR-B1), steroidogeni faktor-1 (SF-1), steroidogeni<br />akutni regulatorni protein (StAR), translokator protein (TSPO), fosfodiesterazu 4B,<br />3&beta;&minus;hidroksisteroid dehidrogenazu (&Eta;SD), CYP17A1 i 17&beta;HSD. Rezultati u okviru ovih<br />istraživanja pokazuju da primena atrazina tokom prepubertalnog perioda razvoja mužjaka<br />pacova dovodi do dozno-zavisnog smanjenja nivoa cAMP i snažne inhibicije androgeneze<br />u prisustvu hCG. Obzirom na blokadu ekspresije LHR, prvog elementa u aktivaciji<br />cAMP-signalnog puta, moglo bi se predpostaviti da je to uzrok blokade androgeneze kod<br />atrazinom-tretiranih životinja. Takođe, rezultati ukazuju na inhibiciju supstrat-stimulisane<br />produkcije androgena paralelno sa redukcijom ekspresije steroidogenih enzima CYP17A1<br />i 17&beta;HSD. U drugom delu ove doktorske disertacije, ispitivan je efekat direktne in vitro<br />primene različitih doza atrazina (1 nM, 1 &mu;M, 20 &mu;M, 50 &mu;M) na ekspresiju i aktivnost<br />steroidogenih enzima u kulturi preči&scaron;ćenih Leydig-ovih ćelija testisa peripubertalnih<br />pacova, pri čemu je zabeleženo stimulatorno dejstvo pomenutog herbicida. Naime,<br />zabeleženo je povećanje bazalne i hCG-stimulisane produkcije testosterona praćeno<br />povećanim nivoom cAMP u medijumu tretiranih ćelija. Pri ispitivanju ekspresije gena za<br />steroidogene enzime i regulatorne proteine, zabeleženo je povećanje ekspresije gena za<br />SF-1, StAR, CYP17A1 i 17&beta;HSD u hCG-stimulisanim uslovima. Takođe, povećana je i<br />produkcija testosterona nakon dodavanja progesterona i androstendiona kao supstrata,<br />kod Leydig-ovih ćelija tretiranih sa atrazinom. Da bi poku&scaron;ali da objasnimo za&scaron;to postoje<br />razlike u efektu atrazina u zavisnosti od načina primene, postavili smo jednokratni in vivo<br />eksperiment sa atrazinom (tretman sa 50 mg/kg i 200 mg/kg telesne mase, životinje<br />tretirane 50. dana starosti). Rezultati ovih eksperimenata ukazali su na up-regulaciju<br />testikularne steroidogeneze, kao i na povećan nivo cAMP kod životinja tretiranih sa<br />atrazinom Stoga, nivo cAMP se pojavljuje kao karika koja povezuje sva tri kori&scaron;ćena<br />eksperimentalna pristupa. Međutim, ostaje otvoreno pitanje na koji način atrazin utiče na<br />modulaciju nivoa cAMP i to pitanje predstavlja motiv za dalja istraživanja. Sumarno,<br />dobijeni rezultati ukazuju da 24-časovni tretman atrazinom izaziva povećanje, a<br />prolongirani tretman snažno smanjenje steroidogenog kapaciteta Leydig-ovih ćelija<br />peripubertalnih pacova.</p> / <p> In the present study, we investigated the effects of oral dosing of atrazine (2-chloro-4-<br /> ethylamino-6-isopropylamino-s-triazine) to peripubertal male rats (50 mg/kg and 200<br /> mg/kg body weight daily from postnatal day 23 to 50) on ex vivo Leydig cell<br /> steroidogenesis. In the presence of<br /> human chorion gonadotropin, the dose-dependent decrease in extra cellular cAMP level<br /> and accordingly strong inhibition of androgenesis were obtained. The transcription of<br /> LHR gene in Leydig cells of atrazine-treated rats was down-regulated in a dose-dependent<br /> manner, which could be the reason for reduction in cAMP level and expression of cAMPdependent<br /> genes. The results also indicated inhibition of substrate-stimulated androgen<br /> production in parallel with reduced expression of steroidogenih enzymes CYP17A1 and<br /> 17&beta;HSD. Obtained<br /> results indicated that 24 h-incubation of peripubertal Leydig cells in the presence of<br /> atrazine increased steroidogenic capacity of that cells. Increased basal and hCGstimulated<br /> testosterone production were accompanied by increasing levels of cAMP in the<br /> medium of treated cells. Also, in comparison to controls, gene expression revealed<br /> increased expression of SF-1, StAR, CYP17A1 and 17&beta;-HSD. When Leydig cells were<br /> challenged with progesterone and &Delta;4&ndash;androstenedione, testosterone production was<br /> increased in atrazine chalenged Leydig cells. To address these two opposite effects of<br /> atrazine we performed 24 h in vivo experiment in which peripubertal male rats (on<br /> postnatal day 50) were exposed to single atrazine treatment (50 mg/kg- and 200 mg/kgbody<br /> weight by gavage), and 24 h later, Leydig cells were isolated and testosterone levels<br /> in medium determined in basal and in hCG-stimulated conditions after 2 h-incubation<br /> period. Obtained results indicated that single in vivo exposure to atrazine was also<br /> accompanied 24 h later by up-regulation of Leydig cell androgenesis and increased cAMP<br /> level. According to the results obtained in this study, it seems that modulation of cAMP<br /> levels appear as a link that connects all three experimental approaches. However, the<br /> question of how atrazine affects the modulation of cAMP levels remains open, and<br /> present a motive for further research. In concluson, obtained results indicated that 24 h<br /> treatment with atrazine caused an increase, while prolonged treatment strongly reduce<br /> steroidogenic capacity of peripubertal Leydig cells.</p>

Fiziološki i molekularni aspekti tolerantnosti šećerne repe prema suši / Physiological and molecular aspects of sugar beet tolerance to drought

<p>Istraživanja su sprovedena na jedanaest genotipova &scaron;ećerne repe (Beta vulgaris ssp. vulgaris, L., označeni brojevima 1-11) koji su u poljskim uslovima ispoljili razlike u opservacionom testu nivoa tolerantnosti prema su&scaron;i (visoko, srednje i nisko tolerantni). U prvom delu eksperimenta biljke su gajene u polukontrolisanim uslovima, u stakleniku u supstratu koji je bio me&scaron;avina zemlje i peska. Biljke su svakodnevno zalivane tokom 90 dana, nakon čega je izazvan vodni deficit prestankom zalivanja, dok je kod kontrolnih biljaka zalivanje nastavljeno. Pet dana po prestanku zalivanja analizirani su parametri koji bi trebalo da ukažu na genotipske razlike u smislu tolerantnosti prema nedostatku vode. Utvrđen je sadržaj vode/suve materije u zemlji&scaron;tu i u biljnim tkivima (koren stablo i list). Lisna povr&scaron;ina i koncentracija pigmenata hloroplasta su ustanovljeni kod kontrolnih biljaka s ciljem utvrđivanja genotipskih specifičnosti. Osim toga, mereni su parametri fluorescencije hlorofila (F0, Fm, Fv, Fv/Fm i t1/2) kako bi se utvrdio efekat stresa izazvanog nedostatkom vode na inhibiciju transporta elektrona kroz PSII, i koncentracija slobodnog prolina, amino kiseline koja se nakuplja u uslovima stresa. U drugom delu eksperimenta ovi genotipovi su testirani u kontrolisanim, in vitro uslovima gajenja. Aksilarni izdanci su gajeni na podlozi za mikropropagaciju sa 0 (kontrola), 3 i 5% polietilen glikola (PEG 6000) pune četiri nedelje, a potom su vr&scaron;ene analize. Utvrđena je sveža masa izdanaka, sadržaj suve materije i koncentracija slobodnog prolina. U trećem delu eksperimenta upoređene su razlike u ekspresiji 13 kandidat-gena, koji su povezani sa reakcijom biljaka na uslove stresa, posebno su&scaron;e, u listovima biljaka gajenih u polukontrolisanim uslovima.<br />Biljke izložene stresu u polukontrolisanim uslovima gajenja su u proseku imale oko tri lista manje, za četiri procenta veći udeo suve materije i sedmostruko veći sadržaj prolina. Koncentracija prolina u listovima je bila vi&scaron;a u uslovima nedostatka vode u polukontrolisanim uslovima kod svih genotipova, a posebno kod nekih iz slabo tolerantne (2, 6) i visoko tolerantne (4) grupe.<br />U in vitro uslovima usled tretmanom PEG-om smanjila se ukupna suva masa i vi&scaron;e nego prepolovio broj aksilarnih izdanaka. PEG u koncentraciji od 3% je doveo do<br />povećanja ukupne sveže mase, a koncentracija prolina se povećala se porastom koncentracije PEG. Koncentracija prolina u uslovima stresa je kod oba eksperimenta bila značajno povećana u odnosu na kontrolu i to u in vitro uslovima &scaron;est puta, a u polukontrolisanim uslovima u stakleniku &scaron;esnaest puta u odnosu na odgovarajuće kontrole. Povećanje sadržaja prolina u biljkama gajenim u polukontrolisanim uslovima je bilo tri puta veće u odnosu na eksperiment sa PEG-om.<br />Koncentracija slobodnog prolina, kao jedan od potencijalnih parametara - pokazatelja tolerantnosti genotipova &scaron;ećerne repe prema su&scaron;i, je adekvatniji od ukupne suve mase. Rezultati su pokazali da je test u in vitro uslovima (posebno tretman 3% PEG-om) efikasiji za ocenu tolerantnosti prema su&scaron;i od eksperimenta u polukontrolisanim uslovima. Grupisanje genotipova prema nivou tolerisanja nedostatka vode na osnovu koncentracije prolina ustanovljene u ekperimentu in vitro dalo je isti rezultat kao i opservacioni test u poljskim uslovima.<br />Utvrđene su promene u ekspresiji kandidat-gena u uslovima su&scaron;e u odnosu na kontrolu, a ustanovljene su i razlike između genotipova. Jedan od analiziranih kandidat-gena može da posluži za dalji razvoj markera.<br />Ovi rezultati mogu da se primene u procesu oplemenjivanja &scaron;ećerne repe koje je usmereno na povećanje tolerantnosti prema ovom abiotičkom činiocu.</p>

Streso baltymo iHsp70 vaidmuo didinant triušio raumens kamieninių ląstelių atsparumą / Role of stress protein ihsp70 in the resistance of rabbit muscle-derived stem cells

Įvairios širdies ligos, kurių metu vyksta kardiomiocitų žuvimas, yra viena iš pagrindinių mirtingumo priežasčių visame pasaulyje. Regeneracinė ląstelių terapija, naudojant pirmines raumeninės kilmės ląsteles širdies ligų gydyme, sulaukia vis didesnio dėmesio. Dėl įvairių patologinių procesų, vykstančių pažeistame audinyje, didelė dalis transplantuotų ląstelių žūva. Todėl viskas, kas gali padėti transplantuotiems mioblastams išgyventi ir geriau prigyti, verta nuodugnesnių tyrimų. Šiame darbe buvo tirtas streso baltymo Hsp70 vaidmuo suaugusio triušio raumens kamieninių ląstelių įvairių viduląstelinių sistemų, atsakingų už ląstelių žuvimą ar išgyvenimą, reguliacijoje. Suaugusio triušio raumens kamieninės ląstelės buvo transfekuotos plazmide, turinčia hsp70 geną. Transfekuotų ląstelių padidėjusio atsparumo toksiniams poveikiams veikimo mechanizmai tirti lyginant jas su paprastomis kamieninėmis miogeninėmis ląstelėmis. Gauti rezultatai parodė, kad padidėjusi Hsp70 baltymo raiška apsaugo triušio raumens kamienines ląsteles nuo apoptozinio žuvimo po poveikio cheminėmis medžiagomis (naftazarinu, vandenilio peroksidu ir natrio prusidu) generuojančiomis reaktyvius deguonies darinius (ROS) ir sukeliančiomis oksidacinį stresą. Padidintas Hsp70 baltymo kiekis po poveikio osidacinį stresą indukuojančiomis medžiagomis keičia mitogenais aktyvinamos kinazės ERK 1,2, streso kinazės JNK trumpalaikį/pro-apoptozinį veikimą paprastose pirminėse miogeninėse ląstelėse į ilgalaikį/ani-apoptozinį... Regenerative stem cell therapy of damaged heart applying muscle-derived stem cells attains more and more attention. Patological environment in the damaged heart is major cause of death of newly transplanted muscle-derived stem cells. Therefore, everything increasing survival of transplanted myogenic cells is of research interest. This work has been designed to investigate role of stress protein Hsp70 in the regulation of intracellular systems responsible for the death or survival of primary myogenic cells. Rabbit muscle-derived stem cells were transfected with the plasmid carrying hsp70 gene. Various intracellular mechanisms responsible for the resistance of transfected cells have been investigated and compared with the regular non transfected primary myogenic cells. Our results show that Hsp70 protein protects rabbit muscle-derived stem cells from the toxic effect of compounds (naftazarin, hydrogen peroxide and sodium pruside) generating reactive oxigen species (ROS) and causing oxidative stress. Increased amount of intracellular Hsp70 protein converts transient/proapototic mode of action of ERK1,2 and JNK in regular primary myogenic cells to sustained/antiapoptotic action in transfected cells and significantly decreased total amount of JNK after the treatment with naftazatin. Klinički i prognostički značaj ekspresije gena EVI1 u akutnoj mijeloidnoj leukemiji / Clinical and Prognostic Significance of EVI1 Expression in Acute Myeloid Leukaemia

<p>UVOD: Akutna mijeloidna leukemija (AML) predstavlja heterogenu grupu oboljenja u odnosu na morfologiju, citogenetiku, molekularnu genetiku, zbog čega se deli na različite kliničke i biolo&scaron;ke entitete, sa različitim odgovorom na terapiju i ishodom lečenja. Humani EVI1 (ecotropic virus integration-1) gen ima ulogu multifunkcionalnog nuklearnog transkripcionog faktora, kako u normalnoj tako i u malignoj hematopoezi. Sve je vi&scaron;e istraživanja koja ističu negativni prognostički značaj visoke ekspresije (overexpression) EVI1 gena u AML.&nbsp; CILJEVI: Ciljevi ovog istraživanja su da se ispita klinički i prognostički značaj ekspresije gena EVI1 u AML, kao i da se utvrdi povezanost visoke ekspresije gena EVI1 sa nalazima citogenetskog ispitivanja i molekularnim markerima: FLT3 mutacijom i nukleofozmin 1 (NPM1) mutacijom. MATERIJAL I METODE: Ovim prospektivnim istraživanjem je obuhvaćena grupa od 38 odraslih novodijagnostikovanih bolesnika sa de novo, non M3 AML, kod kojih je započeto standardno lečenje, a koji su dijagnostikovani i lečeni u Klinici za hematologiju Kliničkog centra Vojvodine u periodu od jula 2012. do marta 2014. Određivanje ekspresije gena EVI1 je vr&scaron;eno pomoću real time kvantitativne PCR (qPCR) metode, tehnikom TaqMan, a relativna ekspresija EVI1 gena je određena primenom &Delta;&Delta;Ct metode.&nbsp; REZULTATI: Medijana starosti bolesnika pri postavljanju dijagnoze AML je bila 52 godine (23-80). Ustanovljena je statistički značajna razlika između ekspresije gena EVI1 kod zdravih osoba (kontrolna grupa) i obolelih od akutne mijeloidne leukemije (p=0.008). Računajući relativnu ekspresiju, 13,2 % bolesnika je imalo visoku ekspresiju (overexpression) gena EVI1. U odnosu na kliničke i laboratorijske karakteristike bolesnika (kao &scaron;to su pol, starost, parametri krvne slike, nivo laktat dehidrogenaze, procenat blasta u perifernoj krvi i ko&scaron;tanoj srži, potom tip akutne mijeloidne leukemije, performans status, komorbiditetni indeks) nije ustanovljena statistički značajna razlika između bolesnika sa visokom ekspresijom EVI1 gena i ostalih bolesnika. Postoji statistički značajna povezanost visoke ekspresije EVI1 gena i nepostojanja NPM1 mutacije (p=0,031), kao i između visoke ekspresije EVI1 gena i prisustva monozomije 7 (p=0,047). Visoka ekspresija EVI1 gena je povezana sa kraćim preživaljvanjem bez dogaĎaja (p=0,004), kao i sa kraćim ukupnim preživljavanjem (p=0,025).&nbsp; ZAKLJUČCI: Postoji značajno povećana ekspresija gena EVI1 kod obolelih od AML u odnosu na zdrave kontrole. Visoka ekspresija EVI1 gena je faktor lo&scaron;e prognoze kod obolelih od akutne mijeloidne leukemije i u kombinaciji sa drugim prognostičkim markerima, doprinosi boljoj risk stratifikaciji ovih bolesnika.</p> / <p>INTRODUCTION: Acute myeloid leukaemia (AML) represents a heterogenous group of diseases in terms of morphology, cytogenetics, molecular genetics, so it can be divided into distinct clinical and biological entities, with variable responsiveness to therapy and different treatment outcome. Human EVI1 (ecotropic virus integration-1) gene plays a role of multifunctional nuclear transcriptional factor, not only in normal, but also in malignant haematopoiesis. There are more and more investigations indicating high EVI1 expression (EVI1 overexpression) as a negative prognostic marker in AML.&nbsp; PURPOSES: The main goal of this investigation was to examine the clinical and prognostic significance of EVI1 expression in AML, as well as to investigate whether there was any association of EVI1 overexpression with cytogenetic abnormalities and other standard molecular prognostic factors, such as FLT3 mutation and nucleophosmin 1 (NPM1) mutation.&nbsp; PATIENTS AND METHODS: This prospective study included 38 adult newly diagnosed patients with de novo nonM3 AML, in whom a standard treatment was started at Clinic of Haematology, Clinical center of Vojvodina in the period from July 2012 to March 2014. EVI1 expression was analyzed by real-time quantitative polymerase chain reaction using TaqMan, and relative EVI1 expression was determined by &Delta;&Delta;Ct method.&nbsp; RESULTS: Median age of patients at diagnosis was 52 (aged 23-80). There has been determined statistically higher EVI1 expression in our AML patients than in healthy volunteers (control group) (p=0.008). The relative EVI1 overexpression was observed in 13.2% of the patients. No significant differences in clinical and laboratory patient data (including sex, age, whole blood counts, lactate dehydrogenase level, peripheral and bone marrow blast percentages, type of AML, performance status, comorbidity index) were observed between patients with high EVI1 expression and patients without high EVI1 expression. Our investigation revealed inverse correlation of high EVI1 expression and nucleophosmin 1 mutation (p=0,031). Also high EVI1 expression was significantly associated with monosomy 7 (p=0,047). Survival analysis revealed significantly inferior event free survival (p=0,004) and overall survival (p=0,025) for patients with high EVI1 expression compared to the other patients.&nbsp; CONCLUSION: EVI1 expression is significantly higher in AML patients compared to healthy controls. High EVI1 expression is a poor prognostic marker for patients with AML, and in combination with other well established prognostic markers, contributes to better risk stratification of these patients.</p>
