Foram utilizados 40 camundongos wild-type (WT) da linhagem C57BL/6. As lesões periapicais foram induzidas nos 1os molares inferiores e após os períodos experimentais de 7, 14, 21 e 42 dias, os animais foram mortos e as mandíbulas foram removidas. Como controles foram usados 1os molares hígidos. A mensuração das lesões periapicais foi efetuada em microtomografia computadorizada. A expressão do RNA mensageiro (mRNA) do EMMPRIN, MMPs -1, -2, e -9 foi determinada por meio de RT-PCR em tempo real. A presença, a localização e a expressão do EMMPRIN e das MMPs nas lesões periapicais foram avaliadas por imunohistoquímica. Os dados foram analisados por meio dos testes ANOVA, Bonferroni e correlação de Spearman com nível de significância de 5%. Os resultados mostraram que a área da lesão periapical aumentou significativamente dos 7 para os 14 dias (83%) e a partir de então houve estabilização. No grupo controle, o EMMPRIN foi expresso fortemente nas células da camada odontoblástica da polpa dentária. Nos períodos de 7, 14, 21 e 42 dias, foram observadas imunomarcações intensas para o EMMPRIN, MMPs -1, -2 e -9 na região adjacente ao forame apical, onde havia infiltrado inflamatório denso e ocasionalmente em outras regiões da lesão periapical. O EMMPRIN, MMP-1, -2 e -9 apresentaram forte expressão em células inflamatórias mononucleares e polimorfonucleares e marcação discreta em fibroblastos. A imunoexpressão do EMMPRIN na região periapical foi maior nos períodos de 7, 14, 21 e 42 dias comparado ao controle. A expressão do gene EMMPRIN apresentou-se maior no grupo controle em relação aos 7, 14, 21 e 42 dias... / This study evaluated the kinetics of expression and location of EMMPRIN and the correlation of its expression with expression of MMPs -1, -2 and -9 and with alveolar bone loss during the development of experimentally induced periapical lesions in mice. A total of 40 C57BL/6 wild-type mice (WT) were used. The periapical lesions were induced in first lower molars. After 7, 14, 21 and 42 days, the animals were killed and the mandibles were removed. Mice without pulp exposure served as controls. Measurement of periapical lesions was performed in micro-computed tomography. mRNA levels of EMMPRIN, MMPs-1, -2, and -9 were detected by real time RT-PCR. Presence, localization, and expression of EMMPRIN and MMPs were evaluated by immunohistochemistry. Data were analyzed using ANOVA, Bonferroni and Spearman correlation tests with significance level of 5%. The results showed that periapical lesion area was significantly increased from 7 to 14 days (83%) and thereafter become stable. In the control group, EMMPRIN was strongly expressed in odontoblastic layer of the dental pulp. At 7, 14, 21 and 42 days there was intense immunostaining for EMMPRIN, MMPs -1, -2 and -9 in the adjacent region of apical foramen, where there was dense inflammatory infiltrate. These proteins were expressed in other parts of the periapical lesion occasionally. EMMPRIN, MMP-1, -2 and -9 were intensely expressed in mononuclear and polymorphonuclear inflammatory cells and discretely expressed in fibroblasts. In the periapical region, the EMMPRIN immunostaining was higher at 7, 14, 21 and 42 days compared with control. The expression of EMMPRIN gene was higher in control group compared with 7, 14, 21 and 42 days. The expression of MMP-1 gene increased at 7 days and stays elevated until 42 days. COMPARAÇÃO DE DOIS MÉTODOS DE AFERIÇÃO DO DESGASTE DENTINÁRIO NA ZONA DE RISCO DE MOLARES INFERIORES / COMPARISON OF TWO MEASUREMENT METHODS OF DENTIN WEAR IN RISK ZONE ON MANDIBULAR MOLARS

The aim of this study was compared two measurement methods of dentin in wear risk of mandibular molars. The distal dentin thickness wall of the mesial canals, at 2 mm of ten molar furcation, was measured before and after the cervical preparation with CPDrill ® through the Cone Beam Tomography (apparent image) and through the Stereomicroscope (real image) in order to verify if the first provided trusted values, even though an examination by image. After analysis in Image Tool computer program, the results were subjected to statistical analysis of Mann-Whitney. The result of this study showed that there was no statistical difference between the values obtained (p ˃ 0.5) in Stereomicroscope and Cone Beam computed tomography. Within the methodology of this study was possible to conclude that the Cone Beam Tomography (Imaging examination) allows to evaluate the dentin wear as similar as real values. / O objetivo deste estudo foi comparar dois métodos de aferição do desgaste dentinário na zona de risco de molares inferiores. A espessura dentinária da parede distal dos canais mesiais, a 2 mm da furca de dez molares inferiores, foi mensurada antes e após o preparo cervical com a broca CPDrill®, por meio da Tomografia Cone Beam (imagem aparente) e por meio de Estereomicroscópio (imagem real) com a finalidade de verificar se o primeiro fornecia valores fidedignos, mesmo sendo um exame por imagem. Após análise no programa de computador Image Tool,os resultados foram submetidos à análise estatística de Mann-Whitney. O resultado deste estudo mostrou que não houve diferença estatística entre os valores obtidos (p˃ 0,5) no Estereomicroscópio e na Tomografia Computadorizada Cone Beam. Dentro da metodologia deste estudo foi possível concluir que a Tomografia Cone Beam (exame por imagem) permite avaliar o desgaste dentinário com valores similares aos valores reais.

INCIDÊNCIA DE DEFEITOS DENTINÁRIOS E FRATURA VERTICAL DA RAIZ APÓS RETRATAMENTO ENDODÔNTICO E CICLAGEM MECÂNICA / INCIDENCE OF DENTIN DEFECTS AND VERTICAL ROOT FRACTURE AFTER ENDODONTIC RETREATMENT AND MECHANICAL CYCLING

The aim of this study was to evaluate the incidence of dentin defects and Vertical Root Fractures (VRF) after endodontic retreatment and mechanical cycling. Methods: Two hundred monorradiculars premolars were divided into five groups. Forty teeth were left unprepared (control group). The other teeth were prepared with ProTaper files up to size F3 and filled with gutta percha and AH Plus. Eighty teeth were filled with the lateral compaction technique (LCT) and eighty teeth were filled with single cone technique (SCT). The filling material of forty teeth of each group was removed with ProTaper retreatment files and the canal was re-prepared with ProTaper files up to size F5 and filled with LCT (LCTR group) and SCT (SCTR group). All teeth were subjected to mechanical cycling (1.000,000 cycles, 130 N, 2.2 Hz and 37°C). The roots were sectioned horizontally at 3, 6 and 9 mm from the apex and observed under a 20X stereomicroscope. The defects were classified as: no defect‟, Vertical Root Fracture‟ and other defects‟. The difference between the group was analyzed using the Fisher‟s exact and Chi-squared tests. Results: The mechanical cycling alone did not promote dentin defects and VRF. The experimental groups presented higher dentin defects than the control group (P = 0.021). Retreatment groups did not present higher amounts of dentin defects that groups subjected to first treatment (P > 0.05). VRF was observed only in the SCTR group. Conclusion: Endodontic treatment and retreatment procedures that induce little dentin defects. The filling techniques do not seem to affect regarding the number of dentin defects in human premolars. / O objetivo deste estudo foi avaliar a incidência de defeitos dentinários e Fratura Vertical da Raiz (FVR) em dentes submetidos ao retratamento endodôntico e ciclagem mecânica. Métodos: Duzentos pré-molares monorradiculares foram divididos em cinco grupos. Quarenta dentes não receberam intervenção endodôntica (grupo controle). O restante dos dentes foram preparados com limas ProTaper até a lima F3 e obturados com guta percha e AH Plus. Oitenta dentes foram obturados com a técnica de compactação lateral (TCL) e oitenta dentes com a técnica do cone único (TCU). O material obturador de quarenta dentes de cada grupo foi removido com limas ProTaper para retratamento, o canal foi repreparado com limas ProTaper até a lima F5 e reobturado com TCL (grupo RTCL) e TCU (grupo RTCU). Todos os grupos foram submetidos à ciclagem mecânica (1.000,000 ciclos, 130 N, 2.2 Hz e 37 ° C). As raízes foram seccionadas horizontalmente em 3, 6 e 9 mm do ápice e observadas em estereomicroscópio com 20X de aumento. As fatias de dentina foram classificadas em: nenhum defeito‟, Fratura Vertical da Raiz‟ e outros defeitos‟. Os dados foram submetidos à análise estatística com nível de significância fixado em α = 0.05. A diferença entre os grupos foi analisada utilizando os testes, Exato de Fisher e Qui-quadrado. Resultados: A ciclagem mecânica por si só não provocou defeitos dentinários e FVR. Os grupos que receberam intervenção endodôntica apresentaram maior quantidade de defeitos dentinários que o grupo controle (P = 0.021). Os grupos submetidos ao retratamento (RTCL e RTCU) apresentaram similaridade com os grupos submetidos ao tratamento primário com relação à presença de defeitos dentinários (P > 0.05). FVR foi observada somente no grupo RTCU. Conclusão: O tratamento e retratamento endodôntico são procedimentos que induzem a poucos defeitos dentinários. As técnicas de obturação parecem não interferir nos resultados com relação ao número de defeitos dentinários e FVR em pré-molares monorradiculares humanos.
