Synthesis and analysis of Eriosema isoflavonoids and derivatives thereof.

Kraussianone 1 and kraussianone 2 were previously isolated as active compounds from the roots of Eriosema kraussianum Meisn., a plant used for the treatment of male impotence and urinary complaints in KwaZulu-Natal. The objectives of this study were firstly, to develop a method for the analysis of metabolites from E. kraussianum and other Eriosema plants that are used for erectile dysfunction and secondly, to develop synthetic methods for kraussianone 1 and structurally related compounds. A reversed-phase HPLC-PDA method was developed for the analysis of the extracts of plants from different sources, two of which were authentic E. kraussianum collected from the Drakensberg and Pietermaritzburg. The roots of other Eriosema species called ubangalala and uqonsi in Isizulu were also analysed. These plants were bought from the local herbal traders. The extracts of the two E. kraussianum plants and one uqonsi sample showed a similar chemical profile, even though there were variations in the relative concentrations of the metabolites within each plant. In these three plants, kraussianone 1, the most active metabolite of E. kraussianum, occurred in relatively low quantities, whereas kraussianone 2 was one of the major constituents. The other commercial plants that were analysed contained different compounds from those found in E. kraussianum. The HPLC method developed herein facilitates rapid identification and relative quantification of metabolites from E. kraussianum. Strategies based on semi-synthesis and total synthesis were employed for the preparation of kraussianone 1. The semi-synthetic route was based on the transformation of the prenyl side chain of kraussianone 2 into a linear dimethylpyran scaffold fused to the A-ring. Two routes were investigated for the semi-synthesis of kraussianone 1 from kraussianone 2. In the first route, the dimethylchromene ring was to be prepared by the acid-catalysed cyclisation of the prenyl group of kraussianone 2, followed by dehydrogenation of the resulting dimethylchroman chromophore. This route was abandoned due to poor regioselectivity of the cyclisation reaction and the difficulty of oxidising the dimethylchroman scaffold on the phloroglucinol moiety into a dimethylchromene. The second strategy involved selective protection of the OH-2', followed by DDQ-mediated oxidative cyclisation of the prenyl group to OH-7. This was the most viable route and kraussianone 1 was prepared in an overall yield of 54% from kraussianone 2. The total synthesis of kraussianone 1, on the other hand, employed the Suzuki-Miyaura reaction for the construction of the isoflavone nucleus and the regioselective introduction of the dimethylpyran scaffolds to the A- and B-rings. The key precursors in this synthesis were 3-iodo-5,7-dimethoxymethoxychromone and a boronic acid coupling partner, 7- benzyloxy-2,2-dimethylchromene-6-boronic acid, already bearing the prerequisite chromene scaffold attached to the B-ring. The isoflavones genistein, 2-hydroxygenistein, eriosemaone D and a geranyl analogue of kraussianone 1 were prepared via the route developed for the total synthesis of kraussianone 1 by structural modifications of rings A and B. Furthermore, this synthetic approach was expanded to the synthesis of the coumarochromones lupinalbin A and lupinalbin H. The development of the feasible semi-synthetic and total synthetic routes described herein for kraussianone 1 is of importance for the production of material for an in depth study of the pharmacological activities and the structure-activity relationship studies of kraussianone 1 and related compounds. / Thesis (Ph.D.)-University of KwaZulu-Natal, 2011.

Filogenia das espécies americanas de Rhynchosia Lour. (Leguminosae, Papilionoideae, Phaseoleae) e estudos taxonômicos do gênero no Brasil

Orientador: Ana Paula Fortuna Perez / Resumo: Rhynchosia Lour. possui distribuição pantropical, apresentando dois principais centros de diversidade para as cerca de 230 espécies, um na África (ca.140 spp.) e outro nas Américas, onde é registrada a ocorrência de 55 espécies, das quais 19 ocorrem no Brasil. O mais recente tratamento taxonômico do gênero foi publicado por Grear em 1978, que tratou apenas as espécies americanas. Neste trabalho, Grear (l.c.) propôs duas seções (R. sect. Copisma (E.Mey.) Endl. e R. sect. Arcyphyllum (Elliott) Torr. & A. Gray. Nenhum trabalho molecular foi realizado para as espécies americanas de Rhynchosia para apoiar esta classificação taxonômica. Estudos de filogenia molecular existentes enfocam a tribo e nestes trabalhos Rhynchosia é apontado como monofilético e mais relacionado com Eriosema, apesar da baixa amostragem utilizada destes gêneros. Do ponto de vista morfológico Rhynchosia é bastante uniforme, dificultando a delimitação das espécies. Pode ser caracterizado por apresentar folhas unifolioladas ou trifolioladas com estípulas persistentes ou caducas, inflorescências racemosas laxas ou congestas, com flores amarelas, cálice com cinco lobos bem desenvolvidos, e fruto do tipo legume, com duas sementes. Considerando o pouco conhecimento sobre as relações filogenéticas em Rhynchosia, enfatizando que nenhum trabalho molecular com as espécies americanas foi realizado, e a riqueza de táxons do gênero existentes no Brasil, um estudo taxonômico para as espécies de Rhynchosia no Brasil e a fil... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Rhynchosia Lour. has a pantropical distribution, with two main centers of diversity for its about 230 species, one in Africa (ca. 140 spp.) and the other in the Americas, where 55 species are recorded, of witch 19 occur in Brazil. The most recent taxonomic treatment of the genus was published by Grear in 1978, which treated only the American species. In this work, Grear (l.c.) proposed two sections (R. sect. Copisma (E. Mey) Endl. and R. sect. Arcyphyllum (Elliott) Torr. & A. Gray. No molecular work was done for the American Rhynchosia species to support this taxonomic classification. Existing molecular phylogenetic studies are those focusing on the tribe and in these works Rhynchosia is designated as monophyletic and more related to Eriosema, despite the low sampling used for these genera. From the morphological point of view Rhynchosia it is quite uniform, making it difficult to delimit the species. It can be characterized by the presence of unifoliolate or trifoliolate leaves with persistent or deciduous stipules, lax or congested racemic inflorescences, yellow flowers, calyx with five well developed lobes, and a legume fruit with two seeds. Tendo em vista a complexidade do processo inflamat?rio, o qual envolve a ativa??o de c?lulas do sistema imune e produ??o de diversos mediadores, o objetivo desse estudo foi realizar uma triagem fitoqu?mica e avaliar a poss?vel atividade anti-inflamat?ria, in vitro, do extrato diclorometano-etan?lico de ra?zes de E. crinitum, atrav?s da avalia??o do efeito do mesmo sobre a prolifera??o de linf?citos, a produ??o de citocinas e da ciclooxigenase 2 (COX-2). A triagem fitoqu?mica do extrato foi realizada por meio de rea??es cromog?nicas, fluorog?nicas e de precipita??o, seguidas da an?lise em cromatografia l?quida de alta efici?ncia acoplada a detector de arranjo de diodos (CLAE-DAD). A citotoxicidade do extrato sobre hem?cias, ap?s 4 horas de cultura, e sobre leuc?citos do sangue perif?rico humano, ap?s 24 horas ou 5 dias de cultura, foi avaliada por meio do teste de atividade hemol?tica e pelo m?todo de exclus?o com azul de Tripan, respectivamente. Essas mesmas culturas foram realizadas no tempo de 36 horas para an?lise da citocina IL-2, por ELISA. Para an?lise da produ??o das citocinas TNF-? e IFN-?, culturas de sangue total foram estimuladas com Miristato Acetato de Forbol (PMA) e tratadas com o extrato nas concentra??es de 50 ?g/mL, 25 ?g/mL e 12,5 ?g/mL. Essas concentra??es foram utilizadas em culturas de sangue total estimuladas com lipopolissacar?deo (LPS) para investiga??o do efeito do extrato sobre a express?o de COX-2. A produ??o de citocinas e de COX-2 foi analisada por citometria de fluxo. Os resultados evidenciaram a presen?a de terpenos e das subclasses dos flavonoides: flavonas e flavon?is no extrato. O extrato, nas concentra??es iguais ou inferiores a 50 ?g/mL, n?o foi t?xico para culturas celulares de 24 horas e, para culturas de 5 dias, as concentra??es n?o t?xicas foram iguais ou inferiores a 25 ?g/mL. Quanto ? a??o anti-inflamat?ria, o extrato n?o inibiu a produ??o das citocinas TNF-? e IFN-? e tamb?m n?o foi capaz de reduzir a express?o de COX-2. No entanto, o extrato inibiu a prolifera??o de linf?citos e suas subpopula??es T CD4 e TCD8, tendo uma efic?cia em rela??o ? Dexametasona de 71,65%, 53,14% e 167,94% para linf?citos totais, T CD4 e T CD8, respectivamente. O extrato tamb?m reduziu os n?veis de IL-2 nas culturas estimuladas com PHA. Esses achados sugerem que o extrato apresenta um efeito inibidor, principalmente sobre a prolifera??o de c?lulas T CD8, associado ? inibi??o na produ??o de IL-2. Don is used in Jequitinhonha Valley as anti-inflammatory plant in the form of decoction of its roots. Given the complexity of the inflammatory process, which involves the activation of immune cells and production of several mediators, the aim of this study was to perform a phytochemical screening and evaluate the potential anti-inflammatory activity, in vitro, the dichloromethane-ethanol extract of the roots of E. crinitum by assessing the effect of the same on lymphocyte proliferation, production of cytokines and cyclooxygenase-2 (COX-2). The phytochemical screening of the extract was performed using chromogenic reactions, fluorogenic and precipitation followed by analysis in high efficiency liquid chromatography coupled with a diode array detector (HPLC-DAD). The cytotoxicity of the extract after 24 hours or 5 days of culture on erythrocytes and leukocytes in human peripheral blood was evaluated by the test of hemolytic activity and the method of exclusion with Trypan blue, respectively. In evaluating of the effect of the extract on the proliferation of lymphocytes by flow cytometry, mononuclear cells from human peripheral blood (PBMC) have been incubated for 5 days in a culture medium containing the mitogen Phytohemagglutinin (PHA) in the presence or absence of extract at concentrations of 25 ?g/mL, 12.5 ?g/mL and 6.25 ?g/mL. These cultures too were performed in the time of 36 hours for analysis of IL -2 by ELISA. To analyze the production of cytokines TNF-? and IFN-?, whole blood cultures were stimulated with Phorbol Myristate acetate (PMA) and treated with the extract at concentrations of 50 ?g/mL, 25 ?g/mL and 12.5 ?g/mL. These concentrations were used in whole blood cultures stimulated with lipopolysaccharide (LPS) to investigate the effect of the extract on the expression of COX-2. Cytokine production and COX-2 was analyzed by flow cytometry. The results showed the presence of terpenes and subclasses of flavonoids: flavones and flavonols in extract. The extract, in concentration equal or greater than 50 ?g/mL was not toxic to cell cultures after 24 hours and in culture after 5 days, non- toxic concentrations were equal to or greater than 25 ?g/mL. In relation to the anti-inflammatory action, the extract did not inhibit the production of TNF-? and IFN-? and also was not able to reduce the expression of COX-2. Moreover, the extract inhibited the proliferation of lymphocytes and their subpopulations CD4 and CD8, and its efficacy compared to dexamethasone was 71.65%, 167.94% and 53.14% for total lymphocytes, CD4 and CD8, respectively. The extract also reduced the levels of IL-2 in cultures stimulated with PHA. These findings suggest that the extract has an inhibitory effect, especially on the proliferation of CD8 T cells associated with the inhibition of IL-2. Considering the presented results, the active molecules present in the plant seem to play their anti-inflammatory functions by regulating the T lymphocytes proliferation,?mainly T CD8 lymphocytes. This function could be exploited in inflammatory lymphoproliferative disorders, as well as in the prevention of transplant rejection. Further trials are needed to investigate the active constituents and the molecular mechanisms involved in the inhibitory action displayed by the extract.

G. Don var. macrostachyum (DC.) G. Don and E. tacuaremboense Arech. Eriosema heterophyllum stands as a new ocurrence for Santa Catarina. The second article is entitled: "The genus Rhynchosia Lour. (Leguminosae-Papilionoideae) in Paraná and Santa Catarina states, Brazil." Esta dissertação trata do estudo taxonômico de dois gêneros de Leguminosae nos estados do Paraná e de Santa Catarina. O primeiro artigo é intitulado "O gênero Eriosema (DC.) Desv. (Leguminosae- Papilionoideae) nos estados do Paraná e de Santa Catarina, Brasil".
