Regulatorische Foxp3+CD4+ T-Zellen sind essentiell für das Gleichgewicht des intestinalen Immunsystems. Eine Einschränkung ihrer Suppressionsfunktion wird bei Patienten mit Immune dysregulation, polyendocrinopathy, enteropathy, X-linked (IPEX)-Syndrom beobachtet und führt im Tiermodell zu lymphoproliferativen Erkrankungen und intestinalen Entzündungen. Von entscheidender Bedeutung für Homöostase und Suppressionsfunktion regulatorischer T-Zellen ist das Signalmolekül Interleukin-2 (IL-2). Im Gegensatz zu Effektor-T-Zellen exprimieren Foxp3+CD4+ T-Zellen den hochaffinen IL-2-Rezeptor αβγ konstitutiv. IL-2 wird von regulatorischen T-Zellen nicht in relevanten Mengen exprimiert. Sie sind somit auf von anderen Zellen sezerniertes IL-2 angewiesen. In der vorliegenden Arbeit wird gezeigt, dass im Tiermodell regulatorische Foxp3+CD4+ T-Zellen durch Applikation eines IL-2/anti-IL-2-Antikörperkomplex nicht nur in mesenterialen Lymphknoten und Milz, sondern auch lokal in der Lamina propria mucosae des Kolons der Versuchstiere expandiert werden. Als relevante Quelle von IL-2 in situ könnten aktivierte proliferierende T-Zellen dienen. Um dies näher zu untersuchen, wurde die Proteinexpression proliferierender Einzelzellen mittels Matrix assisted laser desorption/ionisation-Time of flight-Massenspektrometrie-Imaging (MALDI-Imaging) analysiert. Es gelang die Identifikation präferentiell in lymphoiden Geweben exprimierter Peptidmassen. Obwohl die Einzelzellanalyse mittels MALDI-Imaging prinzipiell möglich erscheint, ist ein Nachweis von Zytokinen wie IL-2 derzeit aufgrund fehlender Sensitivität im Proteinmassebereich zwischen 10kDa und 20kDa nicht möglich. Die therapeutischen Möglichkeiten der Expansion regulatorischer Foxp3+ T-Zellen durch stabile IL-2-Rezeptor-Agonisten und die Rolle von IL-2 für die intestinale Immunregulation sollten weiter untersucht werden.

Regulatory T cells (Treg) represent one of the most important mechanisms of immunoregulation. Treg suppress immune reactions and prevent overactivation of the immune system. There is a lot of ways of Treg action described, here we have focused on Treg interference with macrophages. The suppressor capacity of a highly purified Treg population was demonstrated in proliferation assays. The level of suppression of effector T cell proliferation differs depending on the presence of macrophages in the culture. Treg suppression has been significantly higher in the presence of macrophages. These observations led to hypotesis that Treg affect directly macrophages. However, using flow cytometry, reduction of expression of costimulatory molecules on macrophages after culture with Treg was not observed. Macrophages precultured with Treg showed a comparable functionality as macrophages cultured alone. Neither flow cytometry nor live cell imaging revealed any cytotoxic activity of Treg towards macrophages. Despite the presence of macrophages, Treg did not suppress effector cell proliferation in a model, where stronger activation of effector cells was induced. Mise au point de thérapies anti-tumorales impliquant des vecteurs parvoviraux et la fusion de cellules tumorales et dendritiques

L’immunothérapie anticancéreuse est basée sur la capacité du système immunitaire à reconnaître les cellules tumorales comme étrangères et à les éliminer. Les stratégies immunothérapeutiques abordées dans ce travail, incluent l’activation du système immunitaire par l’expression de facteurs immunomodulateurs (l’interleukine-2) via l’utilisation d’un vecteur dérivé du parvovirus MVM, ou par présentation des antigènes tumoraux par la machinerie des cellules dendritiques (DC), via la génération d’hybrides entre DC et cellules tumorales (TC). L’intérêt majeur du parvovirus autonome MVM en tant que vecteur pour la thérapie génique du cancer vient de son expression préférentielle dans les cellules transformées (oncotropisme) et de son aptitude à lyser celles-ci (oncolyse). Les vecteurs générés au laboratoire conservent l’unité de transcription NS et expriment l’IL2 humaine sous contrôle du promoteur P38, à la place des protéines de capside. Malgré les améliorations apportées à la production de vecteurs recombinants, la faible concentration des stocks reste un problème. Il a été montré que, de nombreux virus sont mieux produits en conditions de faible tension en oxygène (hypoxie). Nous avons tenté d’améliorer les titres des vecteurs en les produisant sous faible tension d’oxygène mais sans y parvenir (annexe 1). Dans un modèle in vivo utilisant la lignée de mélanome K-1735 dans des souris immunocompétentes, des cellules tumorales infectées in vitro avant leur implantation en sous-cutané ont montré un effet anti-tumoral du vecteur MVM/IL2 (annexe 2). Afin de mettre en évidence l’apport de l’oncolyse parvovirale dans l’activité anti-tumorale, nous avons mis au point des expériences, dans le même modèle de tumeur, visant à comparer l’efficacité du vecteur MVM/IL2 à celle d’autres vecteurs, Ad/IL2 et Rétrovirus/IL2, ne possédant pas d’activité oncolytique. Dans le but de mettre en évidence une éventuelle réponse immune in vivo, nous avons utilisé le modèle de tumeur TC-1 mais ce modèle s’est montré moins sensible à l’effet du vecteur MVM/IL2 et nous n’avons pas pu démontrer d’activation de cellules cytotoxiques spécifiques de la tumeur. Il a été proposé d’utiliser des hybrides entre DC/TC pour la vaccination anti-tumorale pour optimaliser la présentation des antigènes tumoraux. Une lignée cellulaire exprimant la protéine fusogène du virus de la leucémie du Gibbon (GaLV-FMG, Gibbon ape leukemia virus) a été dérivée de la lignée cellulaire CHO (cellules ovariennes de hamster chinois) au laboratoire. Cette lignée CHO-FMG, utilisée comme partenaire intermédiaire, a permis la fusion entre cellules tumorales et dendritiques (annexe 3). Nous avons montré que l’expression transitoire après infection par un vecteur AAV-FMG ou après transfection transitoire ne génère pas un pourcentage significatif d’hybrides. En effet, le niveau d’expression ainsi que le pourcentage de cellules transduites exprimant FMG s’est révélé trop faible. Ceci a mis en valeur l’efficacité de la lignée stable CHO-FMG comme intermédiaire de la fusion. De plus, nous avons intégré dans la lignée fusogène, le gène de l’interleukine-2, qui devrait permettre d’augmenter l’efficacité de l’induction de la réponse immune. L’activation des cellules effectrices de l’immunité anti-tumorale via la présentation antigénique et/ou par des cytokines est au centre de ce travail. Une meilleure compréhension des mécanismes sous-jacents devrait permettre d’améliorer ces systèmes de vaccination non-conventionnels.

A Novel in vitro PDE7 Inhibitor Inhibits IL-2 Gene Expression in Activated T Cells and Induces Apoptosis in a B-cell Line and Monocytic Cell Line

Chiles / Elevating intracellular cAMP levels can result in a wide range of anti-inflammatory effects and growth arrest and apoptosis in cancer cells, marking phosphodiesterases (PDEs) as potential targets for inflammatory diseases and cancer treatment. PDE7A is proposed to be a new therapeutic target for its ubiquitous expression in proinflammatory and immune cells. A Barbituric acid based compound, BC12 was identified as an in vitro PDE7 inhibitor in fission-yeast-based high-throughput screen. Analysis of this compound on the activation of Jurkat T lymphocytes, mouse and human primary T cells reveals inhibition of IL-2 production, though cell viability is not significantly impacted. Real-time RT-PCR and mRNA stability assays indicate that the inhibition of IL-2 production by BC12 is attributable to transcriptional repression without accelerating IL-2 mRNA decay. By contrast, compounds of similar structure with that of BC12 exhibit varying effects on IL-2 production that does not correlate with their in vitro PDE7 inhibitory activity, suggesting that the in vivo target of BC12 responsible for these effects may not be PDE7. Our study further reveals that BC12 inhibits IL-2 transcription through targeting NFAT and NFkB-mediated pathways. Preliminary investigation on other T helper cell cytokine secretion indicates that BC12 has a potential to selectively inhibit Th2 cytokines. Our data suggest that BC12 may present a novel anti-inflammatory drug for its immunosuppressive and potential immunomodulatory effects. Analysis of BC12 on a human B-cell line and a monocytic cell line demonstrate its pro-apoptotic effects in a dose-dependent manner. Titration of BC12 on human diffuse large B-cell lymphoma (DLBCL), LY18 cells, and human primary B cells reveals that BC12 induces cell death more effectively in DLBCL LY18 cells than normal B cells, suggesting the anti-cancer potential of this compound. / Thesis (PhD) — Boston College, 2013. / Submitted to: Boston College. The role of JAK1 and JAK3 in CD8⁺ effector T cells

The aim of this project was to explore the role of the tyrosine kinases JAK1 and JAK3 in cytokine signalling, focusing on interleukin-2 signalling in CD8<sup>+</sup> effector T lymphocytes. Initial experiments compared the effects of the pan JAK1/JAK3 inhibitor tofacitinib, the selective JAK1 inhibitor GSK186, and the selective JAK3 inhibitor GSK192 on IL-2 control of effector CD8+ cytotoxic T cells (CTL). On the basis of these preliminary data, a detailed analysis of the effect of tofacitinib on effector CD8<sup>+</sup> T lymphocytes was performed. Phosphorylation events regulated by tofacitinib were identified using mass spectrometry analysis of SILAC (stable isotope labelling with amino acids in cell culture) labelled CTL. Tofacitinib regulated a selective number of phosphorylation sites, with less than 1.2% of the CTL phosphoproteome significantly regulated by tofacitinib treatment following 4hrs tofacitinib treatment. Proteins with downregulated phosphorylation sites were enriched in functions related to the Jak-STAT signalling, regulation of gene expression, and MAPK signalling cascades. Proteins with upregulated phosphorylations were also enriched in functions related to regulation of gene transcription. The proteome of tofacitinib treated CTL was defined by label free mass spectrometry. Approximately 4.5% of the CTL proteome was significantly regulated following 24 hours tofacitinib treatment, suggesting tofacitinib regulates the expression of a selective subset of proteins. Tofacitinib treatment resulted in the downregulation of proteins involved in ribosome biosynthesis, steroid biosynthesis, regulation of transcription and the cell cycle; and the upregulation of proteins with hydrolase activity, and with roles in the lysosome and extracellular exosomes. The phosphoproteomic and proteomic data demonstrates that JAK kinase dependent IL-2 signalling regulates essential processes in CTL by controlling a selective number of phosphorylation events and proteins. Validation of proteins identified as regulated following tofacitinib treatment identified new targets of IL-2 signalling in CTL, including the transcription factor NFIL3. NFIL3 was shown to be upregulated in CD8<sup>+</sup> T lymphocytes following stimulation with IL-2 and regulated perforin and CD62L expression, suggesting a role in the regulation of CTL effector function.

Manipulation of effector and memory cd8+ T cells via IL-2-antibody complexes

Due to the growing burden of malignancy and chronic infections, manipulating CD8+ T cell responses for clinical use has become an important goal for immunologists. CD8+ T cells have the unique capacity to recognize and kill tumor cells and intracellular pathogens. Thus far, failed or only minimally effective T cell vaccines against chronic pathogen infections and tumors have highlighted basic knowledge gaps for eliciting memory CD8+ T cell protection. Defining the immunological mechanisms that determine protective capacity and longevity in T cells will be critical to both therapeutic and prohylactic vaccine efficacy. My studies focus on strategies to manipulate effector and memory CD8+ T cell responses, including their mechanisms of action. Specifically, I show that dendritic cell (DC) immunization coupled with relatively early (days 1-3) or late (days 4-6) administration of enhanced IL-2 signals drive either effector or memory programs. DC + IL-2c administered 4-6 days post-DC transfer is shown to enhance Ag-specific effector CD8+ T cell responses; this approach is further explored in the context of a cancer immunotherapy, demonstrating effective control of tumor burden in multiple murine models of cancer. Temporal alterations of IL-2 signaling from day 4-6 to day 1-3 post-DC immunization is shown to increase memory potential and memory CD8+ T cell numbers long-term. Additional studies reveal CTLA-4-mediated down-regulation of B7-ligands on DCs after IL-2c treatment, demonstrating that weaker or more transient signaling through the CD28-B7 axis may favor memory CD8+ T cell programs. My work contributes valuable concepts in memory CD8 T cell generation to develop T cell vaccines that are both safe and predictable.

Η πρωτεΐνη Tip60 (Tat-interactive protein, 60 kDa) είναι μέλος της οικογένειας πρωτεϊνών MYST και αρχικά προσδιορίστηκε ως αλληλεπιδρώσα πρωτεΐνη με την HIV-1 ΤΑΤ πρωτεΐνη. Πολλά από τα μέλη της οικογένειας MYST μεταξύ αυτών και η Tip60, δρουν ως ακετυλοτρανσφεράσες ιστονών, γεγονός που υποδηλώνει πιθανούς ρόλους στην αναδιαμόρφωση της χρωματίνης και στην γονιδιακή ρύθμιση. Στη παρούσα διπλωματική, στόχος ήταν η μελέτη του προφίλ έκφρασης της ακετυλοτρανσφεράσης Τip60 σε πληθυσμούς Τ βοηθητικών παρθενικών (CD3+CD4+CD45RA+) και Τ βοηθητικών μνημονικών (CD3+CD4+CD45RO+) λεμφοκυττάρων καθώς και η δράση της στην μεταγραφή του HIV-1 και της IL-2. Οι λόγοι που μας οδήγησαν στην μελέτη της Τip60 προκύπτουν από πρόσφατα αποτελέσματα του εργαστηρίου μας, που δείχνουν ότι υπάρχει μια πιθανή αλληλεπίδραση της Tip60 με τον παράγοντα Εts-2, ο οποίος συμμετέχει στη ρύθμιση της μεταγραφής τόσο του γονιδίου της IL-2 όσο και του HIV-1. Επιπλέον η IL-2 και ο HIV-1 παρουσιάζουν κοινή μεταγραφική ρύθμιση που ελέγχεται από την πρόσδεση μεταγραφικών παραγόντων σε κοινά cis στοιχεία. Επίσης, η Tip60 είναι μια Τat-αλληλεπιδρώσα πρωτεΐνη που κωδικοποιείται από το γονίδιο Tat του HIV-1. Τέλος το γεγονός ότι ο HIV-1 παραμένει σε λανθάνουσα κατάσταση στα εν ηρεμία Τ βοηθητικά λεμφοκύτταρα καθώς και ότι ο παράγοντας Εts-2, που πιθανόν αλληλεπιδρά με την Tip60, συμμετέχει στη καταστολή της έκφρασης του ιού, υποδηλώνει μια πιθανή συμμετοχή της Tip60 σε αυτή τη διαδικασία. Αρχικά ελέγξαμε την έκφραση της Tip60 σε υποπληθυσμούς μονοπύρηνων κυττάρων περιφερικού αίματος (PBMCs). Η υψηλότερη έκφραση παρατηρήθηκε στα Τ βοηθητικά λεμφοκύτταρα (CD4+). Για να εξακριβώσουμε το αν και πώς εκφράζεται στους υποπληθυσμούς των Τ λεμφοκυττάρων, απομονώσαμε Τ βοηθητικά μνημονικά λεμφοκύτταρα (CD3+CD4+CD45RO+) από ολικό αίμα ενηλίκων και Τ βοηθητικά παρθενικά λεμφοκύτταρα (CD3+CD4+CD45RA+) από αίμα νεογνών (ομφαλίου λώρου). Τα αποτελέσματά μας έδειξαν ότι σε συνθήκες μη ενεργοποίησης η Τip60 εκφράζεται περισσότερο στα μνημονικά Τh λεμφοκύτταρα σε σχέση με τα παρθενικά Τh λεμφοκύτταρα (στα οποία η έκφραση του παράγοντα παραμένει ουσιαστικά αμετάβλητη). Μετά από την ενεργοποίηση των κυττάρων με PMA/IONO (P/I) η έκφραση της Tip60 αυξάνεται σημαντικά στα μνημονικά Th λεμφοκύτταρα ενώ στα παρθενικά Th παραμένει ουσιαστικά αμετάβλητη. Για να εντοπίσουμε μια λευχαιμική κυτταρική σειρά κατάλληλη για να χρησιμοποιηθεί για πειράματα διαμολύνσεων και ανοσοφθορισμού, ελέγξαμε την έκφραση της Tip60 σε εννέα λευχαιμικές κυτταρικές σειρές και είδαμε ότι η Τ λεμφοκυτταρική λευχαιμική σειρά Jurkat παρουσίαζε την υψηλότερη έκφραση της Τip60. Η κυτταρική σειρά Jurkat παρουσιάζει επίσης παρόμοιο μοτίβο έκφρασης σε επίπεδο mRNA με τα Τ βοηθητικά παρθενικά λεμφοκύτταρα. Την έκφραση της Tip60 την ελέγξαμε και σε πρωτεϊνικό επίπεδο με τη μέθοδο Western blot σε ολικά πρωτεϊνικά εκχυλίσματα που απομονώσαμε από κύτταρα Jurkat. Για να ελέγξουμε τον εντοπισμό της Τip60 στα Jurkat, προχωρήσαμε σε πειράματα ανοσοφθορισμού. Τα αποτελέσματα έδειξαν, ότι σε συνθήκες μη ενεργοποίησης (CM) και ύστερα από ενεργοποίηση των κυττάρων (P/I) η Τip60 εντοπίζεται στον πυρήνα των κυττάρων Jurkat. Επίσης η ποσότητα της πρωτεΐνης Τip60 δεν αλλάζει ύστερα από την ενεργοποίηση των κυττάρων με μιτογόνα, αποτέλεσμα που συμφωνεί με τα πειράματα Western blot. Προκειμένου να ελέγξουμε τη δράση της Τip60 στη μεταγραφική δραστηριότητα του HIV-1 και της IL-2 προχωρήσαμε σε πειράματα διαμόλυνσης Jurkat κυττάρων. Αρχικά διαμολύναμε τα κύτταρα με αυξανόμενες ποσότητες πλασμιδίου υπερέκφρασης της Τip60 (pCMX-tip60) και ελέγξαμε την έκφραση της IL-2 σε μεταγραφικό επίπεδο. Παρατηρήθηκε ότι αυξανομένης της ποσότητας του pCMX-tip60, αυξανόταν και η έκφραση του IL-2 mRNA. Μέσω πειραμάτων CAT-assays σε κύτταρα Jurkat, τα οποία συνδιαμολύναμε με αυξανόμενες ποσότητες πλασμιδίου pCMX-tip60 και πλασμιδίου αναφοράς CAT, που βρίσκεται κάτω από τον έλεγχο του HIV-1-LTR, διαπιστώσαμε ότι η υπερέκφραση της Τip60 αυξάνει και τη μεταγραφική ενεργότητα του υποκινητή του HIV-1. Προκειμένου να διαπιστώσουμε εάν η Tip60 δρα απευθείας στον υποκινητή του HIV-1 και της IL-2 μέσω της πρόσδεσής της σε αυτόν είτε άμεσα, είτε έμμεσα μέσω συμπλόκου με άλλες πρωτεΐνες, προχωρήσαμε σε πειράματα ανοσοκρατακρήμνισης χρωματίνης (ChIP assays) του υποκινητή της IL-2. Παράλληλα επειδή δεν υπάρχει γνωστή θέση πρόσδεσης της Tip60 απευθείας στην LTR αλληλουχία του HIV-1 και στον υποκινητή της IL-2 αναζητήσαμε την πρωτεΐνη με την οποία θα μπορούσε να αλληλεπιδρά προκειμένου να προσδεθεί στα παραπάνω. Με πειράματα συνανοσοκατακρήμνισης διαπιστώσαμε ότι υπάρχει αλληλεπίδραση ανάμεσα στην Tip60 και στον παράγοντα Ets-2, ο οποίος προσδένεται άμεσα στην LTR αλληλουχία του HIV-1 και στον υποκινητή της IL-2 και έτσι επαληθεύσαμε τις αρχικές υποθέσεις για την αλληλεπίδραση τους. Τα πειράματα που πραγματοποιήσαμε μας οδήγησαν στα παρακάτω συμπεράσματα: Υπάρχει διαφορική έκφραση της Tip60 ανάμεσα στα Τ βοηθητικά παρθενικά και μνημονικά λεμφοκύτταρα, αφού σε συνθήκες μη ενεργοποίησης (CM) η έκφραση της Tip60 είναι μεγαλύτερη στα μνημονικά σε σχέση με τα παρθενικά Τh λεμφοκύτταρα. Η ενεργοποίηση των κυττάρων με μιτογόνα προκαλεί επαγωγή της έκφρασης της Tip60 στα μνημονικά Τh λεμφοκύτταρα, ενώ στα παρθενικά δεν προκαλεί καμία ουσιαστική αλλαγή. Η πρωτεΐνη Tip60 δρα σαν συνενεργοποιητής της μεταγραφής του ιού HIV-1 και της IL-2 μιας και προκαλεί επαγωγή στην μεταγραφική τους δραστηριότητα. Η πρωτεΐνη Tip60 αλληλεπιδρά in vivo με τον παράγοντα Εts-2 και παρουσιάζει το ίδιο πρότυπο πρόσδεσης με αυτόν, στον υποκινητή της IL-2 στην κυτταρική σειρά Jurkat. / Tip60 (Tat-interactive protein, 60 kDa) is a member of the MYST protein family and was initially identified as an interacting protein with the HIV-1 TAT protein. Several members of MYST family, Tip60 among them, act as histone acetyltransferases, suggesting their possible roles in chromatin remodeling and gene regulation. We chose to study Tip60 because it is a Tat-interactive protein and Tat is encoded by the Tat gene of HIV-1. Recent results from our laboratory suggest that there is a possible interaction between Tip60 and Εts-2 proteins, which is involved in the regulation of the transcription of both IL-2 and HIV-1. In addition, the transcription of IL-2 and HIV-1 is regulated by common transcription factors that act through binding to common cis-trans elements. We also know that the HIV-1 virus is transcriptionally active in activated CD4 T cells, and inactive in naïve CD4 T cells and that HIV-1 expression is blocked in naive Th cells by the transcriptional factor Ets-2. So the possible interaction between Tip60 and Εts-2 protein led us to study the Tip60 protein. The main purpose of this study was to determine the expression profile of Tip60 acetyltransferase in naive T helper (CD3+CD4+CD45RA+) and memory T helper (CD3+CD4+CD45RO+) lymphocytes and its effect on the transcription of HIV-1 and IL-2 (due to their common transcriptional regulation in T helper cells). The expression of Tip60 mRNA was measured in subpopulations of peripheral blood mononuclear cells (PBMCs) and the highest levels were observed in Th lymphocytes. We therefore isolated from human peripheral blood and cord blood, CD3+CD4+CD45RO+ and CD3+CD4+CD45RA+ Th lymphocytes and we examined the expression of Tip60 at the transcriptional level by RT-PCR. The results showed that Tip60 mRNA expression levels were higher in CD3+CD4+CD45RO+ compared to CD3+CD4+CD45RA+ Th cells. Following cell activation with PMA/IONO (P/I), the transcriptional expression of Tip60 was increased in memory Th cells, whereas it remained unchanged in naive Th cells. To identify a suitable cell line for transfection and immunofluorescence experiments, we measured Tip60 expression in nine cell lines and we observed that the T lymphocytic leukemia Jurkat presented the highest expression of Tip60 mRNA. Jurkat cells also presented a similar pattern of Tip60 mRNA expression levels with naïve Th lymphocytes. In addition to determine the localization of Tip60 into the cells, we proceeded in immunofluorescence experiments. As a result, we observed that when cells were cultured in CM media +/- P/I, Tip60 was identified in the nucleus of Jurkat cells. The amount of Tip60 protein remained unchanged after cell activation (which was also determined by Western blot experiments). To study the effect of Tip60 in transcription of HIV-1 and IL-2 we transfected Jurkat cells with increasing amounts of a Tip60 overexpressing vector (pCMX-tip60) and measured the expression of IL-2 and HIV-1 at the transcriptional level. Overexpression of Tip60, induced the transcription of IL-2 and also increased the transcriptional activity of HIV-1. Co-transfection experiments in Jurkat cells with increasing amounts of PCMX-tip60, led to a gradual increase of HIV1-LTR-CAT Finally, we studied if Tip60 acts directly on the promoter of HIV-1 and IL-2 exercising its effect directly or through complexing with other proteins. According to the literature there is no Tip60 binding site in the promoter of HIV-1 and IL-2, so we searched for a protein that could interact with Tip60. We performed co-immunoprecipitation experiments that showed that there was an interaction between Tip60 and Ets-2, a factor that binds directly both on the LTR of HIV-1 and the IL-2 promoter. In this work we suggest that Tip60 is expressed differentially in naïve and memory helper T cells and participates in the transcriptional activation of both HIV-1 LTR and IL-2. Also Tip60 protein interacts in vivo with Ets-2 protein and Tip60 binding activity is similar with Ets-2 binding activity to the ARRE-2 element of the IL-2 promoter in Jurkat cell line.

A triagem fitoqu?mica do extrato foi realizada por meio de rea??es cromog?nicas, fluorog?nicas e de precipita??o, seguidas da an?lise em cromatografia l?quida de alta efici?ncia acoplada a detector de arranjo de diodos (CLAE-DAD). Para an?lise da produ??o das citocinas TNF-? e IFN-?, culturas de sangue total foram estimuladas com Miristato Acetato de Forbol (PMA) e tratadas com o extrato nas concentra??es de 50 ?g/mL, 25 ?g/mL e 12,5 ?g/mL. Essas concentra??es foram utilizadas em culturas de sangue total estimuladas com lipopolissacar?deo (LPS) para investiga??o do efeito do extrato sobre a express?o de COX-2. A produ??o de citocinas e de COX-2 foi analisada por citometria de fluxo. Os resultados evidenciaram a presen?a de terpenos e das subclasses dos flavonoides: flavonas e flavon?is no extrato. O extrato, nas concentra??es iguais ou inferiores a 50 ?g/mL, n?o foi t?xico para culturas celulares de 24 horas e, para culturas de 5 dias, as concentra??es n?o t?xicas foram iguais ou inferiores a 25 ?g/mL. No entanto, o extrato inibiu a prolifera??o de linf?citos e suas subpopula??es T CD4 e TCD8, tendo uma efic?cia em rela??o ? Dexametasona de 71,65%, 53,14% e 167,94% para linf?citos totais, T CD4 e T CD8, respectivamente. O extrato tamb?m reduziu os n?veis de IL-2 nas culturas estimuladas com PHA. Don is used in Jequitinhonha Valley as anti-inflammatory plant in the form of decoction of its roots. Given the complexity of the inflammatory process, which involves the activation of immune cells and production of several mediators, the aim of this study was to perform a phytochemical screening and evaluate the potential anti-inflammatory activity, in vitro, the dichloromethane-ethanol extract of the roots of E. crinitum by assessing the effect of the same on lymphocyte proliferation, production of cytokines and cyclooxygenase-2 (COX-2). The phytochemical screening of the extract was performed using chromogenic reactions, fluorogenic and precipitation followed by analysis in high efficiency liquid chromatography coupled with a diode array detector (HPLC-DAD). The cytotoxicity of the extract after 24 hours or 5 days of culture on erythrocytes and leukocytes in human peripheral blood was evaluated by the test of hemolytic activity and the method of exclusion with Trypan blue, respectively. In evaluating of the effect of the extract on the proliferation of lymphocytes by flow cytometry, mononuclear cells from human peripheral blood (PBMC) have been incubated for 5 days in a culture medium containing the mitogen Phytohemagglutinin (PHA) in the presence or absence of extract at concentrations of 25 ?g/mL, 12.5 ?g/mL and 6.25 ?g/mL. These cultures too were performed in the time of 36 hours for analysis of IL -2 by ELISA. To analyze the production of cytokines TNF-? and IFN-?, whole blood cultures were stimulated with Phorbol Myristate acetate (PMA) and treated with the extract at concentrations of 50 ?g/mL, 25 ?g/mL and 12.5 ?g/mL. These concentrations were used in whole blood cultures stimulated with lipopolysaccharide (LPS) to investigate the effect of the extract on the expression of COX-2. Cytokine production and COX-2 was analyzed by flow cytometry. The results showed the presence of terpenes and subclasses of flavonoids: flavones and flavonols in extract. The extract, in concentration equal or greater than 50 ?g/mL was not toxic to cell cultures after 24 hours and in culture after 5 days, non- toxic concentrations were equal to or greater than 25 ?g/mL. In relation to the anti-inflammatory action, the extract did not inhibit the production of TNF-? and IFN-? and also was not able to reduce the expression of COX-2. Moreover, the extract inhibited the proliferation of lymphocytes and their subpopulations CD4 and CD8, and its efficacy compared to dexamethasone was 71.65%, 167.94% and 53.14% for total lymphocytes, CD4 and CD8, respectively. The extract also reduced the levels of IL-2 in cultures stimulated with PHA. These findings suggest that the extract has an inhibitory effect, especially on the proliferation of CD8 T cells associated with the inhibition of IL-2. Considering the presented results, the active molecules present in the plant seem to play their anti-inflammatory functions by regulating the T lymphocytes proliferation,?mainly T CD8 lymphocytes. This function could be exploited in inflammatory lymphoproliferative disorders, as well as in the prevention of transplant rejection. Further trials are needed to investigate the active constituents and the molecular mechanisms involved in the inhibitory action displayed by the extract.
