O presente projeto tem como objetivo principal identificar toxinas moduladoras de canais para sódio na peçonha do escorpião Tityus sp. identificando e caracterizando-as por meio de técnicas como Cromatografia líquida de alta eficiência (HPLC) e espectrometria de massa tipo MALDI-TOF/TOF. As sequencias parciais de aminoácidos dos peptídeos purificados foram determinadas por espectrometria de massa (MS/MS) para posteriormente ser testados por meio da técnica electrofisiológica de Patch clamp em canais de sódio expressos em células dissociadas de gânglios da raiz dorsal de ratos (DRG). A peçonha bruta do escorpião Tityus sp. resultou em 54 frações cromatográficas. Três sequencias parciais que correspondem a três NaScTxs foram obtidas, com massas moleculares de 7342 Da, 6971Da, 7970Da. Duas delas ao serem testadas nas células DRG reduziram as correntes de sódio classificando-as como βNaScTxs. ______________________________________________________________________________ ABSTRACT / The venom of scorpions are characterized by presenting pharmacologically active components, including toxins that act on sodium channels (NaScTxs), which are responsible of the strongest neurotoxic effects observed during poisoning caused by the bite of a scorpion. This project aimed to identify new toxins having sodium channels as molecular targets in the venom of the scorpion Tityus sp. identifying and characterizing them by standard techniques such as high performance liquid chromatography (HPLC) and mass spectrometry MALDI-TOF/TOF type. The partial amino acid sequences of the purified peptides were determined by mass spectrometry (MS/MS) to be subsequently tested by electrophysiological patch clamp technique on sodium channels expressed in dissociated cells from dorsal root ganglion of rats (DRG). The crude venom of the scorpion Tityus sp. resulted in 54 chromatography fractions. Three partial sequences corresponding to three NaScTxs were obtained with molecular weights of 7342 Da, 6971Da, 7970Da. Two of them to be tested in DRG cells reduced sodium currents classifying them as βNaScTxs.
