La phylogénie moléculaire du genre nord-américain Eurybia (Asteraceae : Astereae) et ses proches parents (Oreostemma, Herrickia, Triniteurybia)

Eurybia et ses proches parents Oreostemma, Herrickia et Triniteurybia sont appelés le grade des eurybioïdes. Comprenant 31 espèces vivaces, ce grade appartient au clade Nord-américain de la tribu des Astereae. Les analyses moléculaires antérieures ont montré que ce groupe est à la fois paraphylétique aux Machaerantherinae et un groupe frère aux Symphyotrichinae. Les relations infragénériques partiellement résolues et faiblement supportées empêchent d’approfondir l'histoire évolutive des groupes et ce, particulièrement dans le genre principal Eurybia. Le but de cette étude est de reconstruire les relations phylogénétiques au sein des eurybioïdes autant par l'inclusion de toutes les espèces du grade que par l’utilisation de différents types de régions et de méthodes d'inférence phylogénétique. Cette étude présente des phylogénies basées sur l'ADN ribosomal nucléaire (ITS, ETS), de l'ADN chloroplastique (trnL-F, trnS-G, trnC-ycf6) et d’un locus du génome nucléaire à faible nombre de copie (CNGC4). Les données sont analysées séparément et combinées à l’aide des approches de parcimonie, bayesienne et de maximum de vraisemblance. Les données ADNnr n’ont pas permis de résoudre les relations entre les espèces polyploïdes des Eurybia. Les analyses combinées avec des loci d’ADNnr et d’ADNnr+cp ont donc été limitées à des diploïdes. Les analyses combinées ont montré une meilleure résolution et un meilleur support que les analyses séparées. La topologie de l’ADNnr+cp était la mieux résolue et supportée. La relation phylogénétique de genres appartenant au grade des eurybioïdes est comme suit : Oreostemma (Herrickia s.str. (Herrickia kingii (Eurybia (Triniteurybia - Machaerantherinae)))). Basé sur la topologie combinée de l’ADNnr+cp, nous avons effectué des analyses de biogéographie à l’aide des logiciels DIVA et LaGrange. Ces analyses ont révélé une première radiation des eurybioïdes dans l’Ouest de l’Amérique du Nord, suivi de deux migrations indépendantes dans l’Est de l’Amérique du Nord chez les Eurybia. Due au relatif manque de variabilité de l’ADNnr, l’ADNcp et CNGC4, où le triage de lignés incomplet était dominant, l'origine du grade est interprétée comme récente, possiblement du Pliocène. La diversification du groupe a été probablement favorisée par les glaciations Pléistocènes. / Eurybia and it relatives, Oreostemma, Herrickia, and Triniteurybia, are collectively called the eurybioid grade. Comprising 31 perennial species, this grade belongs to the North American clade of the tribe Astereae. Early molecular analyses had inferred that this group is paraphyletic to the Machaerantherinae and sister to the Symphyotrichinae. The partially resolved and poorly supported relationships at the infrageneric level within the group, particularly within the core genus Eurybia, is preventing further insights into the evolutionary history of the group. The aim of this study is to reconstruct the phylogenetic relationships among the eurybioids by including all species of the grade and by using both different types of regions and multiple phylogenetic inference methods. The present study provides phylogenies based on nuclear ribosomal DNA (ITS, ETS), chloroplastic DNA (trnL-F, trnS-G, trnC-ycf6), and a low-copy nuclear locus (CNGC4), in separate and combined datasets analyzed using maximum parsimony, Bayesian and maximum likelihood approaches. In a separate analysis of the nrDNA dataset, the relationships of polyploids in Eurybia proved to be impossible to resolve. The nrDNA and nr+cpDNA combined analyses therefore were restricted to diploids. The combined analyses provided greater resolution and support than separate analyses. The nr+cpDNA phylogeny was the best resolved and supported. The phylogenetic relationship of genera belonging to the eurybioid grade is as follows: Oreostemma (Herrickia s.str. (Herrickia kingii (Eurybia (Triniteurybia – Machaerantherinae)))). Based on the nr+ cpDNA combined topology, we performed biogeographical analyses using DIVA and LaGrange. These analyses revealed an initial radiation of the eurybioids in western North America, with two independent migrations to eastern North America within Eurybia. Based on the relative lack of variation in nrDNA, cpDNA and CNGC4, where incomplete lineage sorting was dominant, the origin of the grade is interpreted as recent, probably from the Pliocene. Background. Esophageal cancer is ranked in the top ten of diagnosed tumors worldwide. Even though improvements in survival could be noticed over the last years, prognosis remains poor. ETS translocation variant 1 (ETV1) is a member of a family of transcription factors and is phosphorylated by mitogen-activated protein kinase (MAPK)-activated protein kinase 2 (MK2). Aim of this study was to evaluate the prognostic role of MK2 and ETV1 in esophageal cancer. Methods. Consecutive patients that underwent surgical resection at the department of surgery at the Medical University of Vienna between 1991 and 2012 were included into this study. After microscopic analysis, tissue micro arrays (TMAs) were created and immunohistochemistry was performed with antibodies against MK2 and ETV1. Results. 323 patients were included in this study. Clinical data was achieved from a prospective patient data base. Nuclear overexpression of MK2 was observed in 143 (44.3%) cases for nuclear staining and in 142 (44.0%) cases a cytoplasmic overexpression of MK2 was observed. Nuclear and cytoplasmic ETV1 overexpression was detected in 20 cases (6.2%) and 30 cases (9.3%), respectively. In univariate survival analysis, cMK2 and nETV1 were found to be significantly associated with patients' overall survival. Whereas overexpression of cMK2 was associated with shorter, nETV1 was associated with longer overall survival. In multivariate survival analysis, both cMK2 and nETV1 were found to be independent prognostic factors for the subgroup of EAC as well. Discussion. Expression of MK2 and ETV1 are prognostic factors in patients, with esophageal adenocarcinoma.

Financial Statement Disclosure of Carbon Footprint Costs in the Airline Industry

Unaccountable corporate polluters profit short term at the expense of global economic sustainability. The purpose of the study was to determine if carbon dioxide (CO2) penalties on the airline emissions would result in financial statement disclosure and emission mitigation. Contributing to environmental accounting, the study was based in corporate social responsibility with a conceptual framework based on economically-centered CO2 studies. A random sample of 69 global airlines, taken from the International Air Transport Association (IATA) and the International Civil Aviation Organization (ICAO) memberships, was stratified between EU bound and non-EU bound airlines. The research questions explored (a) the frequency mean differences in disclosed CO2 costs between the strata based upon the European Union's environmental trading scheme (EU-ETS) and (b) whether international financial reporting standards (IFRS) influenced the financial statement reporting of CO2 emissions costs. Financial statement data were analyzed in a 3-year longitudinal, ex-post, quasi-experimental, repeated measures factorial ANOVA and ANCOVA, pretest-posttest control group design. The results showed significant CO2 disclosure differences between the experimental (EU bound) airlines and control group (non-EU) airlines and for those airlines with IFRS prepared statements. These results should convince accounting practitioners that the quantification and reporting of greenhouse gas pollution can become the catalyst for improved operations and commercial sustainability. Positive social change to mitigate anthropogenic pollution should result and should promote normative accounting practice to hold those responsible to a higher global accountability.

Passive Smoking in Children : The Importance of Parents’ Smoking and Use of Protective Measures

Passive smoking has been recognised as a health hazard, and chidren are especially vulnerable. The general aim of this thesis was to describe and analyse the importance of parents’ smoking and smoking behaviour for children’s tobacco smoke exposure. The studies were conducted in the South-East part of Sweden and pre-school children and their parents constituted the study samples. Five studies are described in six papers. Smoking prevalence among parents (14%) and commonly used measures of protection were surveyed. An instrument designed to measure children’s tobacco smoke exposure in the home was developed and validated. It was used on 687 families with a smoking parent and a child 2½-3 years old, included in a prospective cohort study on environmental variables of importance for immun-mediated diseases ABIS (All Babies in South-East Sweden). Almost 60% of the parents stated that they always smoked outdoors with the door closed, 14% mixed this with smoking near the kitchen fan, 12% near an open door, 7% mixed all these behaviours and 8 % smoked indoors without precautions. The smoking behaviours were related to the children’s creatinine adjusted urine cotinine. All groups had significantly higher values than had children from non-smoking homes, controls. Outdoor smoking with the door closed seemed to be the best, though not a total, measure for tobacco smoke protection in the home. Most parents were aware of the importance of protecting children from tobacco smoke exposure but all were not convinced of the increased risk for disease for exposed children. The majority of parents were not satisfied with the smoking prevention in health-care and 50% did not think that their smoking was of any concern to the child health care nurse. Further research is warranted to describe if the difference in exposure score related to smoking behaviours is related to different prevalence of disease. Efforts are needed to convince those who still smoke indoors that tobacco smoke exposure influence children’s health and that consequent outdoor smoking with the door closed seemed to give the best protection. / Article I: copyright (2003), with permission from Oxford University Press. Type 1 Diabetes is a dangerous and life-long disease for which its prevalence is global. Research has shown that tight glycaemic control of this disease significantly reduces the risks of developing several life threatening diabetic complications. The Ets-Insulin-Bolus Calculator (EIBC), inspired by the Ets concept (Equivalent Teaspoon Sugar), was primarily designed to assist type I diabetics in improving their blood glucose control. The EIBC has shown to improve the average blood glucose level of type 1 diabetics. The need for this study however is to determine whether the ET!3C promotes tighter glycaemic control for type 1 diabetics based on a more-in-depth numerical analysis. With the use of the latest technology in blood glucose monitoring, the CGMS from Medtronic, mathematical models expressing and rating blood glucose control have been proposed and derived in this study. A clinical trial with type 1 diabetics has also been conducted. The use of the models together with the clinical trial results have shown that the EIBC does in fact promote tighter glycaemic control for type 1 diabetics. / Thesis (M.Ing.
