O gênero Brasiliorchis inclui atualmente 14 espécies de orquídeas restritas ao bioma Mata Atlântica, ocorrendo desde Misiones, Argentina, até o Estado da Bahia, na região nordeste do Brasil.Embora estas espécies tenham sido tradicionalmente reconhecidas pelos seus caracteres vegetativos e principalmente florais, elas são morfologicamente muito similares e apresentam caracteres diagnósticos contínuos,assim, ousoexclusivo da morfologiapode não ser útil para identificação das espéciesneste grupo de orquídeas. Um estudo morfométrico anteriormente realizado com seis espécies deste gênero não conseguiu identificar agrupamentos claros. Amaioria dos caracteres analisados apresentou sobreposição entre diferentes espécies, sendo que apenas a espécie B. gracilisse mostrou distinta das demais. O objetivo desse estudo foi contribuir para aidentificação de linhagens evolutivamente distintas dentro do gênero e avaliar se os padrões filogenéticos de diversificação corroboram com o estudo morfométrico anteriormente realizado.Foram utilizados dados de sequências de plastídios (psbj-petA, atpl-atpH)e nucleares (ITS1-2) considerandoampla variação geográfica e morfológica observada no grupo. Foram amostrados 96 indivíduos,originados de 37 localidades de ocorrência no Brasil, incluindo 11 espéciesdo gênero. Análises de máxima parcimônia e bayesiana foram realizadas considerando os dados separadamente e concatenados.Os resultados suportam B. schunkeana como irmã de todas as outras espécies do gênero. B. barbozaefoi reconhecida como um grupo monofilético distinto, corroborando com os caracteres morfológicos diagnósticos desta espécie, e possivelmente irmão das espécies do complexo B. picta.A espécie B. gracilis não foi reconhecida como um grupo monofilético, não corroborando com o estudo de morfometria, na qual essa espécie se apresentou distinta das outras do complexo. Contudo, nas análises concatenadas alguns indivíduos apresentaram-se como irmãos das espécies do complexo B. picta. As espécies morfologicamente homogêneas pertencentes ao complexo B. picta (B. chrysantha, B. marginata, B. porphyrostele, B. ubatubana, B. phoenicanthera e B. picta) não formaram clados monofiléticos distintos. B. kautskyi e B. consanguinea, as quais são morfologicamente distintas, foram incluídas nos clados constituídos pelas espécies do complexo B. picta.Eventos de introgressão e hibridação podem ser potenciais causas da insuficiente resolução das árvores filogenéticas obtidas nesse estudo. Although these species have been traditionally recognized by vegetative andmainly floral traits, they are morphologically very similar and display continuouscharacters, thus the exclusive use of the morphologycannot be useful for species identificationin this group of orchids. A morphometric study previously conducted with six species of this genus could not identify clear groupings. The majority of analyzed characters showed overlap between different species, with only the species B. gracilisbeing distinct from the others. The goals of this study were to contribute to the identification of evolutionary distinct lineages within the genus and assess if the phylogenetic patterns of diversification agree with a previous morphometric study available. Sequence data from plastid (psbj-petA, atpl-atpH) and nuclear (ITS1-2)regions were used considering wide morphological and geographical sampling variation. We sampled 96 individuals, originated from 37 sites of occurrence in Brazil, including 11 species of the genus. Bayesian and parsimony analyses were performed considering data separately and concatenated. The results support B. schunkeana as sister to all other species of the genus. Revision of Juncaceae and Cyperaceae phylogeny based on cpDNA and nDNA

The phylogeny of Juncaceae and Cyperaceae is still not fully understood. The morphology of this group is intricate and without clear homology, and molecular relationships are mostly studied on smaller parts of this large group of Monocots. Proper study is therefore needed. To obtain the most objective view on the phylogeny of these two families, we gathered data on 1174 taxa of rbcL, trnL-trnF, and ITS and analysed them by maximum parsimony, maximum likelihood, and Bayesian inference. Markers of cpDNA appeared much more useful than nDNA (ITS) due to the high rate of mutations in ITS which led to homoplasy and unsure alignment. By considering taxonomical impact of our study, the monophyly of the families and main inner topology of Juncaceae and several tribes of Cyperaceae (Abildgaardieae, Bisboeckelereae, Cariceae, Cypereae, Cryptangieae, Eleocharideae, Rhynchosporeae, Sclerieae, and Trilepideae) were confirmed; some changes in taxonomy were suggested (uniting of Chrysitricheae and Hypolytreae; division of Fuireneae; uniting of Cariceae, Dulicheae, Scirpeae, and Khaosokia caricoides into one tribe, or division of Scirpeae; and separation of Cladieae); changes in the classification of certain taxa were also suggested (the transfer of Distichia, Marsippospermim, Oxychloë, Patosia, and Rostkovia into Juncus, or division of Juncus into more genera; the transfer of Nemum spadiceum into Bulbostylis; Schoenoplectus corymbosus, S. gemmifer, S. hondoensis, and S. multisetus into the genus Schoenoplectiella; and Oreobolopsis into Trichophorum); and the identification of some taxa which possess special combinations of molecular and morphological features and should be studied further was made (Juncus capitatus, J. dregeanus, Bulbostylis juncoides, Crosslandia setifolia, Schoenoplectus litoralis, S. americanus, Cyperus iria, and Amphiscirpus nevadensis).

Speciation, Species Concepts, and Biogeography Illustrated by a Buckwheat Complex (Eriogonum corymbosum)

The focus of this research project is the complex of infraspecific taxa that make up the crisp-leaf buckwheat species Eriogonum corymbosum (Polygonaceae), which is distributed widely across southwestern North America. This complex provides an ideal taxonomic group for research into population relationships and speciation. To avoid unnecessary debates about taxonomic validity or contentious issues regarding appropriate species definitions, the historical evolution of the species concept is first reviewed in detail, demythologizing an often-assumed species problem. Following that review, the E. corymbosum complex is examined specifically. Although eight varieties of E. corymbosum are currently recognized based on morphological characters, this group of large, woody shrubs has a history of revisions that demonstrates the uncertainty inherent in circumscriptions based on morphology alone. The apparent rarity of some E. corymbosum varieties also presents conservation and management challenges, demonstrating the need for taxonomic verification. To bring greater resolution to this group, I genetically tested samples from populations of six of the eight varieties of E. corymbosum, as well as a number of related buckwheat species. With 103 AFLP loci and chloroplast sequence data from 397 samples, I found strong support for the designation of the recently named E. corymbosum var. nilesii. This predominantly yellow-flowered variety had previously been considered part of a more common variety, and thus its management had not been of particular concern. But as a separate variety, its known distribution is quite limited, and management for this rare plant is now advised. An examination of the biogeography of the E. corymbosum complex provides further support for the apparent rarity of var. nilesii, as well as var. aureum. Both taxa are found at the periphery of the complex, and both may represent insipient species. While all other varieties appear more closely related to each other than to varieties aureum and nilesii, with overlapping ranges confined mostly to the Colorado Plateau, both var. aureum and var. nilesii appear to have allopatric ranges largely off the Colorado Plateau. It appears these two peripheral varieties may each entail a separate center of origin for two new taxa.

É rico em endemismos, mas vem sofrendo forte degradação, sendo considerado um dos 34 hotspots mundiais para conservação. O presente estudo analisou populações de duas espécies arbóreas de ampla distribuição no Cerrado, a fim de compreender suas relações filogeográficas e contribuir para sua conservação. As espécies analisadas, Alibertia edulis (Rubiaceae) e Qualea grandiflora (Vochysiaceae), mostraram diferenciação significativa entre as populações quando analisadas por sequenciamento de três regiões plastidiais (matK, psbA-trnH e ndhF). As populações de A. edulis foram divididas por uma barreira norte-sul mais a oeste que a previamente proposta para o Cerrado, sugerindo um gradiente leste-oeste na distribuição da diversidade genética. Para ambas as espécies, as populações de Cerrados considerados "marginais" não são uma expansão recente do Cerrado central, com haplótipos compartilhados ou derivados do mesmo, mas apresentaram haplótipos exclusivos, por vezes ancestrais. Para evitar a perda irreparável de parte da diversidade do Cerrado, é recomendada a implantação de UCs nos Cerrados do Piauí e Pará, mais diferenciados e sob maior risco de degradação. Adicionalmente, o presente estudo desenvolveu microssatélites (SSR) nucleares para a espécie A. edulis, a fim de fornecer marcadores polimórficos para auxiliar em estudos populacionais. Para testes de polimorfismo, indivíduos do Piauí e Mato Grosso foram utilizados, revelando valores mais altos de endocruzamento na primeira população, os quais podem estar associados ao seu longo isolamento. Tais marcadores serão importantes ferramentas para análises populacionais e filogeográficas em A. edulis.

Insights into the Taxonomy and Evolution within an Orchid, Platanthera Dilatata, based on Orphometrics and Molecular Markers

Documenting biodiversity, at and below the species level, is a persistently challenging task for biologists. Poor understanding of biodiversity may lead to incorrect interpretations of observed variation. The underlying basis of variation can be understood by quantifying multiple sources of information. Nine morphometric characters and plastid DNA sequences (2511 bps) were quantified in a highly variable orchid species, Platanthera dilatata, to evaluate taxonomy of the three named varieties and to understand patterns of evolution. Three morphological groups, identified in a cluster analysis, were distinct in multiple floral traits. Additionally, the three clusters were consistently genetically divergent as indicated by infrequent haplotype sharing, significantly different haplotype frequencies, and significant values of the genealogical sorting index. This level of genetic divergence suggests three species rather than varieties in this complex. The divergent floral morphologies suggest that pollinator-mediated selection may be a driving factor for speciation in this complex.

The present work aimed at an investigation of the phylogeography of Psychotria ipecacuanha. The sequence of the chloroplast gene trnT-trnL from 40 individuals of 15 natural populations was obtained for this purpose. Nine of these populations were identified in the southeast and six in the central-western area of Brazil. A total of 959 bases were obtained by the sequence data. These aligned sequences showed the existence of 13 polymorphic sites among them, which differentiated eight haplotypes. All individuals sampled in the central-western area had a single haplotype, while the individuals of the southeast area were each allocated in one of the seven remaining haplotypes. The analysis of the data via nested clade analyses made the achievement of a single net possible and it was inferred that the components of this net are grouped at the level of a total cladogram in agreement with the geographical area of origin. The separation among the populations of the southeast and of the center-west follows a pattern that suggests allopatric fragmentation, while certain populations limited to the southeast suffer restriction of gene flow through isolation by distance. In the elaboration of a program of genetic conservation, the populations from the Rio Doce State Park, from Itaperuna, and from Guaraciaba can be indicated as priority because besides presenting variation within the haplotype number of the population, they also have five private haplotypes. Furthermore, these three populations together hold 75% of the variability of the lineages of the trnT-trnL gene. / Psychotria ipecacuanha é uma espécie medicinal da família Rubiaceae, conhecida popularmente como poaia e que se desenvolve em agregados perenes, denominados reboleiras, em áreas úmidas e sombrias do sub-bosque das matas tropicais do continente americano. O presente trabalho visou investigar a filogeografia da poaia. Para isto, seqüenciou-se o gene cloroplastídico trnT-trnL de 40 indivíduos originários de 15 populações naturais. Nove destas populações foram identificadas na região Sudeste e seis na região Centro-Oeste do Brasil. Os dados de seqüenciamento possibilitaram a obtenção de um total de 959 bases. O alinhamento destas seqüências mostrou a existência de 13 sítios polimórficos entre elas, que diferenciaram oito haplótipos. Todos os indivíduos amostrados na região Centro-Oeste possuíam um único haplótipo, enquanto cada um dos indivíduos da região Sudeste foi alocado em um dos sete haplótipos restantes. A análise dos dados via nested clade analyses possibilitou a obtenção de uma rede única e se inferiu que os componentes desta rede estão agrupados em nível de cladograma total, de acordo com a região geográfica de procedência. A separação entre as populações do Sudeste e do Centro-Oeste segue um padrão que sugere fragmentação alopátrica, enquanto certas populações restritas ao Sudeste estão sofrendo restrição ao fluxo gênico por isolamento à distância. Na elaboração de um programa de conservação genética, as populações do Parque Estadual do Rio Doce, Itaperuna e Guaraciaba podem ser indicadas como prioritárias, pois além de apresentarem variação no número de haplótipos dentro da população, possuem também cinco haplótipos privativos. Além disso, as três populações detêm juntas, 75% da variabilidade das linhagens do gene trnT-trnL.
