As gorduras são componentes importantes em dietas para carnívoros, porém o aproveitamento pelos animais sofre influência de fatores inerentes à composição das dietas, principalmente fibras e minerais, que precisam ser melhor estudados. Os trabalhos que avaliam as perdas endógenas de gordura nas fezes (PEGF) e os efeitos na digestibilidade aparente da gordura são escassos e divergentes. Desta forma, os objetivos destes estudos foram: determinar as PEGF em cães e mink; utilizar as PEGF para obtenção dos valores de digestibilidade verdadeira da gordura; além de avaliar os efeitos da composição de fibras dietéticas e a inclusão de emulsificantes no aproveitamento da gordura por cães. No primeiro estudo, as PEGF foram mensuradas em cães adultos. Dietas com níveis crescentes de gordura de frango foram utilizadas e as PEGF dos cães foram determinadas a partir de análises de regressão. As PEGF foram expressas em relação ao peso corporal (PC) dos animais, 155 mg/kg PV/dia, ou em relação ao consumo de gordura na matéria seca (MS), 7,9 g/kg MS consumida. A digestibilidade aparente total da gordura apresentou aumento conforme o consumo. A digestibilidade verdadeira total da gordura, corrigida para PEGF, não variou mediante o consumo de gordura. A relação entre PEGF e consumo de gordura foi maior em dietas com baixa gordura e explica o aumento na digestibilidade aparente da gordura quando concentrações maiores são consumidas. No segundo estudo, as PEGF foram determinadas em mink. Dietas com níveis crescente de óleo de soja foram formuladas e as PEGF avaliadas por regressão. Foi obtido o valor de 0,5 g/100g MS consumida, próximo ao relatado em cães. No terceiro estudo foi avaliada a influência das fibras dietéticas e da lecitina de soja na digestibilidade aparente da gordura em dietas para cães adultos. Foram avaliadas três fontes de carboidratos, amido de milho, celulose e polpa de beterraba, combinadas ou não à lecitina de soja. A polpa de beterraba reduziu a digestibilidade aparente e verdadeira da gordura em cães, aumentou o teor de umidade e elevou o escore fecal, porém ainda dentro da faixa considerada ideal. A lecitina de soja não apresentou efeito de melhora na digestibilidade dos macronutrientes e da energia das dietas. / Fats are important components of carnivores diets, however inherent factors of diets composition, mainly fibers and minerals, may affect its use by the animals, requiring further studies. Studies evaluating endogenous fat loss (EFL) and the effects on apparent total tract digestibility of fat are limited and divergent. The objectives of these studies were: to determine the EFL in dogs and mink; apply the EFL to calculated the true total tract digestibility of fats; and to evaluate the effects of dietary fiber composition and an emulsifying agent on fats digestibility in dogs. On the first study, the EFL was evaluated in adult dogs. Diets with increasing levels of poultry fat were used and the EFL was determined by regression analysis. The EFL was obtained according to the dogs’ body weight (BW) as 155 mg/kg BW/day, or according to dry matter (DM) fat intake, as 7.9 g/kg DM intake. The apparent total tract digestibility of fat increases according to the intake level. However, the true total tract digestibility of fat, EFL corrected, remained unaffected by fat intake. The relation between EFL and fat intake is higher for low fat diets and explains why the apparent digestibility increases according to intake. On the second study, the EFL was measured in mink. Diets with graded levels of soybean oil were offered and the EFL evaluated by regression. The value of 0.5 g/100 g DM intake was obtained, close to the reported with dogs. The third study assessed the influences of dietary fibers and soy lecithin on apparent total tract digestibility of fat in adult dog diets. Were evaluated three carbohydrate sources; corn starch, cellulose and beet pulp; combined or not with soy lecithin. The beet pulp decreased apparent and true total tract digestibility of fat in dogs, increased the moisture level of faeces and the faecal score, yet the faeces still had good quality. The soy lecithin did not improve digestibility of macronutrients and energy of diets.

Validity and Reliability of a Photographic Method of Assessing Body CompositionMacDonald, Elizabeth Z. 01 June 2016 (has links)
The LeanScreenTM app uses photographs and touchscreen technology of an iPad or iPhone to estimate body composition using the Department of Defense (DoD) prediction equations that use cirumference measurements of the neck, abdomen, waist, and hips. The purpose of this study was to determine the validity and reliability of the LeanScreenTM app in 148 weight-stable adults (82 men, 66 women) who were normal weight, overweight, or obese as defined by body mass index. The percent body fat (%BF) of each subject was estimated during one visit using dual-energy x-ray absorptiometery (DXA) as the criterion measure, and three field methods: the LeanScreenTM app, manually measured circumferences, and an OMRON bioelectical impedance (BIA) device. The %BF of each subject was determined once using DXA. Each of two administrators assessed the %BF of each subject twice using the LeanScreenTM app, manually measured circumferences, and the OMRON BIA device. When using the LeanScreenTM app, administrators assessed body composition using photographs they had taken and the photographs taken by the other administrator. Validity was established by comparing estimates of %BF from the LeanScreenTM app, manually measured circumferences, and the OMRON BIA device to %BF values obtained from DXA. Inter- and intrarater reliability was determined using mutliple measurements taken by each of two administrators. The three field methods were compared to DXA using mixed model ANOVA and Bland-Altman analyses. Analysis of the data revealed that the LeanScreenTM app, manually measured circumferences, and the OMRON BIA device significantly underestimated (p < 0.05) the %BF determined by DXA by an average of -3.26 ± 3.57 %BF, -4.82 ± 3.45 %BF, and -8.45 ± 3.48 %BF, respectively. Limits of agreement (LOA) for the LeanScreenTM app (6.99 %BF), manually measured circumferences (6.76 %BF), and the OMRON BIA device (6.82 %BF) were large. Slopes of the line-of-best-fit through the data in the Bland-Altman plots indicate that bias of %BF estimates using the LeanScreenTM app (slope = 0.06; p = 0.008) and the OMRON BIA device (slope = 0.15; p < 0.0001) increased as %BF increased. For each method of assessment, minimal variance could be attributed to different administrators peforming the assessment and each administrator performing multiple assessments. All inter- and intrarater reliability coefficients of the LeanScreenTM app, manually measured circumferences, and OMRON BIA estimates of %BF exceeded 0.99. The results of this study indicate that all three field methods of body composition assessments were highly reliable, however, these field measures are not recommended for use in the assessments of %BF due to a significant bias and large limits of agreements.

Being able to effectively match energy intake to energy expenditure (EE) is an important aspect in preventing weight re-gain in the post-obese. Although it is generally agreed upon that resting EE decreases concomitantly with weight loss, there is no set standard comparing the deviations with differing weight loss protocols and additionally, controversy remains as to whether this decrease is greater than can predicted. In order to address these issues 2977 subjects were analyzed using a systematic review and the differences of both the protocol and length of various interventions in addition to sex were compared. Next, data was selected from this systematic review and 815 subjects were analyzed for weight loss-induced changes in resting EE, FM and FFM. Another subgroup of studies (n = 1450) was analyzed and compared against the Harris-Benedict prediction equation to determine whether the changes in resting EE were greater than what was expected. Finally, in order to determine which factors may be involved in regulating changes in resting EE during weight loss, a secondary analysis was performed on 28 post-menopausal women (age= 50.4 ± 2.0 yrs; BMI= 32.4 ± 5.2 kg/m²) who were submitted to a 6-month caloric restriction. Body composition (DXA), resting EE (indirect calorimetry), physical activity EE (PAEE) and total EE (TEE) (doubly-labelled water) were measured before and after the 6 month weight loss. Blood samples were collected before and after to measure leptin and peptide YY. The results indicate that there was indeed a depression in resting EE during weight loss regardless of the type of intervention utilized. Furthermore, these findings suggest that the changes could not fully be explained by changes of FM and FFM alone and that leptin may be an important contributor to the changes of resting EE during weight loss.

Association analysis of MC1R, MC4R and AGRP in beef cattleMcLean, Kim Lauren 13 January 2010
Three interrelated genes postulated to affect economically important traits related to growth and/or carcass quality of beef cattle were chosen to characterize and perform association analyses for this study. Melanocortin 1 receptor (MC1R), melanocortin 4 receptor (MC4R) and agouti related protein (AGRP) play an integral role in the appetite pathway and in fat deposition.<p>
We genotyped 328 crossbred steers of various coat colours that were purchased at weaning and fed until slaughter for the previously published alleles ED, E+ and e. The E+ allele was present at five percent in this population and therefore was not included in further analysis. Black cattle of ED/ED or ED/e genotype had increased backfat (P<0.05) and required significantly fewer days (15-25) (P<0.01) on feed to reach a target fat level for slaughter compared to the red cattle. Red cattle of e/e genotype were found to have significantly larger longissimus dorsi (l. dorsi) area, shipping weight and hot carcass weight. Subsequent analysis revealed that the differences were comparable whether black versus red coat colour or MC1R genotype was used as the criteria for the group of cattle.<p>
MC4R sequence was obtained from 20 random crossbred steers. In addition to several previously published polymorphisms, a novel Ser330Asn polymorphism was detected. A population of 382 crossbred Canadian steers and 985 crossbred American steers was genotyped for this Ser330Asn polymorphism. A minor allele frequency of 0.01 was observed in the Canadian and 0.02 in the American steer populations. No homozygous g.989AA cattle were detected. In the Canadian population, heterozygous steers had increased grade fat (P=0.036) and decreased lean meat yield (P=0.032). Similarly in the American population, steers of the g.989GA genotype had increased backfat (P=0.031) and less desirable yield grades (P=0.022,) but also lower ribeye area measurements (P=0.031). These results suggest that genotyping for the Ser330Asn polymorphism may lead to increased quality of carcasses either through lean meat production or backfat measurements, depending on the goal of the beef operation.<p>
Sequence data obtained from 38 Bos taurus beef cattle, 4 Holsteins and 4 Bos indicus cattle revealed six polymorphisms in the AGRP gene. No polymorphisms that altered amino acids were detected in Bos taurus cattle. Genotyping of 382 crossbred beef steers was performed for two polymorphisms, an intronic deletion (g.439_440delTC) and a base pair substitution in exon 4 that did not alter an amino acid (g.715G>A). An ANOVA analysis, using PROC Mixed, was performed for both polymorphisms on several growth and carcass traits. No significant differences were observed.<p>
Polymorphisms in MC1R and MC4R could be used as genetic tests which may be beneficial for beef producers in North America. The significant differences observed in this study in relation to cattle growth and fat deposition would represent savings for producers when used for sorting feedlot cattle or in selection of breeding cattle.

Direct Water and Fat Determination in Two-Point Dixon ImagingRambow, Olen 16 September 2013 (has links)
The Dixon technique is a well-established method in magnetic resonance imaging for obtaining separate images of water and fat. Here we present a generalized solution to the two-point Dixon problem with a geometric interpretation, allowing for flexible echo times and a multi-peak fat model. By simulation and experiment, we have analyzed the dependence on the echo times of the error in the water, fat, and relative background phasor values due to both signal noise and T2* decay. Furthermore, we have demonstrated that broken symmetry due to the multi-peak nature of fat enables direct water and fat determination without phase correction, and we have quantified the reliability of this technique as a function of the echo times. The results may provide valuable guidance for selecting scan parameters to balance the objectives of optimizing fat-water identification, minimizing error in the pixel values, and minimizing total scan time.

The aim of this essay is to discuss some of the shared legacies of oppression between queerness, femininity, and fatness in order to theorize a form of activism that can do justice to these intersecting identities. A key component of this is to discuss the complexities of negotiating the shame and pride that go hand in hand with stigmatized identities, a project recently taken up by queer theorists that has yet to be well represented in fat studies or activist circles. This essay will engage with conversations happening in queer theory and fat studies about shame as it relates to the politics of attachment. I hope to begin a conversation about how to organize effective activist circles that can do justice to queer fat femmes' complex relationships with visibility, embodiment and community building. / text

Anticarcinogenic and Immunomodulatory Properties of the Milk Fat Globule MembraneZanabria Eyzaguirre, Romina 10 May 2013 (has links)
The milk fat globule membrane (MFGM) obtained from bovine milk is a source of bioactive compounds. In this research, the hypothesis that the MFGM possesses anticarcinogenic capacity and immune modulatory properties was tested, along with the hypothesis that processing history of the fat globules will affect the bio-functionality of the MFGM.
Throughout all experiments, lipopolysaccharide (LPS) was minimized by extracting the milk using a catheter, working under aseptic conditions and avoiding contamination via reagents. To study the anticarcinogenic capacity, native MFGM was tested in vitro using two colon adenocarcinoma cell lines (HT-29, Caco-2). Stimulated cells showed a dose-dependent decrease in cell proliferation, starting at very low concentrations (1 μg/mL based on protein) and its efficiency was comparable or even superior to the commercial anticarcinogenic drugs melphalan and C2-ceramide. Cytotoxicity and apoptosis were found to be mechanisms responsible for this bioactivity, as shown by the lactate dehydrogenase enzyme activity and the increase in caspase-3 production.
MFGM- immune modulatory capacity was also tested by studying its effects on splenocyte proliferation, apoptosis and cytokine production. While splenocyte proliferation was not affected when the MFGM isolate was used alone, it suppressed cell division in the presence of polyclonal activators (LPS, Concanavalin A) used to simulate inflammatory conditions. Cytokine production suggests inhibition of the splenocytes’ activation process as the mechanism behind its bioactivity.
Milk heating caused a significant decrease in bioactivity, indicating that protein interactions and denaturation as well as the structural changes caused by this treatment, directly affect the MFGM bio-functionality. Partial hydrolysis (by trypsin and phospholipase-A2) caused a similar effect, suggesting that not only the phospholipids are involved in the anticarcinogenic capacity, but also, the complex mixture of proteins which form part of the MFGM complete system.
In summary, the bioactivity of the MFGM extracted from milk is not limited to its anticarcinogenic but also to its immune regulatory ability, by helping control the response of the immune system when inflammatory conditions arise. However, pre-treatment of the raw material greatly affects this bioactivity, suggesting that greater care may be needed in processing of fat globules to maintain such important characteristics of their components. / Canadian Dairy Council (CDC), Ontario Dairy Council (ODC), Natural Sciences and Engineering Research Council of Canada (NSERC).

You have Nothing to Lose! Using Culturally Relevant Pedagogy in Secondary Education to Make Space for Body AcceptanceFullbrook, Ashley C. 28 November 2012 (has links)
Schools are sites of great power and influence where the “obesity” discourse is often taken uncritically as truth and reproduced, to the detriment of young people. The purpose of this thesis was to investigate how theories of fatness can inform theories of culturally relevant pedagogy with the goal of helping teachers create spaces where increased size acceptance is possible for secondary students. Literature from both these areas of study was reviewed and applied to the Ontario secondary curriculum documents for science and physical education. This analysis demonstrated a body acceptance orientation in teaching these disciplines, and that doing so can mitigate many of the negative effects of living in a fat hating world.
