Consequences of intraspecific genetic variation for population dynamics and niche expansion

Intraspecific genetic diversity is an important attribute of natural populations and is deemed critical for their adaptive potential and persistence. However, we have limited empirical understanding of the impact of genetic diversity on population performance under different conditions. For my dissertation, I conducted long-term laboratory experiments with populations of the flour beetle Tribolium castaneum to test the consequences of genetic variation for population dynamic stability and niche evolution. In Chapter 1, I show that genetic variation prevented population extinction in a novel habitat. In addition, genetically diverse populations were more stable, both in a novel heterogeneous habitat and in their ancestral habitat. In the ancestral habitat, alleles from a single founding lineage dominated the dynamics, leading to increased stability of genetically diverse populations. However, such as selective effect was not observed in the novel heterogeneous habitat. Therefore, while genetic variation within populations increased their stability and persistence, the magnitude of the impact and its mechanism depended on the selective habitat. In Chapter 2, I ask whether genetic variation also facilitates resource niche expansion, i.e., use of a novel resource. Using stable carbon isotopes, I analyzed diets of beetles sampled from the above experiment and quantified the rate of change in resource use. Contrary to theoretical predictions, I found that genetic variation for resource use had no effect on the rate of niche evolution. Furthermore, behavioral niche expansion accounted for most of the adaptation to the novel resource, and the behavioral change hindered subsequent evolutionary change in resource use. It is thus apparent that in the short term, behavioral plasticity in niche use may impose far greater constraints on niche evolution than the amount of standing genetic variation. Mathematical models predict that intraspecific competition generates selection for niche evolution, and that genetic variation increases the response to selection. Therefore, I hypothesized that the impact of genetic variation on resource niche evolution may depend on the degree of intraspecific competition. In the final chapter of this thesis, I describe results of an experiment to test this hypothesis. I found that genetic variation and competition indeed interacted to increase the rate of niche expansion in T. castaneum, but that their impacts were temporally variable. Furthermore, the two factors acted on different components of niche evolution: while competition only affected the degree of niche expansion, genetic variation also promoted maintenance of individual variation in resource use. In summary, my thesis describes experiments to test for the ecological and evolutionary impacts of intraspecific genetic variation; and its interaction with behavioral plasticity, intraspecific competition, and resource availability. Genetic diversity and behavioral plasticity are common features of living organisms, and therefore it is vital to understand their combined consequences for population ecological and evolutionary dynamics. In addition, natural populations often face intense competition for limited resources. Hence the experimental results presented here can help us to better understand how populations overcome these resource constraints, given their specific genetic composition. Biologists are increasingly aware that the intricate connection between ecological and evolutionary dynamics is important to gain a more complete understanding of population biology. The work described here represents one of the few experiments providing such detailed mechanistic understanding of the interactions between- and consequences of - key ecological and evolutionary parameters. Finally, the results have important implications for conservation biology, because they show that the effects of genetic diversity can vary greatly depending on a number of population and environmental parameters. / text

Οι καταναλωτές τα τελευταία χρόνια ενδιαφέρονται ολοένα και περισσότερο για την προέλευση και την ποιότητα των τροφίμων, συμπεριλαμβανομένης και της προέλευσης των Ά υλών. Η ασφάλεια, η θρεπτική αξία και πιο πρόσφατα οι περιβαλλοντικές επιπτώσεις των συστημάτων παραγωγής και διανομής των τροφίμων επίσης αντιμετωπίζονται με ανάλογο ενδιαφέρον. Το αλεύρι είναι το κύριο συστατικό του ψωμιού που θεωρείται ως η βάση της υγιούς διατροφής παγκοσμίως, των προϊόντων ζαχαροπλαστικής και άλλων αρτοσκευασμάτων. Επιπλέον στις μεσογειακές χώρες τα προϊόντα αυτά βρίσκουν μεγάλη αποδοχή από τους καταναλωτές. Στην Ελλάδα το αλεύρι καλλιεργείται σε σημαντικές ποσότητες και οι αλευροβιομηχανίες εφοδιάζουν τις βιομηχανίες τροφίμων και τους φούρνους αρτοποιίας με πολλούς τύπους αλεύρου. Οι ανάγκες της χώρας για τα διάφορα προϊόντα αλεύρου παρουσιάζουν ένα μέσο ετήσιο ρυθμό αύξησης περίπου 1,5% για τη διετία 1999-2000, ενώ οι ανάγκες για το έτος 2000 ήταν 753.000 τόνοι. Για το ίδιο έτος η εθνική παραγωγή ανέρχεται σε 825.000 τόνους. Η Ανάλυση Κύκλου Ζωής (ΑΚΖ) είναι ένα σχετικά νέο περιβαλλοντικό εργαλείο λήψης αποφάσεων που πρόσφατα τυποποιήθηκε από το Διεθνή Οργανισμό Προτύπων (International Standardisation Organisation (ISO)). Η ΑΚΖ είναι ένα εξαιρετικό εργαλείο για την περιβαλλοντική διαχείριση, καθώς παρέχει σημαντικές πληροφορίες για τους περιβαλλοντικούς κινδύνους που σχετίζονται με ένα προϊόν, μια διαδικασία ή οποιαδήποτε ανθρώπινη δραστηριότητα γενικότερα. Σε αυτή τη μελέτη μια απλουστευμένη ΑΚΖ εφαρμόζεται με μια προσέγγιση «από τη γέννηση ως την πύλη» (“cradle-to-gate”) για να αναγνωρισθούν τα κρίσιμα σημεία της διαδικασίας παραγωγής του αλεύρου στην Ελλάδα. Το λογισμικό SimaPro χρησιμοποιήθηκε για την ανάλυση των δεδομένων της απογραφής και της εκτίμησης των επιπτώσεων του συστήματος συνολικά (από τους σπόρους σιταριού ως την παραγωγή του αλεύρου). Το σύστημα που αναλύθηκε περιλαμβάνει την καλλιέργεια μαλακού σιταριού, τη μεταφορά, τη βιομηχανική άλεση και τη συσκευασία του αλεύρου. Η χρήση ενέργειας και υλικών, καθώς και οι εκπομπές στο περιβάλλον υπολογίσθηκαν και εκτιμήθηκαν τα κύρια δυναμικά των περιβαλλοντικών επιπτώσεων. Η εφαρμογή των λιπασμάτων, η μεταφορά και η χρήση ηλεκτρικής ενέργειας βρέθηκαν να είναι τα πιο σημαντικά σημεία για τις περισσότερες, όχι όμως για όλες, τις κατηγορίες επιπτώσεων και συγκεκριμένες ενέργειες προτάθηκαν με στόχο τη βελτίωση του περιβαλλοντικού προφίλ του τελικού προϊόντος. / Consumers have become increasingly interested in the origin and the quality of their food, including the source of food’s raw materials, its safety and nutritional value and most recently the environmental impacts of the production systems adopted in producing and delivering the food. Flour is the main ingredient of bread, cakes, pastries and other bakery products which are considered as the base of modern healthy diet worldwide. Moreover, in Mediterranean countries these products have an outstanding acceptance by consumers. In Greece, wheat is cultivated in significant quantities and a number of milling industries supply the food industries and small bakeries with many different types of flour. The national demand for various products of flour-producing industries presented a mean annual increase rate of about 1.5% for the years 1999-2000, while the demand for the year 2000 was 753.000 tons. For the same year the national production ranged up to 825.000 tons. Life Cycle Assessment (LCA) is a relatively new, cutting edge environmental decision support tool recently standardised by the International Standardisation Organisation (ISO). LCA is an excellent tool for environmental management, as it provides important information about the environmental burdens associated to a product, a process or any human activity. In this study, a simplified LCA following the “cradle-to-gate” approach has been applied to identify the critical points of the flour production in Greece. The SimaPro software was used for analyzing the main inventory data and estimating the environmental impacts of the overall system (from wheat grain to flour production). The system investigated includes agricultural production of soft wheat, transportation, industrial milling and packaging of the flour. Energy use, material use and emissions were quantified and the main potential environmental effects were assessed. The application of fertilizers, transportation and electrical energy consumption were found to be the ‘hot spots’ for many, but not all, of the impact categories investigated and specific actions were proposed in the aim to improve the environmental profile of the final product.

Effects Of Different Batter Formulations On Quality Of Deep-fat Fried Chicken Nuggets

The main objective of this study was to evaluate the effects of different flour and protein types on quality of deep-fat fried chicken nuggets. Additionally, the rheological properties of batters were studied and the batter viscosity was correlated with fried product quality. In the first part of the study, the effects of soy and rice flour (5%) addition to the batter formulation on product quality were studied. Coating pick-up of batters, and moisture content, oil content, texture, color, volume, porosity, and cooking yield of nuggets were determined for 3, 6, 9 and 12 minutes of frying times at 180&ordm / C. In the second part of the study, the effects of protein types (soy protein isolate, whey protein isolate and egg albumen) at different concentrations (1 and 3%) on quality attributes were studied. A batter formulation with no flour or protein addition was used as control. In both parts of the study, flow behavior of batters prepared using different flour and protein types were determined. Soy flour and soy protein isolate (SPI) provided the highest apparent viscosity. Batter viscosity was found to be correlated with coating pick-up. All batters were found to show thixotropic behavior. The batters were modeled as power-law fluid and all of them turned out to be shear-thinning except egg albumen added batter, which was shear-thickening. As a result of the study, among the flour and protein types used, 3% whey protein isolate (WPI) was found to be the most effective ingredient on improving quality parameters of deep-fat fried chicken nuggets. 3% WPI added batters provided the hardest and crunchiest product with the darkest color. It also reduced the oil content of fried nuggets significantly. However, low cooking yield values were observed for batters with 3% WPI. On the other hand, soy flour containing batters provided high cooking yield. Therefore, if high cooking yield with low oil content is desired, soy flour can be advised to be used in batter formulations for chicken nuggets.

Use of marker assisted selection for the introgression of quality traits from Australian into Chinese wheats

[Abstract]Quantitative Trait Loci (QTLs) for polyphenol oxidase and xanthophyll have a significant impact on variation in wheat flour for noodle colour and colour stability. QTLs from two Australian wheat cultivars, Sunco and Tasman, have been backcrossed into two Chinese wheat varieties, Chuanmai 22 and Mianyang 11, to assess marker predictability for these important traits in significantly different genetic backgrounds. The concept of Marker-Assisted-Selection (MAS) is being trialled in this study as a proposed method for wheat improvement. In this approach molecular markers are used in conjunction with backcross breeding methods to introgress specific characters into elite breeding materials, with the goal of improving the quality attributes of wheat for the Asian noodle market. After three single seed descent generations, the backcross populations generated allow four QTLs to be investigated. These include two for polyphenol oxidase (chromosome 2A and 2D) and two for xanthophyll (chromosome 3B and 7A). This research was successful in identifying microsatellite markers that are capable of predicting PPO activity levels and Xanthophyll content within the backcross populations. These microsatellites were validated as useful markers for these quality traits, as they have also found to be important in the Sunco x Tasman doubled haploid population. The combination of marker assisted selection and backcrossing has generated three lines that contain different combinations of the PPO activity and Xanthophyll content QTLs. These lines have been found to produce low levels of PPO activity and have a low Xanthophyll content. This improvement in flour colour and colour stability highlights the potential of marker assisted selection as a useful tool in wheat breeding.
