A knowledgebase of stress reponsive gene regulatory elements in arabidopsis Thaliana

<p>Stress responsive genes play a key role in shaping the manner in which plants process and respond to environmental stress. Their gene products are linked to DNA transcription and its consequent translation into a response product. However, whilst these genes play a significant role in manufacturing responses to stressful stimuli, transcription factors coordinate access to these genes, specifically by accessing a gene&rsquo / s promoter region which houses transcription factor binding sites. Here transcriptional elements play a key role in mediating responses to environmental stress where each transcription factor binding site may constitute a potential response to a stress signal. Arabidopsis thaliana, a model organism, can be used to identify the mechanism of how transcription factors shape a plant&rsquo / s survival in a stressful environment. Whilst there are numerous plant stress research groups, globally there is a shortage of publicly available stress responsive gene databases. In addition a number of previous databases such as the Generation Challenge Programme&rsquo / s comparative plant stressresponsive gene catalogue, Stresslink and DRASTIC have become defunct whilst others have stagnated. There is currently a single Arabidopsis thaliana stress response database called STIFDB which was launched in 2008 and only covers abiotic stresses as handled by major abiotic stress responsive transcription factor families. Its data was sourced from microarray expression databases, contains numerous omissions as well as numerous erroneous entries and has not been updated since its inception.The Dragon Arabidopsis Stress Transcription Factor database (DASTF) was developed in response to the current lack of stress response gene resources. A total of 2333 entries were downloaded from SWISSPROT, manually curated and imported into DASTF. The entries represent 424 transcription factor families. Each entry has a corresponding SWISSPROT, ENTREZ GENBANK and TAIR accession number. The 5&rsquo / untranslated regions (UTR) of 417 families were scanned against TRANSFAC&rsquo / s binding site catalogue to identify binding sites. The relational database consists of two tables, namely a transcription factor table and a transcription factor family table called DASTF_TF and TF_Family respectively. Using a two-tier client-server architecture, a webserver was built with PHP, APACHE and MYSQL and the data was loaded into these tables with a PYTHON script. The DASTF database contains 60 entries which correspond to biotic stress and 167 correspond to abiotic stress while 2106 respond to biotic and/or abiotic stress. Users can search the database using text, family, chromosome and stress type search options. Online tools have been integrated into the DASTF&nbsp / database, such as HMMER, CLUSTALW, BLAST and HYDROCALCULATOR. User&rsquo / s can upload sequences to identify which transcription factor family their sequences belong to by using HMMER. The website can be accessed at http://apps.sanbi.ac.za/dastf/ and two updates per year are envisaged.</p>

Job and family stress amongst firefighters

Firefighters providing emergency services to the public are involved with some of the most tragic aspects of the boundary between life and death, often in a context over which they have little or no control. The outcome of this may be that stress at work and at home are without doubt the reason that highly qualified and loyal firefighters give themselves over to alcohol or drug abuse, extramarital relationships and suicidal thoughts. The general aim of the research is to evaluate job and family stress amongst firefighters in the South African context, and to use the results in developing a developmental/counselling programme for firefighters and their families. The research is quantitative and qualitative, consisting of a survey design and a phenomenological design. Three measuring instruments were used, namely the Biographical questionnaire, the Experience of Work and Life Circumstances questionnaire, and the Stress questionnaire. Task characteristics, organisational functioning, physical working conditions and job equipment, career and social matters, remuneration, fringe benefits and personnel policy were identified as causes of job stress originating within the work situation. Interviews were conducted to determine how these firefighters experience job and family stress. Marital dysfunction and divorce, limited time with the family, problems with children, alcohol and drug abuse, lack of exercise, suicide, anger aimed at family members, physical and emotional exhaustion, lonely marital partners, unavailability to help the family when needed and depression were identified as causes of family stress arising outside the work situation. The main recommendation is to implement a developmental/counselling job and family stress programme. The programme can be instituted to enhance the wellness and psychological health of firefighters and their families, or for counselling of firefighters and their families who are experiencing job and/or family stress. Relationship between work and non-work stressors and work-life balance amongst global market trading professionals

In the financial industry, there is a high demand for employees to possess all the necessary skills and motivation to perform highly specialized functions, and handle demands that accompany increasing stress levels. The aim of the research was to determine whether there is a relationship between work and non-work stressors and work-life balance in the Global Market Trading industry. A cross-sectional survey design was used, with a sample of 72 global market trading professionals drawn from a financial institution in Gauteng. Two questionnaires and a biographical questionnaire were administered. The questionnaires proved to be reliable. The power of the study was calculated. Descriptive statistics and Spearman correlation was used to organise, summarise and describe the data. The findings of the study show that as global market trading professional’s intrinsic factors at work and stress regarding their personal time increases, their experience of negative WHI increase as well. / Industrial and Organisational Psychology / M.Com. Text in Afrikaans / In this study an instrument is suggested for implementation by the Educational Psychologist to design a support programme, aimed at handling family members' stress where a child has sustained a brain injury. Attention was paid to the phenomenon "brain injured child" to ascertain demands and needs (physical, cognitive, psychological and emotional). Specific note was taken of the toll on each family member in their observance, experience, assistance and giving· meaning to the child. Reference was made to existing support programmes for such family members from the acute care phase to the final acceptance and readjustment of the family. With this study the need for a continuous support programme and the contents of such a programme was addressed. Using these guidelines the Educational Psychologist will be able to prepare the family for the stress possibilities in dealing with the brain injured child. / Met hierdie studie is 'n instrument daargestel vir die ontwerp van 'n hulpverleningsprogram vir gebruik deur die Opvoedkundige Sielkundige. Hierdie hulpprogram het as doel, die hantering van stres, deur die gesin van 'n kind, wat 'n breinbesering opgedoen het. In die studie is aandag gegee aan die tipiese gedrag wat oor die algemeen van 'n breinbeseerde kind verwag kan word. Daar is ook gepoog om te bepaal hoe elke lid van die gesin die breinbeseerde kind beleef, aan hom betekenis gee, en hom probeer help ten opsigte van die eise (fisiek en emosioneel) wat hy stel. Verder is daar gekyk na bestaande hulpverlening (gerig op die hantering van stres) aan die gesinslede van 'n breinbeseerde kind vanaf die akute versorgingsfase tot en met die aanpassing en herorganisering van die gesin. Parental human investment : economic stress and time allocation in Russia

A decade of growth and wealth generation in Russia ended in 2009 with the collapse in GDP and rising unemployment. This Great Recession added new economic challenges to the ‘old’ problems facing children and families, including widening income inequalities and the phenomenon of social orphanage. One question is how the new and existing material pressures affect parent–child relationships. This research contributes to the answer by examining, in aggregate terms, the role poverty plays in the allocation of parental time in this emerging economy. Utilising a nationally representative sample of children, it explores how child interactions with parents are affected by aggregate and idiosyncratic shocks. Drawing on the rational choice paradigm and its critique, we put forward the Parental Time Equilibrium as an analytical guide to the study. This theoretical approach presents individual decisions concerning time spent with children over the long term as the product of a defined equilibrium between resources and demands for involvement. We test this approach through pooled cross-sectional and panel analyses based on the Russian Longitudinal Monitoring Survey dataset from 2007 to 2009. Children in low-income households face the double disadvantage of a lack of money and time investments at home, with both persistent and transient poverty being associated with lower than average parental time inputs in the sample. Moreover, while on average, we find that children do maintain the amount of time they spend with their parents under conditions of severe financial strain, low–income children lose out on play time with the mother. Material resources cannot be considered in isolation from structural disadvantages, of which rural location in particular is detrimental for parent–child time together. The study demonstrates that the cumulative stress of adverse macro-economic conditions and depleted material resources makes it difficult for parents to sustain their human investment in children. The evidence this study provides on the associations between economic stress and pa-rental time allocations advances our knowledge of the disparities of in the childhood experience in modern Russian society. The findings strongly support the equal importance of available resources and basic demand for involvement, thus drawing policy attention to the need to address both in the best interests of children.

Effects of insecure attachment on marital interactions : examining the family stress model

This exploratory study posited that being in an insecure marriage would increase marital conflict and perceived relational aggression when the couple is experiencing economic constraints. The cross-sectional design assessed two groups of marriages: secure and insecure. A structural equation model using multiple group comparison tested the effect of each type of marriage, controlling for husband and wife education, number of children, and length of marriage. Results indicated that husbands who identify as insecure experience greater marital conflict and perceive their spouses to be more psychologically aggressive. There was also evidence of a cross-lag effect indicating husbands' perceived marital conflict positively influences wives' perceptions of greater spousal psychological aggression for insecure couples, but not secure couples. This is the first study to use attachment as a moderating variable in the family stress model. Future research should look to include greater measures of attachment, with a focus on longitudinal designs. Implications are also discussed. / Graduation date: 2013
