Effects of exercise and dietary intervention on metabolic syndrome markers of inactive premenopausal women

The effects of exercise and dietary intervention on metabolic syndrome (Mets) markers of inactive premenopausal women were investigated. In Study I, early markers of MetS were examined in young, (21.2 ?? 0.3 years), healthy but unfit women (N = 66) consuming a processed food diet. A second aim was to examine the relationship between ethnic influences (European versus Chinese) and early markers of MetS (e.g., hyperinsulinemia). Study II compared the hormonal and metabolic responses to steady state exercise (SSE) and high intensity interval exercise (HIIE) in untrained young women (N = 18, 21.7 ?? 0.8 years). Women were further divided into high and low fasting insulin levels to determine if hyperinsulinemia influenced hormonal and metabolic response to SSE and HIIE. Study III examined the hormonal and metabolic response to a randomized controlled intervention named the Fish oil, Exercise and Mediterranean diet (FEM) trial. The FEM trial was conducted with overweight, inactive young (23.5 ?? 0.6 years) women (N = 56) for 12 weeks. Characteristics of a subset of women (N = 34, 23.5 ?? 0.7 years) in the FEM trial, non-responders (NRES), who did not lose weight (??? 1% loss in mass), were also examined. Metabolic profiles were developed based on body composition, aerobic fitness, blood markers, diet, resting metabolic rate, medical history, blood pressure, and autonomic function. Body composition was measured by skinfolds and girths, bioimpedance (Tanita, Japan), and Dual Energy X-Ray Absorptiometry. Peak oxygen uptake was assessed using an open circuit spirometer, TrueMax 2400 Metabolic Cart. Venepuncture and cannulation techniques were used for collecting blood samples that were also centrifuged and frozen for later analysis. In Study I young women who were physically unfit and consumed a processed food diet possessed high levels of fasting insulin, HOMA-IR (an insulin resistance index), and C-reactive protein (CRP). Insulin, p < .001, and HOMA-IR, p < .05, were significantly greater in Chinese Australians compared to European Australians, whereas plasma CRP levels were significantly, p < .05, lower. Significant differences, p < .05, existed between the groups with the Chinese Australians possessing lower body composition indices. However, ethnic differences still existed for insulin, HOMA-IR, and CRP after adjusting for body composition. Both groups consumed significantly high protein relative to their body mass. Thus, it appears that hyperinsulinemia is one of the earliest markers of MetS in young inactive females of both European and Asian descent who are unfit and consume a high level of dietary protein. In Study II, HIIE compared to SSE proved to be more effective at preventing an increase in insulin levels in the two hours after exercise. Resting respiratory quotient (RQ) was significantly lower, p < .05, following both HIIE and SSE, whereas plasma glycerol levels were higher, p = .06, suggesting greater lipolysis following HIIE. The women were divided by baseline fasting plasma insulin (> 9.98 ??IU/ml) into high insulin (HI) and low insulin groups (LI). The fasting plasma human growth hormone levels of the HI women were significantly lower at baseline compared to that of LI women. Baseline RQ was correlated with baseline glycerol, r = - .54, p < .05. Insulin levels at one hour post HIIE was related to fasting plasma adrenocorticotropic hormone (ACTH) one hour post exercise, r = .52, p < .05. Fasting plasma leptin at one, r = .56, p < .05, and two, r = .53, p < .05, hours post exercise was associated with 2 hour post insulin levels. Interestingly, fasting ACTH was significantly elevated in the 2 hours post exercise in the HI women compared to LI. All diet data between the groups were similar and lipids were in the healthy range with no significant differences between the women possessing high or low fasting plasma insulin. Thus, young women who completed one session of short duration HIIE compared to SSE improved certain aspects of their metabolic profile (e.g., reduced insulin levels) and enhanced their fat oxidation in the immediate two-hour exercise recovery period. Following FEM (a 12-week multi-component lifestyle intervention) overweight women recorded significantly lower, p < .05, body composition (mass, fat mass, percent body fat, waist circumference), insulin, inflammation (CRP), blood pressure, and lipids. The improvements within the Mediterranean diet (Mediet) were related to the reductions in body weight, fat mass, and insulin. Consumption of saturated fats, legumes, meat, poultry, and egg were also significantly decreased, p < .05, following the trial. Adherence to the Mediet and fish oil consumption, measured through a Mediet score, was significantly increased, p < .001, and was associated with reduced levels of fat mass, r = .43, p < .05. Autonomic function (measured by power frequency analysis), aerobic fitness, and fat oxidation were all significantly, p < .05, enhanced. Therefore, an intervention incorporating fish oil consumption, HIIE, and Mediet significantly reduced body fat, fasting insulin, inflammatory markers, and some blood lipids. Interestingly, some women did not lose fat mass following the FEM intervention, despite experiencing significant reductions in insulin, inflammation, waist circumference, blood pressure, and an increase in aerobic power. The major differences between those women who lost fat and those that did not was that the non-responders possessed significantly lower, p < .05, systolic blood pressure, lower resting heart rate, and a higher resting RQ. In summary, in Study I, young, unfit women consuming a processed diet, demonstrated hyperinsulinemia and low grade inflammation. The high levels of fasting insulin suggest that these women are at a higher risk for developing MetS and type 2 diabetes. The results of Study II suggests that one bout of HIIE compared to a longer bout of SSE was more effective at preventing a rise in post-exercise insulin levels. Finally, results of Study III indicate that a 12-week lifestyle intervention, encompassing HIIE, fish oil ingestion, and a Mediet positively influenced early MetS markers (e.g., hyperinsulinemia), aerobic and anaerobic fitness, low grade inflammation, and body composition in young women.

As gorduras são componentes importantes em dietas para carnívoros, porém o aproveitamento pelos animais sofre influência de fatores inerentes à composição das dietas, principalmente fibras e minerais, que precisam ser melhor estudados. Os trabalhos que avaliam as perdas endógenas de gordura nas fezes (PEGF) e os efeitos na digestibilidade aparente da gordura são escassos e divergentes. Desta forma, os objetivos destes estudos foram: determinar as PEGF em cães e mink; utilizar as PEGF para obtenção dos valores de digestibilidade verdadeira da gordura; além de avaliar os efeitos da composição de fibras dietéticas e a inclusão de emulsificantes no aproveitamento da gordura por cães. No primeiro estudo, as PEGF foram mensuradas em cães adultos. Dietas com níveis crescentes de gordura de frango foram utilizadas e as PEGF dos cães foram determinadas a partir de análises de regressão. As PEGF foram expressas em relação ao peso corporal (PC) dos animais, 155 mg/kg PV/dia, ou em relação ao consumo de gordura na matéria seca (MS), 7,9 g/kg MS consumida. A digestibilidade aparente total da gordura apresentou aumento conforme o consumo. A digestibilidade verdadeira total da gordura, corrigida para PEGF, não variou mediante o consumo de gordura. A relação entre PEGF e consumo de gordura foi maior em dietas com baixa gordura e explica o aumento na digestibilidade aparente da gordura quando concentrações maiores são consumidas. No segundo estudo, as PEGF foram determinadas em mink. Dietas com níveis crescente de óleo de soja foram formuladas e as PEGF avaliadas por regressão. Foi obtido o valor de 0,5 g/100g MS consumida, próximo ao relatado em cães. No terceiro estudo foi avaliada a influência das fibras dietéticas e da lecitina de soja na digestibilidade aparente da gordura em dietas para cães adultos. Foram avaliadas três fontes de carboidratos, amido de milho, celulose e polpa de beterraba, combinadas ou não à lecitina de soja. A polpa de beterraba reduziu a digestibilidade aparente e verdadeira da gordura em cães, aumentou o teor de umidade e elevou o escore fecal, porém ainda dentro da faixa considerada ideal. Strategies for the Reduction of Adipose Tissue and Retention of Muscle Mass in Overweight Individuals

Purpose: The purpose of this study was to determine whether time-restricted feeding (TRF) was an effective dietary strategy for reducing fat mass and preserving fat-free mass while evaluating potential changes in cardiometabolic biomarkers, hormones, muscle performance, and energy and macronutrient intake after eight weeks of aerobic exercise and resistance training in overweight and obese adults. Methods: This study was a randomized, controlled trial. Sedentary, overweight and obese adults (mean ± SD; age: 44.48 ± 7.28 years; BMI: 29.61 ± 2.62 kg/m2; females: 85.71%; males: 14.29%) were randomly assigned to a TRF or normal feeding (NF) dietary strategy group. The TRF group consumed all calories between 1200 and 2000 hours, whereas the NF group ate their typical diet. All groups completed eight weeks of aerobic exercise and supervised resistance training. Body composition, muscle performance, energy and macronutrient intake, physical activity, and physiological variables were assessed week zero and week nine. Results: A total of 21 participants completed the study (NF: n = 10; TRF: n = 11). A mild energy restriction was seen for the TRF (~300 kcal/day, 14.0%) and NF (~250 kcal/d, 11.0%) groups between baseline and week seven. Losses of total body mass were significantly greater for TRF (3.3%) relative to NF (0.2%), of which TRF had significantly greater losses of fat mass (9.0%) compared to NF (3.3%) despite similar reductions in energy intake. Lean mass increased across the intervention for both TRF (0.6%) and NF (1.9%), with no group differences. Conclusion: These data support the use of TRF and concurrent exercise training as a short-term dietary strategy for reducing fat mass and preserving lean mass in overweight and obese adults.

Déterminisme biologique de la variabilité de la fonte lipidique à la cuisson du foie gras de canard / Biological determinism of the variability of fat loss during cooking of duck 'foie gras'

Les objectifs de ce travail sont d'identifier les mécanismes biologiques impliqués dans le déterminisme de la variabilité de la fonte lipidique à la cuisson du foie gras de canard, et de définir des marqueurs de la fonte. Pour répondre à ces objectifs, nous avons choisi de conduire une caractérisation du tissu hépatique, selon son comportement ultérieur à la cuisson, basée sur une approche intégrée combinant de la biochimie, de la protéomique et de l'histologie. D'une part, l'expression différentielle des protéines hépatiques à l'abattage des animaux et au cours de la réfrigération, semble indiquer que le niveau de fonte pourrait être relié à des stades de développement de stéatose différents. Les animaux du groupe ‘Fonte faible' se distinguent de ceux du groupe ‘Fonte élevée' par un ‘profil d'accrétion' avec la surexpression de protéines du métabolisme énergétique par exemple, indiquant des mécanismes de synthèse et d'accumulation de lipides. A l'inverse, les animaux du groupe ‘Fonte élevée' présentent la surexpression de protéines de réponse au stress, ce qui pourrait indiquer un stade de stéatose plus avancé. Au cours de la réfrigération, la diminution globale de l'expression des protéines semble plus marquée dans le groupe ‘Fonte faible'. Dans le groupe ‘Fonte élevée', la surexpression de protéines du cytosquelette après réfrigération suggère un niveau de protéolyse plus important. D'autre part, les observations histologiques montrent des modifications de la morphologie des gouttelettes lipidiques dans le foie gras cru et au cours de la cuisson. Il semble que la fonte soit d'autant plus importante quand des fusions de gouttelettes lipidiques surviennent dans le produit cru. De plus, lors de la cuisson, les observations sur foies gras du groupe ‘Fonte élevée' révèlent une densification de la matrice non lipidique plus importante. Ces changements morphologiques et structuraux pourraient correspondre à un tissu plus fragile et donc plus sensible à la fonte lipidique. L'ensemble des résultats acquis dans ce projet permet de formuler une hypothèse sur le déterminisme biologique de la fonte lipidique même si certains mécanismes identifiés restent à confirmer. De plus, la relation entre l'aptitude à la stéatose et la qualité technologique du foie gras est une piste à explorer afin de mieux comprendre la variabilité de la fonte lipidique à la cuisson observée lors de la cuisson du foie gras de canard. / This work aims to identify the biological mechanisms involved in the determinism of variability of fat loss during cooking of duck ‘foie gras'. To achieve this objective, we carried out a characterization of liver tissue, according to its later behaviour during cooking, and based on an integrated approach combining biochemistry, proteomic and histology. On the one hand, the differential expressions of proteins early post mortem and during chilling indicate a different state of steatosis between the low and high fat loss groups. Livers with low fat loss during cooking were still in anabolic processes with regards to energy metabolism and protein synthesis, whereas livers with high fat loss during cooking developed cell protection mechanisms. The overall expression of proteins in the low ionic strength fraction was lower after chilling which revealed a down regulating effect of chilling on biological processes. In the high ionic strength fraction, the results showed an over expression after chilling of proteins from cytoskeleton and their associated proteins. This suggests that the variability of technological yield observed in processing plants could be explained by different state of ageing of fatty livers during chilling, most likely associated to different proteolytic pattern. On the other hand, the histological observations showed that, in raw livers, the lipid droplets were nearly spherical while after cooking, they were larger and lost their spherical shape. We also observed a decrease in the number of droplets after cooking, probably due to droplet fusion caused by the heat treatment. Fat loss during cooking was higher when there was more fusion of lipid droplets before cooking. Furthermore, it seemed that the effect of cooking on liver tissue was different between the two fat loss groups. These morphological modifications could mark a fragility of the tissue associated with a higher fat loss during cooking. All the results acquired in this work allowed us to draw a hypothesis on the biological determinism of cooking losses eventhough some of the identified mechanisms still need to be confirmed. The relationship between the potential to develop steatosis and the technological quality of ‘foie gras' deserves further studies in order to better understand the variability in fat loss during cooking observed in processing plants.
