An appraisal of warm temperate mangrove estuaries as food patches using zooplankton and RNA: DNA ratios of Gilchristella aestuaria larvae as indicators

Mangrove habitats are considered as the ideal fish nursery as they are known to increase the growth and survival of juvenile fishes by providing enhanced food availability and protection. However, most studies have focused on tropical mangroves with a few recent warm temperate studies finding conflicting results. Furthermore, the nursery value of South African mangroves to fishes remain understudied in subtropical areas, while warm temperate mangroves are yet to be evaluated. This study aimed to assess whether mangrove presence leads to any advantage to the larvae of an important estuarine resident fish species, Gilchristella aestuaria, by comparing the food patch quality of South African warm temperate mangrove and non-mangrove estuaries. Results indicate that larvae fed primarily on the dominant prey species, Pseudodiaptomus hessei, Paracrtia longipatella, and Acartiella natalensis. However, postflexion larvae consumed more of the larger species, P. hessei, within the two mangrove estuaries (16.09 %V in Nahoon and 13.79 %V in Xhora) than the two nonmangrove estuaries (12.20 %V in Gonubie and 7.05 %V in Qora), despite other prey species occurring at similar densities. Results indicate that mangrove habitats acted as sediment sinks, slightly reducing the turbidity of these estuaries which resulted in postflexion larvae actively selecting larger, more nutritious prey, which in turn, significantly increased their individual instantaneous growth rates (0.11 ± 0.21 Gi) when compared to postflexion larvae in non-mangrove estuaries (0.09 ± 0.12 Gi). This study found that mangrove presence was significantly related to postflexion larval densities when coupled with abiotic (such as temperature and turbidity) and biotic factors (such as predator-prey interactions). Understanding the spatial and temporal dynamics, predator-prey interactions as well as the growth and survival of G. aestuaria is particularly important as they are key zooplanktivores that are prey to other species in estuarine food webs.

Cranial ontogeny of the sole, dagetichthys marginatus (soleidae), with considerations on the feeding ability of larvae and early juveniles

The overall aim of this study was to gain a better understanding of the feeding mechanisms of the larvae and early juveniles of the sole, Dagetichthys marginatus (Soleidae), with which to assess the suitability of current feeding protocols and to facilitate the development of an appropriate pelleted feed. This was achieved by examining the ontogeny of the cranium of laboratory reared sole, with particular emphasis on those elements associated with feeding and by comparing the cranium of juvenile fish with that of adult fish. At 4 dah (days after hatch) Dagetichthys marginatus larvae develop the first rudimentary branchial arches that facilitates the capture and ingestion of food items. Subsequent development of cranial structures, such as the oral jaws, suspensorium, neurocranium, hyoid and branchial arches and the opercular apparatus enables the larvae, at 16 dah, to switch from ram feeding to suction feeding on live prey. The use of live Artemia nauplii from 4 to 16 dah is therefore appropriate. The first morphological asymmetries developed at 16 dah in the dentaries and at 22 dah the maxillae and the premaxillae began to show asymmetries. Teeth were present only on the blind side of the oral jaw elements and during this period (16 to 22 dah) the existing elements began to ossify. From 16 to 35 dah the standard feeding protocol consists of a combination of pelagic (Artemia metanauplii) and benthic prey (dead, frozen Artemia nauplii) and from 25 dah onwards a sinking pellet is provided. The time (dah) at which frozen Artemia and sinking pellets were provided, appropriately corresponded to the initiation of benthic feeding behaviour. However at this stage the use of pelagic Artemia metanauplii is inappropriate and unnecessary. At 31 dah the cranial morphology resembled that of adult fish. Adult D. marginatus display extreme asymmetries among the elements of the oral jaws, the suspensorium and certain elements of the neurocranium. Elements on the blind side are larger and more robust than those on the ocular side and are adapted for feeding, while those on the ocular side appear to have a respiratory function. From 31 dah the cranial elements are identical to those of adult fish, suggesting that no further feeding behavioural changes occurred and that a sinking pellet, of which the nutrient composition meets the requirements of the fish, would be appropriate for ongrowing.

A study of coeval sibling cannibalism in larval and juvenile fishes and its control under culture conditions

The primary objective of this study was to examine environmental parameters thought to affect cannibalism in certain fish species. It was found that environmental, behavioural, genetic and physiological factors all affect cannibalism in the species exhibiting the phenomenon. The diversity of factors nfluencing cannibalism served to illustrate the complexity of this behaviour pattern. Feeding to satiation was found to suppress cannibalism in catfish, trout and koi carp. High population densities were found to increase the rate of cannibalism, thereby acting as a population regulation mechanism for catfish, trout and the common and koi carps. Live food, as compared with dry pelletized feed was found to significantly suppress cannibalistic aggression. Catfish grown in total darkness, provided with refuges and living in turbid conditions were found to exhibit lowered cannibalistic and territorial aggression. Various lines of evidence suggested that cannibalism has a genetic basis, as was shown by the differences in cannibalistic behaviour of the two strains of Cyprinus carpio, viz. common and koi carp, with cannibalism being higher in the latter. This finding substantiates the hypothesis that cannibalism is genetically controlled and therefore open to evolutionary change. It is concluded that cannibalism is adaptive in times of food limitation, but that it is merely a by-product of normal feeding behaviour when food is abundant. Since cannibalism is advantageous and thus adaptive, the question arises ai to whether selection is occurring at the individual or the population level (or both). It was concluded that it is acting at the individual level, and that any benefits accruing at the population level iv were simply the effect of the initial cause, viz. individual selection. One of the aims of this study was to determine whether cannibalistic tendencies in fish are influenced by differing life history style trajectories. Based on the results of this study it is hypothesized that cannibalism is an r-selected trait. wi th the understanding gained from the knowl edge of the fundamental principles governing cannibalism, certain recommendations for its control in fish culture could be made. It is, however, imperative that further intensive studies be carried out to understand more fully this complex subject. General "rules" for regulating cannibalism could be helpful for any given cannibalistic species. However, in considering the differing life-history styles of each species, it becomes evident that species-specific guidelines need to be worked out. Until then, any suggestions for cannibalistic control offered to the aquaculturist can only serve as unrefined tools.

Development of artificial diets for delivery of water-soluble nutrients to altricial fish larvae

In the present study, the potential of low-melting point lipids, namely methyl palmitate (MP) and menhaden stearine (MS), were studied for delivery of water-soluble materials to fish larvae. Addition of the solid emulsifier, sorbitan monopalmitate (SMP) resulted in higher RE compared to the liquid emulsifier sorbitan sesquioleate (SSO). Additions of SMP were found to have a positive effect on the stability of lipid spray beads (LSB) at concentrations >10% (w/w) of lipid. LSB were shown to retain riboflavin with the highest RE when the lipid matrix was composed of a mixture of MP+10% SMP+1O% ethyl cellulose (w/w). Use of MP facilitated digestive breakdown of LSB by larvae of zebrafish, Brachydanio rerio, and glowlight tetra, Hemigrammus erythrozonus, as indicated by release of riboflavin from ingested LSB. LSB composed of MS performed better than LSB composed of MP when particulate glycine was incorporated within LSB. Highest retention efficiencies (RE) and delivery efficiencies (DE) were achieved by LSB composed of 100% MS. RE and DE of LSB were further improved when glycine was encapsulated as aqueous droplets rather than in a particulate form. The overall performance of LSB containing tyrosine showed that nutrients with similar water-solubilities to tyrosine can be very effectively delivered to early fish larvae. Visual observation indicated that LSB composed of 100% MS were broken down by larvae of clownfish, Amphiprion percula. Zein-bound particles (ZBP) were not efficient in delivering riboflavin to fish larvae due to high leaching rates. In an attempt to improve ZBP, we developed complex particles (CP) containing LSB incorporated in a dietary mixture bound by zein, to obtain a single particle type. The potential of LSB for delivery of free amino acids (FAA) was evaluated when LSB were incorporated within CP. RE and DE for tyrosine were significantly greater than those for alanine, glycine and serine. There was a significant inverse correlation between RE for FAA and their solubilities where FAA with low solubilites resulted in higher RE. Development of CP with high RE and DE for amino acids is an important step towards formulating a diet to allow early weaning of fish larvae onto artificial diets. / Graduation date: 2003

A contribution towards the taxonomy of the ichthyoplankton species community and an understanding of its dynamics along the south-east coast of South Africa

This study was prompted by the need to remedy the situation that existed with respect to the poor status of our knowledge regarding the ichthyoplankton assemblage of the nearshore region along the south-east Cape coast of South Africa. The first chapter provides a brief introduction to the field of ichthyoplankton research and includes a summary of the status of research in southern Africa and an explanation of early life history terminology. The selection of all sample sites, times and strategies is also outlined. The study area along the south-east Cape coast with respect to its location, climate and physical oceanography is described in the second chapter, as is the gear used, bongo nets and an RMT1x6. A sampling protocol for the use of bongos from a small ski-boat, and the RMT from the research vessels, and for the handling and processing of samples was established. The selection of Middlebank as the main monthly sampling site within the Tsitsikamma National Park (TNP) was based on taxonomic diversity as well as logistical and safety constraints. The effect of mesh size and time of sampling with bongo nets on the catchability of ichthyoplankton was investigated in chapter three. Most data was accumulated during Sea Fisheries research cruises, with additional collections coming from the National Parks vessel. Although the differences were not significant, the 505 μ mesh nets captured larger larvae, with catches comprising higher percentages of flexion and postflexion larvae. Larval concentration and size were consistently greater in samples from periods of reduced light intensity, but significant differences were the exception. It was decided that sampling with 505 μ mesh nets during daylight would provide a representative sample of the available ichthyoplankton assemblage, while at the same time being the most practical and least time consuming with respect to handling, clogging and backflushing. In chapter four, the early life history stages of thiny of the seventy-five taxa sampled are described, reflecting the paucity of information which existed on the ichthyoplankton of the nearshore zone in the south-east Cape. These descriptions are seen as an important contribution towards any future research efforts in the region, but as many of these descriptions are based on few or single specimens, it is realised that the description of egg and larval stages will be an ongoing process. Based upon the data collected during this study, an ichthyoplankton species checklist was established in chapter five. Seventy-five taxa of fish larvae were identified to either family, genus or species level. A number of squid para-larvae were also encountered. Similarities and discrepancies with a previous survey in the region are presented. The temporal distribution of eggs and larvae between August 1993 and October 1996 was established, and the spatial distribution of ichthyoplankton along an offshore transect was determined between January 1995 and May 1996. Only 7 species from Middlebank and twelve from all stations combined displayed seasonal trends, with most of these being prevalent during winter months. Egg production, both over Middlebank and from all stations combined, appeared to be consistent, with no seasonal trends. Based upon the results from the offshore transect samples, it would appear that a single ichthyoplankton assemblage exists from Storms River out to fifteen nautical miles. Although a variety of statistical methods were applied to the data during this study, low egg and larval concentrations and a low sampling frequency meant that results had to be interpreted carefully. Chapter six describes the preliminary investigation into the vertical distribution of eggs and larvae. During two research cruises, discrete depth sampling using an RMT1x6 net was performed, with the majority of samples being accompanied by physical data provided by a CTD rosette sampler. No definite patterns could be seen as larval concentrations were low, and the short time scale did not allow for the identification of any diel migratory patterns. The relationship between total larvae and the physical environment was poor. The only possible relationship was that between plankton volume and total egg and total larval concentrations. The TNP may play an important role in the conservation of reef fish and the seeding of nearby fishing grounds through the export of pelagic eggs and larvae. Chapter seven describes a preliminary investigation into the dispersal potential of ichthyoplankton from the TNP. Based upon longshore currents determined from drogues, ADCP vectors and current meter readings, it was clear that if larvae were passive drifters, the potential for their dispersal from the TNP did exist. This pilot study showed that future work should concentrate not only on the oceanographic aspects, but on behavioural aspects of larvae which may enhance or retard dispersal. In the final discussion, it is emphasised that while this study went a long way to increasing our knowledge of the nearshore ichthyoplankton community, the true picture of the temporal and spatial dynamics of the species assemblage may only be revealed once more intensive sampling has been performed. The resolution of the early life history stages of the sparids and the identification of eggs to species level are seen as priorities for the near future. A complete species checklist for the TNP is provided based upon previous surveys (both on adult and larval fishes), this work and personal observations. A total of 171 species of fish from 70 families were identified, illustrating that the eggs and larvae of many species in the TNP have yet to be sampled. A brief description of the collaborative effort which is envisaged for the TNP over the next few years is provided.

The ontogeny of respiration in herring and plaice larvae

The study of larvae, their behaviour & physiology has gathered momentum in recent years due to the development of improved techniques of rearing during the past decade. Many marine larvae have been successfully reared in the laboratory (Shelbourne, 1964; Schumann, 1967; Blaxter, 1968,1969; Houde & Palko, 1970; Futch & Detwyler, 1970; Lasker, Feder, Theilacker & May, 1970). Fish larvae, in particular those of most marine species form an important tool for research in that at hatching they only possess the rudiments of most organs. Thus they are ideal material for ontogenetical studies. Although the respiratory mechanisms of adult fish have been the subject of a great deal of investigation from the point of view of gas exchange (see Randall 1970) gill structure (Hughes, 1966; Hughes & Grimstone, 1965; Newstead, 1967; to name a few) and dimensions (see Muir, 1969) gill ventilation and perfusion (see Shelton, 1970) circulatory systems (see Randall, 1970) bioenergetics, (see Brett, 1970) the respiratory systems of larvae have not been investigated in any great detail. Apart from a few studies on oxygen uptake (see Blaxter, 1969) and Harder (1954) on the development of branchial elements, no detailed study of the development of respiratory mechanisms have been made in marine fish larvae. The purpose of this study was to investigate the development of respiration in two species of marine larvae viz. the herring (Clupea harengus L.) and the plaice (Pleuronectes plates sa L.) These two species are well separated taxonomically and both adults and young have very different life histories. Herring lay demersal eggs, the plaice pelagic ones. The yolk-sac larvae of both species are planktonic, later feeding on diatoms and copepod nauplii and much later copepods. Adult herring are pelagic, living in mid water as juveniles and moving into deeper water with age, ranging from offshore to about 200m. They perform migrations partly caused by the distribution and density of food organisms. In contrast to this plaice at metamorphosis show an interesting asymmetry in that one eye migrates over the head and comes to lie against its opposite number. At this stage pelagic life ceases and the young fish assumes a bottom-living existence. Other features associated with asymmetry are secondary to the migration of the eye and follow on from the adoption of the benthic mode of life. They range from the shoreline when young to lOOOm feeding on bottom living organisms. During development the mechanism of respiration changes from a cutaneous one to gill respiration typical of the adult form. There is apparently no respiratory pigment in the early stages but the blood becomes pink at metamorphosis. The problem was approached from a morphological and a physiological viewpoint. The main parts of the study are as follows. (1) The survival times in water of low oxygen concentrations. (2) The oxygen uptake at normal oxygen concentrations. (3) The oxygen uptake at low oxygen concentrations. (4) Measurement of the body surface area as well as the gill area available for respiration. (5) The appearance of haemoglobin and its quantitative measurement.

The dynamics of larval fish demographics in nearshore southern Lake Michigan

Nearshore larval fishes were collected with an oblique trawl tow during day and night from mid-May to August 2010 and 2011 in the Indiana waters of Lake Michigan to determine diel differences in the distribution, depth, and abundance of larval fishes. Alewife, spottail shiner, yellow perch and round goby were the prevailing larval species. Alewives dominated the catch and were in highest abundance near East Chicago. In general, night catch rates were greater when compared to day catch rates, suggesting a diurnal difference in trawl susceptibility. Stratified larval trawling was subsequently conducted during June and July 2011 to detect whether diurnal vertical migration existed, potentially affecting day and night catch rates. Vertical migration was not detected in the stratified larval sampling, eliminating it as a factor in higher nighttime catch rates. Further, the most common fish, alewife, was measured (TL) to determine whether size was a factor in trawl avoidance and to provide information regarding trawl selectivity. Night trawls yielded larger size classes of alewife, potentially explaining higher nighttime catch rates. These data suggests a size bias of our larval trawl exists when comparing day and night samples, as well as the limited efficiency of our trawl to catch larger larval size classes. / Department of Biology

Diel Changes in the Vertical Distributions of Some Common Fish Larvae in Southern Kaneohe Bay, Oahu, Hawaii

Nine series of vertically-stratified zooplankton tows were made with a closing net at a single station in southern Kaneohe Bay, Oahu, Hawaii, between 31 August 1973 and 11 April 1974. Sampling periods occupied from 12 to 26 hours, with tows usually taken at about 3 m intervals between the surface and a maximum depth of 10 m. A total of 21,254 fish larvae of 49 kinds was collected. Six species were abundant: Foa brachygrammus, Omobranchus elongatus, Callionymus decoratus, Caranx mate, Stolephorus purpureus, and Abudefduf abdominalis. Blennius sp. and Gnathanodon speciosus were commonly taken in small numbers. These common larvae displayed five "distribution patterns: 1. F. brachygrammus and the smallest S. purpureus were most abundant near the surface at night and at depths below 4 m during the day; 2. C. mate and G. speciosus were dispersed throughout the water column at night and usually most abundant between 5 m and 6 m depth during the day; 3. Blennius sp., o. elongatus, and A. abdominalis were dispersed throughout the water column at night and concentrated near the surface during the day; 4. the larger ~. purpureus maintained a level of maximum abundance below 6 m day and night; 5. C. decoratus was taken at all depths at all times. Patterns 1, 2, and 3 are shown to be light-related. Pattern 4 is shown to be partially attributable to avoidance of the towed net by S. purpureus larvae larger than about 6mm, and a feeding-related migration is proposed to account for pattern 5. The observed patterns are analogous to those shown for fish larvae in the open ocean on scales of from 50 m to 200 m. It is proposed that Kaneohe Bay represents a vertically compressed ocean with respect to the vertical distribution of fish larvae. / Typescript. Bibliography: leaves 130-134.
