Os estudos na literatura dos gangliosídios abrangem desde suas propriedades e funções nas membranas neuronais assim como a sua farmacologia no sistema nervoso central, quando injetados na forma pura em animais e humanos. Entretanto, o estudo de seus efeitos no sistema imunológico, quando associados aos lipossomas, ainda é escasso, o que motivou o desenvolvimento dessa pesquisa. Este trabalho compreendeu a preparação e caracterização de lipossomas multilamelares compostos de dipalmitoilfosfatidilcolina, colesterol, dihexadecilfosfato e gangliosídios, monossialogangliosídio (GM1) e Mist (mistura de GM1 21%, dissialogangliosídio (GD1a) 40%, dissialogangliosídio (GD1b) 16%, trissialogangliosídio (GT1b) 19%) e avaliação do efeito imunológico destes lipossomas quando injetados em camundongos. Os animais testados foram camundongos do tipo Balb/c, os quais receberam seis doses de lipossomas a cada duas semanas, perfazendo um total de doze semanas. Após este período os animais foram sacrificados e seus soros submetidos a testes ELlSA para avaliação da produção de anticorpos antigangliosídios, classes IgM e IgG. Em termos de significância estatística os lipossomas livres de gangliosídios e os associados à Mist não apresentaram imunogenicidade e alguns soros de animais tratados com lipossomas associados ao GM1 apresentaram algumas classes de imunoglobulinas. Porém, em termos de avaliação para diagnóstico de anticorpos, todas as formulações foram consideradas não imunogênicas quando considerada as unidades arbitrárias / Abstract: The pharmacological importance of gangliosides has increased in the last few years due to the identification of these compounds as antigens, producing antiganglioside antibodies, which may be involved in autoimmune diseases or neuropathy. Studies in ganglioside literature encompass their properties and functions in the neuronal membranes as well as their pharmacology in the central nervous system upon injection in pure ganglioside form in animais and humans. However, as associated to the liposomes, the studies of their effects on the immunological system are still scarce, which motivated the development of this research. This work includes the preparation and characterization of multilamellar liposomes consisting of dipalmitoyl phosphatidyl choline, cholesterol, dihexadecylphosphate and gangliosides, monosialoganglioside (GM1) and Mist (mixture of GM1 21%, disialoganglioside (GD1a) 40%, disialoganglioside (GD1b) 16%, trisialoganglioside (GT1b) 19%) and evaluation of immunologic effect of hese liposomes as administered in mice. The animais tested were Balb/c mice, which were treated with six liposome doses at 2 week-intervals during twelve weeks. After this period the animais were sacrificed and their sera were assayed by ELlSA tests in order to evaluate the production of antiganglioside antibodies of IgM and IgG classes. With respect to the statistical significance, the liposomes without gangliosides and the ones associated to the Mist were non immunogenic and some of animal sera, which were treated with liposomes associated to the GM1, showed some Immunoglobulin classes. Brainstem Gangliosides in Suddden Infant Death Syndrome

Recent studies have shown that the Sudden Infant Death Syndrome (SIDS) is related to abnormal control of respiration (Ischemic degeneration of the brainstem may play an important role in altered respiratory control leading to death). In our studies we have examined brainstem ganglioside compositions in samples derived from SIDS victims and appropriate controls. Gangliosides are acidic glycosphingolipids that contain sialic acid. The high concentration of gangliosides in the central nervous system (CNS) implies that these lipids play an important role in CNS function. Some studies have indicated that gangliosides may function as receptor site determinants or modifiers, and in neural transmission. In our studies we used the Tettamanti, et al methodology to extract gangliosides, and High Performance Thin Layer Chromatography (HPTLC) and laser densitometry techniques for ganglioside analysis. The results of these analyses are being employed to establish lipid profile patterns to determine if there are significant variations in these lipid patterns between SIDS and control groups.
