Detection Of Genetically Modified Insect Resistant Tomato Via Polymerase Chain Reaction

Tomato, which is one of the most important component of human diet, has been genetically modified to develop some properties like delayed ripening and insect resistance. In order to give a choice to the consumer, it is necessary to detect and label GM foods. This study was carried out to detect genetically modified tomato samples purchased from different food markets of Turkey. PCR method was used to detect genetically modified insect resistant tomatoes. The DNAs of collected samples were isolated according to CTAB DNA extraction protocol and the amplification capacity of isolated samples were checked with patatin specific control PCR. Screening tests of tomatoes were done by targeting 35S promoter, Nos terminator and NptII kanamycin resistance gene with four primer sets. It was aimed to detect Cry1A and Cry1Ac genes with three PCR systems, in order to identify insect resistant samples. From twenty-eight samples, twenty-two gave positive amplification signal in NptII specific PCR system and results were confirmed with sequence analysis. Additionally, we observed seventeen and ten DNA fragments with Cry1A-F/Cry1A-R and Cry1Ac-F/Cry1Ac-R primer sets respectively, it is necessary to confirm these results with DNA sequencing.

Screening For Genetically Modified Tomatoes &amp / Tomato Seeds And Identification Of Cry1ac And Sam-k Specific Modifications Using Gene And Construct Specific Pcr

This study was carried out to analyze tomato samples and tomato seeds, purchased from different food markets of Turkey randomly, for the presence of genetic modification by using PCR method as it allows more specific detection. The DNAs of collected samples were isolated according to CTAB DNA extraction protocol and also with extraction kits. Screening tests of tomatoes were done by targeting 35S promoter, NOS terminator and NptII kanamycin resistance gene with eight different primer sets. Real time PCR is used to confirm 35S and NOS positives results obtained from conventional PCR. In this study, it was observed that 14 out of 35 seed samples, and 14 out of 40 fresh tomato samples which were screened had at least one transgenic element of 35S promoter, NOS terminator and NPTII kanamycin resistance gene indicating the possible presence of genetic modifications. After screening, gene specific studies were carried out for PG, sam-k indicating F type ripening delayed tomato and the 35 1 N lines respectively and cry1Ac genes inserted in 5345-1 insect resistant tomato line. PG and sam-k specific primers were not amplified in any of the samples investigated whereas 18 out of 75 samples were cry1Ac positive and 1 out of 75 samples was sam-k positive. Positives were confirmed by sequence analysis. Additionally, construct specific primers specific to 5345-1 and 35 1 N lines were designed. PCR amplicons indicate the existence of the construct sequence. In order to verify the results, PCR products were sent to sequence analysis

Magistro baigiamajame darbe atskleista, kad daugelis respondentų yra neapsisprendę dėl savo pozicijos GMO gamybos bei naudojimo atžvilgiu, tačiau beveik trečdalis asmenų GMO gamybai bei naudojimui nepritaria. Dauguma gyventojų mano, kad GMO ne tik kenkia žmogaus sveikatai, bet ir yra pavojingi, taip pat kenkia aplinkai, o GMO turinčių produktų naudojimas gali negrįžtamai pakeisti genetinį žmogaus kodą ir augalų bei gyvūnų populiacijose gali sukelti negrįžtamus mutacinius pokyčius. Vis dėlto GMO produkciją kai kurie gyventojai linkę naudoti dėl mažesnės jų kainos ir ilgesnio vartojimo galiojimo termino. Nors kai kurie respondentai ir buvo įsitikinę GMO teikiama nauda bei tolimesniu GMO gamybos bei naudojimo plitimu ateityje, tačiau beveik visi respondentai buvo įsitikinę ir tuo, jog GMO gamyba bei naudojimu labiausiai suinteresuotos yra kuo didesnio pelno siekiančios monopolinės kampanijos. Vienareikšmiškai galima teigti, jog nors gyventojai apie GMO informacijos daugiausiai ir gauna iš televizijos laidų, pokalbių su draugais pažįstamais ar giminaičiais bei iš spaudos leidinių arba naršydami internete, tačiau tos informacijos kiekis yra nepakankamas ir daugiau kaip pusė respondentų norėtų, jog informacijos apie GMO būtų pateikiama daugiau. / The Master’s final paper on GMO discovered that the majority of respondents have no strong opinion about production and use of GMO, but one third of responses on production and use of GMO were negative. Many people consider that GMO not only negatively affect human health, but also damage environment. Moreover, eating genetically-modified food can irreversibly change human’s genetic code as well as irreversible mutations in plants’ and animals’ populations. However, some people tend to use GM foods because they are cheaper and can be stored longer than other foods. Even though some of the respondents see benefits of GMO production and usage, almost all of the respondents claim that genetically engineered products interest mostly large companies that are seeking to profit from GMO. The most obvious finding to emerge from this study is that people learn about GMO from mass media and communicating with friends and relatives mostly, but they also admit the lack of credible information on GMO. More than a half of the respondents would like to receive more information about genetically modified organisms.

Os Organismos Geneticamente Modificados (OGM) surgiram há poucos anos e estão sendo discutidos por toda a comunidade científica por ter implicações ambientais e na saúde pública. Por ser um tema vasto, foi estudado apenas a liberação para o plantio comercial dos OGM, mostrando suas vantagens e desvantagens. Neste trabalho são apresentados vários dados das plantações comerciais dos OGM no Brasil e no restante do mundo, mostrando o aumento do uso destes organismos na agricultura mundial. Esta dissertação apresenta um breve histórico da liberação para o plantio comercial dos OGM no Brasil e os seus aspectos legais. Foram pesquisados os aspectos legais para essa liberação, levando em conta a necessidade de ser feito o Estudo de Impacto Ambiental (EIA), que é um instrumento de aplicação do Princípio da Precaução existente no ordenamento jurídico brasileiro, do qual a Avaliação de Impacto Ambiental (AIA) é uma de suas ferramentas. Nesse contexto foi estudada a legislação brasileira sobre o meio ambiente e sobre os OGM. Para se decidir quais são os melhores métodos de AIA, para liberação comercial do plantio dos OGM, de acordo com as exigências legais, foi feita uma pesquisa revisando esses métodos. Foi verificado para cada Método se ele apresenta características de identificar satisfatoriamente os impactos ambientais em relação aos seus efeitos, espaço e tempo. Sabendo-se quais as exigências de avaliação de impacto necessárias para a liberação do plantio dos OGM foi possível identificar as metodologias de AIA mais indicadas para o caso. Concluiu-se que os métodos mais indicados para a liberação comercial do plantio dos OGM são a Matriz de Interação e o AMBITEC-AGRO / Genetically Modified Organisms (GMO) have only recently appeared and are being discussed by all of the scientific community for having environmental and public health implications. By being a vast subject, only the release for the commercial plantation of the OGM was studied, showing its advantages and disadvantages. In the following work, data of GMO commercial plantations in both Brazil and globally are presented, showing an increase of the use of these organisms in world-wide agriculture. Also included is a brief historical account of the approval and release of GMOs for commercial plantation in Brazil. It was taking in account the necessity of conducting the Environmental Impact Study (EIA), a component of the Environmental Impact Assessment (AIA) that is an instrument of the application of the Precaution Principal in the Brazilian legal system. Additionally, Brazilian environmental regulations and regulations pertaining to GMOs was studied. An analysis of the best methods of AIA legally required for commercial authorization of planting GMOs was performed. Each method was verified if it met satisfactory characteristic identifying the environmental impacts concerning to its effect, time, and space. It was possible to determine the best AIA methodology for this case by matching the necessary requirements for evaluating the impact of authorizing the planting of GMOs. It was concluded that the Interaction Matrix and the AMBITEC-AGRO are the best methods for the commercial authorization of planting GMOs
