Swelling and protein adsorption characteristics of stimuli-responsive hydrogel gradients

<p>In this work, a gradient of interpenetrating polymer networks, consisting of anionic</p><p>and cationic polymers, has been investigated with respect to protein resistant</p><p>properties and swelling characteristics at different pH and ionic strength</p><p>conditions.</p><p> </p><p>The swelling and protein adsorption have been studied using <em>in situ </em>spectroscopic</p><p>ellipsometry(SE) and imaging surface plasmon resonance(iSPR) respectively.</p><p>It has been shown that, by altering the buffer pH, the region of lowest</p><p>protein adsorption on the surface could be moved laterally. The swelling has</p><p>similarly been shown to respond to both changes in pH and ionic strength. Additionally,</p><p>the arise of surface charge and the polymer swelling in solution, both a</p><p>consequence of the ionisation of fixed charges on the polymer, have been indicated</p><p>to occur at different buffer pH.</p><p> </p><p>The studied polymer systems show promising properties for future applications</p><p>in, for example, the biosensor area, where the surface chemistry can be</p><p>tailor-made to work optimally in a given environment.</p>

Ecosystem Restoration and Subtropical Seagrass Fishes: Insights into Salinity Effects from Habitat Selection and Preference Tests

The work of this Master of Science thesis project is an analysis of salinity effects on nearshore epifauna along the western shore of Biscayne Bay in southeast Florida, USA. Field collection surveys have found a high probability of occurrence of bigeye mojarra (Eucinostomus havana) in salinities near 25 ppt. In a salinity gradient observation experiment test subjects of the same species and size class were also observed more frequently at 24 ppt. In this analysis presence and abundance patterns found in field surveys were compared with behavioral results obtained in the observation tank. This apparatus provided insight into distribution patterns of the bigeye mojarra (Eucinostomus havana) and possible changes in distribution that may result from habitat changes in the future. Historically, the western shore of Biscayne Bay was more freshwater marsh than the mangrove dominated marine environment that prevails today. Changes to fresh water inputs into the Bay are planned through projects of the Comprehensive Everglades Restoration Plan (CERP). CERP is a joint Florida state and U.S. federal effort to redesign surface water flow through the canal system of South Florida, replenish the Everglades ecosystem, and restore a more natural quantity, timing, and distribution of flow into Biscayne and Florida Bays. Approved by the U.S. Congress as part of the Water Resources Development Act of 2000, CERP will be implemented by the South Florida Water Management District (SFWMD) and the U.S. Army Corps of Engineers (USACE). This plan is designed to restore the ecosystem from its freshwater core to the coastal wetlands recreating a condition close to that existing before the current system of flood control drainage canals was begun in 1903 and continued by the federal Central and Southern Florida Project in 1948 (www.evergladesplan.org). Changes are planned to divert a portion of canal flows to Biscayne Bay into coastal wetlands as sheet flow and surface runoff. Planned changes to freshwater delivery may change the habitat along the shoreline and thus the distribution of prey organisms living in this habitat. This may in turn affect predator fish important to local recreational and commercial fisheries as well as other predators such as wading birds. The analysis and prediction provided in this thesis work is important for better understanding the effects of restoration efforts on the Bay nearshore habitat and its condition as essential fish habitat, which is federally regulated by the Magnuson-Stevens Fishery Conservation and Management Act (Magnuson-Stevens, 1996). The distribution of small fish and invertebrate inhabitants of the nearshore environment and habitat environmental qualities have been recorded over the past five years from throw-trap surveys of the western shoreline of Biscayne Bay as part of a CERP-sponsored monitoring program. This pattern is correlated with salinity, but there may be other factors affecting the distribution of this species.

Comparison between two different cryoprotectants for human sperm, with emphasis on survival

The increasing number of patients undergoing treatment with assisted reproductive techniques (ART) during the past years have led to the need of developing different methods for separation of spermatozoa that can be used for different fertilisation procedures and for freezing. Cryopreservation of spermatozoa includes preparation, freezing, storage and thawing. In this study two different cryomedia (Cryo Protec I and Cryo Protec II) for human spermatozoa were compared. The main outcome was spermsurvival rate for spermatozoa after freezing. Sperm viability was assessed using the Hypo-osmotic swelling test which is based on osmolality. A total of 86 samples of semen were used in this study (Cryo Protec I=38, Cryo Protec II=48). The survival rate between the two cryomedia did not differ much but Cryo Protectant I showed a small increase in survival for the spermatozoa after freezing. The Hypo-osmotic swelling test also showed similar values of viable spermatozoa for the two cryomedia both before and after freezing.

Upper gradients and Sobolev spaces on metric spaces

The Laplace equation and the related p-Laplace equation are closely associated with Sobolev spaces. During the last 15 years people have been exploring the possibility of solving partial differential equations in general metric spaces by generalizing the concept of Sobolev spaces. One such generalization is the Newtonian space where one uses upper gradients to compensate for the lack of a derivative. All papers on this topic are written for an audience of fellow researchers and people with graduate level mathematical skills. In this thesis we give an introduction to the Newtonian spaces accessible also for senior undergraduate students with only basic knowledge of functional analysis. We also give an introduction to the tools needed to deal with the Newtonian spaces. This includes measure theory and curves in general metric spaces. Many of the properties of ordinary Sobolev spaces also apply in the generalized setting of the Newtonian spaces. This thesis includes proofs of the fact that the Newtonian spaces are Banach spaces and that under mild additional assumptions Lipschitz functions are dense there. To make them more accessible, the proofs have been extended with comments and details previously omitted. Examples are given to illustrate new concepts. This thesis also includes my own result on the capacity associated with Newtonian spaces. This is the theorem that if a set has p-capacity zero, then the capacity of that set is zero for all smaller values of p.

Vascular Smooth Muscle Precursor Cell Behavior in Non-Uniform Stretch Environments

Cells in the body respond to mechanical loads in ways that are crucial to normal and disease physiology. Understanding these processes is difficult due to the complex mechanical environment in vivo. In this research, we have developed several cell-stretching devices capable of subjecting cell cultures to non-uniform stretch distributions in order to investigate pathological responses of vascular smooth muscle cells to physiologic stretches. 10T1/2 cells were cyclically stretched with these devices for 24 hours upon silicone membranes, PDMS tubes, and within 3D PEGDA hydrogels. After stretching, altered cell behaviors were measured, including orientation, proliferation (quantified by BrdU incorporation), and gene expression (quantified by real-time, RT-PCR). Cells demonstrated marked changes in orientation, proliferation, and mRNA expression, which all varied with cellular location in the non-uniform environment. More specifically, increased orientation, increased proliferation, and more dramatically altered mRNA expression were found in regions of high, uniaxial stretch, relative to regions of low, near-equibiaxial stretch. These findings demonstrate the capabilities of graded stretch distributions to produce graded cell responses, indicating potentially localized smooth muscle cell behavior in a diseased artery. The novel devices employed herein will hopefully improve our understanding of these complicated cellular pathways, ultimately allowing for improved treatment or prevention of vascular disease.

Rates of diagnosis and treatment of Attention Deficit/Hyperactivity Disorder in Manitoba children: considering the socioeconomic gradient

This project investigated the diagnosis and psychostimulant treatment rates of Attention Deficit/Hyperactivity Disorder (ADHD) in Manitoba children. These rates were considered according to sex, age, socioeconomic status (SES), geographical region, and comorbidity with learning disabilities (LD) and behavioral disturbances (BD). Data came from the Manitoba Population Health Research Data Repository, a comprehensive collection of administrative, registry, survey and other databases. The research population included all children aged 0 to 19 years in Manitoba (n = 319,506) with a diagnosis of ADHD (n= 9,233), during two Fiscal years (2003/2004 and 2004/2005). The term “gradient” refers to the relationship between SES and health and emphasizes the idea that the change in outcomes is gradual and occurs across the full range of SES. Results from this study indicate that region of residence (urban versus rural) and comorbid BD moderate the SES gradient, as low income, urban dwelling children with a comorbid diagnosis of BD had the highest rates of ADHD diagnoses and prescriptions. Furthermore, whereas age did not moderate the SES gradient, the crude rates indicated that the SES gradient for ADHD diagnoses and prescriptions was most pronounced in urban children 0 to 13 years of age. Otherwise, all main effects tested (sex, age, socioeconomic status, geographical region, and comorbid BD and LD) were significant in both the diagnosis and prescription models for ADHD. Policy considerations that arise out of this study include more stringent diagnostic and prescription treatment practices, additional support resources for children who are most at risk of having ADHD, and increased information about alternate treatment implementation for ADHD. / February 2008

Model Predictive Control of a Tricopter / Modellprediktiv reglering av en tricopter

In this master thesis, a real-time control system that stabilizes the rotational rates of a tri-copter, has been studied. The tricopter is a rotorcraft with three rotors. The tricopter has been modelled and identified, using system identification algorithms. The model has been used in a Kalman filter to estimate the state of the system and for design ofa model based controller. The control approach used in this thesis is a model predictive controller, which is a multi-variable controller that uses a quadratic optimization problem to compute the optimal con-trol signal. The problem is solved subject to a linear model of the system and the physicallimitations of the system. Two different types of algorithms that solves the MPC problem have been studied. These are explicit MPC and the fast gradient method. Explicit MPC is a pre-computed solution to the problem, while the fast gradient method is an online solution. The algorithms have been simulated with the Kalman filter and were implemented on themicrocontroller of the tricopter.

Lake lowering in sulphide-rich areas is currently a major environmental impact for surface water. This study focuses on whether there is a relationship between a gradient of lake lowering and surface water impacts in areas of sulphide-rich sediments, in order to better understand their contribution of heavy metals and sulfuric acid. Also, is it a reasonable method to use the reduced lake area in order to quantify the gradient? The survey was conducted by collecting water samples from reference lakes and lowered lakes from south to north in coastal areas within the county of Västerbotten. Water samples were then analyzed for TOC, pH, conductivity, anions, base cations, alkalinity, acidity, sulfate, Al, Cd, Fe, Mn, Cu, Zn and Pb. These parameters were then compared statistically using regression analysis and t-tests. The results show that no gradient was discernible in response to the reduced lake area. However, significant differences (p &lt; 0,05) between the reference lakes and all the reduced lakes were visible for pH, conductivity, Cd, Cu, Zn and Al. The study shows that there is a correlation between lake lowering and negative impacts on surface water. However, to calculate a gradient from the reduced lake area is deficient as it is limited by the lakes volume reduction, and how the area around the lake has been affected.
