O reator de pesquisas IEA-R1 opera em regime de 40 h semanais à potência de 4,5 MW. Nestas condições, os cestos disponíveis para o armazenamento dos elementos combustíveis irradiados possuem menos de metade da sua capacidade inicial. Assim, nestas condições de operação, teremos apenas cerca de seis anos de capacidade para armazenamento. Considerando que a vida útil desejada do IEA-R1 seja de pelo menos mais 20 anos, será necessário aumentar a capacidade de armazenamento de combustível irradiado. Dr. Henrik Grahn, especialista da Agência Internacional de Energia Atômica sobre o armazenamento molhado (em piscinas de estocagem), ao visitar o reator IEA-R1 (setembro/2012) fez algumas recomendações. Entre elas, a concepção e instalação de cestos fabricados com aço inoxidável borado e internamente revestidos com uma película de alumínio, de modo que a corrosão dos elementos combustíveis não ocorresse. Após uma revisão da literatura sobre opções de materiais disponíveis para esse tipo de aplicação chegamos ao BoralcanTM fabricado pela 3M devido suas propriedades. Este trabalho apresenta estudos sobre a análise de criticalidade com o código computacional MCNP-5 utilizando duas bibliotecas americanas de dados nucleares avaliados (ENDF/B-VI e ENDF/B-VII) comparativamente. Estas análises demonstraram a possibilidade de dobrar a capacidade de armazenamento de elementos combustíveis, no mesmo espaço ocupado pelos cestos atuais, atendendo a demanda do reator de pesquisas IEA-R1 e também satisfazendo os requisitos de segurança da Comissão Nacional de Energia Nuclear (CNEN) e da Agência Internacional de Energia Atômica (IAEA). / The IEA-R1 research reactor on business under the 40 h weekly to the power of 4.5 MW. Under these conditions, the racks available for the storage of spent fuel elements have less than half of its initial capacity. Thus, in these operating conditions, we will have only about six years of capacity for storage. Whereas the desired service life of the IEA-R1 is at least another 20 years it will be necessary to increase the storage capacity of spent fuel. Dr. Henrik Grahn, an expert of the International Atomic Energy Agency on the wet storage (in storage pools), to visit the IEA-R1 reactor (September/2012) made some recommendations. Among them, the design and installation of baskets made of borated stainless steel and internally lined with an aluminum film, so that corrosion of the fuel elements does not occur. After a literature review of material options available for this type of use, we got to BoralcanTM manufactured by 3M due to its properties. This work presents studies on the criticality analysis with the computer code MCNP-5 using two American libraries of the Evaluated Nuclear Data (ENDF/B-VI and ENDF/BVII), and compare results based on each database. Development of a Human Mesenchymal Stem Cell and Pluripotent Stem Cell Derived Cardiomyocyte Seeded Biological Suture for Cell Delivery to Cardiac Tissue for Cardiac Regeneration Applications

"Recent data show that 7.6 million Americans have survived a myocardial infarction (MI), and 5.1 million Americans suffer from severe heart failure. Stem cell therapy has the potential to improve cardiac function after MI. Two promising cells for cardiovascular regeneration therapies include human mesenchymal stem cells (hMSCs) and pluripotent stem cell derived cardiomyocytes (hPS-CM) each with their own unique method for improving cardiac function post-infarct. However, a limiting factor to cell therapies is that the methods currently used to deliver cells to the myocardium, including intramyocardial injection (considered the gold standard), suffer from low retention rates. To promote localization of delivered cells to the infarct and increase retention rates, our lab has developed a fibrin biological suture that can deliver human mesenchymal stem cells (hMSCs) with an efficiency of 64% compared to just 11% with intramyocardial injection in the normal rat heart. In this dissertation we sought to examine the functionality of hMSC and hPS-CM seeded sutures and their impact on cardiovascular regeneration applications. We began by delivering hMSC seeded fibrin sutures to an infarcted rat heart and found that the sutures are an effective method to deliver cells to the infarcted myocardium and demonstrated a trend towards improved regional mechanical function in the infarct region over infarct alone. Next, we transitioned to using hPS-CM and developed methods to seed the sutures, as well as a method to measure hPS-CM contractility with high spatial and temporal resolution, while concurrently capturing calcium transients. This technique allowed us to examine the contractile behavior in terms of contractile strain and conduction velocity of hPS-CM seeded on fibrin microthreads over 21 days in culture. We found that the fibrin microthread is a suitable scaffold for hPS-CM attachment and contraction and that extended culture promotes cell alignment along the length of the suture as well as improvements in contractile function in terms of increases in contractile strain and conduction velocity. Finally, we delivered the hPS-CM seeded microthreads to an uninjured rat heart and found a delivery efficiency of 67%. Overall, we further demonstrated the technology of the fibrin suture to deliver cells to an infarct as well as the ability to support the attachment, contraction and delivery of hPS-CM to cardiac tissue. Characterization of oxysterols produced in macrophages and mechanisms of regulation / Caractérisation des oxystérols produites dans les macrophages et les mécanismes de régulation

Les macrophages jouent un rôle clé dans l'athérosclérose. Après la captation massive des LDL oxydées (oxLDL), les macrophages sous-endothéliaux sont chargés en cholestérol et se transforment ainsi en cellules spumeuses qui contribuent à la formation de la plaque d'athérome. Les oxystérols, produits d'oxydation du cholestérol, sont retrouvés en quantité importante dans les oxLDL. Au niveau cellulaire, ils sont impliqués dans la régulation de l'homéostasie du cholestérol, l'induction du stress oxydatif cellulaire et de la cytotoxicité. Notre travail montre que le cholestérol, associé aux LDL et d'origine cellulaire, est fortement oxydé par les macrophages lors d'une exposition aux oxLDL. Les principaux produits d'oxydation sont le 7-cétocholesterol et 7α/β-hydroxycholestérol. De plus, nous démontrons que ces oxystérols sont exportés hors des macrophages via les HDL, mais pas l'apoA1. Nous avons aussi caractérisé les oxystérols formés dans les HDL suite à des modifications oxydatives et les HDL issues de patients diabétiques. Nous avons en outre montré que ces modifications sont associées à une diminution de la capacité des HDL à exporter les oxystérols. / Macrophages play a key role in atherosclerosis. After massive uptake of oxidized LDL (oxLDL), subendothelial macrophages are overloaded with cholesterol thereby leading to the formation of foam cells, which is one characteristic of atherogenesis. Oxysterols, the oxidation products of cholesterol, are one of major components of oxLDL; they are involved in the regulation of cholesterol homeostasis, induction of cellular oxidative stress and cytotoxicity. Our works show that both LDL derived-cholesterol and cellular cholesterol can be strongly oxidized in human THP1 and murine RAW macrophages, especially during exposure of oxLDL. The major oxidative products are 7-ketocholesterol and 7α/β-hydroxycholesterol. Moreover, we demonstrate that both oxysterols derived from LDL cholesterol and cellular cholesterol can be exported to HDL, whereas not to apoA1. Then, we studied the functionality of modified HDL and diabetic HDL on oxysterols efflux. A decrease of oxysterols efflux was observed with oxidized and glycoxidized HDL. Compared to the HDL of healthy controls, the HDL of diabetic subjects are less efficient to efflux oxysterols. Taken together, the increased production of oxysterols in presence of oxLDL and their lower efflux by modified HDL support the detrimental role of these oxidative compounds in pathophysiological conditions like diabetes.

Effects of welding parameters on the integrity and structure of HDPE pipe butt fusion welds

Butt fusion welding process is an extensively used method of joining for high density polyethylene (HDPE) pipe. With the increasing number of HDPE resin and pipe manufacturers and the diversity of industries utilising HDPE pipes, a wide range of different standards have evolved to specify the butt fusion welding parameters with inspection and testing methods, to maintain quality and structural integrity of welds. There is a lack of understanding and cohesion in these standards for the selection of welding parameters; effectiveness, accuracy, and selection of the test methods and; correlation of the mechanical properties to the micro and macro joint structure. The common standards (WIS 4-32-08, DVS 2207-1, ASTM F2620, and ISO 21307) for butt fusion welding were used to derive the six welding procedures. A total of 48 welds were produced using 180 mm outer diameter SDR 11 HDPE pipe manufactured from BorSafe™ HE3490-LS black bimodal PE100 resin. Three short term coupon mechanical tests were conducted. The waisted tensile test was able to differentiate the quality of welds using the energy to break parameter. The tensile impact test due to specimen geometry caused the failure to occur in the parent material. The guided side bend specimen geometry proved to be too ductile to be able to cause failures. A statistical t-test was used to analyse the results of the short term mechanical tests. The circumferential positon of the test specimen had no impact on their performance. Finite element analysis (FEA) study was conducted for the long term whole pipe tensile creep rupture (WPTCR) test to find the minimum length of pipe required for testing based on pipe geometry parameters of outer diameter and SDR. Macrographs of the weld beads supplemented with heat treatment were used to derive several weld bead parameters. The FEA modelling of the weld bead parameters identified the length to be a key parameter and provided insight into the relationship between the geometry of the weld beads and the stresses in the weld region. The realistic bead geometry digitised using the macrographs contributed a 30% increase in pipe wall stress due to the stress concentration effect of the notches formed between the weld beads and the pipe wall. The circumferential position of the weld bead had no impact on the pipe wall stresses in a similar manner to the results of the different mechanical tests. IV Nanoindentation (NI) and differential scanning calorimetry (DSC) techniques were used to study the weld microstructure and variation of mechanical properties across the weld at the resolutions of 100 and 50 microns, respectively. NI revealed signature 'twin-peaks and a valley' distribution of hardness and elastic modulus across the weld. The degrees of crystallinity obtained from DSC followed the NI pattern as crystallinity positively correlates with the material properties. Both techniques confirm annealing of the heat affected zone (HAZ) material towards the MZ from the parent material. The transmission light microscopy (TLM) was used to provide dimensions of the melt zone (MZ) which displays an hour glass figure widening to the size of the weld bead root length towards the pipe surfaces. Thermal FEA modelling was validated using both NI and TLM data to predict the HAZ size. The HAZ-parent boundary temperature was calculated to be 105 ⁰C. The 1st contribution of the study is to prove the existence of a positive correlation between the heat input calculated from FEA and the energy to break values obtained from the waisted tensile test. The 2nd contribution providing the minimum length of pipe for WPTCR based on the pipe dimensions. The 3rd contribution is the recommendation for the waisted tensile test with the test using the geometry designed to minimise deformation of the loading pin holes. The 4th contribution related the weld bead parameters to pipe wall stresses and the effect of notches as stress concentrators. The 5th contribution is a new method of visualising a welding procedure that can be used to not only compare the welding procedures but also predict the size of the MZ and the HAZ. The 6th contribution of the study is the proposal of new weld bead geometry that consist of the MZ bounded by the HAZ, for butt fusion welded joints of HDPE pipes.

Advancements in the Understanding of Nonlinear Optics and Their Use in Material Analysis

Adhesion, heterogeneous catalysis, electrochemistry, and many other important processes and properties are driven by interactions at surfaces and interfaces. Vibrational sum frequency generation spectroscopy (VSFG) is an increasingly popular analytical technique because it can provide information about the nature and physical orientation of functional groups at these surfaces and interfaces. Analysis of VSFG data can be complicated by the presence of SFG signal that is not associated with a resonant vibration. This nonresonant sum frequency generation (NR-SFG) signal can interfere with the resonant signal and influence the detected spectrum. Methods have been developed to remove NR-SFG signal; however, these methods tend to be complicated and expensive. In fact many SFG practitioners do not have the ability to remove NR-SFG signal components, and systems designed to remove NR-SFG signal contributions may not be able to do so for some materials. We have worked to help develop a better understanding of NR-SFG. As part of this work, a better understanding of the temporal and phase behavior of NR-SFG signal has been developed, based on the behavior of NR-SFG signal from Si(111) wafers. This work calls into question some assumptions underlying nonresonant suppression methods based on time-domain detection. A new method for nondestructively testing (NDT) materials has been developed that uses nonresonant second harmonic generation, the degenerate form of SFG. This new NDT technology has the potential to detect several forms of material damage, such as aluminum sensitization, and plastic deformation of materials, which are largely invisible to current NDT technologies. Methods for extracting functional group orientation from VSFG data that contains NR-SFG contributions are also demonstrated and used to investigate how the surface of high density polyethylene changes in response to mechanical deformation. This work shows that the inability to remove NR-SFG contributions from VSFG spectra does not mean that these instruments cannot be used to make important discoveries. It simply means that NR-SFG contributions must be properly understood and accounted for during experimental design, and kept in mind during the analysis of VSFG spectra.

The design metrics for the five-transistor cell are discussed in detail and performance and stability are evaluated. Finally a comparison is done between a 128Kb memory of an existing six-transistor technology and the proposed technology. The comparisons include area, performance and stability of the memories. It is shown that the area of the memory array can be reduced by 23% while maintaining comparable performance. The new cell also has 43% lower total leakage current. As a trade-off for these advantages some of the stability margin is lost but the cell is still stable in all process corners. The performance and stability has been validated through post-layout simulations using Cadence Spectre.</p>

An Investigation into the Household and Resident Composition of Higher Density Residential Districts in the Greater Toronto Area

The intensification of existing urban areas has become a common strategy used by planners to combat the negative aspects associated with unrestrained urban growth. This paper investigates the cultural and socio-economic characteristics of higher density households and residents of both owned and rented tenures in the Greater Toronto Area’s three constituent urban zones, the central city, the inner suburbs and the outer suburbs, between 2001 and 2006. Canada census data at the dissemination area level is used to produce descriptive statistics for the 100 variables included in the analysis. Although research relating to higher density housing is abundant, the consideration and affirmation of higher density housing sub-markets in the Greater Toronto Area based on location and tenure makes this study unique. It becomes clear that the diversity of the higher density housing market must be regarded during the planning process. The findings will be useful to planners for the purposes of infrastructure planning, community planning and aid in the implementation of urban intensification strategies in the Greater Toronto Area.
