La cartographie comparative des interactions E2-hôte révèle de nouveaux rôles de E2 dans la pathogénie associée aux papillomavirus humain

Les HPV sont les agents d'infections latentes, d'hyperplasies bénignes ou encore de cancer. Afin de mieux comprendre leur pathogenèse, nous avons cartographié les réseaux d'interactions de protéines de régulation virale E2 pour 12 génotypes HPV. Par double hybride, nous avons procédé à l'identification des interacteurs des protéines E2 suivi par une validation en cellule de mammifère par une méthode basée sur la reconstitution d'une luciferase. Le regroupement des profiles d'interaction montre une corrélation avec la phylogénie, établissant ainsi la contribution de E2 dans la pathogénie associée aux HPV. L'étude des réseaux d'interaction a révélé le ciblage préférentiel de protéines cellulaires hautement connectées, impliquées dans 5 catégories fonctionnelles récapitulant les principales fonctions de E2 mais aussi ouvrant de nouvelles perspectives quant au rôle de cette protéine virale dans les mécanismes d'infection. Dans un deuxième temps, ce travail s'est focalisé sur l'étude d'une interaction impliquant spécifiquement la protéine E2 d'HPV16, le virus le plus représenté dans les cancers cervicaux, et une protéine cellulaire, CCHCR1. En identifiant la surface d'interaction de CCHCR1 sur E2, il s'est avéré qu'elle induisait une compétition avec BRD4, un interacteur majeur de E2, se traduisant par une diminution des capacités transcriptionelle de E2. De même, nous avons montré que CCHCR1 induisait la délocalisation de E2 du noyau vers le cytoplasme. Enfin, nos résultats indiquent qu'en présence de CCHCR1, HPV16 E2 est moins apte à induire la différentiation précoce des kératinocytes, ce qui pourrait potentiellement avoir un effet important sur le cycle viral.

Differential Effects of Chronic Fluoxetine on the Behaviour of Dominant and Subordinate Naked Mole-rats

Naked mole-rats are eusocial rodents that live in subterranean colonies with a strict reproductive and social hierarchy. Breeders are socially dominant and other colony members are non-reproductive subordinates. The effects of manipulating the serotonergic system on aggression are well studied in many species, but not in eusocial rodents like the naked mole-rat. For the current study, the effects of fluoxetine hydrochloride (FLX) on status-specific behaviours of subordinates (Experiment 1) and queens (Experiment 2) were evaluated both in-colony and in a social-pairing paradigm to investigate how the serotonergic system influences aggression in this species. In accordance with our main hypothesis, chronic treatment of FLX attenuated the frequency and duration of aggression in queens, but not subordinates, when paired with an unfamiliar conspecific. Further exploration of pharmacological manipulation on status-specific behaviours of this eusocial species may elucidate the neurobiological mechanisms underlying their unique and rigid social hierarchy.

Some aspects of the presentation of industrial relations and race relations in some major British news media

Firstly, the social functions of mass media are analysed, with critical assessment of some significant sociological and psychological literature on mass communication and related topics. It is concluded they operate principally to maintain definitional social categories in the context of change and to sustain a given level of public discussion. Secondly, leading theories of race relations and industrial relations are analysed critically; it is concluded that different theoretical approaches, typically applying to different levels of analysis, tend to be mutually contributive, not exclusive. Both sets of relationships are then briefly compared and contrasted. The nature of racial and industrial conflict in contemporary Britain is then surveyed, with reference to dominant perceptions of the situation and the objective character of these conflicts. Next, content analysis as a research technique is discussed, and the precise methods employed in this study axe described. Selected television and press coverage of the 1970 docks strike is then analysed, with a particular focus on levels of explanation, the degree of public participation in discussion, and the definitions provided of the situation. Certain related data are appended. The same task is subsequently performed in relation to immigration, domestic race relations, the sale of arms to South Africa, and some other topics. Finally, these data are placed in their sociological context in Britain, by integrating them with an analysis of the social construction of consensus in that society. After discussion of certain theories interrelating culture and social divisions, together with analysis of certain institutions bearing on the construction of consensus, the particular role of the British news media, and the special functions of British ideologies of nationalism and objectivity, are surveyed. Conclusions for the relation between consensus and conflict in modern Britain are then stated.

Urban transportation problems in the Muscat area, Sultanate of Oman

The economy of Oman, was traditional in character before the development of the oil sector which set in motion the present structural transformation. Economic prosperity gave the opportunity to the public to possess their own private means of transport, thus resulting in a rapid increase in the number of vehicles in the country, particularly in the Muscat Area the capital city of the country. Free essential services such as health and education, and no taxes and duties have led to high disposable household income. Therefore, Muscat is witnessing rapid and successive land-use changes, expansion of the urban area and multi-car owning households. In the last two decades the population of Muscat increased considerably. This population growth was accompanied by a substantial expansion of Muscat's boundaries to provide homes, work places and other facilities. The topographical nature of the area limited the flat land available for housing, shops, schools, and other elements of the infrastructure. The resulting competition for space had as one of its consequences that insufficient land was allocated for car parking in the major activity centres, and the result was an acute shortage of parking spaces in these areas. The expansion of the Muscat Area was accompanied by changes in the employment and residential pattern. This resulted in a significant increase in number of vehicles, trips and commuting, and so the need for effective transport services and facilities became greater than ever before. The topographical features made it more difficult to provide sufficient transportation facilities. Development planning neglected the importance of arranging urban activities in such a way that the need for vehicle movements would be greatly reduced. It also resulted in low density population areas with street patterns mostly not designed for public transport services. This study sets out to discuss the problems of urban transportation in the Muscat Area and seeks to answer the following questions: What are the trip characteristics? How far do the natural topographical features inhibit the development of the Muscat road network? How does the existing network serve the needs of the area? How far can it cope with the traffic movement? Will the proposed major roads solve the present problems of traffic congestion and alleviate future ones? What are the main causes for traffic accidents? To what extent are car parking facilities adequate at the major activity centres? What is the role of Oman National Transport Company buses within the public transport system? The Muscat Area faces problems of traffic congestion and accidents, high demand on parking facilities and inadequate public transport. The situation is liable to deteriorate sharply in the next few years, unless effective action is taken. There is a need for a study that can provide immediate practical solutions and propose guidelines for future policy to ensure that the transport system is expanded and improved to cope with the needs arising from future growth. This study identifies factors contributing to the existing traffic problems with the intention of providing useful information which can help traffic planners and decision makers in understanding the nature of the problems, and finding solutions and guidelines for future policy. As far as methodology is concerned, a literature review is supported by fieldwork involving questionnaires and contacts with relevant authorities. Four types of surveys were conducted in order to collect information that can identify some of the factors that are contributory to the present problems. On the basis of the analysis of the data collected, urban transport problems are identified and discussed together with some possible solutions and recommendations.

Transformation Of Public Space: The Case Of Migros Akkopru Shopping Center

Today, it is observed that shopping centers that are increasingly growing in number, especially in metropolitan cities, are used as public spaces by many urban inhabitants. Shopping centers have become places where social life is experienced and leisure time is spent through a wide range of activities offered besides shopping. On the other side, city centers that can be regarded as the most essential public spaces of urban areas are in a process of deterioration both in terms of physical quality and functional features. These two developments are essential indicators of a change and transformation concerning the public space&rsquo / s role and its features like openness to everyone&rsquo / s use, high accessibility and social integration. Within this context, in this study, finding the direction of this transformation and the extent that shopping centers possess the characteristics of public space are aimed. In this respect, after a literature review about public space, city center and shopping center within the context of the aim, the development of city centers and public spaces of Ankara is examined. After the development of shopping centers that flourished within the last ten-fifteen years in Ankara and their socio-spatial effects upon the city are discussed, hypothesis is tested through the findings of the questionnaire survey made in Migros Akk&ouml / pr&uuml / Shopping Center. Lastly, findings and conclusions are summarized and interpretations for future and policy proposals are made concerning the changing functions and uses of public spaces and shopping centers.

Housing Management Models And Household Behaviour

A significantly large stock of housing has been realised In Turkey during the past five decades, building the cities almost entirely anew. This has shifted the central concern from production, design and ownership issues in the housing sector to that of housing management. The major problem in housing for the coming decades is not how to maintain the growth of the stock further, but how to efficiently use and improve the existing assets. Currently, no central or local authority is responsible for the management, running or control at any scale, but only the residents and property owners responsibilities exist at the individual plot scale. There is evidence of greater efficiency however, for the need of housing management at supra-plot scales. The hypothesis of the study in this context is that no part of the stock is without problems in terms of management. This is empirically investigated by two complementary analyses based on two distinct surveys. The first analysis demonstrated that the role of tenure and income on expenditures on housing, especially expenditures for repairs and maintenance are dominant. A three-fold difference is observed between tenants and owner-occupiers, and 10 times between households of highest and lowest incomes. Expenditures for repairs and maintenance is 1.9 times greater in the apartment stock and 1.2 times more in the &amp / #8216 / gecekondu&amp / #8217 / dwellings than in individual &amp / #8216 / houses&amp / #8217 / . Lowest levels of expenditures are observed in oldest part of the stock, in less developed neighbourhoods, and in stock with lowest rental values. According to the results of the second analysis, organisational tendencies of household groups varying in their characteristics are not sharply differentiated as in their expenditures. One most significant factor is tenure. Tenants are observed to have a weaker sense of dedication and identity in the dwellings they occupy and in neighbourhoods they live. Current management problem issues could then be identified as: low-income households, tenant households, stock with low rental values, aged stock and undeveloped neighbourhoods. Some of the most significant policy tools for tackling these problems are credit opportunities to be made available to households for repairs and maintenance in such problem areas, subsidies in terms of tax deductions, material incentives, technical support and public investments in degraded localities to boost economic activities which are eventually to initiate private investments.

In this thesis, changing regional development policies and effects of these in Turkey is discussed. Regional inequalities have been one of the important concerns of the Republic of Turkey. However, the restructuring of state and capitalism during 1980&amp / #8217 / s led to a change in regional policies and Turkish regional polices have surely begun to be affected from new policies. Yet, these new policies do not seem to offer much about promoting development in underdeveloped regions. This study aims to analyze the effects of new regional policies with their successes and failures in order to find out whether they can offer a suitable policy framework for promoting development in underdeveloped regions of Turkey. With this aim, changing policies in Southern Eastern Anatolia Project (GAP) will be discussed. Transformation of GAP, from a project of irrigation and energy based on previous allocative regional policies to a sustainable human development project will be examined. According to findings of the study, some remarks will be made about policy framework that should be taken as a basis in GAP.

This study aims to analyze the emergence and evaluation of a new type of suburbanization in T&uuml / rkiye, which are concomitant with the rise of new middle class having a high purchasing power. It examines different urbanization and suburbanization processes in various societies and demonstrates that the suburbanization of T&uuml / rkiye does not exactly fit in these models. Such a suburban expansion is taking place under the prevailing impact of political economy in the world and leads to a social segregation within metropolitan areas, which is argued to become permanent. Upper middle class have developed privatized, enclosed, and monitored exclusive spaces of residence, work, leisure, and consumption. The main focus of the study, therefore, is the gated communities, which is the last extension of suburbanization. In this thesis, the gated communities are mainly residential in character and offer a new concept of life-style, which is based upon the idea of total security and retreat from the illnesses of the urban core in terms of noise, dust, disorder, crowds and related issues. It is argued that, in the specific case of Angora Evleri-Kooperatif-18, gated communities can be viewed as a further theme of fragmentation of the city of Ankara. The new urban fragmentation indicates a dual process of increasing social and spatial polarization on the urban land. These dualities have been identified in society. One part of the society has experienced affluence, and success while the other has suffered degradation. The economic growth has been at the expense of sharp increases both at the top and bottom ends of the income distribution. Social inequality, in return, has manifested itself spatially.

Askew and unsystematic urbanization that occurred after rapid increase and migration in 1950&rsquo / s in T&uuml / rkiye, caused destruction of green spaces. In this way, green spaces in urban pattern that are insufficient now are going to decrease day by day. As a result, human beings that are living in cities have to survive their life in a condition of less green and much building. Urban populace has physical and psychological problems caused by noise, pollution, stress together with not meeting their strolling, resting and being comfortable. For this reason, importance of green spaces is increasing day by day. Being under a disaster of earthquake doubles this importance in istanbul. In this research, in which green spaces in Kartal and Sariyer are taken into consideration, one-by-one fixing and photographing technique is used. Results are evaluated in terms of ownership, opening year, classification, size, activity and distribution of green spaces. Research is consisting of eight chapters. Aim and extension of research is taken into consideration in first chapter. In second chapter, urban open and green spaces are examined in terms of definition, classifications, hierarchy and design criteria. Third chapter include urbanization and green pace relation. In this chapter, historical development of urban open spaces, importance of urban open and green spaces and need of green space issues are mentioned. Green space concept in development plans are taken into consideration in fourth chapter. Firstly, green space concept in development plans / then, procuring methods of green spaces and at last, problems of application decisions of green spaces are examined. Fifth chapter includes green space norms in T&uuml / rkiye and in other countries and comparison of these norms. In sixth chapter, green space situation of istanbul, Kartal and Sariyer are given. In this chapter, firstly, urbanization and green space problem / then, existing situation of Kartal and Sariyer are researched. In case study, which is observed in seventh chapter, green space situation of Kartal and Sariyer is investigated. At last chapter, results of these researches are attained by comparison of green space situations of these two districts in a heading of conclusion.

Urban Spaces Re-defined In Daily Practices: The Case Of

This study, preconceives space as a social phenomenon, and emphasizes the fact that the urban space cannot be separated from its inhabitants. Accordingly, it suggests that the investigation of both the city and its inhabitants is crucial with respect to everyday life and practice. Hence, the study questions how inhabitants create their spaces following their needs and demands, and how the urban space is re-defined and re-produced through appropriation. Moreover, the study aims to understand how the inhabitants express themselves and how they resist through the spaces they produce in their daily practices. With this aim, the thesis investigates a spatial activity performed by young people in Ankara, the case of &lsquo / Minibar&rsquo / , for understanding the process explained as &lsquo / re-definition&rsquo / of urban space. The research reveals that these spaces become possible through their spatial characteristics. These spaces transgress the established space, yet they are sustained due to their ephemerality, impermanency and flexibility. Furthermore these spaces are discovered to be a medium of expression for the inhabitants. In conclusion, this study asserts an approach towards the city and explains that through looking to the &lsquo / lived spaces&rsquo / rather than rhetorics, calculations and presumptions, we can obtain a clear and actual picture about the city and the inhabitants.
