Fabrication and use of new solid state phosphate ion selective electrodes for monitoring phosphorylation and dephosphorylation reactions

Highly selective and sensitive phosphate sensors have been fabricated by constructing a solid membrane disk consisting of variable mixtures of aluminium powder (Al), aluminium phosphate (AlPO4) and powdered copper (Cu). Both binary and ternary electrode systems are produced depending on their composition. The ternary membranes exhibit greater selectivity over a wide range of concentrations. The ternary electrode with the composition 25% AlPO4, 25% Cu and 50% Al was selected as our preferred electrode. The newly fabricated ternary membrane phosphate selective electrodes exhibited linear potential response in the concentration range of 1.0 × 10−6 to 1.0 × 10−1 mol L−1. The electrodes also exhibit a fast response time of <60 s. Their detection limit is 1.0 × 10−6 mol L−1. The unique feature of the described electrodes is their ability to maintain a steady and reproducible response in the absence of an ionic strength control. The electrodes have a long lifetime and can be stored in air when not in use. The selectivity of the new phosphate selective electrodes with respect to other common ions is excellent. The results obtained provide further insight into the working principles of the newly fabricated phosphate selective electrodes. Dephosphorylation and phosphorylation reactions were monitored using the preferred phosphate selective electrode. The following reactions were studied and inferences drawn; (a) the reactions between *[{CoN4(OH)(OH2)}]2+ and *[OH(PO2O)]2- for 1:1, 2:1 and 3:1 *[{CoN4(OH)(OH2)}]2+ to *[OH(PO2O)]2- ratios.(b) the reactions between *[{CoN4(OH)(OH2)}]2+ and *[O2NC6H4PO2(O)(OH)]- for 1:1, 2:1 and 3:1 *[{CoN4(OH)(OH2)}]2+ to *[O2NC6H4PO2(O)(OH)]- ratios. (c) the reactions between *[{CoN4(OH)(OH2)}]2+ and *[(OH)2(PO2)2O]2- for 1:1, 2:1 and 3:1 [{CoN4(OH)(OH2)}]2+ to *[(OH)2(PO2)2O]2- ratios, and (d) the reactions between *[{CoN4(OH)(OH2)}]2+ and *[(OH)2(PO2)3O2]3- for the 1:1, 2:1 and 3:1 [{CoN4(OH)(OH2)}]2+ to *[(OH)2(PO2)3O2]3- ratios. Design, Synthesis, and Evaluation of Bicyclic Peptides as Ammonium Ionophores

A series of bicyclic peptides have been designed and synthesized to provide ammonium ion complexation sites via hydrogen bonding in a tetrahedral geometry. Molecular modeling dynamics and electrostatics studies indicate that target compounds 1d-6d may provide better selectivity for ammonium ions over potassium ions than the ammonium ionophore currently used for blood analysis applications, nonactin. Attempts to synthesize 1d, cyclo(L-Glu1�D-Val2�L-Ala3�D-Lys4�D-Val5�L-Val6)-cyclo-(1ã-4å), were unsuccessful due to poor solubility of the synthetic intermediates. This led to the design of 2d-6d in which specific amino acid residues were chosen to provide higher solubility. Compound 2d, cyclo(L-Glu1�D-Ala2�D-Ala3�L-Lys4�D-Ala5�L-Ala6)-cyclo-(1ã-4å), was successfully synthesized, but was also too insoluble for characterization or testing in an ion selective electrode (ISE) sensor format. Compound 6d, cyclo(L-Glu1�D-Leu2�Aib3�L-Lys4�D-Leu5�D-Ala6)-cyclo-(1ã-4å), was successfully synthesized and characterized. When 6d was incorporated into an ISE sensor and tested as an ammonium ionophore, results indicated that the bicyclic peptide lacked solubility in the ISE membrane. A 13C-NMR study has been initiated in order to evaluate selectivity of 6d for ammonium over potassium and sodium cations in solution. Preliminary results with the potassium ionophore valinomycin as a control have been completed.

ION-SELECTIVE ELECTRODES FOR ORGANIC CATIONS AND PHARMACEUTICALS (ISE, POLYMER MEMBRANE CALCIUM CHANNEL BLOCKERS).

Three areas related to Polymer Membrane Ion-Selective Electrodes were investigated. The first two concered (1) their application to analysis of organic cations and pharmaceuticals, and (2) the use of electrochemically polymerized films for potentiometric sensors. The third area focused on the development of a computer controlled data acquisition system for studying Ion Transport at the Interface of Two Immiscible Electrolyte Solutions (ITTIES). Coated wire Polymer Membrane Ion-Selective Electrodes (PMISEs) based on dinonylnapthalenesulfonic acid (DNNS) were prepared for several alkylammonium ions and pharmaceuticals. Selectivity coefficients and other operational characteristics were predictable from calculated distribution constants. Specifically, Log k ᵖᵒᵗ(i)(j) values increased linearly with carbon number in a homologous series of alkylammonium ions. Among cations of the same carbon number, interference increased tenfold with each additional substitution in going from quaternary, tertiary, secondary and primary alkylammonium ions. Addition of hydrophilic subtituent groups to cyclohexylammonium interferents further diminished electrode response. Electrodes for several important pharmaceuticals, namely cocaine, methamphetamine, methadone, protriptyline, cimetidine, lidoflazine, verapamil, acebutalol, diltiazem, and nicardipine, displayed nearly theoretical response down to micromolar concentrations. Their selectivity was consistent with calculated distribution constants. Selectivity was independent of sensitivity, suggesting that the magnitude of each was determined by the distribution of both the free cation and the association complex of this cation with DNNS. The polymerization of phenol derivatives onto copper rendered the resulting films responsive to counter-ions of the electroactive species present in the polymerization medium. Those incorporating Aliquat 336 (tricaprylmethylammonium chloride) showed response to various anions, while others prepared using a sulfonated phenol derivative (m-hydroxybenzenesulfonic acid) responded to tributylammonium. The films had short lifetimes due to poor adhesion to the copper substrate. Construction of a computer controlled data acquisition system provided a rapid method for obtaining electrocapillary data simultaneously with the current-scan polarogram. The system was evaluated by observation of ion-transfer of a variety of alkylammonium ions.

Fabricação de sensores eletroquímicos para a determinação de espécies químicas de relevância ambiental / Fabrication of electrochemical sensors for determination of chemical species of environmental relevance

Neste trabalho são apresentados resultados relacionados à determinação de nitrato, nitrito e amônio por técnicas eletroquímicas. Para a determinação de nitrato foi utilizada uma lâmina de cobre cuja superfície é renovada continuamente empregando um processo de ativação (oxidação do cobre e subseqüente redução de íons Cu(II)) com potencial controlado em sistema FIA. As condições ótimas foram alcançadas por meio de 60 s para a etapa de ativação, vazão do transportador 3,0 mL min-1 e volume de amostra 150 &#181;L. A repetibilidade foi estimada em 4,7% (n=9) com freqüência de amostragem de 60 amostras h-1. Trabalhando nas condições otimizadas um aumento na concentração de nitrato gerou um aumento proporcional na corrente, resultando em curva analítica em um intervalo de 0,1 2,5 mmol L-1 (-Ip (&#181;A) = 0,13 + 4,6 [NO3-] (mmol L-1), R2 = 0,9991). Os limites de detecção e quantificação foram estimados em 4,2 (S/N = 3) e 14 (S/N = 10) &#181;mol L-1, respectivamente. O mesmo sensor foi utilizado para determinações de nitrito por voltametria de pulso diferencial e a otimização dos parâmetros relativos ao processo de ativação (tempos de dissolução e redeposição) foi efetuada com o intuito de aumentar a sensibilidade. As condições ótimas foram de 30 s para a etapa de ativação, com uma repetibilidade de 1,9% (n=10), e o aumento nas concentrações de nitrito gerou um aumento proporcional na corrente, resultando em curva analítica em um intervalo de 50 &#181;mol L-1 - 1,44 mmol L-1 (-Ip (&#181;A) = -0,13 + 53,52 [NO2-] (mmol L-1 ), R2 = 0,9996). Os limites de detecção e quantificação foram estimados em 2,8 (S/N = 3) e 9,4 (S/N = 10) &#181;mol L-1, respectivamente. O estudo da morfologia da superfície do eletrodo de cobre durante o processo de ativação foi realizado por AFM com o objetivo de compreender o impacto das mudanças microestruturais no sinal de corrente para nitrato e nitrito. Os resultados indicam que após a ativação do eletrodo a superfície apresenta cristais com uma textura rugosa e ocorre aumento da área superficial, justificando o aumento de sinal de corrente. Para a determinação de amônio foram fabricados eletrodos íon seletivo (ISE) com membrana polimérica de forma tubular, os quais foram acoplados em um sistema de injeção em fluxo. A influência de parâmetros como vazão do transportador e alça de amostragem foi investigada e melhores resultados foram obtidos em 0,5 mL min-1 e 250 &#181;L, respectivamente. Nas condições otimizadas a repetibilidade das determinações foi de 1,3 % (n=10) com freqüência de amostragem de 13 amostras h-1. Synthesis of an Ammonium Ion-selective Fluoroionophore

The drawbacks of nonactin, the current commercial standard receptor for ammonium ion necessitate the development of new ammonium ionophores. We have designed and attempted to synthesize fluoroionophores, I- III. Molecular modeling of I suggests superior selectivity over that of nonactin. III was synthesized as a selective ionophore for optical detection of ammonium ion. The synthetic strategies for III are two-fold: solid phase and solution phase. Solution phase synthesis was performed with two different protecting groups (t-butyl ester and benzyl ester). A methyl-amino substituted anthracene molecule will be covalently coupled to the secondary amine group to provide an optical signaling moiety that operates on the basis of an“off-on" fluorescence emission mechanism. Compound IV was also synthesized in order to provide a sample reaction for the covalent coupling of the chromophore and to provide a fast route to an ammonium fluoroionophore.

Construção e avaliação de eletrodos de membrana sólida cristalina seletivos a cobre e prata / Construction and evolution copperand silver ion selective electrode based on crystalline solid membranes

O presente trabalho visou a construção e avaliação de eletrodos de membrana sólida cristalina seletivos a cobre e prata. Obteve-se o sulfeto de prata, material eletroativo usado na preparação de membranas seletivas a íons Ag+, por precipitação com gás sulfídrico em meio ácido. Eletrodos com membranas de 1,0 e 0,50 mm de espessura apresentaram respectivamente limites nernstianos de 10-4 e 10-5 Mol.L-1 e precisão de &#177; 0,1 e 1 mV nas leituras de potencial. Os sulfetos de cobre e prata, usados na preparação de membranas seletivas para determinação de cobre, foram obtidos por precipitação simultânea com gás sulfídrico, tioacetamida em meio ácido e tiouréia em meio amoniacal. Difratogramas de Raios X revelaram que o material obtido com tiouréia corresponde ao sulfeto ternário, Cu2S.3Ag2S ou Ag1,55Cu0,45S, responsável pela resposta seletiva do sensor em soluções de Cu2+. O eletrodo construído a partir deste sulfeto, onde o cobre aparece no estado monovalente, apresentou limite nernstiano da ordem de10-7Mol.L-1, fornecendo resultados satisfatórios na determinação deste metal em amostras de aguardente e álcool. Cálculos termodinâmicos mostraram que quando membrana é constituída de CuS, um contacto elétrico efetuado através de cobre ou prata é instável, uma vez que ocorre a formação de Cu2S. Os resultados experimentais conduziram a um novo modelo para explicar o mecanismo de resposta do eletrodo, o qual se baseia na reação: Cu2S (memb) + Cu2+ (sol) &#8596; CuS (memb) + 2 Cu + (sol). A atividade de Cu+ na interface eletrodo / solução é controlada pela atividade de Cu2+ nas soluções de medida, de modo que o sensor funciona como um eletrodo de Cu2S/CuS em contacto com o sistema Cu2+/Cu+. / Present work deals with the assembling and testing of solid membrane electrodes for copper and silver. The silver sulphide used to prepare the membrane selective to Ag+ was precipitated with hydrogen sulphide in acidic medium. Electrodes 1,0 and 0,50 mm thick present Nernstian limits of 10-4 and 10-5 Mol.L-1 order with a &plusmn; 0,1 and 1 mV respectively. A mixture of silver sulphide and copper sulphides were used for the copper (II) selective electrode. This solid was prepared from Cu2+ and Ag+ in the same solution with hydrogen sulphide or thioacetamide both in acid medium and with thiourea in ammoniacal medium. X-Ray diffractograms shows that the solid prepared from thiourea solutions presents significant contribution of jalpaite Ag1,55Cu0, 45S (in fact Cu2S.3Ag2S). An important finding was the presence of copper (I) in the matrix and this has been also found to the responsible for the higher performance of this electrode with a limiting response to 10-7 Mol.L-1 copper (II). In fact thermodinamic calculation has show instability of a contact of Ag or Cu with CuS solid, as Cu2S must be formed in any case. On this basis, the theory of the electrode is based in the following reaction which takes place on the electrode surface: Cu2S (memb) +CU2+ (sol) &#8596; CuS (memb) + 2 Cu+(aq) K = 10-13 M. On this basis the Cu+ activity at the electrode / solution interface is controled by the activity of Cu2+ of the solution and the Cu2+/ Cu+ system in contact with the solid matrix is defined.

Study of Disposable Silicon-based Chlorine and Ammonium Micro Ion Sensors

Human blood reflects the health of internal organs and tissues, the constituent can be affected on account of abnormal health status. Moreover, the ion concentration of chloride and ammonium in human blood relates to the functionality of our kidney and liver, which is one of the most important health indicators. For real-time monitoring and detecting applications, this study uses micro-electromechanical systems (MEMS) surface micromachining process technology to develop an extended-gate field-effect transistors (EGFET), and by combining two different ion-selective membranes (ISM), a disposable silicon-based chloride and ammonium micro ion sensor was fabricated. The main process steps include four photolithography process and two thin film deposition. In order to probe into the influences on modulating three designing parameters: (i) channel width to length ratio; (ii) channel shape; (iii) mixing ratio of the sensing film additives, the size of the sensing area is fixed to 1¡Ñ1 mm2 while the device is fixed to 6 mm3. In this thesis, the optimization of the development chloride and ammonium micro ion sensors, according to the measuring results from the commercial semiconductor analyzer (Agilent B1500A), when measuring range of 10-1~10-5 mol/L, the sensitivity and linearity for the chlorine ion sensor is 47.5 mV/pCl and 99.13%, as for the ammonium ion sensor are 41.2 mV/pNH4 and 99.28%, the interfering ion selectivity coefficients (log KCl,OH, log KNH ,Na) are -4.71 and 0.53 respectively.

The lifetime of the polymer membrane ion-selective electrode is, in general, short when compared to that of the glass pH electrode. It is the manner in which the elctroactive materials are entrapped in the ion-selective membranes which accounts for this lifetime difference. In the polymer membrane electrode a high molecular weight electroactive material is dissolved in the plasticized polymer membrane. Because it is hydrophobic, this material is much more compatible with the low dielectric membrane phase than with the aqueous analyte phase. It, of course, has some solubility in the aqueous phase and, in time, leaches from the membrane. In the glass pH electrode, the electroactive material is the SiOH group which is covalently attached to the insoluble glass membrane. Because it is attached, it cannot be leached from the membrane. The purpose of this study was to investigate the possibility of covalently attaching the electroactive material to the polymer membrane in polymer membrane electrodes. In this weay, it was hoped that electrodes with much longer lifetimes could be obtained. Cation selective electrodes based on sulfonated polystyrene were investigated first. Sulfonation was accomplished by dipping polystyrene membranes into chlorosulfonic acid. Electrodes of both the conventional (i.e., with internal reference) and coated-wire types were prepared. Neither gave satisfactory response. Cation selective electrodes based on the perfluorinated, sulfonic acid containing ion exchange polymer NAFION 120® were investigated next. This material produced well-behaved Cs⁺ electrodes but electrodes responsive to larger cations (e.g., tetrapropylammonium and dodecyltri-methylammonium) could not be obtained. This may be due to ion-pairing in the membrane phase between these larger cations and the polymer bound sulfonate groups. To help answer some of the questions encountered during these investigations of electrodes based on covalently attached sulfonate groups, polymer membrane electrodes based on dinonylnaphthalenesulfonic acid were prepared and investigated. These electrodes were found to have very great selectivity for high molecular weight organic cations relative to inorganic and smaller organic cations. This type of selectivity is analogous to that obtained in ion pair solvent extraction of cations with a high molecular weight anionic species. Since a number of species of clinical, biological and toxicological interest are, at physiological pH, high molecular weight organic cations, electrodes based on dinonylnaphthalenesulfonic acid show great promise for determination of such species. An electrode for the determination of the drug phencyclidine was prepared to demonstrate the potentialities of drug analysis with this type of electrode. A microcomputer-based potentiometric analysis system was used to collect and analyze the data in this study. This system was one of the first stand-alone microcomputer systems employing a high level computer language to be described. The IMSAI 8080 Microcomputer and the computer language CONVERS were used.

Desenvolvimento de membranas íon-seletivas com poliestireno sulfonado e polianilina dopada para a aplicação em eletrodiálise

Atualmente, a preocupação com a redução da poluição industrial tem motivado os pesquisadores na busca de novas tecnologias para o tratamento de resíduos industriais. Tecnologias limpas, como a eletrodiálise, são capazes de tratar alguns destes resíduos, como por exemplo o efluente da indústria de galvanoplastia, minimizando os impactos que ocorreriam caso eles fossem descartados diretamente no meio ambiente. O componente principal desta técnica é a membrana na qual ocorre a etapa de retirada dos íons da solução. Atualmente estas membranas são importadas e caras, o que justifica o desenvolvimento de membranas eficientes e acessíveis. Neste sentido, no presente trabalho membranas de poliestireno sulfonado/ poliestireno de alto impacto (SPS/HIPS), polianilina dopada com ácido canforsulfônico/ poliestireno sulfonado/ poliestireno de alto impacto (PAniCSA/SPS/HIPS), polianilina sulfonada/ poliestireno sulfonado/ poliestireno de alto impacto (SPAN/SPS/HIPS), e polianilina dopada com ácido p-tolueno sulfônico/ poliestireno sulfonado/ poliestireno de alto impacto (PAniTSA/SPS/HIPS) foram desenvolvidas usando o método de mistura química. As membranas foram caracterizadas utilizando as técnicas Análise termogravimétrica (TGA), Análise dinâmico Mecânica (DMA), e Microscopia Eletrônica de Varredura (MEV). Membranas foram submetidas a curvas corrente-potencial e ensaios de eletrodiálise em soluções de NaCl e KCl, a fim de determinar o transporte iônico através das mesmas. Os resultados foram comparados com uma membrana comercial Selemion CMT. A extração percentual média para íons de Na+ obtidos pelas membranas desenvolvidas foi superior a 20%. / Nowadays the concern with the reduction of industrial pollution has motivated researchers to found out new technologies for treatment of industrial waste. The clean technologies, as electrodialysis, are capable of treating some these residues, as for example the galvanoplasty’s waste, minimizing the impacts that would happen to them if they were discarded directly on the environment. The main component of this technique is the membrane on which occurs the ions removal stage of the solution. The membranes are imported and expensive what justifies the development of efficient and accessible membranes. In this sense, in the present work membranes of sulfonated polystyrene / high impact polystyrene (SPS/HIPS), polyaniline doped with camphorsulfonic acid / sulfonated polystyrene / high impact polystyrene (PAniCSA/SPS/HIPS), sulfonated polyaniline/ sulfonated polystyrene / high impact polystyrene (SPAN/SPS/HIPS), and polyaniline doped with p-toluenesulfonic acid / sulfonated polystyrene / high impact polystyrene (PAniTSA/SPS/HIPS) were developed using chemical mixture method. Membranes were characterized by Infrared Spectroscopy (FTIR), Thermogravimetric Analysis (TGA), Dynamic Mechanical Analysis (DMA) and Scanning Electronic Microscopy (SEM). Membranes were submitted to current-voltage curves and electrodialysis experiments with NaCl and KCl solutions, in order to determine ionic transport through them. Results were compared with a commercial membrane, Selemion CMT. The average percent extraction for Na+ ions obtained by membranes developed were beyond 20%.
