Evaluación del efecto antibacteriano de los irrigantes endodonticos contra cepas del Enterococcus Faecalis (ATCC29212)

Objetivo: Comparar in vitro el efecto antibacteriano de tres irrrigantes endodónticos (Hipoclorito de sodio al 5%, Clorhexidina al 2% y EDTA al 17%) sobre cepas del Enterococcus faecalis (ATCC29212). Materiales y Métodos: Se trabajó con una muestra de 10 pocillos por grupo para comparar el efecto antibacteriano en 24, 48 y 72 horas. Se realizaron cuatro pruebas: la actividad antibacteriana se evaluó mediante el método de difusión en agar utilizando medio Brain Heart infusión Broth (BHI). Por otro lado, para la concentración mínima inhibitoria (CMI), y el efecto bactericida y bacteriostático (CMB) se empleó el método de dilución en medio BHI. Para evaluar la prueba de citotoxicidad se utilizó la línea celular MDCK mediante la prueba colorimétrica del MTT. Resultados: Se observó a las 72 horas que la Clorhexidina al 2% tuvo efecto antibacteriano sobre cepas del Enterococcus faecalis (ATCC29212) con una media de 30.45 + 8.54 mm, mientras que el NaOCl al 5% y el EDTA al 17% tuvieron una media de 19.3+ 1.82 mm y 30.40 + 8.77 mm respectivamente. La CMI de la clorhexidina fue de 1:25 μg/ml, el CMB de la Clorhexidina fue de 1:125 μg/ml y la citotoxicidad de la Clorhexidina fue de 65.6%. Conclusiones: Al comparar los tres irrigantes endodónticos se demostró que la CHX al 2% es el irrigante con mayor efecto antibacteriano y el que posee menos efecto citotóxico. / Objective: To compare, in vitro, the antibacterial effect of three endodontic irrrigants (5% Sodium Hypochlorite, 2% chlorhexidine and 17% EDTA) on strains of Enterococcus faecalis (ATCC29212). Materials and Methods: It was worked with a sample of 10 wells per group to compare the antibacterial effect in 24, 48 and 72 hours. Four tests were conducted: the antibacterial activity was evaluated with the agar diffusion method in Brain Heart Infusion Broth (BHI). On the other hand, for the minimum inhibitory concentration (MIC) and bactericidal and bacteriostatic effect (MBC), dilution in BHI method was used. To evaluate the cytotoxicity test MDCK cell line was used by the MTT colorimetric assay. Results: It was observed at 72 hours the 2% chlorhexidine had antibacterial effect on strains of Enterococcus faecalis (ATCC29212) with an average of 30.45 + 8.54 mm, while the 5% NaOCl and 17% EDTA had an average of 19.3 + 1.82 mm and 30.40 + 8.77 mm respectively. The MIC of chlorhexidine was 1:25 ug/ml, the MBC Chlorhexidine was 1: 125 ug/ml and the cytotoxicity of Chlorhexidine was 65.6%. Conclusion: When comparing the three endodontic irrigants it was demonstrated that the 2% CHX is the irrigant with more antibacterial effect and has less cytotoxic effect. / Tesis
