Jaburetox, peptídeo tóxico derivado da urease : estudos de estrutura e função

Ureases (E.C. 3.5.1.5) são metaloenzimas dependentes de níquel, que estão envolvidas na biodisponibilidade de nitrogênio e em mecanismos de defesa em plantas. Nosso grupo descreveu a atividade inseticida da Canatoxina, uma isoforma da urease. Esta toxicidade envolve a liberação de um peptídeo interno de 10 kDa (pepcanatox) da proteína, por ação hidrólitica de catepsinas encontradas no sistema digestivo de insetos suscetíveis. Baseado na sequência N-terminal do pepcanatox, um fragmento de 270 pb correspondente (jaburetox-2Ec) foi clonado, a partir da sequência da JBURE-II, e expresso em Escherichia coli. Este peptídeo recombinante Jaburetox-2Ec (carregando epítopo V5 e cauda His) foi testado contra os insetos Dysdercus peruvianus, Rhodnius prolixus e Spodoptera frugiperda e 100% de mortalidade foi observado em todos os modelos após a ingestão de microgramas do peptídeo. Outros dados mostram que jaburetox-2Ec tem capacidade de interagir com bicamadas lipídicas acídicas, permeabilizando lipossomas, além de atividade antifúngica. Estudos de modelagem do peptídeo revelaram a presença de um grampo beta, que poderia estar envolvido nesta toxicidade. A fim de estudar os motivos envolvidos nestas atividades biológicas, escolhemos uma abordagem de mutagênese dirigida. Para isto, a sequência de DNA do jaburetox foi clonado em plasmídeo pET-23a, obtendo-se a expressão do jaburetox, contendo apenas cauda de histidina, e a partir deste, obtivemos diferentes mutantes: 1) deleção de todo o grampo beta (aminoácidos 61-75); 2) deleção da metade N-terminal do peptídeo, que corresponde a uma região ausente nas ureases bacterianas; 3) deleção da metade C-terminal do peptídeo. Ensaios de formação de canais iônicos em bicamadas lipídicas artificiais com o jaburetox e mutantes mostraram que todos formam canais iônicos, ainda que com diferentes características. Planar lipid bilayers experiments were performed with jaburetox and all mutants, and we observed channels activity in all cases. Bioassays with Oncopeltus fasciatus and Rhodnius prolixus showed that the region of β-hairpin is not involved in the effects of the peptide in these organisms, and the N-terminal region of the Jbtx carries the most important entomotoxic domain which is fully active in the absence of the β-hairpin motif.

Urease (EC 3.5.1.5) é uma metaloenzima dependente de níquel que catalisa a hidrólise da ureia à amônia e dióxido de carbono. A Soja (Glycine max) produz duas isoformas de urease: a urease ubíqua (uSBU) e a urease embrião-específica (eSBU). Nosso grupo demonstrou que eSBU apresenta propriedades biológicas independentes da sua atividade ureolítica, como ativação de plaquetas, atividade inseticida e inibição do crescimento de fungos fitopatogênicos, estes últimos sugestivos de envolvimento das SBUs em mecanismos de defesa da planta. Outras ureases também estudadas pelo nosso grupo, são as de Feijão-de-porco (Canavalia ensiformis). O Feijão-de-porco produz três isoformas de urease: Canatoxina, JBU e JBURE-II, que também apresentam atividades biológicas, não relacionadas à sua atividade enzimática. Dentre elas, a atividade inseticida tem sido a mais estudada. A toxicidade das ureases a insetos envolve a liberação de um peptídeo interno tóxico, mediante à hidrólise por enzimas digestivas do inseto. Um peptídeo equivalente foi clonado em Escherichia coli e denominado Jaburetox. Este peptídeo apresenta um amplo espectro de ação contra insetos e toxicidade contra fungos. Nossos objetivos nessa tese foram estudar: 1) as propriedades biológicas da urease ubíqua de soja, fusionada à Glutationa S-Transferase (GST-uSBU), e 2) as propriedades estruturais do peptídeo Jaburetox. No capítulo 1 mostramos que a GST-uSBU é tóxica contra fungos filamentosos, interferindo no metabolismo secundário fúngico e na produção de pigmentos, e também foi tóxica a leveduras, induzindo mudanças morfológicas. A proteína de fusão apresentou ainda atividade inseticida e induziu agregação de plaquetas de coelho e de hemócitos de insetos. Os dados sugerem que GST-uSBU serve de modelo para o estudos futuros de propriedades biológicas de ureases de plantas. No captítulo 2, o peptídeo Jaburetox foi estabilizado em sua forma monomérica, com a utilização do agente redutor TCEP (tris (2-carboxietil) fosfina). O Jaburetox apresentou elevado raio hidrodinâmico, característico de uma proteína intrinsicamente desordenada. Estes dados foram confirmados através de análises de Dicroísmo Circular, de preditores de desordem (PONDR) e por Ressonância Magnética Nuclear. Jack bean produces three isoforms of ureases, Canatoxin, JBU and JBURE-II, which also have biological properties unrelated to enzymatic activity. Among them, the insecticidal activity has been the most studied. The toxicity of ureases against insects involves hydrolysis by insect digestive enzymes and the release of a toxic internal peptide. An equivalent peptide was cloned in Escherichia coli and called Jaburetox. This peptide presents a broad spectrum of activity against insects and is also fungitoxic. In this thesis, our objectives were to study: 1) the biological properties of the soybean ubiquitous urease fused to Glutathione S- Transferase (GST-uSBU), and 2) the structural properties of the peptide Jaburetox. In chapter 1, we show that GST-uSBU is toxic against filamentous fungi, interfering on the fungal secondary metabolism, in the pigments production, and also affected yeasts, inducing morphological changes. GST-uSBU also displayed insecticidal activity and induced aggregation of rabbit platelets and insect hemocytes. Thus, this fusion protein could serve as model for future studies related to biological properties on plant ureases. In chapter 2, the peptide Jaburetox was stabilized in its monomeric form by using the reducing agent TCEP (tris (2-carboxyethyl) phosphine). Jaburetox showed a large hydrodynamic radius, which is characteristic of an intrinsically disordered protein. These data were confirmed by Circular Dichroism analysis, disorder predictors (PONDR) and Nuclear Magnetic Resonance. The structure of the peptide in solution was elucidated, showing a α-helix motif in the N-terminal region and two turn-like structures, one located in the central region and another near the C-terminal of the peptide. The knowledge of Jaburetox’s three-dimensional structure is an important step towards the understanding of its insectidal and fungitoxic mode of action.

Nossos objetivos nessa tese foram estudar: 1) as propriedades biológicas da urease ubíqua de soja, fusionada à Glutationa S-Transferase (GST-uSBU), e 2) as propriedades estruturais do peptídeo Jaburetox. No capítulo 1 mostramos que a GST-uSBU é tóxica contra fungos filamentosos, interferindo no metabolismo secundário fúngico e na produção de pigmentos, e também foi tóxica a leveduras, induzindo mudanças morfológicas. A proteína de fusão apresentou ainda atividade inseticida e induziu agregação de plaquetas de coelho e de hemócitos de insetos. Os dados sugerem que GST-uSBU serve de modelo para o estudos futuros de propriedades biológicas de ureases de plantas. No captítulo 2, o peptídeo Jaburetox foi estabilizado em sua forma monomérica, com a utilização do agente redutor TCEP (tris (2-carboxietil) fosfina). O Jaburetox apresentou elevado raio hidrodinâmico, característico de uma proteína intrinsicamente desordenada. Estes dados foram confirmados através de análises de Dicroísmo Circular, de preditores de desordem (PONDR) e por Ressonância Magnética Nuclear. A estrutura do peptídeo em solução foi elucidada, apresentando um motivo α-hélice na região N-terminal e duas estruturas do tipo volta, uma na região central e outra próxima da região C-terminal do peptídeo. O conhecimento da estrutura tridimensional do peptídeo será de grande importância para o entendimento do mecanismo de ação inseticida e fungitóxico do Jaburetox. / Urease (EC 3.5.1.5) is a nickel-dependent metalloenzyme that catalyzes the hydrolysis of urea to form ammonia and carbon dioxide. Soybean (Glycine max) produces two urease isoforms: ubiquitous urease (uSBU) and the embryo-specific urease (eSBU). Our group demonstrated that eSBU displays biological properties independent of its ureolytic activity, such as platelets activation, insecticidal activity and inhibition of phytopathogenic fungi growth. These data suggested that soybean ureases could be involved in plant defense. Other ureases also studied by our group are jack bean (Canavalia ensiformis) ureases. Jack bean produces three isoforms of ureases, Canatoxin, JBU and JBURE-II, which also have biological properties unrelated to enzymatic activity. Among them, the insecticidal activity has been the most studied. The toxicity of ureases against insects involves hydrolysis by insect digestive enzymes and the release of a toxic internal peptide. An equivalent peptide was cloned in Escherichia coli and called Jaburetox. This peptide presents a broad spectrum of activity against insects and is also fungitoxic. In this thesis, our objectives were to study: 1) the biological properties of the soybean ubiquitous urease fused to Glutathione S- Transferase (GST-uSBU), and 2) the structural properties of the peptide Jaburetox. In chapter 1, we show that GST-uSBU is toxic against filamentous fungi, interfering on the fungal secondary metabolism, in the pigments production, and also affected yeasts, inducing morphological changes. GST-uSBU also displayed insecticidal activity and induced aggregation of rabbit platelets and insect hemocytes. Thus, this fusion protein could serve as model for future studies related to biological properties on plant ureases. In chapter 2, the peptide Jaburetox was stabilized in its monomeric form by using the reducing agent TCEP (tris (2-carboxyethyl) phosphine). Jaburetox showed a large hydrodynamic radius, which is characteristic of an intrinsically disordered protein. These data were confirmed by Circular Dichroism analysis, disorder predictors (PONDR) and Nuclear Magnetic Resonance. The structure of the peptide in solution was elucidated, showing a α-helix motif in the N-terminal region and two turn-like structures, one located in the central region and another near the C-terminal of the peptide. The knowledge of Jaburetox’s three-dimensional structure is an important step towards the understanding of its insectidal and fungitoxic mode of action.

O Jbtx e o Jbtx-C-ter mantiveram-se em conformação muito similar à proposta por modelagem por homologia, enquanto que o Jbtx-N-ter apresentou-se completamente desenovelado e o Jbtx-Db, como duas a-hélices antiparalelas. Quando inseridos em bicamada, diversos comportamentos foram observados. Em configuração transmembrana, o Jbtx foi capaz de atrair cabeças polares dos fosfolípídeos para dentro da bicamada, evidenciando um mecanismo do tipo toroidal. A importância de ácidos nucleicos extracelulares para o sistema imune de Rhodnius prolixus : possível envolvimento na resposta contra a toxina derivada de urease, jaburetox

Ureases são enzimas que catalisam a hidrólise de ureia para formar amônia e dióxido carbono, sendo produzidas por plantas, fungos e bactérias. A toxina de estudo, Jaburetox, é um peptídeo recombinante derivado de uma das isoformas de urease presentes em Canavalia ensiformis. Esta molécula mostrou toxicidade a insetos de diferentes ordens quando administrada oralmente ou por injeção e, além disso, apresenta atividades antifúngica e bacteriostática, não exibindo efeitos tóxicos para mamíferos, demonstrando um enorme potencial biotecnológico. Em estudos realizados com Rhodnius prolixus, um dos principais vetores da Doença de Chagas na América Latina, observamos que o Jaburetox é capaz de interagir com hemócitos, células do sistema imune. Concomitante a esta interação, também demonstramos que o peptídeo é capaz de induzir a agregação de hemócitos, característica da resposta celular, e também é capaz de aumentar a atividade da fenoloxidase (PO), enzima envolvida na resposta humoral dos insetos. Estes efeitos sugerem que o peptídeo é capaz de ativar o sistema imune dos insetos. Os insetos apresentam somente imunidade inata, sendo esta subdividida em resposta celular e humoral. A resposta celular, mediada por hemócitos, é caracterizada pela fagocitose, agregação e encapsulação de patógenos. Em contraste, a resposta humoral é representada pela produção de peptídeos antimicrobianos, melanização desencadeada pela enzima PO e produção de espécies reativas de oxigênio (ROS) e nitrogênio (RNS). Recentemente, outro mecanismo de resposta imune inata tem sido estudado: as armadilhas extracelulares - Extracellular Traps (ETs), sendo bem caracterizadas em mamíferos, porém pouco estudadas em insetos. As ETs são armadilhas de ácidos nucleicos extracelulares associados a proteínas citoplasmáticas, que podem auxiliar na captura e na morte de patógenos. Essa resposta é desencadeada por receptores extracelulares de células granulares que induzem o aumento de Ca2+ intracelular, levando à produção de ROS, que parece ser parte fundamental para liberação dos ácidos nucleicos, permitindo com que estes passem para o citoplasma e associem-se com as proteínas. O presente estudo tem como objetivo avaliar o papel de ácidos nucleicos extracelulares na resposta imune e na proteção contra o peptídeo entomotóxico Jaburetox. As respostas imunológicas foram avaliadas por meio da contagem de agregados, através de hemocitômetro, e por ensaio colorimétrico para dosar alterações da resposta humoral, 6 e 18 h após injeção de Jaburetox e/ou RNA extracelular (RNAet). A liberação de RNA e DNA foi avaliada 6 h pós injeção de tratamentos para ensaios in vivo e 1 h pós incubação para ensaios in vitro, através do kit Qubit RNA HS e Qubit dsDNA HS. Para avaliar a liberação de ROS e viabilidade celular, realizamos culturas de hemócitos, que foram tratados por 24 h com Jaburetox. Por fim, avaliamos a imunocompetência dos insetos após tratamentos com Jaburetox, RNAet ou Jaburetox + RNAet e desafiamos o inseto contra uma bactéria patogênica. A hemolinfa dos insetos foi coletada e unidades formadoras de colônia foram mensuradas pelo método de drop-plate. Os resultados obtidos neste trabalho indicam que o RNAet é capaz de ativar as defesas do organismo, tanto celular quanto humoral e, quando associado à toxina, apresentam um efeito protetor. Jaburetox não foi capaz de induzir alterações na liberação de ácidos nucleicos em nenhuma das abordagens, mas modificou a liberação de ROS na dose de 6 M. A viabilidade celular não foi alterada em nenhuma das condições testadas. Nos ensaios de imunocompetência, observamos que o peptídeo modula a resposta do organismo, deixando-o mais susceptível a infecções. Os tratamentos com RNAet desencadeiam uma resposta protetora. Nossos dados sugerem que o RNAet modula as defesas do organismo de forma benéfica para combater patógenos, enquanto que o Jaburetox, nas condições testadas, não induz resposta imune através de ETs, mas torna o inseto mais susceptível a infecções por patógenos. Os presentes achados enfatizam a importância dos ácidos nucleicos extracelulares para as defesas imunológicas em insetos. Também elucidamos os efeitos desencadeados pelo peptídeo, aprimorando os conhecimentos referentes a esta toxina, enfatizando o potencial uso deste composto como um bioinseticida. / Ureases are enzymes that catalyze the hydrolysis of urea to form ammonia and carbon dioxide, being produced by plants, fungi and bacteria. The studied toxin, Jaburetox, is a recombinant peptide derived from one of the urease isoforms present in Canavalia ensiformis. This molecule showed toxicity to insects of different orders when administered orally or by injection and, in addition, presents antifungal and bacteriostatic activities, not exhibiting toxic effects to mammals, demonstrating an enormous biotechnological potential. Studies with Rhodnius prolixus, one of the main vectors of Chagas' disease in Latin America, have shown that Jaburetox is capable of interacting with hemocytes, the cells of the immune system. Concomitant with this interaction, we also demonstrated that the peptide induces hemocyte aggregation, a characteristic of the cellular response, and is also capable of increasing the activity of phenoloxidase (PO), an enzyme involved in the humoral response in insects. These effects suggest that the peptide is able of activating the insect immune system. Insects present only innate immunity, which is subdivided into cellular and humoral responses. The cellular response, mediated by hemocytes, is characterized by phagocytosis, aggregation and encapsulation of pathogens. In contrast, the humoral response is represented by the production of antimicrobial peptides, melanization triggered by the PO enzyme and the production of reactive oxygen species (ROS) and reactive nitrogen species (RNS). Recently, another mechanism of innate immune response has been studied: the Extracellular Traps (ETs), being well characterized in mammals, but little studied in insects. ETs consist of extracellular nucleic acids associated with cytoplasmic proteins that may aid in the capture and killing of pathogens. This response is triggered by extracellular granular cell receptors that induce the increase of intracellular Ca2+, leading to the production of ROS, which appear to play a key role in the release of nucleic acids, allowing them to pass into the cytoplasm and associate with proteins. The present study aims to evaluate the role of extracellular nucleic acids in the immune response and protection against the entomotoxic peptide Jaburetox. Immunological responses were evaluated by means of hemocytometer counting and by colorimetric assays to measure changes in the humoral response, 6 and 18 h after injection of Jaburetox and/or extracellular RNA (RNAet). The release of RNA and DNA was evaluated 6 h post injection of treatments for in vivo assays and 1 h post incubation for in vitro assays, using the Qubit RNA HS and Qubit dsDNA HS kits. In order to evaluate the release of ROS and 14 the cell viability, hemocyte cultures were treated for 24 h with Jaburetox. Finally, the immunocompetence was evaluated after treatment with Jaburetox, RNAet or Jaburetox + RNAet and challenging the insects with a pathogenic bacterium. Hemolymph of the insects was collected and colony forming units were measured by the drop-plate method. The results obtained in this work indicated that RNAet is able to activate the organism defenses, both cellular and humoral, and displays a protective effect when associated with the toxin. Jaburetox was not able to induce changes in the release of nucleic acids in any of the approaches, but modified the release of ROS at the dose of 6 μM. Cell viability was not altered in any of the conditions tested. In the immunocompetence assays, we observed that the peptide modulates response of the organism, making it more susceptible to infections. RNAet treatments elicit a protective response. Our data suggest that RNAet modulates the insect defenses in a beneficial way to fight pathogens, while Jaburetox, in the conditions tested, does not induce an immune response through ETs, but makes the insect more susceptible to pathogen infections. The present findings emphasize the importance of extracellular nucleic acids for immune defenses in insects. We also elucidate the effects triggered by the peptide, improving the knowledge regarding this toxin, reinforcing its potential use as a bioinsecticide.
