Effect of reducing agents on batter consistency and physical characteristics of bread from sorghum flour

Tolerance to harsh climates and low moisture conditions are unique traits making sorghum an economical choice in an era of global water scarcity. In recent years, sorghum has gained greater recognition as a gluten-free grain and is a safe alternative for individuals suffering from gluten sensitivities or celiac disease. Still, the lack of gluten proteins does not allow sorghum to form a viscoelastic dough. In this study reducing agents were added to improve functional properties of sorghum kafirins for bread baking. Study objectives were to determine the effect of reducing agents on protein body structure of sorghum kafirins, investigate the influence on the sorghum batter consistency, and evaluate the effects on the physical characteristics of sorghum bread. Protein analysis, accomplished using RP-HPLC, showed reducing agents, L-cysteine and sodium metabisulfite, reduced protein structure; increasing RP-HPLC total peak area up to 747% and 681%, respectively. Batter consistency was obtained using a RVA. Treatments of L-cysteine (2.5% fwb) expressed increased RVA peak viscosity and decreased final viscosity. Samples treated with sodium metabisulfite (500 ppm fwb) had increased peak viscosity, holding strength and final viscosity. Yeast activity of batter treated with ≥3000 ppm (fwb) sodium metabisulfite caused volume loss of 95% yet at 500 ppm (fwb) sodium metabisulfite did not have an effect. Batter with 2.5% (fwb) L-cysteine experienced reduced yeast activity after 20 min. Sorghum bread characteristics were altered. Loaf volume and crumb grain characteristics of bread produced using sodium metabisulfite (500 ppm) were equal to that of the control, while initial texture and staling were improved. The addition of L-cysteine (2.5% fwb) to breads lowered loaf volume but produced softer initial crumb texture and improved in-vitro protein digestibility by 18.8%.

A cana-de-açúcar é um alimento bem aceito por toda a população em geral, podendo ser consumido na forma de toletes (in natura) ou na forma de caldo, podendo ser apreciado com gelo somente, ou adicionado de sucos cítricos, mas nem sempre tratada com algum sanitizante ou outro produto para manutenção de sua qualidade. A cana-de-açúcar também pode ser consumida na forma de minimamente processada, sanitizada e embalada, facilitando sua utilização em redes de alimentação. Embora o processamento mínimo de vegetais aumente a suscetibilidade ao ataque de microrganismos e escurecimento enzimático, essas alterações podem ser reduzidas com o uso de tratamentos adequados, como sanitizantes e agentes antioxidantes. Em vista disso, procurou-se nesse trabalho avaliar a qualidade da cana-de-açúcar minimamente processada, tratada com antioxidantes e radiação gama. Os tratamentos aplicados para a conservação dos toletes de cana-de-açúcar foram: ácido cítrico nas concentrações de 1,5% e 3,0%; cloridato de L-cisteína a 1,0% e 2,0%; radiação gama com as doses de 2,0 e 4,0 kGy. Antes de receberem os tratamentos, os toletes de cana-de-açúcar foram processados e sanitizados com solução de hipoclorito de sódio. Após a drenagem, os toletes foram acondicionados em embalagens de polietileno de alta densidade e refrigerados a 4°C. Os produtos foram analisados após 1, 7 e 14 dias de armazenamento. Todos os tratamentos apresentaram-se em condições sanitárias adequadas para o consumo, independente das concentrações e dosagens. Os toletes de canas-de-açúcar tratados com ácido cítrico e cloridrato de L-cisteína apresentaram os melhores resultados quanto à inibição das enzimas de escurecimento, polifenoloxidase e peroxidase, sendo a cana-de-açúcar tratada com L-cisteína a que apresentou o caldo com melhores resultados quanto à inibição de seu escurecimento e níveis de açúcares. Todos os tratamentos inibiram o desenvolvimento de microrganismos / Sugar cane is a well-accepted food for the general population and can be consumed in the form of straw (in natura) or in the form of juice, which can be enjoyed with ice only, or added with citrus juice, but always treated with some sanitizing agent or other product to maintain its quality. Sugarcane can also be consumed in the form of minimally processed, sanitized and packaged sugarcane, improving its use in food networks. Unless, minimal processing of vegetables increases susceptibility to microorganism attack and enzymatic browning, but such changes can be reduced with the use of appropriate treatments such as sanitizers and antioxidants. The aim of this work was to evaluate the quality of minimally processed sugar cane treated with antioxidants or gamma radiation. The treatments applied for the preservation of sugarcane straw were: citric acid at concentrations of 1.5% and 3.0%; L-cysteine at concentrations of 1.0% and 2.0%, and gamma irradiation at doses of 2.0 and 4.0 kGy. Before receiving the treatments, the sugar cane straws were processed and sanitized with sodium hypochlorite solution. After draining, the straws were packed in high-density polyethylene packages and refrigerated at 4°C. The products were analyzed after 1, 7 and 14 days of storage. All treatments were in sanitary conditions suitable for the consumption, independent of the concentrations and dosages. The sugarcane treated with citric acid and L-cysteine presented the best results regarding the inhibition of the darkening enzymes, polyphenoloxidase and peroxidase. Sugarcane treated with L-cysteine presented the juice with better sugar levels and inhibition of browning. All treatments showed effective inhibiton of the microorganisms development

The Effects of <em>L</em>-Cysteine on Alzheimer's Disease Pathology in <em>APOE2</em>, <em>APOE3</em>, and <em>APOE4</em> Homozygous Mice

The APOE gene is of profound importance regarding the onset of Alzheimer's disease (AD). From the small physical differences among the protein products of the isoforms of this gene arises a profound difference in their physiologies. For example, the APOE2 isoform confers resistance to AD, the APOE3 isoform confers neutral susceptibility to AD, and the APOE4 isoform confers proneness to AD. L-cysteine is an amino acid that has several anti-AD properties, among which are its ability to sequester iron and form glutathione – a powerful antioxidant – and therefore may be a promising potential dietary supplement for ameliorating AD pathology. In our experiment, we fed Mus musculus (mice) homozygous for APOE2, APOE3, and APOE4 either a control diet or a diet high in L-cysteine. Using Western blotting analysis, we quantified Amyloid β (Aβ), hyper-phosphorylated Tau (HP-Tau), and the three APOE proteins that we extracted from post-mortem brains of APOE2, APOE3, and APOE4 homozygous mice of 3-, 6-, 9-, and 12-month ages. We calculated a three-way ANOVA on a sample of 86 mice to examine the effect of age, genotype, and diet on protein quantities. We found that administration of L-cysteine trends towards lowering levels of Aβ in each cohort, but this effect is statistically insignificant. On the other hand, L-cysteine caused a significant decrease in APOE production with regard to diet [F(1,62) = 6.17, p=0.02], indicating that less APOE is produced due to the decrease in Aβ burden. Furthermore, administration of L-cysteine revealed no significant impact on or trends regarding HP-Tau deposition between diet types for each cohort. However, we observed that L-cysteine appeared to nullify the increasing trend in HP-Tau deposition between APOE2 and APOE4 cohorts. Thus, L-cysteine may be weakly affecting HP-Tau deposition via its ability to somewhat reduce Aβ burden and consequently prevent the shutdown of the proteosomes responsible for the degradation and clearance of HP-Tau. Taken together, these data suggest that L-cysteine should be considered as an intervention for AD pathology.

Transformed Legionella for Application in Engineering Process Validation in the Built Environment

abstract: Legionella pneumophila is a waterborne pathogen that causes Legionnaires' disease, an infection which can lead to potentially fatal pneumonia. In a culture-based technique, Legionella is detected using buffered charcoal-yeast extract (BCYE) agar supplemented with L-cysteine, Iron salt and antibiotics. These supplements provide essential and complex nutrient requirements and help in the suppression of non-target bacteria in Legionella analysis. Legionella occurs naturally in freshwater environments and for their detection; a sample is plated on solid agar media and then incubated for several days. There are many challenges in the detection of Legionella in environmental waters and the built environments. A common challenge is that a variety of environmental bacteria can be presumptively identified as Legionella using the culture-based method. In addition, proper identification of Legionella requires long incubation period (3-9 days) while antibiotics used in BCYE agar have relatively short half-life time. In order to overcome some of the challenges, Legionella has been genetically modified to express reporter genes such Green Fluorescent Protein (GFP) that can facilitate its detection in process validation studies under controlled laboratory conditions. However, such studies had limited success due to the instability of genetically modified Legionella strains. The development of a genetically modified Legionella with a much rapid growth rate (1-2 days) in simulated environmental systems (tightly-controlled water distribution system) is achieved. The mutant Legionella is engineered by transforming with a specific plasmid encoding CymR, LacZ and TetR genes. The newly engineered Legionella can grow on conventional BCYE agar media without L-Cysteine, Iron salt and only require one antibiotic (Tetracycline) to suppress the growth of other microorganisms in media. To the best of our knowledge, this is the first report of L. pneumophila strain capable of growing without L-Cysteine. Design and analysis of an electronically switchable ion exchange system

Metal contamination is a considerable environmental problem because metals are persistent contaminants. Ion exchange is one of the most commonly used treatment options for trace metal removal. This research develops and evaluates a redox active modified ion exchange system that has the potential to reduce the ionic strength of ion exchange regeneration streams. Poly-L-cysteine (PLC) was selected as the redox active, adsorbing functional group on the surface of a reticulated vitreous carbon (RVC) electrode. PLC is an excellent soft acid metal chelator and is unique in that its thiol groups can form disulfide bonds with each other. The reduction of available thiols changes the metal binding capacity of the peptide since the thiol is the primary binding group. RVC provides a macroporous conductive monolithic resin to support the peptide. An experimental apparatus was designed to study the properties of this system and estimate performance. Distinct oxidized and reduced states of PLC on the surface of the RVC were confirmed by changes in metal binding characteristics. Adsorption edges showed a sharper pH dependence for the reduced electrode compared to the oxidized electrode from pH 3-7. Adsorption isotherms performed at pH 7 showed increased capacity for the reduced electrode. The change was reversible by chemical and electrical reduction. This difference was confirmed at the molecular level with Cd- EXAFS of oxidized and reduced electrodes. A greater degree of cadmium-sulfur coordination was observed on the reduced electrode and a greater cadmium-oxygen coordination was apparant on an oxidized electrode. A multidentate adsorption model was developed to model the pH dependent behavior of cadmium adsorption on the PLC-RVC surface. Nickel adsorption showed increased adsorption in the oxidized state. The most likely explanation is increased carboxylate complexation. The electronically switchable ion exchange system (ESIE) provides a framework for modifying traditional ion exchange processes. The system has 5 to 10 times less specifc capacity than current ion exchange systems, but uses solutions 10-100 times lower in ionic strength for regeneration. Further studies on the effect of ionic strength on adsorption and current usage are necessary to compare the cost of the ESIE process to traditional ion exchange. / text

The Role of N-acetyl-L-Cysteine (NAC) as an Adjuvant to Opioid Treatment in Patients with Inadequately Controlled Chronic Neuropathic Pain

Introduction. While opioid medications are commonly prescribed for management of neuropathic pain (NP), long-term use has been associated with increased risk for overdose, drug interactions and addiction. New strategies are necessary to better manage chronic pain, thereby reducing need for opioid medications and their associated adverse consequences. N-acetyl-L-cysteine (NAC), an over-the-counter supplement, has shown promise in the treatment of psychiatric and addictive disorders. In addition, NAC has shown promise for reducing physiological signs of NP in laboratory rat models, prompting this study. Purpose. The present study was an open-label clinical trial of NAC as an adjuvant to opioid treatment for poorly controlled, chronic NP. It examined whether 1200 mg NAC twice daily for 4 weeks was associated with: lower ratings of patient-reported pain; reductions in PRN opioid medication for breakthrough pain; and improvements in physical and mental health quality of life (QoL). The study also examined whether appraisal of pain impacts response to medication. Method. Participants were N=28 chronic NP patients who consented to study participation. This consisted of 2 baseline assessments, 4 weeks of NAC and 1 post-trial follow-up visit. The majority (N=17) dropped out or were excluded during baseline. Of the remaining participants, N = 11 started the study medication and N=10 completed the study, with daily recordings of pain severity ratings and use of PRN opioid medication. Small sample size limited analyses to qualitative case reviews and effect sizes. Results. Over 90% of participants receiving NAC completed the study. Case review found varied results. While 4 of 10 participants showed decrease in average pain ratings during NAC, estimated effect sizes for the whole sample were small, bordering on negligible (ω² from .003 to .027) as were those for PRN opioids (Partial Eta-Squared=.0003). Effect size for mental health QoL was medium (Cohen's d=.421). Conclusions. With N=10, findings must be interpreted with caution. Nonetheless, the study found some albeit small evidence supporting NAC for improving mental health QoL and pain ratings. Several participants reported improvements in pain and mental health domains while taking NAC. NAC was well tolerated with minimal side effects. Lessons from this study will inform design and implementation of future NAC studies.

Este estudo teve como objetivo avaliar o efeito da N-acetil-L-cisteína (NAC), adicionada ao diluente composto de leite em pó desnatado, sobre a viabilidade espermática e o estresse oxidativo do sêmen equino resfriado a 5°C. Ejaculados de 8 pôneis da raça Brasileira foram coletados em triplicata resultando em 24 ejaculados. O sêmen foi distribuído em 4 grupos: Equidil® + 0,00 mM (controle), 0,5 mM, 1,0 mM ou 2,5 mM de NAC. As amostras foram armazenadas em tubos de 15mL e mantidas em caixas de transporte de sêmen BotuFLEX® (Botupharma, Botucatu-SP, Brasil). Parâmetros como motilidade total (MT), motilidade progressiva (MP), vigor, pH, resposta ao teste hiposmótico (HOST) e atividade mitocondrial (MTT) foram avaliados nas 24 e 48 h, bem como no sêmen fresco, após a diluição. O vigor, MT e MP foram também avaliados após teste de termorresistência (TTR), na ausência e presença de peróxido. A MT, a MP, o vigor espermático e a MTT foram similares (P > 0.05) entre as concentrações de NAC, nas 24 e 48 h. A resposta ao HOST foi semelhante entre as concentrações de NAC (P > 0,05) nas 24 h de resfriamento, porém nas 48 h ocorreu diminuição da funcionalidade da membrana no grupo NAC 2,5 mM em comparação ao grupo EQUIDIL, sem adição de NAC. A MT, a MP e vigor, das amostras resfriadas por 24 h e submetidas ao TTR, diferiu entre sem e com peróxido (P < 0,05) nos grupos Equidil, 0,5 mM e 1,0 mM, mas foi similar na concentração de 2,5 mM de NAC. Quando o sêmen foi resfriado por 48 h, houve diferença no vigor e MT entre amostras com e sem peróxido (P < 0,05), em todos os grupos testados, mas a MP foi similar entre amostras com e sem peróxido, na concentração 2,5 mM. Ejaculate of 8 ponies of the Brazilian breed were collected in triplicate, resulting in 24 ejaculates, distributed in 4 groups: Equidil® extender + 0.00mm (control), 0.5mM, 1.0mM and 2.5mM of NAC with the semen samples were The samples were stored in 15mL tubes and kept in BotuFLEX® semen transport boxes (Botupharm, Botucatu/SP/Brazil). Parameters, such as motility, vigor, pH, osmolarity, HOST, and MTT were evaluated at 24 and 48 h of cooling also in fresh semen. Vigor, total (TM) and progressive motility (PM) were also evaluated after thermoresistance test (TTR), in the absence and presence of peroxide. TM, PM, sperm vigor and MTT were similar (P > 0.05) between NAC concentrations at 24 and 48 h. The response to HOST was similar between NAC concentrations (P > 0.05) at 24 h cooling, but at 48 h there was a decreased in membrane functionality in 2.5mM NAC group compared to the EQUIDIL group. TM, PM, and vigor of the samples cooled by 24 h and submitted to TTR differed between without and with peroxide (P< 0.05) in the EQUIDIL, 0.5mM and 1.0mM groups, but was similar in 2.5mM NAC. Este estudo teve como objetivo avaliar o efeito da N-acetil-L-cisteína (NAC), adicionada ao diluente composto de leite em pó desnatado, sobre a viabilidade espermática e o estresse oxidativo do sêmen equino resfriado a 5°C. Ejaculados de 8 pôneis da raça Brasileira foram coletados em triplicata resultando em 24 ejaculados. O sêmen foi distribuído em 4 grupos: Equidil® + 0,00 mM (controle), 0,5 mM, 1,0 mM ou 2,5 mM de NAC. As amostras foram armazenadas em tubos de 15mL e mantidas em caixas de transporte de sêmen BotuFLEX® (Botupharma, Botucatu-SP, Brasil). Parâmetros como motilidade total (MT), motilidade progressiva (MP), vigor, pH, resposta ao teste hiposmótico (HOST) e atividade mitocondrial (MTT) foram avaliados nas 24 e 48 h, bem como no sêmen fresco, após a diluição. O vigor, MT e MP foram também avaliados após teste de termorresistência (TTR), na ausência e presença de peróxido. A MT, a MP, o vigor espermático e a MTT foram similares (P > 0.05) entre as concentrações de NAC, nas 24 e 48 h. A resposta ao HOST foi semelhante entre as concentrações de NAC (P > 0,05) nas 24 h de resfriamento, porém nas 48 h ocorreu diminuição da funcionalidade da membrana no grupo NAC 2,5 mM em comparação ao grupo EQUIDIL, sem adição de NAC. A MT, a MP e vigor, das amostras resfriadas por 24 h e submetidas ao TTR, diferiu entre sem e com peróxido (P < 0,05) nos grupos Equidil, 0,5 mM e 1,0 mM, mas foi similar na concentração de 2,5 mM de NAC. Quando o sêmen foi resfriado por 48 h, houve diferença no vigor e MT entre amostras com e sem peróxido (P < 0,05), em todos os grupos testados, mas a MP foi similar entre amostras com e sem peróxido, na concentração 2,5 mM. O pH do diluente Equidil® foi o maior e os grupos Equidil + 0,5 mM e 1,0 mM tiveram valores intermediários, enquanto a concentração de 2,5 mM de NAC gerou valores mais baixos, nos três momentos avaliados (P < 0,05). Não houve variação significativa de pH entre os momentos 0 e 24 h (P= 0,7075) e entre 0 e 48 h (P= 0,4617), em todos os grupos testados. As concentrações de NAC testadas não melhoraram a motilidade, integridade da membrana plasmática, atividade mitocondrial e resposta ao HOST de espermatozoides equinos resfriados a 5°C e armazenados por 48 h. Após TTR, as concentrações de NAC testadas não evitaram a diminuição da motilidade e vigor espermático, na presença de peróxido. / The aim of this study was to evaluate the effect of N-acetyl-L-cysteine (NAC) based on skim milk powder extender on sperm viability and oxidative stress of equine semen cooled to 5°C. Ejaculate of 8 ponies of the Brazilian breed were collected in triplicate, resulting in 24 ejaculates, distributed in 4 groups: Equidil® extender + 0.00mm (control), 0.5mM, 1.0mM and 2.5mM of NAC with the semen samples were The samples were stored in 15mL tubes and kept in BotuFLEX® semen transport boxes (Botupharm, Botucatu/SP/Brazil). Parameters, such as motility, vigor, pH, osmolarity, HOST, and MTT were evaluated at 24 and 48 h of cooling also in fresh semen. Vigor, total (TM) and progressive motility (PM) were also evaluated after thermoresistance test (TTR), in the absence and presence of peroxide. TM, PM, sperm vigor and MTT were similar (P > 0.05) between NAC concentrations at 24 and 48 h. The response to HOST was similar between NAC concentrations (P > 0.05) at 24 h cooling, but at 48 h there was a decreased in membrane functionality in 2.5mM NAC group compared to the EQUIDIL group. TM, PM, and vigor of the samples cooled by 24 h and submitted to TTR differed between without and with peroxide (P< 0.05) in the EQUIDIL, 0.5mM and 1.0mM groups, but was similar in 2.5mM NAC. After cooling for 48 h, there was difference in vigor and TM between samples with and without peroxide (P < 0.05) in all groups tested, but the PM was similar between samples with and without peroxide at concentration 2.5mM of NAC. The pH of the EQUIDIL extender was higher and the EQUIDIL + 0.5mM and 1.0mM groups had intermediate values, while the 2.5mM NAC concentration generated lower values in the three evaluated periods (P < 0.05). There was no significant variation of pH between 0 and 24 h (P=0.7075) and between 0 and 48 h (P=0.4617) in all groups tested. The concentrations of NAC tested did not improve motility, plasma membrane integrity, mitochondrial activity and response to HOST equine spermatozoa cooled to 5°C and stored for 48 h. After TTR, the concentrations of NAC tested did not prevent the decrease of motility and sperm vigor in the presence of peroxide.
