DETERMINISTIC TREND OR STOCHASTIC TREND ¡Ð EMPRICAL ANALYSIS OF PRACIFIC BASIN CONTRIES

In this paper, we assume structural changes occur in macroeconomic variables of eight countries in Asia Pacific Basin, including Indonesia, Japan, Malaysia, Philippines, Singapore, Thailand, and Taiwan. These variables are: current account, consumer price index, exchange rate, industrial product index, money supply (M1 and M2), interest rate, and real gross domestic product. In addition to performing conventional unit root test, we also perform Lee and Strazicich (2003) minimum LM test for unit root with breaks. When comparing two test results, we find that the test conclusions are diffident. Empirical results show that by conventional unit roots test, almost all macroeconomic variables are non-stationary. By minimum LM test for unit root with breaks, however, we increase the possibility to reject the null hypothesis that macroeconomic variables are all non-stationary. In estimating the time points when structural changes occur, Oil crisis(1973¡B1979¡B1990), Financial liberalization and De-regulation, Plaza Accord(1985)and Asia financial crisis(1997) remain the primary time point. They determine weather the macroeconomic variables of Asia Pacific basin countries have structural changes.

Abstract With a rapid market opening and structural reforms, Chinese economy has made great progress since 1978. A shortage of the necessities in product markets has been changed into overproduction and insufficient demand. An issuing of national debts in capital markets and then an opening of stock and foreign exchange tradings initiate the operations of stock, mutual fund, and futures markets. Thus applicability of IS-LM model in the analysis of Chinese economy is more plausible at the present time for the rapid development of Chinese capital market and the rising financial innovation activity than in the past. The objective of this thesis is to investigate the effectiveness of monetary policy by Chinese central bank with its economic reforms for 25 years. The important results of this study are as follows. First, unit roots of money supply, real gross domestic product and interest rates are present using the ADF test and the PP test. From Johansen-Juselius MLE test, we find a long-run equilibrium co-integration relationship among these three variables examined. Second, for 1978~2001, 1991~1992 and 1996~2000 periods, what actually causes different slopes of the LM curve is the income factor, and for the other periods, that is the interest rate factor, which means that the income factor first moves vertically and the interest rate factor moves then horizontally as increases in money supply shifts the LM curve outward. Finally, in the estimation of money demand elasticities with respect to income and interest rate, the long-run estimates of income elasticity are far greater than one, which implies money is an extreme luxury good in China. The interest rate elasticity estimates, however, exhibit a huge volatility and with positive sign unexpected, which can possibly be explained by deflation and tremendous savings in China since 1993.

Finite-different frequency-domain analysis of a dielectric waveguide crossing

Multiple dielectric crossing waveguides are indispensable in building a complex optical integrated circuit. Since each input/output waveguide will have many crossings, it is important to design a low-loss waveguide crossing to ensure the overall radiation loss is kept at a minimum. The beam propagation method (BPM) is usually the method of choice for modeling large but low-index-contrast waveguide devices. BPM assumes one-way propagation and adopts the paraxial approximation. It is neither able to consider reflection of electromagnetic (EM) fields nor to perform wide angle propagation of forward fields. Therefore, it can not be used to analyze perpendicular dielectric crossing waveguides. At a maximum 0.5 dB power loss per crossing, the difficulty of simulation a waveguide crossing is how to compute the complex coupling waves with high enough precision. In this thesis, two-dimensional planar integrated optical waveguide crossing is studied in detail for the through and cross power coupling coefficients with the finite-difference frequency-domain (FD-FD) method. By exploiting the dual symmetries: the ¡§+¡¨ symmetry and the ¡§X¡¨ symmetry in the perpendicular crossing waveguide, we are able to compute the EM fields and their power coefficients without using artificial absorbing boundary conditions (ABC) nor using the perfectly matching layer (PML). We develop the layer-mode based transparent boundary condition (LM-TBC) [1] for launching the fundamental incident mode as well as transmitting the reflected and scattered wave fields off the crossing area. Numerical results including the field distribution, power coefficients are carefully verified and the convergent comparisons are also studied in the thesis.

Valstybės išlaidų politikos ir visuminės paklausos Lietuvoje analizė 1995-2007 / Analysis of Fiscal Policy and Aggregate Demand in Lithuania in 1995 - 2007

Šiame darbe nagrinėjama vyriausybės vykdomos fiskalinės politikos įtaką visuminei paklausai Lietuvoje. Darbe yra apžvelgiama vyriausybės vartojimo išlaidų bei visuminės paklausos komponentų pokyčiai nuo 1995 iki 2007 metų. Teorinėje dalyje aptariamos pagrindinės Keinsistinės teorijos, kurios nagrinėja fiskalinės politikos įtaką visuminei paklausai, naudojant IS-LM ir AD-AS modelius. Taip pat teorinėje dalyje yra parodoma biudžeto deficito mažinimo įtaka ekonomikai. Darbe atliekama regresinė analizė, kuri įrodo, kad egzistuoja statistiškai reišmingas ryšys tarp visuminės paklausos ir valdžios sektoriaus vartojimo išlaidų. Pabaigoje pateikiamos rekomendacijos, kaip sustiprinti biudžeto išlaidų poveikį visuminei paklausai. / In this thesis the relationship between fiscal policy and aggregate demand in Lithuania is investigated. The first part of the work shows changes of government consumption expenditure and components of aggregate demand in period 1995 - 2007. The theoretical part analyses the main Keynesian ideas which research relationship between aggregate demand and fiscal policy, using IS-LM and AD-AS models. Also theoretical part shows how the reduction of budged deficit influences interest rates, aggregate demand, prices. In the last part aggregate demand is regressed against government consumption expenditure, interest rates, inflation and income. Results suggest that there are statistically significant relationship between aggregate demand and government consumption expenditure. Finally some recommendations are made in the end of the work.

Caracterização do padrão de integração do retrovírus pMFG-FVIII-P140K em linhagens celulares humanas produtoras de fator VIII recombinante / Characterization of the integration pattern of pMFG-FVIII-P140K retroviral vector in human cell lines producer of recombinant factor VIII

A hemofilia A é uma doença caracterizada pela deficiência do fator VIII (FVIII) de coagulação sanguínea e atinge 1 em 5000 homens. Países desenvolvidos, utilizam como terapia o FVIII recombinante (rFVIII), por apresentar maior segurança e consistir uma fonte ilimitada. Estudos realizados em nosso laboratório, utilizando o vetor retroviral pMFG-FVIII-P140K, originaram as linhagens celulares humanas, HepG2FVIIIdB/P140K (hepática) e Hek293FVIIIdB/P140K (renal), que foram selecionadas pelo mesmo sistema de seleção, o tratamento in vitro com as drogas Benzilguanina e Temozolomide. O presente trabalho teve por objetivo caracterizar e detalhar o padrão de integração do vetor retroviral pMFG-FVIII-P140K nas duas linhagens celulares humanas, e verificar se o perfil de integração está relacionado ao tipo celular específico ou se este sofre influência da estratégia de seleção a qual as células foram submetidas anteriormente. Foi utilizada a técnica de LM-PCR (ligação-mediada por PCR) que permite localizar o sítio de integração do vetor viral, por meio do sequênciamento do produto de PCR obtido após a digestão com enzimas de restrição e ligação de uma sequência adaptadora (LINKER) ao DNA genômico. Após o seqüenciamento as sequências foram submetidas a análises em bancos de dados de genomas (Human BLAT, QuickMap e DAVID/EASE) para caracterização dos respectivos clones. Também foi realizada uma análise da expressão relativa do mRNA relativo ao rFVIII por RT-PCR e da atividade biológica pelo teste de TTPA. Dessa forma foi possível observar que ambas as linhagens modificadas expressam o mRNA relativo ao rFVIII e que as células HepG2 e Hek293 apresentam níveis de secreção da proteína recombinante biologicamente ativa da ordem de 7,9 UI/mL e 2,1 UI/mL, respectivamente. Foram sequenciados um total de 201 clones da HepG2, sendo 123 similares ao genoma humano e dentre estes 73 considerados verdadeiros e 50 ambíguos. Da Hek293 foram sequenciados 221 clones, sendo 179 similares ao genoma humano e dentre estes 64 verdadeiros e 115 ambíguos. Observamos que o vetor retroviral pMFG-FVIII-P140K apresenta um perfil de integração não randômico e diferente entre as células estudadas, uma vez que na linhagem HepG2 houve uma preferência pelos cromossomos 19, 17 e 11, e na Hek293 pelo cromossomo 9. Em relação a regiões genômicas tais como distância de ilhas CpGs e de sítios de ligação de fatores de transcrição não houve diferença no perfil de integração em ambas as linhagens celulares, visto que nas duas células o vetor se inseriu preferencialmente a uma distância de ± 30-60Kb dessas regiões. Observamos também que houve uma integração dentro de genes codificadores de proteínas da ordem de 52% e 44% nas células HepG2 e Hek293, respectivamente, contudo em ambos os casos mais de 90% dessas inserções ocorreram em regiões intrônicas. Houve 20% de integrações em sítios frágeis do genoma na linhagem HepG2 e 17% na Hek293. Em suma este trabalho mostrou a integração do vetor retroviral pMFG-FVIII-P140K específica para as duas linhagens em estudo. Além disso, pudemos observar que em ambas as linhagens celulares, HepG2FVIIIdB/P140K e Hek293FVIIIdB/P140K, existem regiões gênicas dentro dos cromossomos na qual o vetor exibe uma preferência. O perfil de integração caracterizado aqui difere de outros trabalhos da literatura que utilizaram retrovírus derivados de MLV. Isso nos leva a crer que o padrão de inserção descrito pode estar relacionado ao fato de que essas células foram tratadas anteriormente com drogas quimioterapêuticas para seleção de um clone celular com maior nível de produção de rFVIII, levando consequentemente a seleção de um perfil de integração semelhante entre as linhagens celulares, mesmo estas tendo origens teciduais diferentes. / Hemophilia A is a disease characterized by deficiency of blood clotting factor VIII (FVIII) and affects 1 in 5000 men. Developed countries use as therapy for Hemophilia A recombinant FVIII (rFVIII) because rFVIII concentrates have proven efficacy and safety and this therapy consist of an unlimited supply.. Studies in our laboratory using the retroviral vector pMFG-FVIII-P140K, originate the human recombinant cell lines, HepG2FVIIIdB/P140K (liver origen) and Hek293FVIIIdB/P140K (renal origen) which were selected with high doses of Benzyladenine and Temozolomide. This study aimed to characterize the integration pattern of retroviral vector pMFG-FVIII-P140K in these human cell lines, and verify if the profile of integration is related to specific cell type or it was influenced by the selection strategy which cells have been previously submitted. We used the technique of LM-PCR (ligation-mediated PCR) that allows to locate the site of viral vector integration by the sequencing of PCR products. The sequences were obtained after genomic DNA digestion with restriction enzymes and subsequent connection of an adapter (LINKER) to 5or 3 of the DNA fragment. The sequences were analyzed in genomic databases (Human BLAT, and Quickmap DAVID / EASE) for characterization of the respective clones. It was also performed an analysis of relative expression of rFVIII mRNA by RT-PCR and the biological activity was measured by APTT test. Thus it was observed that both cell lines express the mRNA relative to rFVIII and HepG2 and HEK293 cells have levels biologically active recombinant protein approximately 7.9 IU / mL and 2.1 IU / mL, respectively . We sequenced a total of 201 clones of HepG2FVIIIdB/P140K, 123 of these sequences match to the human genome and, among those 73 were considered true and 50 were ambiguous. In Hek293FVIIIdB/P140K 221 clones were sequenced, these total, 179 were similar to the human genome. 64 were true and 115 were ambiguous. We note that the retroviral vector pMFG-FVIII-P140K presents a profile of integration different and non-random between the studied cells lines, in HepG2FVIIIdB/P140K was observed a preference of integration for chromosomes 19, 17 and 11, and in Hek293FVIIIdB/P140K for the chromosome 9. In summary this study showed the integration profile of retroviral vector pMFG -FVIII-P140K in two different cell lines that produces FVIIIr protein are very similar. Furthermore, we observed that in both cell lines, HepG2FVIIIdB/P140K and Hek293FVIIIdB/P140K, there are genetic regions within the chromosomes in which the vector displays a preference. The integration profile featured described here differs in CpGs island and TFBS from other studies in the literature that used retroviruses derived from MLV. This leads us to believe that the integration pattern described here may be related to the fact that these cells were previously treated with chemotherapeutic drugs for selecting a cell clone with the highest level of production of rFVIII, consequently leading to selection of a similar profile integration between cell lines, even those having different tissue origins.

As atuais tecnologias de informação permitem a manipulação de um volume grande de dados por diversas pessoas simultaneamente, representando uma valiosa ajuda aos profissionais de AEC (Arquitetura, Engenharia Civil e Construção). Para servir de base a essa manipulação é imprescindível que os dados estejam organizados em um conjunto harmônico, consistente e compartilhável. Os benefícios são numerosos, principalmente advindos da maior facilidade, rapidez e segurança na comunicação entre os indivíduos diretamente atuantes no ciclo de vida da edificação. A complexidade de um conjunto de informações desse tipo é muito grande, tanto pelo número de informações necessárias e de pessoas envolvidas, quanto pela quantidade e diversidade de interesses relacionados ao uso dessas informações. Nos últimos anos surgiram diversas propostas desses conjuntos de informações, em vários níveis de complexidade. Algumas propostas tiveram continuidade chegando a dar suporte a ferramentas computacionais muito utilizadas hoje em dia. É o caso do IES LM-63, sigla utilizada para se referir ao arquivo padrão de intercâmbio de dados relacionados às luminárias (IES Standard File Format for Electronic Transfer of Photometric Data and Related Information), elaborado pela IESNA (Illuminating Engineering Society of North America) e aprovado pelo ANSI (American National Standards Institute), e do IFC (Industry Foundation Classes), que é um modelo amplo dos elementos do ciclo de vida de uma edificação elaborado pela IAI (International Alliance for Interoperability) e aprovado pela ISO (International Organization for Standardization). Esta tese apresenta uma proposta de ampliação do modelo IFC na qual se inclui uma especificação mais ampla e eficiente do elemento luminária, baseada na descrição do IES LM-63. Analisam-se os caminhos possíveis para essa proposta e demonstra-se a viabilidade e as vantagens dessa ampliação ao utilizá-la no cálculo de iluminação de ambientes. / Recent information technologies allow handling of a large data volume by a number of people concurrently, representing a valuable help to AEC (Architecture, Engineering and Construction) professionals. Organizing data in a harmonic, consistent and shareable set is essential to base the handling. Benefits are numerous, mainly due to easier, faster and securer communication between directly acting people in building lifecycle. This kind of information set is very complex, caused by the number of necessary information and involved people, and by multiple and distinct aspirations related with the use of this information. In the last years, many and diverse information set proposals took place, in different level of complexity. Some proposals have continuity and even reached support computational tools broadly utilized. It is the case of IES LM-63, sequence that designate a standard file to luminaire data exchange (IES Standard File Format for Electronic Transfer of Photometric Data and Related Information), elaborated by IESNA (Illuminating Engineering Society of North America) and approved by ANSI (American National Standards Institute), and IFC (Industry Foundation Classes), a wide range building lifecycle elements model elaborated by IAI (International Alliance for Interoperability) and approved by ISO (International Standardization Organization). This doctoral thesis presents a proposal to extend IFC model in which is included a wider and more efficient specification of the luminaire element, based in IES LM-63 description. Possible ways to implement the proposal are analyzed and the feasibility and advantages of the extended data set are demonstrated.

This leads us to believe that the integration pattern described here may be related to the fact that these cells were previously treated with chemotherapeutic drugs for selecting a cell clone with the highest level of production of rFVIII, consequently leading to selection of a similar profile integration between cell lines, even those having different tissue origins.

As atuais tecnologias de informação permitem a manipulação de um volume grande de dados por diversas pessoas simultaneamente, representando uma valiosa ajuda aos profissionais de AEC (Arquitetura, Engenharia Civil e Construção). Para servir de base a essa manipulação é imprescindível que os dados estejam organizados em um conjunto harmônico, consistente e compartilhável. Os benefícios são numerosos, principalmente advindos da maior facilidade, rapidez e segurança na comunicação entre os indivíduos diretamente atuantes no ciclo de vida da edificação. A complexidade de um conjunto de informações desse tipo é muito grande, tanto pelo número de informações necessárias e de pessoas envolvidas, quanto pela quantidade e diversidade de interesses relacionados ao uso dessas informações. Nos últimos anos surgiram diversas propostas desses conjuntos de informações, em vários níveis de complexidade. Algumas propostas tiveram continuidade chegando a dar suporte a ferramentas computacionais muito utilizadas hoje em dia. É o caso do IES LM-63, sigla utilizada para se referir ao arquivo padrão de intercâmbio de dados relacionados às luminárias (IES Standard File Format for Electronic Transfer of Photometric Data and Related Information), elaborado pela IESNA (Illuminating Engineering Society of North America) e aprovado pelo ANSI (American National Standards Institute), e do IFC (Industry Foundation Classes), que é um modelo amplo dos elementos do ciclo de vida de uma edificação elaborado pela IAI (International Alliance for Interoperability) e aprovado pela ISO (International Organization for Standardization). Esta tese apresenta uma proposta de ampliação do modelo IFC na qual se inclui uma especificação mais ampla e eficiente do elemento luminária, baseada na descrição do IES LM-63. Analisam-se os caminhos possíveis para essa proposta e demonstra-se a viabilidade e as vantagens dessa ampliação ao utilizá-la no cálculo de iluminação de ambientes. / Recent information technologies allow handling of a large data volume by a number of people concurrently, representing a valuable help to AEC (Architecture, Engineering and Construction) professionals. Organizing data in a harmonic, consistent and shareable set is essential to base the handling. Benefits are numerous, mainly due to easier, faster and securer communication between directly acting people in building lifecycle. This kind of information set is very complex, caused by the number of necessary information and involved people, and by multiple and distinct aspirations related with the use of this information. In the last years, many and diverse information set proposals took place, in different level of complexity. Some proposals have continuity and even reached support computational tools broadly utilized. It is the case of IES LM-63, sequence that designate a standard file to luminaire data exchange (IES Standard File Format for Electronic Transfer of Photometric Data and Related Information), elaborated by IESNA (Illuminating Engineering Society of North America) and approved by ANSI (American National Standards Institute), and IFC (Industry Foundation Classes), a wide range building lifecycle elements model elaborated by IAI (International Alliance for Interoperability) and approved by ISO (International Standardization Organization). This doctoral thesis presents a proposal to extend IFC model in which is included a wider and more efficient specification of the luminaire element, based in IES LM-63 description. Possible ways to implement the proposal are analyzed and the feasibility and advantages of the extended data set are demonstrated.
