SUPPLEMENTAL YEAST FERMENTATION PRODUCTS EFFECT ON SOW LACTATION PERFORMANCE AND POST-PARTUM RECOVERY BASED ON UTERINE FLUIDS AND BLOOD PARAMETERS

The longevity of high productivity sows in the herd has become a challenge in pig production. Several factors may contribute to increased mortality rates observed over the past few years as well as lower retention rates of young sows. Chronic inflammation and metabolic disorders are conditions that sows had evolved over the years together with the greater productivity. This dissertation underlines the immunomodulatory effects of using yeast fermentation product fed to lactating sows. In the interest of determining patterns of local and systemic immune response, a new methodology to access cytokine profiles in puerperium sows was developed. In Chapter 2, one hundred-forty sows were used to evaluate the effects of two different <i>Saccharomyces cerevisiae</i> fermentation product (SCFP), a liquid source (LIQ) and a dry source (XPC®; Diamond V), on sow and litter performance. Sows were fed a common gestation diet until d 112 of pregnancy and then allotted to one of four treatments: 1) Control diet (CON), 2) CON + 15 mL/d of LIQ from d 112 to weaning (LIQ), 3) CON + 0.20% of XPC from d 112 to weaning (DRY), and 4) DRY + 15 mL/d of LIQ from d 112 to d 7 post-farrowing (D+L). Colostrum immunoglobulin concentrations were estimated using Brix refractometer. Plasma of piglets (2/sow) was collected 24 h after birth for immunocrit ratio analysis and for determination of plasma IgA and IgG concentrations. Lactation water and feed intake (ADFI) were recorded daily. Post-weaning follicle growth was evaluated by transrectal ultrasonography. Sows had the same initial BW (P > 0.13) but those fed any SCFP were heavier at weaning (P = 0.03) while not affecting sow backfat and loin depth (P>0.05). Overall, sows fed SCFP had greater ADFI than CON fed pigs (P < 0.01) while water intake, reproductive performance (total born, stillborn, weaned) did not differ among treatments (P > 0.05). Sows fed LIQ had the greatest ADFI on weeks 1, 2, 3, and overall compared to CON (P < 0.05). Litter ADG from SCFP treatments tended to be greater than CON (P = 0.10) and litter weight variability was lower (P = 0.10). No treatment effects were observed in colostrum Brix values (P > 0.77), in the piglet plasma IgG and IgA, and serum immunocrit ratio (P > 0.21). The average daily post-weaning follicle growth was greater for SCFP treatments than CON (P = 0.05). The wean to estrus interval was shorter for sows fed LIQ than CON and DRY (P < 0.01).<br><div><br></div><div>In Chapter 3 a non-invasive methodology to assess cytokine profiles from post-partum uterine lavage is described. The uteri of fourteen second and third parity sows were flushed with sterile saline solution (0.9%) on days 2, 4, and 14 post-parturition. Uterine fluid collected was immediately centrifuged and the supernatant stored at -20°C. Samples were freeze-dried, re-suspended in sterile saline (2 mL), and stored at -80°C. Cytokine profiles of the uterine fluid were evaluated using a multiplex ELISA panel including interleukin (IL)-1β, IL-4, IL-6, IL-8, IL-10, tumor necrosis factor-α (TNF-α), and interferon-γ (IFN-γ). Cytokine concentrations were calculated relative to protein content (pg/mg of protein). IFN-γ and TNF-α were lower than the limit of detection in most samples (5/38 and 1/38, respectively). IL-4 and IL-10 concentrations did not differ among days of collection (P>0.14). IL-8 was greater on day 4 than on days 2 or 14 (P<0.05). IL-1β and IL-6 were greater on days 2 and 4 than on day 14 (P<0.05).<br></div><div><br></div><div>The study presented in Chapter 4 refers to a subsample of sows (n=40) from the entire group of sows used in the study presented in Chapter 2. In this case, the methodology presented in Chapter 3 was used to evaluate SCFP effects on blood and uterine cytokine profiles in sows. A similar set of cytokines from Chapter 3 were evaluated on d 112 of gestation, d 2 and 6 post-farrowing in the plasma, and from uterine fluid collected on d 2, 4, and 6 post-farrowing. Serum C-Reactive protein (CRP) and haptoglobin concentrations were evaluated. No interactions between treatments and day of collection were observed (P>0.13). LIQ and D+L sows had the greatest serum IL-10 concentration (P<0.001) and sows fed CON tended to have lower serum concentration of IL-8 (P<0.06) vs. other treatments. Serum CRP concentrations were greatest on d 2 (P<0.001), serum IL-10 (P<0.04) and IL-4 (P<0.07) linearly decreased while serum haptoglobin (P<0.02) and IFN-γ (P<0.001) linearly increased post-farrowing. In the uterine fluid, LIQ and D+L sows had greater IFN-γ (P=0.04) concentrations and CON tended to have the least concentration of TNF-α (P=0.08). Uterine fluid IL-1 tended to linearly increase (P<0.07) and IL-6 linearly decrease (P<0.01) post-farrowing. LIQ sows had the greatest daily feed intake and CON the least during the first week of lactation (P=0.04).<br></div><div><br></div><div>In conclusion, feeding SCFP to lactating sows improved feed intake and litter growth while not affecting milk yield and colostrum quality. Besides improvements on litter ADG, the uniformity was better for all sources of SCFP. The liquid sources had slightly better results over the other sources and CON, including the greatest feed intake, less body weight mobilization, and a reduction in WEI. The method proposed to evaluate cytokine profiles in the uterine fluids of sows after farrowing, accomplished the objective of being a non-invasive procedure to be applied in puerperium sows. This new procedure was applied to analyze the immunomodulatory effects of SCFP. The correlations observed between the uterine and serum cytokines lead to a refined description of immune response in puerperium sows. Feeding SCFP to lactating sows stimulates the immune system allowing sows to build a desirable immune responses. Thus, the quicker resolution of acute phase reaction as demonstrated by greater daily feed intake in the first week post-farrowing can be attributed to SCFP immunomodulatory effects, ensuring better lactation performance.<br></div>

Circulating glucose responses in early lactation dairy cows to dietary restriction and rbST treatment

Galactopoietic effects of somatotropin are the result of IGF-I and require high-quality nutrient intake. This study investigated short-term partitioning effects during recombinant bovine somatotropin (bST) administration in high yielding early lactation dairy cows. Administration of recombinant bST has been shown generally to alter results of metabolic tests in the face of unchanged basal glucose and insulin concentrations. Ten multiparous Holstein cows were subjected to rbST (Lactotropin®) and/or feed intake restriction to 80% of predicted ME requirement (80% ME). Responses to insulin challenge (0.1 IU porcine insulin/kg BW. 210 min) and hyperglycaemic clamp (+50 mg/dL whole blood, 120 min) were tested during weeks 8 (control), 9 (rbST ), 11 (80% ME) and 12 (rbST + 80% ME) postpartum. Plasma and whole blood samples were assayed for glucose concentrations. The rbST treatment decreased fasting whole-blood glucose concentration by 9.4% (P<0.0001), which was likely a remnant of control hyperglycaemia. Maximum glucose response was 4.0 mg/dL (21.7%) lower (P<0.0038) and took 6.5 minutes longer to attain (P<0.0037). Steady-state glucose infusion rate (SSGIR) decreased by 8.1 % (P<0.0001). The 80% ME treatment decreased glucose availability by 5 to 6% (P<0.0100), while no glucose responses were affected. Restricted energy intake during treatment with rbST resulted in plasma glucose increase by 5.5% (P<0.0001). Peripheral uptake and utilization of glucose increased by 5.1 % (P<0.0005). Compared to energy restriction, 80%ME + rbST did not alter effects of nutrient restriction on responses to exogenous insulin challenge. Effects were small and inconsistent. SSGIR decreased by 5.0% in the 80% ME + rbST compared to the 80% ME period (P<0.0004) and the change in the hyperglycaemic clamp in the absence of an effect in the insulin challenge may be due to differences in endogenous insulin secretion. Lactogenesis II is critical stage of lactation and it is hindered by conditions like caesarean section, premature delivery, obesity, age, and parity. Previous studies have shown a connection between morphological characteristics of breast and nipple-areola complex and lactation onset. Higher number of areolar glands and their non-uniform distribution has been linked to earlier onset of lactation. Extremely large breasts and short nipples has been linked to breastfeeding difficulty. Aim of this study was to investigate relationship between morphological characteristics of breast and nipple-areola complex (number and distribution of areolar glands, nipple, areola, and breast area, protrusion of nipple and areola, and contrast of breast and areola) and lactation onset. For this purpose, we collected breasts and areolae photographs of 141 mothers one day postpartum in maternity ward in Prague and compared the morphological traits to lactogenesis II onset. We have found a non-uniform distribution of areolar glands with the highest abundance on upper lateral quadrant of areola. We found that right breast area tends to be bigger than left, and left nipple and areola tends to be longer than right. Parity had significant effect on lactation onset. The aim of the diploma thesis was to cover the changes in body composition by bioimpedance spectroscopy in Czech breastfeeding women during three periods - 3 weeks postpartum, 3 months and 6 months postpartum. Eight healthy breastfeeding women were involved in the study. At each examination there were measured anthropometric parameters and bioimpedance spectroscopy parameters by the Body Composition Monitor. Breast milk was also extracted during the study. We observed a gradual decrease in body weight of monitored women. The median weight loss between the first and third examination was 2,2 kg, the median weight in 6 months postpartum was 1,3 kg higher than the median of prepregnancy body weight, two of the women had lower weight 6 months postpartum than before the pregnancy, the body weight of other six women was higher in a scale between 0,1 kg to 3,1 kg. There were not proven statistically significant differences in body composition during lactation. A trend of decrease of TBW was observed during all lactation periods. A trend of slight increase of ICW was observed between the 3rd week and 3rd month postpartum, and then the decrease of ICW in the 6th month postpartum. Nutrient intake and nutrition knowledge of lactating women (0-6) months postpartum) in a low socio-economic area in Nairobi, Kenya

Breastfeeding is at its best when both the mother and infant benefit from the experience. Aim: To obtain data on the adequacy of the diet and nutrition knowledge of lactating women (0-6 months postpartum) living in a low socio-economic area in Nairobi. The findings could be used to plan future nutrition intervention programs. Research design: A cross-sectional survey in the quantitative and qualitative research paradigms. Health status indicators, socio-economic and cultural factors were additionally investigated as interactive factors that could influence the nutrient intake and nutrition knowledge of the lactating women. Setting: Mbagathi District Hospital, located at the edge of Kibera slum area in Nairobi. Sample: Lactating mothers (0-6 months postpartum), who were visiting the Maternal-Child Health Clinic to bring their children for immunisation, were recruited. Convenience sampling was used (N=120). Informed consent was obtained from the mothers in their home tongue. Methodology: Individual interviews in Swahili, using structured questionnaires (Socio biodemographic questionnaire, Hunger Scale, 24 Hour-recall, and Quantitative Food Frequency Questionnaire), and anthropometry (according to standard procedure) were done in the quantitative domain. Seven structured focus group discussions were employed in the qualitative domain. Descriptive and inferential statistics were used on the quantitative data, and Krueger‘s framework of analysis on the qualitative data. Ethical approval was obtained from the Ethics Committee, Faculty of Natural and Agricultural Sciences, University of Pretoria; Ref no EC 080922-039, and research permission was granted by the National Council for Science and Technology, Nairobi; Permit no NCST/5/002/R/355. Main findings: Majority of women (92%) had energy intakes between 5040 – 10080 kJ/day with the mean energy intake of 6975.5 kJ/day which was lower than the recommended 11340 kJ/day. Macronutrient intake was within the acceptable macronutrient distribution ranges, whereas the micronutrients were consumed below the recommended dietary allowances (WHO). The mean adequacy ratio (excluding niacin) was 0.74. The highest frequency of consumption (100%) was from cereals, while the lowest were for vitamin A rich vegetables/fruits (6.6%) and dairy products (1.7%). Overall, the mean Food Variety Score (FVS) was 6.6 ( ± 2.0) and the Dietary Diversity Score (DDS) was 4.3 ( ± 1.0). Hunger assessment revealed 43 households that were food secure, whereas 77 households were either hungry or at risk of hunger. Majority of women (93%) had good nutrition knowledge but the rationale for applying the knowledge was lacking for most of the nutrition concepts. Socio-economic status, cultural beliefs and practices, health status and nutrition knowledge were all identified as probable factors that influenced the dietary intake of the lactating women. Recommendation: There seems to be a need for well-designed nutrition intervention programs focusing on nutrient intake from culturally acceptable and affordable foods to increase dietary diversity and food variety of lactating women in this low socio-economic area. Lactational Performance and Energy Partitioning of Dairy Cows Supplemented with N-Acetyl-L-Methionine During Mid to Late Lactation

The N-acetyl-L-methionine (NALM) molecule is a methionine (Met) derivative produced via acetylation of the L-Met α-amino group with an N-acetyl group. This molecule has been shown to be bioavailable and capable of fulfilling the dietary requirement for Met in animals and humans. The current experiment was conducted to test a hypothesis that lactating dairy cows fed with NALM would increase milk production by increasing N and energy utilization efficiencies in a dose dependent manner. Eight multiparous Holstein cows that were mid lactation (124 ± 13 days-in-milk) with similar milk production were used in a 4 x 4 Latin square design for 84 d. A developmental NALM product from CJ CheilJedang (Seoul, South Korea) was used as the supplemental source of rumen-protected Met in the present study. Four dietary treatments included 0 g (control), 15 g, 30 g, and 45 g/d/cow of NALM supplementation. Supplementing NALM significantly increased dry matter intake (linear effect; P < 0.01), while milk yield tended to increase quadratically (P = 0.07). A linear decrease in milk fat concentration was seen due to supplementation of NALM in relation to the control ration (P = 0.02). However, milk fat yield was similar across treatments. A trend toward an increase in milk protein yield was observed between the control ration and the ration supplemented with 45 g of NALM (1.18 vs. 1.21 kg/d; P = 0.10). There were no differences in energy-corrected or 3.5% fat-corrected milk yields in response to treatments. It is likely that the supplementation of NALM to mid to late lactating dairy cows may have shifted nutrient and energy utilization toward tissue gain and lactation, which resulted in a decrease in feed efficiency for lactation (P = 0.02). Overall results from the present study suggest that supplementing NALM to mid to late lactating cows can increase milk yield in a dose dependent manner with a shift of net energy partitioning toward milk production and body weight gain. In addition, supplementing NALM increased milk nitrogen (N) output without affecting urinary N excretion.

Lactation and Weaning Weight Relationships in Hereford and Simmental-Hereford Cows in Southern Utah

Lactation and weaning weight relationships were studied in 37 cows grazing Southern Utah irrigated pastures during the summer of 1979. The herd included 23 Hereford cows ranging in age from 2 to 12 years and 14 Simmental-Hereford half-blood cows aged 3 to 9 years. The suckling calves were sired by Hereford bulls. The average daily milk yield of 6.16 kg was affected by the cow breed. Simmental-Hereford cows produced 7.11 kg of milk per day, while the Herefords produced 5.21 kg per day. Milk production declined with increased days in lactation. The decline was linear in the Simmental- Hereford cows and quadratic in the Herefords. Milk production was also affected by the sex of the suckling calf. Heifer calves received 6.44 kg per day compared t o 5.80 kg per day for bull calves. Calf birth weight and test day weight influenced milk yield as cows nursing heavier calves produced more milk. Cow breed affected milk composition. Simmental-Hereford cows had higher percent solids-not-fat, while Herefords were higher for percent protein. Calf weights were affected by sex of calf. Bull calves weighed 33 .89 kg at birth, while heifers weighed 32.14 kg. At weaning, bulls weighed 188.63 kg, while heifers weighed 177.14 kg. While no breed difference was evident for birth weight, calves from Simmental-Hereford dams weighed more over the duration of the study than calves from Hereford dams. The difference between the mean 205-day adjusted weights for the calves from the Simmental-Hereford and Hereford cows was 24.96 kg (230.50 kg and 205.54 kg, respectively). Milk yield was highly correlated to calf gain (r 0. 71). The breed of dam influence upon weaning weight was largely a result of differences .in quantity of milk produced.
