<p>Denna processbeskrivning undersöker hur en plantering i parkmiljö i Gävle skulle kunna planeras baserat på systemet matrix planting. Begreppet matrix planting beskrivs, samt grunderna för hur ett växtsamhälle byggs upp med passande växter. Fortsättningsvis redovisas inventeringen av den aktuella platsen, med beskrivning av de ekologiska förutsättningarna och förvaltarens önskemål. Ett förslag presenteras, där tankarna bakom formgivningen förklaras. En förteckning på lämpliga växter och deras egenskaper ingår. Därefter anges övergripande skötselanvisningar för matrix-planteringar. I processbeskrivningen konstateras slutligen att det är oerhört svårt att förutse hur ett växtsamhälle kommer att utvecklas, och att det inte går att lämna någon garanti för att en matrix-plantering kommer att uppnå stabilitet. Likväl kan skötselbehovet minskas genom att använda matrix planting som metod och genom noggranna växtval. En illustrationsplan i tre delar bifogas till processbeskrivningen.</p>

Polarization-sensitive Mueller-matrix optical coherence tomography

Measuring the Mueller matrix with optical coherence tomography (OCT) makes it possible to acquire the complete polarization properties of scattering media with three-dimensional spatial resolution. We first proved that the measured degree-of-polarization (DOP) of the backscattered light by OCT remains unity-a conclusion that validated the use of Jones calculus in OCT. A multi-channel Mueller-matrix OCT system was then built to measure the Jones-matrix, which can be transformed into a Mueller matrix, images of scattering biological tissues accurately with single depth scan. We showed that when diattenuation is negligible, the round-trip Jones matrix represents a linear retarder, which is the foundation of conventional PS-OCT, and can be calculated with a single incident polarization state although the one-way Jones matrix generally represents an elliptical retarder; otherwise, two incident polarization states are needed. We discovered the transpose symmetry in the roundtrip Jones matrix, which is critical for eliminating the arbitrary phase difference between the two measured Jones vectors corresponding to the two incident polarization states to yield the correct Jones matrix. We investigated the various contrast mechanisms provided by Mueller-matrix OCT. Our OCT system for the first time offers simultaneously comprehensive polarization contrast mechanisms including the amplitude of birefringence, the orientation of birefringence, and the diattenuation in addition to the polarization-independent intensity contrast, all of which can be extracted from the measured Jones or the equivalent Mueller matrix. The experimental results obtained from rat skin samples, show that Mueller OCT provides complementary structural and functional information on biological samples and reveal that polarization contrast is more sensitive to thermal degeneration of biological tissues than amplitude-based contrast. Finally, an optical-fiber-based multi-channel Mueller-matrix OCT was built and a new rigorous algorithm was developed to retrieve the calibrated polarization properties of a sample. For the first time to our knowledge, fiber-based polarization-sensitive OCT was dynamically calibrated to eliminate the polarization distortion caused by the single-mode optical fiber in the sample arm, thereby overcoming a key technical impediment to the application of optical fibers in this technology.

Correlation Between MMP-2 and -9 Levels and Local Stresses in Arteries Using a Heterogeneous Mechanical Model

The mechanical environment influences vascular smooth muscle cell (VSMC) functions related to the vascular remodeling. However, the relationships are not appropriately addressed by most mechanical models of arteries assuming homogeneity. Accounting for the effects of heterogeneity is expected to be important to our understanding of VSMC functions. We hypothesized that local stresses computed using a heterogeneous mechanical model of arteries positively correlate to the levels of matrix metalloproteinase (MMP)-2 and -9 in situ. We developed a mathematical model of an arterial wall accounting for nonlinearity, residual strain, anisotropy, and structural heterogeneity. The distributions of elastin and collagen fibers, quantified using their optical properties, showed significant structural heterogeneity. Anisotropy was represented by the direction of collagen fibers, which was measured by the helical angle of VSMC nuclei. The recruiting points of collagen fibers were computed assuming a uniform strain of collagen fibers under physiological loading conditions; an assumption motivated by the morphology. This was supported by observed uniform length and orientation of VSMC nuclei under physiological loading. The distributions of circumferential stresses computed using both heterogeneous and corresponding homogeneous models were correlated to the distributions of expression and activation of MMP-2 and -9 in porcine common carotid arteries, which were incubated in an ex vivo perfusion organ culture system under either normotensive or hypertensive conditions for 48 hours. While strains computed using incompressibility were identical in both models, the heterogeneous model, unlike the homogeneous model, predicted higher circumferential stresses in the outer layer. The tissue levels of MMP-2 and -9 were positively correlated to circumferential stresses computed using the heterogeneous model, which implies that areas of high stress are expected to be sites of localized remodeling and agrees with results from cell culture studies. The results support the role of mechanical stress in vascular remodeling and suggest the importance of structural heterogeneity in studying mechanobiological responses.

Einfluss von modifizierter extrazellulärer Matrix auf die Proteinexpression von Fibroblasten

Der humanen dermalen Wundheilung liegt ein komplexes Zusammenspiel verschiedener Faktoren zugrunde. Die Bedeutung dieses fein regulierten Gleichgewichts wird deutlich, wenn es durch Fehlregulationen oder Störungen zu chronischen Wundheilungsstörungen oder lokaler Fibrose mit überschießender Narbenbildung kommt. Eine der möglichen Methoden zur Prävention und Behandlung ist die Deckung der Wunde mit einem Hautersatz. Dabei werden zunehmend sogenannte Biomaterialien aus natürlichen Substanzen mit hoher Biokompatibilität und der Möglichkeit zur Interaktion mit dem nativen Gewebe verwendet. In Studien wurde gezeigt, dass vor allem sulfatierte Glykosaminoglykan-Derivate durch die Interaktion ihrer negativ geladenen Sulfatgruppen mit Zytokinen, Wachstumsfaktoren und dermalen Zellen einen positiven Einfluss auf den Wundheilungsprozess haben können. In der vorliegenden Arbeit wurden daher kollagenbasierte artifizielle extrazelluläre Matrizes mit unsulfatierter oder sulfatierter Hyaluronsäure hinsichtlich ihres Einflusses auf humane dermale Fibroblasten als Komponenten der Wundheilung untersucht. Dermale Fibroblasten spielen im Ablauf der Wundheilung eine tragende Rolle und interagieren eng mit der umgebenden Matrix. Anhand ihrer Proteinexpression lassen sich Rückschlüsse auf wichtige Funktionen wie Adhäsion, Proliferation, Differenzierung und Matrixsynthese ziehen. In den durchgeführten Experimenten zeigte sich, dass sulfatierte Matrix in der Kultur mit dermalen Fibroblasten kein entzündliches Milieu förderte. Die Proliferation, Differenzierung und Migration der Fibroblasten schienen gesteigert, während sich die Matrix-Synthese und ihr Remodeling weder pathologisch gehemmt noch überschießend zeigten. Daher wäre die weitere Untersuchung dieses Biomaterials ein vielversprechender Ansatz, um langfristig dem Risiko von Wundheilungsstörungen wie chronischen Wunden oder fibroproliferativen Wundheilungsstörungen effektiv entgegenzuwirken.

Rational Realizations of the Minimum Rank of a Sign Pattern Matrix

A sign pattern matrix is a matrix whose entries are from the set {+,-,0}. The minimum rank of a sign pattern matrix A is the minimum of the rank of the real matrices whose entries have signs equal to the corresponding entries of A. It is conjectured that the minimum rank of every sign pattern matrix can be realized by a rational matrix. The equivalence of this conjecture to several seemingly unrelated statements are established. For some special cases, such as when A is entrywise nonzero, or the minimum rank of A is at most 2, or the minimum rank of A is at least n - 1,(where A is mxn), the conjecture is shown to hold.Connections between this conjecture and the existence of positive rational solutions of certain systems of homogeneous quadratic polynomial equations with each coefficient equal to either -1 or 1 are explored. Sign patterns that almost require unique rank are also investigated.
