[en] HYDROTHERMAL LIQUEFACTION OF MALT BAGASSE BIOMASS FOR BIO-OIL AND BIOCHAR PRODUCTION / [pt] LIQUEFAÇÃO HIDROTÉRMICA DA BIOMASSA DE BAGAÇO DE MALTE PARA A PRODUÇÃO DE BIO-ÓLEO E BIOCARVÃO

[pt] A necessidade mundial de energia tem aumentado exponencialmente, no entanto, as reservas de combustíveis fósseis, além de produzirem sérios impactos ambientais, estão se esgotando ao longo dos anos. Por estas razões, muitos estudos vêm sendo feitos na busca de novas fontes renováveis de energia, como o reaproveitamento de resíduos de biomassa. Desta forma, o objetivo deste trabalho é estudar o processo de liquefação hidrotérmica (HTL) do bagaço de malte, gerado ao final do processo cervejeiro, para a produção de bio-óleo e biocarvão como potenciais combustíveis renováveis. A caracterização inicial da biomassa apresentou significativa quantidade de celulose e hemicelulose, alto teor de umidade e pequeno tamanho de partícula, sendo ideal para o processo. A HTL foi conduzida em um reator sob alta pressão em diferentes faixas de temperatura e tempos de residência. Uma modelagem cinética e termodinâmica foi realizada para a etapa inicial da liquefação, apresentando 62,08 kJ.mol−1 de energia de ativação e caráter endotérmico. Bio-óleo apresentou melhor rendimento, 18,2 por cento, a 300 C e 30 min, já o biocarvão atingiu 21,0 por cento de rendimento a 250 C e 5 min. A pequena diferença de valores, ao longo do tempo, comprovou que a maior produtividade ocorre sempre em 5 min, sendo este o tempo ótimo de reação. A análise do poder calorífico superior (PCS) demonstrou que altas temperaturas elevam a energia produzida. Em 5 min e a 300 C, melhores condições de operação, a HTL gerou um bio-óleo com PCS de 33,6 MJ.kg−1, sendo 27,8 por cento inferior a gasolina e um biocarvão com 26,7 MJ.kg−1, sendo 11,4 por cento superior ao carvão tradicional. Através das caracterizações finais, foi possível observar alta degradação da estrutura lignocelulósica da biomassa e identificar os compostos presentes no bio-óleo, indicando que os produtos da HTL apresentam alto potencial de utilização como combustíveis renováveis. / [en] The global energy needs have increased exponentially; however, fossil fuel reserves, in addition to producing serious environmental impacts, are running out over the years. For these reasons, many studies have been done in the search for new renewable energy sources, such as the reuse of biomass wastes. In this way, the purposes of this study are associated with the hydrothermal liquefaction process (HTL) of the malt bagasse, generated at the end of the brewing process, for the production of bio-oil and biochar as potential renewable fuels. The initial biomass characterization presented a significant amount of cellulose and hemicellulose, high moisture content and small particle size, ideal for the process. The HTL was conducted in a high pressure reactor in different temperature ranges and residence times. A kinetic and thermodynamic modeling was performed for the initial stage of liquefaction, presenting 62.08 kJ.mol−1 of activation energy and endothermic behavior. Bio-oil presented a better yield, 18.2 percent, at 300 C and 30 min, while the biochar reached 21.0 percent yield at 250 C and 5 min. The small distintion between values, over time, proved that the highest productivity always occurs at 5 min, which is the optimal reaction time. The higher heating value analysis (HHV) showed that high temperatures increase the energy produced. At 5 min and 300 C, better operating conditions, HTL generated a bio-oil with HHV of 33.6 MJ.kg−1, with 27.8 percent less than gasoline and a biochar with 26.7 MJ.kg−1, being 11.4 percent higher than traditional coal. Through the final characterization, it was possible to observe high degradation of the lignocellulosic structure of the biomass and to identify the compounds present in the bio-oil, indicating that the HTL products present high potential for use as renewable fuels.

Technological Innovations for Mid-Atlantic Cropping Systems

Greater projected demand for food, fuel, and fiber will require substantial increases in global agricultural production over the next three decades. Climate change is also forecasted to make weather events more extreme and variable. Efficiency will become more important as demand for food products increases and the availability of fertilizer and land decreases. Technology may be of paramount importance for pushing the boundaries of production while remaining sustainable for generations to come. The first chapter of this dissertation investigated the importance of rate and timing of the plant growth regulator trinexapac-ethyl to malting barley in Virginia. Plant growth regulators can help plants remain upright during strong winds, thereby preserving grain quality and yield. However, this study demonstrated that risks of plant injury also exist. Application should be restricted to fields with greater risk of lodging and made only after the barley crop has broken dormancy and a substantial increase in air temperature is not forecasted in the week following application. Chapter two compared the efficacy of eight vegetation indices calculated from three satellites (Landsat 8, Sentinel 2, and Planet) for estimating cover crop biomass. Cover crops can have beneficial effects on agricultural land as well as groundwater and surface water, but only when adequate biomass is established to reduce erosion and nutrient leaching. Satellite imagery was able to estimate multi-species cover crop biomass more accurately than field-based sensors, although the most accurate vegetation index was dependent upon which satellite was being tested. Chapter three investigated the potential of Arabidopsis thaliana ipk1-, a loss-of-function mutant which exhibits decreased growth at elevated phosphorus concentration, for serving as in indicator of plant available phosphorus. An indicator crop could provide greater spatial resolution compared to soil testing, as well as represent plant available nutrients as opposed to chemically extracted nutrient estimations. Plant response exhibited a quadratic relationship with media P concentration in the range of fertilizer decision making for maize, providing valuable insight for potential yield response in agricultural fields below 'very high' phosphorus concentration. / Doctor of Philosophy / Climate change, increased demand for locally sourced ingredients, and elevated pressure for environmentally responsible practices will make meeting the growing demand for food difficult for farmers to achieve over the next few decades. Similar to many other industries, implementation of advanced technology may be necessary to keep up with agricultural demand. Plant growth regulators are one such technology which when applied to plants can cause them to remain short, decreasing the chance of blowing over during windstorms. However, chapter one of this dissertation concluded that risks of plant injury also exist when applying plant growth regulator on malting barley (for brewing or distilling). Application should be restricted to fields with greater risk of wind damage (e.g. taller barley) and made only after the barley crop begins spring growth and a decrease in air temperature is not forecasted in the week following application. Chapter two compared eight spectral vegetation indices across three satellites with different image resolution for their ability to estimate cover crop biomass. Cover crops protect groundwater and surface water quality, but only when adequate growth is achieved. Satellite imagery was able to estimate multi-species cover crop biomass more accurately than field-based sensors, although the most accurate vegetation index was dependent upon which satellite was being tested. Chapter three investigated the potential of Arabidopsis thaliana ipk1-, a loss-of-function mutant which exhibits decreased growth at elevated phosphorus concentration, as in indicator of plant available phosphorus in soil. An indicator crop could help determine which areas of a field are likely to have increased crop yield if fertilized and which are not. The mutant tested could be useful as an indicator crop given its response to phosphorus concentration, warranting further research with other plant species more appropriate for field use.

Lymphocytes T et vieillissement : lymphopénie ou redistribution ? / Lymphocytes T and Ageing : Lymphopenia or Redistribution ?

L’atteinte de l’âge sur les populations lymphocytaires T conventionnelles CD4 et CD8 avec l’avancée en âge est relativement bien décrite en périphérie lymphoïde secondaire chez la souris, et dans le sang périphérique chez l’homme. Deux paramètres sont observés : réduction du nombre de ces cellules et altération du ratio naïve/mémoire. À l’inverse, l’évaluation des tissus lymphoïdes tertiaires et des tissus extra lymphoïdes dans les réponses immunes, reste à affiner. Notre étude au cours du vieillissement physiologique du compartiment T fut menée dans des tissus lymphoïdes et non lymphoïdes de souris C57BL/6 wild-type, âgées entre 2 et 6 mois, entre 10 et 14 mois et entre 22 et 26 mois. Nous avons démontré que la lymphopénie T classiquement décrite liée au vieillissement dans les organes lymphoïdes secondaires ne s’applique pas à tout l’organisme : les compartiments intestinaux étudiés présentent une accumulation de cellules TCRαβ+ CD4+ (TCD4) et CD8+ (TCD8). Nos résultats dévoilent un impact différentiel du vieillissement sur le nombre absolu des différents compartiments cellulaires TCRαβ+ dans les organes lymphoïdes et la muqueuse intestinale. Ces résultats suggèrent donc que la lymphopénie T décrite dans les organes lymphoïdes s’établissant au cours du vieillissement pourrait être essentiellement liée à une redistribution des lymphocytes. A l’inverse, la persistance des cellules T régulatrices dans les organes lymphoïdes secondaires pourrait être liée à une production locale dans la muqueuse intestinale. Il semble donc que l’équilibre TCD8/TCD4 peut être différemment affecté selon le site considéré et cette observation peut fournir une justification pour la plus grande susceptibilité aux infections observée avec l’âge. / Consequences of ageing on conventional CD4 and CD8 T lymphocytes populations is relatively well described in murine secondary lymphoid organs and in human peripheral blood: reduction the number of these cells and alteration of naïve/effector-memory ratio in favour of effector-memory cells. Conversely, evaluation in tertiary lymphoid tissues and non-lymphoid tissues remains to be refined. We conducted an exhaustive analysis of T cell compartments during physiological aging in lymphoid and non-lymphoid tissues isolated from wild-type C57BL/6 mice aged of 2 to 6 months, 10 to 14 months and 22 to 26 months. We demonstrated that T lymphopenia described classically associated with aging in the secondary lymphoid organs does not apply to the whole organism: intestinal compartments studied show an accumulation of TCRαβ+ CD4+ cells (TCD4) and CD8+ (TCD8). Our results reveal a differential impact of aging on the absolute number of different TCRαβ+ cellular compartments in lymphoid organs and intestinal mucosa. T cell lymphopenia in secondary lymphoid organs currently associated to ageing may essentially reflect T cell redistribution. TCD8/TCD4 balance may be affected differently depending on the site considered and this observation may provide a rationale for the greater susceptibility to infection observed with age.

The first part deals with general description of all production stages of malting and brief description about the Research Institute of Brewering and Malting. It also generally characterizes the used hardware and software for automation control system. The second part of this thesis describes the types of programmable logic controllers and the selection of optimal variant for application of the operationally experimental malt-house. Master thesis in the third and fourth part contains a brief description about the methodology and the creation of control program and visualisation for automatic control system of the operationally experimental malt-house. The output of the master thesis is a complete control program and visualisation of the controlled application of the operating malt-house.

Nowadays there are hundreds of breweries scattered throughout Bohemia, Moravia and Silesia, waiting for their new future. They are either abandoned or in private property. In the worst case they have been deteriorating with no hope of recovery. There are not many left. It is very difficult to find a new function and especially enlightened investor. Important thing is how we will treat with these structures in the future. Will we use this unique potential, or leave this place for a new function? Which way we can choose? What is the potential of these buildings and what they need? This thesis attempts to briefly describe and demonstrate the value of brewery architecture which due to dramatic changes in society became endangered. And to offer possible solutions for the preservation this unique buildings their charm and atmosphere for the next generations.

Stanovení thiaminu v ječmeni, sladu a potravních doplňcích / HPLC determination of thiamine in barley, malt and food supplements

The aim of this diploma thesis is focused on determination of B vitamin in food especially in malting barley and malt. This diploma thesis is consists of theoretical and experimental parts including the conclusion and supplements. Theoretical part describes thiamine characteristic, chemical structure, chemical and physical properties, function of thiamin in organism, impact it´s insufficiency to the human health, occurrence in common food and methods for it´s determination. Further are describing theoretical principles of high-efficiency liquid chromatography, chromatography on reverse phase, derivatization and ways of validation. In the Experimental section is detailed description of used method for determination of thiamine in barley and malt, fresh mash and beer but also in common food supplements as are B-kompex, Pangamin and Thiamin. Determination was made by high-efficiency liquid chromatography on reverse phase with isocratic elution and fluorescent detection. Next part introduce the results of theses, discussion of experimental work, volume of thiamine in analyzed food and validation parameters of used method. In the conclusion are valorized results of experimental work. In the attachment are graphs of standard curves, chromatogram and calculations of validation parameters. Diploma thesis was realized in laboratory of Chemistry and Technology of Foodstuff, Faculty of chemistry, Brno University of Technology.

Závislost kvalitativních ukazatelů sladu na teplotě rmutování při přípravě sladiny / Dependence of quality parameters of malt on the temperature of mashing at sweet wort preparation

The Masters thesis deals with dependence of quality parameters of malt on the temperature of mashing at sweet wort preparation. The theoretical part focuses on the technology of malt production including description of ingredients, production parts, some kinds of malt and the quality parameters. There is also described the technology of sweet wort preparation and the methods of determination of the malt quality parameters in the sweet wort there. The experimental part focuses on determination of malt extract, viscosity, haze, colour, time of starch cleavage, pH, degree of fermentation and beta-glucans content. Based on temperature dependence of parameters, the individual samples were compared and the effects of temperature were discussed. The biggest extract of light and caramel malts was explored at 70 °C, of coloured malt at 50 °C. The lowest viscosity of light and coloured malts was explored at 65 °C, of caramel malt at 50 °C. The lowest haze of wort was at 65 °C. Ideal colour of all malts was at all mashing temperatures. Ideal pH of all malts was explored at 70 °C. Ideal time of starch cleavage was explored at 70-85 °C. The biggest achievable degree of fermentation and the hugest increase of beta-glucans content were explored at 65 °C.

Stanovení netěkavých N-nitrosaminů ve sladu / Determination of non-volatile N-nitrosamines in malt

The aim of diploma thesis has been development and optimization of method determination of N- nitrososarcosine and N-nitrosoproline in malt by gas chromatography with chemiluminiscence detector. Optimization of extraction method has been performed by response surface method. Quantification has been performed by internal standard method (N-nitrosopipecolic acid, in which matrix effects has been studied. These has been verified in münchen and pilsen malt, therefore matrix-matched calibration has been constructed. The developed method has been aplicated on wheat, münchen and pilsen malt. N- nitrosoproline was detected only in münchen malt and in other cases has been under limit of detection (LOD=4,0 µg/kg). N-nitrososarcosine was in all cases under limit of detection (LOD=3,7 µg/kg). [pt] Resíduos de biomassas são de grande interesse por serem matérias primas para a geração de energia renovável. Neste trabalho foram estudados os processos de conversão térmica de pirólise lenta e de carbonização hidrotermal, empregando os resíduos de bagaço de cana-de-açúcar, bagaço de malte e casca de coco. A cinética e a termodinâmica das reações foram determinadas experimentalmente pelo cálculo da energia de ativação, fator de frequência, energia livre de Gibbs, entalpia e entropia do complexo ativado. A pirólise foi avaliada empregando a termogravimetria em atmosfera inerte, para uma faixa de temperatura do ambiente até 1.000 graus Celsius e aplicando diferentes taxas de aquecimento. A carbonização hidrotermal procedeu em um reator tipo autoclave Parr modelo 452HC2, em diferentes temperaturas e tempos de operação. Modelos cinéticos da pirólise para métodos Model-free, denominados Kissinger, Flynn- WallOzawa (FWO) e Kissinger-Akahira-Sunose (KAS), e Model-fitting, intitulado Coats-Redfern, foram investigados. Para os modelos KAS, FWO e Coats-Redfern foram analisados 19 tipos de mecanismos reacionais e respectivos coeficientes de determinação (R2) foram determinados. O bagaço de cana-de-açúcar se ajustou ao método de Kissinger (R2 de 0,9973) e ao Coats-Redfern, entretanto os métodos FWO e KAS não se adequam a este material. Os testes com bagaço de malte e casca de coco se ajustaram a todos os métodos aplicados, apresentando valores de R2 elevados (0,9 a 0,9999). O modelo para a cinética da carbonização hidrotermal foi aplicado, onde as três biomassas apresentaram a ordem variando de 3 a 3,49, valor compatível com a literatura. Adicionalmente foram feitas caracterizações físico-químicas nos resíduos de biomassa e nos biocarvões produzidos na carbonização hidrotermal, incluindo análise elementar e imediata, microscopia eletrônica de varredura, espectroscopia de infravermelho, densidade aparente, pH, condutividade e poder calorífico. / [en] Biomass residues are of great interest because they are raw materials for the generation of renewable energy. In this work, the processes of thermal conversion of slow pyrolysis and hydrothermal carbonization were studied using the residues of sugarcane bagasse, malt bagasse and green coconut shell. The kinetics and thermodynamics of the reactions were determined experimentally by calculating the activation energy, frequency factor, Gibbs free energy, enthalpy and entropy of the activated complex. Pyrolysis was evaluated using thermogravimetry in an inert atmosphere, from a room temperature up to 1,000 Celsius and applying different heating rates. Hydrothermal carbonization was carried out in a Parr autoclave type 452HC2 reactor, at different temperatures and times of operation. Kinetic models of pyrolysis for Model-free methods, called Kissinger, Flynn-WallOzawa (FWO) and Kissinger-Akahira-Sunose (KAS), and Model-fitting, entitled Coats-Redfern, were investigated. For the KAS, FWO and Coats-Redfern models, 19 types of reaction mechanisms were analyzed and their determination coefficients (R2) were evaluated. Sugarcane bagasse was adjusted to the Kissinger method (R2 equal to 0.9973) and Coats-Redfern, however the FWO and KAS methods are not suitable for this material. The tests with malt bagasse and coconut fiber were adjusted to all applied methods, showing high R2 values (0.9 to 0.9999). The model for the kinetics of hydrothermal carbonization was applied, where the three biomasses presented the order varying from 3 to 3.49, a value compatible with the literature. In addition, physical-chemical characterizations were carried out for the biomass residues and biochar produced by hydrothermal carbonization, including elementary and immediate analysis, scanning electron microscopy, infrared spectroscopy, apparent density, pH, conductivity and calorific value.

Kilning invokes oxidative changes in malt proteins

Beneath glycation, oxidation reactions may take place at cereal proteins during production of malt. The extent of oxidative chemical changes at malt proteins has not yet been studied. In the present short communication, malt protein was characterized by the determination of free thiol groups and degree of methionine oxidation as well as the sites that are reactive to covalent modification by 2,4-dinitrophenylhydrazine (DNPH, “protein carbonylation”). Protein carbonylation in pale malts was around 1.5 nmol/mg protein and increased with increasing malt colour. Investigations on the protein pellet isolated for determination of carbonylation revealed that solubility and colour may disturb the quantification of carbonyl sites in roasted malts. Free thiols decreased with increasing malt colour already in pale malts (EBC < 10). The formation of methionine sulfoxide (MetSO) was intensified with increasing malt colour. An amount of 7–20% of methionine was converted to MetSO in pale and dark malt, whereas nearly 60% of methionine was oxidized to MetSO in roasted malts. The formation of methionine sulfone was negligible. This study shows that malt proteins suffer from oxidation during kilning, and future studies will have to show whether this supports the pro- or antioxidant activity of malt.
