The synthesis and characterisation of Langmuir-Blodgett film forming TCNQ adducts

Substitution reactions of TCNQ (7,7,8,8-tetracyanoquinodimethane) with suitable electron donor moieties extended the range of gamma-bridged adducts R(4)Q3CNQ and R(4)P3CNQ of which the short chain Z-beta-(l-butyl-4'-quinolinium)-alpha-cyano-4-styryldicyanomethanide (C[4]H[9](4)Q3CNQ) is a typical example. These zwitterionic D-pi-A materials (where D and A are electron donors and acceptors respectively) exhibit properties such as solvatochromism, molecular rectification and second harmonic generation. Further synthetic work concentrated on modifying the donor, the acceptor and the substitution position within these zwitterions to create a series of diverse materials for non-linear optic research. Modification of the picolinium and quinolinium systems where the TCNQ substitution is in the alpha-position, has created the extensive analogous series-R(2)Q3CNQ and R(2)P3CNQ, their properties being compared and contrasted to the original gamma-bridged adducts. The behaviour of the materials on the subphase and their resultant Langmuir-Blodgett (LB) film fabrication was studied. The abrupt change in molecular orientation in LB films of the gamma-bridged adducts occurring at R= C[15]H[31] and above was not seen in the more flatly orientated alpha-bridged adducts. Further characterisation of the LB films was performed using reflectance-absorption infrared spectroscopy (RAIRS) and surface plasmon resonance (SPR).A number of N-alkylpyridinium benzimidazolate betaine derivatives and the related betaine-TCNQ adducts were prepared and their LB film forming properties were studied.

Uma das propriedades interessantes de tal vidro é o elevado índice de refração. De acordo com estudos realizados, o incremento de óxidos modificadores de rede nos vidros teluritos provoca o aumento do índice de refração em tal material, devido à mudanças estruturais ocorridas. Com esse intuito, sintetizamos amostras de vidros teluritos com nióbio, de composição 80TeO2-(20-x)Li2O-xNb2O5 (x = 0, 5, 10 e 15). Para caracterização das amostras foram realizadas medidas de espectroscopia na região do infravermelho (FTIR) e UV-Vis, espectroscopia Raman e Difratometria de Raios-X. Foram realizadas medidas de índice de refração das amostras para verificar a influência da concentração de óxido de nióbio acerca deste índice. A difratometria de Raios-X confirmou o estado amorfo das amostras. Os estudos de caracterização (por espectroscopia no infravermelho e Raman) mostraram que a inserção de óxido de nióbio deslocou, para maiores freqüências, as bandas de absorção características dos grupos TeO4 e NbO4 provocadas por mudanças estruturais e pela incorporação de oxigênios não-ligantes. A espectroscopia na região ultravioleta-visível mostrou uma tendência no aumento do comprimento de onda de corte com o acréscimo de óxido de nióbio, o qual pode estar associado a alguma mudança estrutural. As medidas de índice de refração mostraram que, de acordo com o incremento de óxidos modificadores, este índice aumentou. Tal fato se deve à transição de oxigênios nãoligantes para ligantes e à polarização das ligações Te–O dos grupos TeO4 e ligações Nb–O dos grupos NbO4, pois essas ligações apresentam maiores momentos de dipolo elétrico que as demais. / Tellurite glasses are important materials which have interesting optical properties application in photonics mainly due to its high refractive index. According to studies, the increase of network modifier oxides in tellurite glasses can increase the refractive index in such material, due to structural changes. For this purpose, samples with niobium tellurite glasses of composition 80TeO2-(20-x)Li2O-xNb2O5 (x = 0, 5, 10 and 15) were synthesized and characterized by (FTIR) and UV-Vis spectroscopy, Raman spectroscopy and X-ray diffraction. The influence of niobium oxide concentration to the refractive index was also investigated. The characterization studies (infrared spectroscopy and Raman) showed that the insertion of niobium oxide shifted the characteristic absorption bands of groups TeO4 and NbO4 to higher frequencies. The structural changes are due to incorporation of non-bridging oxygen. The UV-Vis spectra showed a trend of increasing the wavelength of UV-Vis cut-off with the addition of niobium oxide and this increase may be linked to structural changes. The X-ray diffraction confirmed the amorphous state of samples. The measurement of refractive index showed that, in accordance with the increment of oxide modifiers, this index increased. This is due to the transition from non-bridging oxygen to bridging oxygen and the polarization of the Te-O bonds of the groups TeO4 and links Nb-O of the NbO4 groups because these bonds have higher dipole moments than the others.

Análise por modelagem e dinâmica molecular da interação entre a integrina α6β1 e a laminina 111 humana / Molecular modelling and dynamics analisys of human α6β1 integrin and laminin 111 interaction

A matriz extracelular (ECM) é definida como um complexo de proteínas e glicoproteínas que envolve as células nos mais diversos tecidos. A laminina é uma glicoproteína que é formada por três cadeias polipeptídicas dispostas em cruz. Sua função é a de ancorar as células epiteliais à ECM, da qual faz parte, através de associações a outras proteínas como as integrinas, o colágeno, a elastina e a fibronectina. As integrinas são receptores de adesão localizadas na superfície celular, que medeiam a interação célula-ECM. Estas interações são cruciais para diversos processos biológicos, tais como a diferenciação celular, a transdução de sinais, a resposta imunológica, a cicatrização de ferimentos e a formação de metástases. Porém a forma estrutural com que a interação entre a integrina e a laminina ocorre ainda não foi esclarecida. Neste contexto este trabalho visa analisar, em escala molecular, a forma com que a integrina α6β1 e a laminina 111 humanas interagem. Assim, foram conduzidos vários estudos, entre eles: i) alinhamentos estruturais e seqüenciais das regiões β-propeller e βA de integrinas não-possuidoras de domínio I; ii) construção do modelo das regiões β-propeller e βA da integrina α6β1 em complexo com pequenos inibidores peptídicos do tipo ECD ou RGD; iii) alinhamento entre os domínios LG de lamininas; iv) construção do modelo do domínio LG1 da laminina 111; e v) construção do primeiro modelo descrito do complexo formado pela porção N- terminal da integrina α6β1 e o domínio LG1 da laminina 111. Para tanto, foram aplicadas as técnicas de modelagem por homologia e dinâmica molecular, além de alinhamentos entre as cadeias α e β de integrinas, e dos domínios LG de lamininas. Inicialmente os resultados mostraram que o loop que corresponde à região entre os subdomínios D2 e D3 da cadeia α6 discrimina ligantes por interações eletrostáticas, e a partir disso, que a integrina α6β1 possui interação preferencialmente com peptídeos do tipo ECDF. Foi mostrado que o domínio LG1 de laminina 111 interage com a integrina α6β1 pelo contato da fita β H com o β-propeller de α6. Além disso, por seu caráter eletrostático e proximidade à fita β H, o resíduo Asp82 de LG1 se adere ao íon Mg+2 do MIDAS da integrina α6β1, e que esta interação é indispensável à ligação entre as duas proteínas. / The extracelullar matrix (ECM) is formed by an assembly of proteins and glycoproteins which surrounds the cells, in various tissues. The laminin is a glycoprotein localized in the ECM that consists of three polypeptidic chains cross- shaped. It functions by anchoring epithelial cells to basal lamin, through associations with integrins, collagen, elastin and fibronectin. Integrins are adhesion receptors localized on cellular surface, that mediate interactions between cells and ECM. The interactions between proteins of ECM and cellular proteins are crucial for many normal biological processes such as cell differentiation, signal transduction, immune responses, wound healing and metastasis formation. The nature of interactions between laminin and integrin has not been fully identified yet. The present work aims to analyze, in an molecular scale, the interaction between human α6β1 integrin and laminin 111. In this work we conducted many studies, including: i) structural and sequential alignments of β-propeller and βA regions in lacking I-domain integrins; ii) model building of β-propeller e βA regions of α6β1 integrin complexed with small ECD or RGD peptide antagonists; iii) sequential alignment of various laminin LG domains; iv) model building of laminin 111 LG1 domain; and v) model building of the first described complex between the N-terminal portion of α6β1 integrin and laminin 111 LG1 domain. In order to do this, homology modeling and molecular dynamics techniques were applied, together with alignments between integrins α and β chains, and laminin LG domains. Our initial results show that the loop between blades 3 and 4 of α6 integrin subunit discriminates ligands by electrostatic interactions. Therefore we assumed that α6β1 integrin preferentially interacts with ECDF based peptides. It was demonstrated that the laminin 111 LG1 domain interacts with α6β1 integrin by the contact of this H β strand and α6 β-propeller. Further, by its electrostatic function and proximity to H β strand, LG1 residue Asp82 adheres to Mg+2 containing MIDAS in α6β1 integrin. This interaction appears to be indispensable for α6β1 and laminin 111 binding.
