DNA Molecules Stretching in Torus-type MicrochannelsLin, Ci-jie 05 August 2010 (has links)
In this study, we design different inscribed/circumscribed circular torus-type microchannels to investigate the stretching behavior of DNA molecules. Strain rate and relaxation time play an important role in DNA stretching. In order to perform an analysis of the coil-stretch transition of DNA, we develop a method of stretching DNA molecules by using £gPIV and CLSM measurements. £gPIV is designed to measure the velocity distribution, after which the local strain rate can be estimated. The hydrodynamic stretching of DNA molecules in the elongation flow is observed using a confocal laser scanning microscope (CLSM). The relaxation time of the DNA molecules is then estimated according to the CLSM images analysis. At present, our experiments using the electro-osmotic flow (EOF) driven at various electric fields and viscosities to stretch DNA molecules show how one can investigate the influence of hydrodynamic interactions in the case of stretching of DNA molecules.

The First Hyperpolarizability of Charge-Transfer Molecules Stuidied by Hyper-Rayleigh ScatteringTai, Yung-Hui 26 July 2000 (has links)
Abstract
The first hyperpolarizability(£]) of five charge-transfer molecules are determined using the hyper-Rayleigh scattering (HRS) technique at two excitation wavelengths : 1064nm and 1907nm. The 1064nm excitation wavelength is derived from a Nd: YAG pulsed laser, and the 1907nm excitation wavelength is obtained by shifting the 1064nm laser light by stimulated Raman scattering of pressurized H2 gas. Four of the five samples contains thiophene and thiazole are synthetized by Prof. Shu Ching-Fong at the National Chiao Tung University (NCTU) and the other sample is synthetized by Prof. Hong, Jin-Long at the National Sun Yat-sen University (NSYSU). The measured £] values are used to calculate the intrinsic molecular hyperpolarizabilities using the two-level model previously developed by Oudar and Chemla. The results are related to molecular structure.

Cadherin-Based Adhesion Molecules for Classification of Melanoma with Aqua TechnologyGraff, Gretchen Melaine 14 February 2008 (has links)
Cadherin and catenin-family proteins regulate adhesion in malignant melanoma. Using AQUA (Automated Quantitative Analysis) to quantitate the levels of alpha-catenin, beta-catenin, p120-catenin, N-cadherin, E-cadherin, and P-cadherin in melanoma on tissue microarrays (TMAs), we classified 513 patients by protein expression using hierarchical clustering and regression analysis. The dendrogram supported positive correlations seen upon Spearman rho analysis of P-cadherin and beta-catenin (r=0.5238, p<0.0001) and negative, weak association of N-cadherin with other markers. Patients with high expression of N-cadherin had the highest 20-year survival rate (p=0.0003). Our adherens protein molecular classification of melanoma defines at least two distinctive sub-populations of melanoma patients, those with high expression of N-cadherin and those with low expression who have decreased survival. These findings extend previous cDNA array-based findings of an epithelioid class and neural crest class of melanomas.
