Abstract The dissertation addresses the issue of architecture in municipal housing built between the two wars in the town of Ústí nad Labem. From mid-nineteenth century the North Bohemian industrial agglomeration, populated mostly by Germans, experienced unprecedented urban and demographic growth. Thus, between 1919 and 1938, the municipality developed a comprehensive project of approximately one hundred buildings. The thesis identifies relations and balanced proportions between a formal analysis of the individual stages of development and a study of their broader historical context. The method of art-historical analysis is applied, whereby the projects are chronologically classified and their urban, typological and chronological aspects are assayed and assessed. The thesis also focuses on the identity of the author, the architect of German origin Franz J. Arnold. The architecture of the Ústí municipal housing complex was designed as a concretization of the social democratic political programme and of the state housing policy, it carries therefore a contents of political significance. Background: The tradition of specialised institutional alcohol treatment in what is now the Czech Republic dates back over a hundred years. The first modern institution aimed at treating alcohol dependency began to operate on 7 September 1948. While formally constituting an organisational unit of the Psychiatric Clinic, the "U Apolináře" facility, headed by Dr. Jaroslav Skála, was an independent workplace which gave rise to a distinct treatment approach. Becoming known as the "Apolinar" treatment model, this approach was adopted by all the residential alcohol treatment facilities which came into existence or operated in Czechoslovakia from 1948 to 1989. Before the establishment of this department, three similar treatment facilities existed on the historical territory of Czechoslovakia - Velké Kunčice (1911 to 1915), Tuchlov (1923 to 1938), and Istebné nad Oravou (1937 to 1939/1949). No / NAD(P)H:quinone oxidoreductase-1 (NQO1) is a potential target for therapeutic intervention but attempts to exploit NQO1 using quinone-based bioreductive prodrugs have been largely compromised by toxicity to organs that inherently express high levels of NQO1. In an attempt to circumvent this problem, this study describes the development of a tripartite quinone-based drug delivery system, the ultimate objective of which is to release a targeted therapeutic agent following the reduction of a quinone "trigger" by NQO1. Molecular modeling of drug/NQO1 interactions were conducted prior to the synthesis of N-{4-[bis-(2-chloroethyl)-amino]-phenyl}-beta,beta,2,4,5-pentamethyl-3,6-dioxo-1,4-cyclohexadiene-1-propanamide (prodrug 1). Prodrug 1 is a good substrate for purified NQO1 (V(max) and K(m) values of 11.86 +/- 3.09 micromol/min/mg and 2.70 +/- 1.14 micromol/L, respectively) and liquid chromatography-mass spectrometry analysis of the metabolites generated showed that lactone 3 and aniline mustard 4 were generated in a time- and NQO1-dependent manner. Chemosensitivity studies showed that prodrug 1 is selectively toxic to cells that overexpress NQO1 under aerobic conditions, and comet assay analysis confirmed the presence of elevated interstrand cross-links in NQO1-rich compared with NQO1-deficient cells. Hypoxic sensitization (hypoxic cytotoxicity ratio = 15.8) was observed in T47D cells that overexpress cytochrome P450 reductase. In conclusion, the results of this study provide mechanistic proof of principle that a tripartite benzoquinone drug delivery system is enzymatically reduced to release an active therapeutic agent. Further development of this concept to fine-tune substrate specificity for specific reductases and/or the inclusion of alternative therapeutic agents is warranted.

The dissertation is dealing with work of Viennese architect and builder from the first half of the 18th century Antonio Ehorto Martinelli in the Schwarzenberk's service in the years 1722-1732. It contains an index of buildings, that Martinelli was working on for Schwarzenberks, including contentious and mistakenly ascribed buildings. The collected information is based on study of extant archival collections, on field research of separate buildings and on study of the relevant literature. The main goal was an evaluation of separate Martinelli's architectural fruitions relating to history of art, and furthuremore an attempt to compare them both within the bounds of the whole architect's work and within the bounds of contemporary Czech and Austrian architecture. The knowledge of work of A. E. Martinelli in the Shwarzenberk's service was markedly extended on the occasion of dealing with the settled topic and moreover a few little and even a few bigger new pieces of knowledge appeared, e.g. information about concerned gangers, building development of the studied objects out of the settled period etc.
