QSPR/SAR em derivados 5-nitro-heterocíclicos com atividades antichagásica. Estudo das relações entre o potencial de redução do grupo nitro e propriedades físico-químicas / QSPR/SAR in 5-nitro-heterocyclic derivatives with antichagasic activity. Study of relationships between the reduction potential of nitro group and the physicochemical properties

Alguns nitrocompostos apresentam atividade antichagásica resultante do processo de redução do grupo nitro com conseqüente formação de radical nitro ânion e de intermediários tóxicos ao parasita. A variação estrutural dos derivados 5-nitroheterocíclicos pode interferir no processo de redução destes compostos e, também, na atividade biológica. Neste aspecto, o estudo da redução dos nitrocompostos por meio de técnicas voltamétricas, como a voltametria cíclica e a voltametria de onda quadrada é uma forma de avaliar ou até simular o mecanismo de ação destes derivados. Assim, a avaliação de processos eletródicos e a determinação dos potenciais de redução dos nitrocompostos fornecem subsídios para a compreensão do mecanismo da atividade antiparasitária. Neste trabalho, desenvolveram-se procedimentos eletroquímicos utilizados para a determinação do potencial de redução e do potencial de meia onda de vinte e três nitrocompostos (5-nitro-2-tiofilideno 4-R-benzidrazidas e 5-nitro-2-furfurilideno 4-R-benzidrazidas), visando o emprego destes em estudos de QSPR e SAR. As determinações foram realizadas empregando-se Voltametria Cíclica e Voltametria de Onda Quadrada em célula eletroquímica de 10 mL nos meios prótico (solução tampão PIPES com eletrólito de suporte NaNO3 0,1 mol L-1), misto de DMSO/solução tampão PIPES (50% v/v) e em meio aprótico de DMSO (com eletrólito de suporte Bu4NH4BF4). O eletrodos de referência e auxiliar utilizados foram Ag/AgCl saturado e platina. Os eletrodos de trabalho utilizados foram o carbono vítreo (meio aprótico e misto), pasta de carbono (meio misto e prótico) e pasta de carbono modificada com nitrocompostos (meio prótico). Foram realizados estudos teóricos, de modelagem molecular, e experimental, espectrofotometria UV/visível, com o objetivo de determinar a conformação de menor energia mínima global e confirmar a ocorrência de disposição estrutural co-planar. No estudo teórico, utilizou-se as metodologias de otimização de geometria, e análise conformacional dos nitrocompostos calculadas por meio de AM1 e HF3-21G* e HF6- 31G* para obtenção dos confôrmeros de menor energia mínima. Estes confôrmeros foram submetidos a cálculo de carga de ponto único (AM1), onde determinou-se os valores de energia de diversas propriedades de caráter eletrônico (energias de formação em vácuo e meio solvatado, de HOMO, de LUMO, momento de dipolo, dureza, potencial químico, afinidade eletrônica, potencial de ionização, cargas de potencial eletrostático), geométrica (volume molecular) e mapas 3D de densidade de potencial eletrostático, de HOMO e de LUMO. Todos os cálculos foram realizados empregando-se os pacotes computacionais Spartan O2 for Linux, Spartan O4 for Windows e ClogP 4.0 (para determinar os valores de hidrofobicidade). Os valores do potencial de pico catódico (Epc 1) e de meia onda bem como as propriedades físico-químicas, obtidos a partir de cálculos de química quântica, de σp e σR, ClogP e π) foram utilizados em análise de QSPR aplicando-se a Análise de Hansch e regressão multivariada, PLS. Avaliou-se ainda, a atividade antichagásica de dez nitrocompostos (oito derivados tiofilidênicos e dois derivados fufurilidênicos) visando investigar a possibilidade de influência do processo de redução dos nitrocompostos sobre a atividade biológica. Realizaram-se ensaios que permitiram avaliar o efeito antiproliferativo dos nitrocompostos sobre o parasita, em 24 horas, por meio de contagem do número de unidades viáveis em câmara de Neubauer. Os métodos voltamétricos utilizados mostraram-se adequados para a avaliação da redução eletroquímica dos nitrocompostos, e foram apropriados para a obtenção dos potenciais de redução de pico catódico e dos potenciais de meia onda de todos os derivados em meio aprótico e misto. Em meio aquoso, obtiveram-se os valores dos potenciais de redução de pico catódico apenas para os derivados tiofilidênicos. A partir dos resultados obtidos na análise conformacional, observou-se a quebra de efeito co-planar estrutural na região do anel benzênico substituído e porção hidrazídica molecular. Adicionalmente, nos mapas de densidade de potencial eletrostático, registrou-se a ocorrência de efeito mesomérico conjugado reduzido entre as porções moleculares citadas acima. De posse destes dados, em conjunto com as visualizações de sinais característicos para grupos cromóforos distintos em espectros UV/visível, sugere-se a ausência de efeito conjugado molecular e, como conseqüência, de influência dos grupos substituintes em posição para do anel benzênico sobre o grupo nitro ligado ao anel heterocíclico. Nos resultados obtidos a partir das análises de QSPR, observou-se que não ocorre correlação entre as propriedades físico-químicas, determinadas para os nitrocompostos, e os descritores eletroquímicos, obtidos em diferentes meios de reação. Diante deste fato, sugere-se que a quebra do efeito co-planar interfere diretamente na intensidade da influência das propriedades físico-químicas de grupos substituintes sobre o potencial de redução dos nitrocompostos. Observou-se, também, que a maioria dos compostos apresentou atividade antichagásica superior ao fármaco de referência, o benznidazol. Verificou-se que os compostos não substituído e cloro derivado da série dos 5-nitro-2-furfurilidênicos são mais ativos que os correspondentes análogos das 5-nitro-2-tiofilideno benzidrazidas. Em análise preliminar, observou-se que o potencial de redução e o potencial de meia onda não exercem influência sobre a atividade antichagásica dos nitrocompostos. / Some nitrocompounds have show activity against to Trypanosoma cruzi. Those biological activity are resulting from nitro group reduction with subsequent formation of radical nitro anion and other reaction\'s intermediates, which are toxic to the parasite. The structural difference among the 5-nitro-heterocyclic derivatives might exert influence on these compounds reduction and also on the antichagasic activity. In this way, the study of reduction process by voltammetric techniques, such as cyclic voltammetry and square wave voltammetry, is a mode of evaluate the biological action mechanism. Therefore, the evaluation of the electrodic process and the determination of nitrocompounds reduction potential provide information for elucidating the antitrypanosomal activity mechanism. In the present work, the procedures employed to determinate the cathodic reduction and reduction potentials were carried out on twenty three nitrocompounds (5-nitro-2-tiofilidenic benzhidrazides and 5-nitro-2-furfurilidenic benzhidrazides) with the aim using those data in QSPR and SAR analysis. Cyclic voltammetry and square wave voltammetry techniques were used to determined the reduction potential in electrochemical cell of 10 mL in the protic media (PIPES buffer and electrolyte of support NaNO3 0.1 mol L-1), mixed media of DMSO and PIPES buffer 50:50 v/v (electrolyte of support NaNO3 0.1 mol L-1), and aprotic media of DMSO (electrolyte of support Bu4NH4BF4 0.1 mol L-1). The reference and counter electrodes used were Ag/AgCl (with KCl saturated) and platinum, respectively. In these studies, the work electrodes were glassy carbon (aprotic and mixed media), carbon paste (mixed and protic media) and carbon paste modified with nitrocompounds (protic media). Molecular modeling studies and UV/visible spectrophotometry were investigated with the aim for determining the lowest conformational energy and the occurrence of co-planarity molecular structure. In this work were performed the geometry optimization (using the quantum chemistry AM1, ab initio HF3-21G* and HF6-31G* level of theory), conformational analysis(AM1 and ab initio HF6-31G*) and single point calculations (AM1). All the lowest energy conformers from conformational analysis were submitted to single point calculations. These models were used to determine the physicochemical properties (energy of formation in vacuum and solvated media, energy of HOMO, energy of LUMO, dipole moment, hardness, chemistry potential, electronic affinity, ionization potential and charges of electrostatic potential) and the molecular electrostatic potential, HOMO and LUMO maps, respectively. All calculations were performed using the computational software Spartan O2 for Linux, Spartan O4 for Windows, and the ClogP 4.0 (only for hydrophobicity). The cathodic reduction and half-wave potentials values and the set of physicochemical properties, which are obtained from quantum chemistry calculations, as well as the electronic effect (σp and σR derived of σ Hammett) and the hydrophobicity property (ClogP, and π of Hansch) were used in QSPR analysis applying Hansch analysis and PLS methodologies. Afterwards, the antichagasic activity of ten compounds (eight thiofilidenic and two furfurylidenic derivatives) was evaluated considering the influence of nitrocompounds reduction on biological activity. This assay allows to analyse the antiproliferative effect of nitrocompounds on the parasites growing, in twenty-four (24) hours, reading the number of trypanosomes in Haemocytometer. The voltammetric methodologies allowed to determinate the cathodic reduction and half-wave potential values in aprotic and mixed media to all nitrocompounds. In protic media, however, the cathodic reduction potential values were obtained only for the 5-nitro-2-tiofilidenic benzhidrazides. Those procedures seen to be appropriated for perform the electrochemistry reduction evaluation. The geometry optimization and conformational analysis allows determining a lack of structural planarity in all derivatives located at the bond between the carbonilic carbon and benzhoyl group. Additionally, the electrostatic potential maps presented a decreased on the electronic conjugate effect of the structures of nitrocompounds investigated. The conformational analysis allows us to determine the nitroheterocyclic biosostere conformations dependence on the reaction phase studied. The UV/visible spectra presented two waves, which are indicating responses related to different chromophores. These results suggest the absence of conjugate effect in nitrocompound structure, indicating that the presence of groups attached to benzene ring do not exert influence on the nitro group attached to heterocyclic ring. In the QSPR analysis was not detected any correlations between thel physicochemical properties and the cathodic reduction and reduction potentials, in all reaction media investigated. Those results might be occurred due to the lack of molecular co-planarity, which is reducing the influence of any property on the nitrocompounds reduction. Almost all nitrocompounds investigated show higher antichagasic activity than the reference drug, benznidazol. In a preliminary analysis it was verified that the cathodic reduction and half-wave potentials do not exert influence on the nitrocompounds antichagasic activity.

A incidência de infecções hospitalares tem aumentado substancialmente, resultando no aumento de morbidade e mortalidade nas últimas décadas. Embora as infecções hospitalares estejam intimamente ligadas à assepsia e à higiene do ambiente nosocomial, não se pode deixar de considerar a susceptibilidade já instaurada dos doentes acometidos por estas infecções. Os avanços tecnológicos relacionados à procedimentos invasivos, diagnósticos ou terapêuticos, ajudaram na disseminação das infecções microbianas. Em decorrência desta evolução, surgiram micro-organismos multirresistentes ao atual arsenal de agentes farmacológicos utilizados rotineiramente na quimioterapia, tornando estas infecções grave problema de Saúde Pública. A necessidade de pesquisar novas alternativas para o combate destes micro-organismos é evidente, sendo que derivados nitro-heterocíclicos apresentam-se como alternativa bastante viável para estudos de novos compostos com potencial atividade antimicrobiana. Baseado nestes fatos o presente trabalho propõe o planejamento, síntese e avaliação da atividade antimicrobiana de derivados tiofilidênicos com estruturas azometínica e oxadiazolínica frente a micro-organismos responsáveis por algumas das principais infecções hospitalares instauradas em hospitais em todo o mundo. A escolha dos grupos substituintes dos compostos planejados foi baseada no digrama de Craig, tendo sido planejado e sintetizados vinte derivados tiofilidênicos, estruturalmente identificados e avaliados quanto à atividade antimicrobiana frente às cepas padrão de Staphylococcus aureus (ATCC 29213) e multirressitente (VISA 3), Escherichia coli (ATCC 25922), Candida albicans (ATCC 573Y), Klebsiella pneumoniae (ATCC 70060), Enterococcus faecalis (ATCC 29212), Enterobacter cloacae (ATCC 23355) e Serratia marcescens (ATCC 14756). Observou-se que os compostos sintetizados apresentaram atividade bastante promissora frente às bactérias Gram positivas (S. aureus ATCC 29213, S. aureus VISA 3 e E. faecalis ATCC 29212) e para o fungo (C. albicans ATCC 573Y), enquanto que para bactérias Gram negativas não foi observado atividade antibacteriana significativa. Estudos de Relação Estrutura Atividade (REA) foram realizados, com objetivo de determinar quais propriedades físico-químicas auxiliam na atividade biológica. Pode-se observar que o equilíbrio lipo-hidrofílico, juntamente com as propriedades eletrônicas, estereoquímicas e topológicas foram as propriedades que contribuíram para a atividade antimicrobiana dos compostos estudados. Revelando esta classe de compostos como promissora opção no delineamento de novos fármacos no combate às infecções hospitalares com caráter de multirresistência. / Incidence of nosocomial infections has increased substantially, resulting in increased morbidity and mortality in recent decades. Although nosocomial infections are closely connected to asepsis and hygiene of the nosocomial environment, one cannot fail to consider the already established susceptibility of patients affected by these infections. Technological progress related to invasive procedures, diagnostic or therapeutic, helped in spreading microbial infections. However, as a consequence of this advancement, microorganisms that are multidrug resistant to the current arsenal of pharmacologic agents used in antibiotic therapy emerged and these infections became a serious public health problem. The need for new alternatives to fight these microorganisms is evident and nitro-heterocyclic derivatives are considered as viable for studies of new compounds with potential antimicrobial activity. Based on those events the present study proposes the design, synthesis and antimicrobial activity of thiophylidene derivatives with azomethine and oxadiazole structures against microorganisms responsible for some of the main nosocomial infections observed in hospitals worldwide. The choice of the substituent groups of compounds was based on the Craig diagram, having been designed and synthesized twenty thiophylidene derivatives, structurally identified and evaluated for antimicrobial activity against the strains standard Staphylococcus aureus (ATCC 29213) and mutiresistant (VISA 3), Escherichia coli (ATCC 25922), Candida albicans (ATCC 573Y), Klebsiella pneumoniae (ATCC 70060), Enterococcus faecalis (ATCC 29212), Enterobacter cloacae (ATCC 23355) and Serratia marcescens (ATCC 14756). It was observed that the synthesized compounds show very promising activity on the Gram positive bacteria (S. aureus ATCC 29213, S. aureus VISA 3 and E. faecalis ATCC 29212) and against the fungus (C. albicans ATCC 573Y), while for Gram negative bacteria significant antibacterial activity was not observed. Structure Activity Relationship (SAR) Studies were performed with the objective of determining which physicochemical properties help in the biological activity. It can be observed that the lipophilic-hydrophilic balance, together with the stereochemistry, electronic and topological properties were contributed to the antimicrobial activity of the studied substances. Revealing this class of compounds as a promising option in designing of new drugs to fight nosocomial infections with multidrug resistance character.

Planejamento, síntese e avaliação da atividade antimicrobiana de derivados furfurilidênicos frente a micro-organismos causadores de infecções hospitalares / Design, synthesis and evaluation of antimicrobial activity of novel furfuryliden derivatives against nosocomial infections

Infecções hospitalares, causadas por fungos e bactérias, são responsáveis por milhares de mortes anuais e disseminam-se cada vez mais rápido em ambiente hospitalar. Estas infecções afetam principalmente pacientes que são submetidos a procedimentos invasivos ou que apresentam baixa imunidade. Este quadro tende a agravar-se devido ao surgimento de micro-organismos resistentes aos tratamentos atualmente disponíveis, decorrentes, principalmente, de excessivas prescrições, automedicação, pela utilização errônea de antibióticos bem como pela aplicação de métodos de profilaxia inadequados determinando, desta forma, a necessidade de pesquisar e de identificar novos medicamentos para esta finalidade. Neste contexto, ressalta-se que os nitrocompostos que vêm demonstrando bons resultados como agentes antimicrobianos e, sendo assim, este trabalho se propõe a planejar, sintetizar e avaliar duas séries de compostos análogos à nifuroxazida frente a bactérias Gram-positivas, Gram-negativas e fungos de importância em infecções hospitalares com caráter de multirresistência. O planejamento de modificações moleculares na estrutura da série de análogos à nifuroxazida foi auxiliado pelo diagrama de Craig, que se aplica à seleção dos grupos substituintes. A identificação dos compostos obtidos foi realizada por análise espectrofotométrica RMN 1H e RMN 13C e, como critério de pureza, foi realizado análise elementar de CHN. A melhor atividade dos análogos foi frente à S. aureus, onde 16 dos 23 compostos planejados apresentaram atividade superior aos fármacos de referência, estas quais, e composto protótipo. Vale salientar também que os compostos mais ativos foram os análogos 4-butil-N\'-[(5-nitrofuran-2-il) metileno] benzidrazida e 3-acetil-2-[5-nitro-furan-2-il]-5-[4-ciano-fenil]-2,3-diidro-1,3,4-oxadiazolínico com IC90 = 1,8 ± 0,04 µM e 3,89 ± 0,07 µM, respectivamente. Nos estudos de relações estrutura-atividade, a propriedade eletrônica se mostrou com importância fundamental para a atividade dos compostos frente aos micro-organismos considerados. Os resultados encontrados são bastantes promissores e sinalizam para a possibilidade de identificação de um novo candidato para fármaco antimicrobiano frente à infecções hospitalares com caráter de multirresistência. / Nosocomial infections are caused by bacteria and fungi and are the main reason for thousands of deaths every year in the hospital environment. The most susceptible individuals to these type of infection are the immunocompromised, highlighting old-aged and immunodeficiency people, as also patients who passed by invasive procedures. Not only do this situation is very serious, but also a huge number of multidrug-resistant microorganisms are reported worldwide, basically due to excess of prescription and wrong use of antibiotics. If this situation doesn\'t be maintained today, there will not be a bright sight in the future. In this context, there is the necessity for research and development of new antimicrobial agents, and the nitrocompounds, highlighting nifuroxazide\'s analogs, are showing excellent activity against several pathogens related to multidrug resistant nosocomial infections. Therefore, this work aims to design, to synthesize and to evaluate two series of nifuroxazide\'s analogs against strains of gram-positive and gram-negative bacteria, also fungi strains. The sets of analogs were designed based on the replacement of hydroxyl group of nifuroxazide by different substituent groups according to Craig\'s diagram, also a modification in the structure core; identification of these compounds was carried out through 1H and 13C NMR. Melting point and elementary analysis were analyzed for purity criteria. Until the moment, about 23 compounds were evaluated and the best observed activity was against S. aureus strains, which 16 analogs showed better activity than the lead compound and several other compounds, used as referential drug. It is noteworthy that the best activity were analogs 4-butyl-N\'-((5-nitrofuran-2-yl) methylene) benzohydrazide and 3-acetyl-2-[5-nitro-furan-2-yl]-5-[4-cyano-phenil]-2,3-dihydro-1,3,4 oxadiazole with IC90 = 1,8 ± 0,04 µM and 3,89 ± 0,07 µM, respectively. The chemometrics studies suggest that electronic properties are most related to the biological activity on these microorganisms. All the available results shows the potential of nitrofurans to be a new candidate for an antimicrobial drug against multidrug resistant nosocomial infections.

It was observed that the synthesized compounds show very promising activity on the Gram positive bacteria (S. aureus ATCC 29213, S. aureus VISA 3 and E. faecalis ATCC 29212) and against the fungus (C. albicans ATCC 573Y), while for Gram negative bacteria significant antibacterial activity was not observed. Structure Activity Relationship (SAR) Studies were performed with the objective of determining which physicochemical properties help in the biological activity. It can be observed that the lipophilic-hydrophilic balance, together with the stereochemistry, electronic and topological properties were contributed to the antimicrobial activity of the studied substances. Revealing this class of compounds as a promising option in designing of new drugs to fight nosocomial infections with multidrug resistance character.

Planejamento, síntese e avaliação da atividade antimicrobiana de derivados furfurilidênicos frente a micro-organismos causadores de infecções hospitalares / Design, synthesis and evaluation of antimicrobial activity of novel furfuryliden derivatives against nosocomial infections

Infecções hospitalares, causadas por fungos e bactérias, são responsáveis por milhares de mortes anuais e disseminam-se cada vez mais rápido em ambiente hospitalar. Estas infecções afetam principalmente pacientes que são submetidos a procedimentos invasivos ou que apresentam baixa imunidade. Este quadro tende a agravar-se devido ao surgimento de micro-organismos resistentes aos tratamentos atualmente disponíveis, decorrentes, principalmente, de excessivas prescrições, automedicação, pela utilização errônea de antibióticos bem como pela aplicação de métodos de profilaxia inadequados determinando, desta forma, a necessidade de pesquisar e de identificar novos medicamentos para esta finalidade. Neste contexto, ressalta-se que os nitrocompostos que vêm demonstrando bons resultados como agentes antimicrobianos e, sendo assim, este trabalho se propõe a planejar, sintetizar e avaliar duas séries de compostos análogos à nifuroxazida frente a bactérias Gram-positivas, Gram-negativas e fungos de importância em infecções hospitalares com caráter de multirresistência. O planejamento de modificações moleculares na estrutura da série de análogos à nifuroxazida foi auxiliado pelo diagrama de Craig, que se aplica à seleção dos grupos substituintes. A identificação dos compostos obtidos foi realizada por análise espectrofotométrica RMN 1H e RMN 13C e, como critério de pureza, foi realizado análise elementar de CHN. A melhor atividade dos análogos foi frente à S. aureus, onde 16 dos 23 compostos planejados apresentaram atividade superior aos fármacos de referência, estas quais, e composto protótipo. Vale salientar também que os compostos mais ativos foram os análogos 4-butil-N\'-[(5-nitrofuran-2-il) metileno] benzidrazida e 3-acetil-2-[5-nitro-furan-2-il]-5-[4-ciano-fenil]-2,3-diidro-1,3,4-oxadiazolínico com IC90 = 1,8 ± 0,04 µM e 3,89 ± 0,07 µM, respectivamente. Nos estudos de relações estrutura-atividade, a propriedade eletrônica se mostrou com importância fundamental para a atividade dos compostos frente aos micro-organismos considerados. Os resultados encontrados são bastantes promissores e sinalizam para a possibilidade de identificação de um novo candidato para fármaco antimicrobiano frente à infecções hospitalares com caráter de multirresistência. / Nosocomial infections are caused by bacteria and fungi and are the main reason for thousands of deaths every year in the hospital environment. The most susceptible individuals to these type of infection are the immunocompromised, highlighting old-aged and immunodeficiency people, as also patients who passed by invasive procedures. Not only do this situation is very serious, but also a huge number of multidrug-resistant microorganisms are reported worldwide, basically due to excess of prescription and wrong use of antibiotics. If this situation doesn\'t be maintained today, there will not be a bright sight in the future. In this context, there is the necessity for research and development of new antimicrobial agents, and the nitrocompounds, highlighting nifuroxazide\'s analogs, are showing excellent activity against several pathogens related to multidrug resistant nosocomial infections. Therefore, this work aims to design, to synthesize and to evaluate two series of nifuroxazide\'s analogs against strains of gram-positive and gram-negative bacteria, also fungi strains. The sets of analogs were designed based on the replacement of hydroxyl group of nifuroxazide by different substituent groups according to Craig\'s diagram, also a modification in the structure core; identification of these compounds was carried out through 1H and 13C NMR. Melting point and elementary analysis were analyzed for purity criteria. Until the moment, about 23 compounds were evaluated and the best observed activity was against S. aureus strains, which 16 analogs showed better activity than the lead compound and several other compounds, used as referential drug. It is noteworthy that the best activity were analogs 4-butyl-N\'-((5-nitrofuran-2-yl) methylene) benzohydrazide and 3-acetyl-2-[5-nitro-furan-2-yl]-5-[4-cyano-phenil]-2,3-dihydro-1,3,4 oxadiazole with IC90 = 1,8 ± 0,04 µM and 3,89 ± 0,07 µM, respectively. The chemometrics studies suggest that electronic properties are most related to the biological activity on these microorganisms. All the available results shows the potential of nitrofurans to be a new candidate for an antimicrobial drug against multidrug resistant nosocomial infections.

Molecular Design Of Some Potential Explosives

With existing energetic materials, the on going demand from the user for increased performance with reduced vulnerability induces the synthesis of novel explosives. The need to control costs ensured that theoretical methods can help to eliminate any poor candidates due to performance and safety problems. Thus, this study is aimed to design of some potential high explosive molecules which have high detonation properties and are also insensitive. Presently 65 molecules were studied and 54 of them were handled for the first time in the literature. The detonation properties have been evaluated within the limitations of the Kamlet-Jacobs equations, based on the quantum chemically calculated densities and heat of formation values. It is found that there might be some candidates of relatively insensitive and high energy density materials among these 54 studied molecules so far not mentioned in the literature.

The crystal structures of xenobiotic reductase A and B from pseudomonas putida II-B and pseudomonas fluorescens I-C: structural insight into regiospecific reactions with nitrocompounds

Nitrochemicals are currently widely used as solvents, drugs, biocides, fuels and explosives and are consequently widely distributed in the environment. The reductive nitrite elimination from explosive compounds is catalyzed by two FMN-dependent, xenobiotic reductases (XenA or XenB). These genes for these regiospecific enzymes were cloned from Pseudomonas putida and P. fluorescens I-C respectively and isolated from the soil of a contaminated World War II munitions manufacturing plant. These enzymes enable the microbes to fulfill their nitrogen requirements from nitroglycerin by catalyzing the regiospecific, NADPH dependent, reductive denitration of nitroglycerin with differing selectivities. The two enzymes also transform a number of additional nitrocompounds in vitro, e.g. TNT and metronidazole, a leading drug in the treatment of Helicobacter pylori, a causative agent of human ulcers. Single crystals were obtained for XenA and XenB and complete X-ray diffraction datasets have been collected and analyzed to better understand these characteristics. The 1.6 Å resolution structure of XenA reveals a dimer of β/α)₈-TIM barrels, but the 2.3 Å resolution structure for XenB is a monomer. The (β/α)₈-TIM barrel protein fold is the most common fold in the PDB. However, the XenA structure exhibits a unique, C-terminal domain-swapped topology. Thus a portion of each active site is comprised of residues from the neighboring monomer. To probe the reaction cycle, crystal structures of ligand complexes and the reduced enzyme have been refined. For example, our structure of the XenA-metronidazole complex shows that ligands bind parallel to the FMN si-face. Our 1.5 Å resolution structure for reduced XenA reveals an FMN isoalloxazine ring with an angle of ~165° along the N5-N10 axis. We have also generated models of the reduced enzyme-nitroglycerin complexes by molecular dynamics. The results with both XenA and XenB reveal differences in enzyme-ligand hydrogen bonding. These differences correlate remarkably well with the regiospecific differences observed for nitrite elimination from nitroglycerin and reduction of TNT by the two enzymes.

Síntese e avaliação biológica de chalconas preparadas a partir do produto natural safrol

Este grupo tem despertado o interesse dos pesquisadores por apresentarem várias atividades biológicas incluindo: antitumoral, antiparasitárias, antimalária, anti-inflamatória, entre outras. Os flavonoides tem sido alvo de estudos por apresentarem grande potencial farmacológico e, do ponto de vista sintético, há um grande interesse para o desenvolvimento de estruturas análogas, buscar novas moléculas e realizar uma investigação biológica para o tratamento de diversas patologias. A busca de novos medicamentos, despertou nosso interesse em desenvolver alguns compostos nitroderivados para serem testados em diversas doenças parasitárias. No entanto, a síntese das chalconas com o grupo nitro (NO2) ligado ao anel aromático, é de grande interesse e amplamente utilizadas como fármacos. Portanto, para viabilizar o processo de síntese das chalconas, foi utilizado o safrol como matéria-prima, e depois seguida de uma reação de isomerização e convertido em piperonal que depois foi submetido a uma reação de nitração aromática. A síntese da chalcona é baseada na reação de condensação de Claisen-Schimidt, empregando um catalisador básico (NaOH), solvente polar (EtOH), acetofenona e nitropiperonal. Todas as chalconas foram caracterizadas por análise de ressonância magnética nuclear (RMN), obtido o ponto de fusão e realizados os ensaios biológicos. A chalcona que tem o grupo nitro (NO2) ligado ao anel aromático apresentou um melhor resultado contra o parasita Schistosoma mansoni, responsável pela doença esquistossomose. / The chalcones are a class of natural products belonging to the group of flavonoids. This group has aroused the interest of researchers because they have several biological activities including: anti-tumor, anti-parasitic, anti-malarial, anti-inflammatory, among others. Flavonoids have been investigated for having great pharmacological potential, and the synthetic point of view, there is great interest for the development of similar structures, seek new molecules and perform a biological research for the treatment of various diseases. The search for new drugs, aroused our interest in developing some nitroderivatives compounds to be tested in several parasitic diseases. However, the synthesis of chalcones with the nitro group (NO2) attached to the aromatic ring, is of great interest and widely used as drugs. Therefore, to facilitate the process of synthesis of chalcones it was used safrole as a raw material, and then followed by an isomerization reaction and converted into piperonal which was then subjected to a reaction for aromatic nitration. The synthesis of chalcone is based on the reaction Claisen-Schmidt Condensation, using a basic catalyst (NaOH), polar solvent (EtOH), acetophenone and nitropiperonal. All chalcones were characterized by nuclear magnetic resonance analysis (NMR), the melting point obtained and biological tests conducted. The chalcone having nitro group (NO2) attached to the aromatic ring presented a better result against the parasite Schistosoma mansoni, responsible for the disease schistosomiasis. In this sense, the analysis by spectroscopy in the infrared region, indicated that no changes in the structure of the nanostructured chemical pulp, after sonication. Thermal analysis showed stability of nanostructured sonicated above the pulp fiber and untreated fiber obtained via chemical hydrolysis, indicating the possibility of use as matrix polymers with melting temperatures higher than those polyolefins. Since the morphological analysis showed a reduction in fiber dimensions and good dispersion of the fibers after compression molding cycles. Thus, the mechanical traction results are comparable to those observed in the literature for similar composites obtained by the extrusion process which allows better mixing and dispersion of the composite. The nanostructured cellulose obtained proved to be interesting for application in polymeric composites.

Alguns nitrocompostos apresentam atividade antichagásica resultante do processo de redução do grupo nitro com conseqüente formação de radical nitro ânion e de intermediários tóxicos ao parasita. A variação estrutural dos derivados 5-nitroheterocíclicos pode interferir no processo de redução destes compostos e, também, na atividade biológica. Neste aspecto, o estudo da redução dos nitrocompostos por meio de técnicas voltamétricas, como a voltametria cíclica e a voltametria de onda quadrada é uma forma de avaliar ou até simular o mecanismo de ação destes derivados. Assim, a avaliação de processos eletródicos e a determinação dos potenciais de redução dos nitrocompostos fornecem subsídios para a compreensão do mecanismo da atividade antiparasitária. Neste trabalho, desenvolveram-se procedimentos eletroquímicos utilizados para a determinação do potencial de redução e do potencial de meia onda de vinte e três nitrocompostos (5-nitro-2-tiofilideno 4-R-benzidrazidas e 5-nitro-2-furfurilideno 4-R-benzidrazidas), visando o emprego destes em estudos de QSPR e SAR. As determinações foram realizadas empregando-se Voltametria Cíclica e Voltametria de Onda Quadrada em célula eletroquímica de 10 mL nos meios prótico (solução tampão PIPES com eletrólito de suporte NaNO3 0,1 mol L-1), misto de DMSO/solução tampão PIPES (50% v/v) e em meio aprótico de DMSO (com eletrólito de suporte Bu4NH4BF4). O eletrodos de referência e auxiliar utilizados foram Ag/AgCl saturado e platina. Os eletrodos de trabalho utilizados foram o carbono vítreo (meio aprótico e misto), pasta de carbono (meio misto e prótico) e pasta de carbono modificada com nitrocompostos (meio prótico). Foram realizados estudos teóricos, de modelagem molecular, e experimental, espectrofotometria UV/visível, com o objetivo de determinar a conformação de menor energia mínima global e confirmar a ocorrência de disposição estrutural co-planar. No estudo teórico, utilizou-se as metodologias de otimização de geometria, e análise conformacional dos nitrocompostos calculadas por meio de AM1 e HF3-21G* e HF6- 31G* para obtenção dos confôrmeros de menor energia mínima. Estes confôrmeros foram submetidos a cálculo de carga de ponto único (AM1), onde determinou-se os valores de energia de diversas propriedades de caráter eletrônico (energias de formação em vácuo e meio solvatado, de HOMO, de LUMO, momento de dipolo, dureza, potencial químico, afinidade eletrônica, potencial de ionização, cargas de potencial eletrostático), geométrica (volume molecular) e mapas 3D de densidade de potencial eletrostático, de HOMO e de LUMO. Todos os cálculos foram realizados empregando-se os pacotes computacionais Spartan O2 for Linux, Spartan O4 for Windows e ClogP 4.0 (para determinar os valores de hidrofobicidade). Os valores do potencial de pico catódico (Epc 1) e de meia onda bem como as propriedades físico-químicas, obtidos a partir de cálculos de química quântica, de σp e σR, ClogP e π) foram utilizados em análise de QSPR aplicando-se a Análise de Hansch e regressão multivariada, PLS. Avaliou-se ainda, a atividade antichagásica de dez nitrocompostos (oito derivados tiofilidênicos e dois derivados fufurilidênicos) visando investigar a possibilidade de influência do processo de redução dos nitrocompostos sobre a atividade biológica. Realizaram-se ensaios que permitiram avaliar o efeito antiproliferativo dos nitrocompostos sobre o parasita, em 24 horas, por meio de contagem do número de unidades viáveis em câmara de Neubauer. Os métodos voltamétricos utilizados mostraram-se adequados para a avaliação da redução eletroquímica dos nitrocompostos, e foram apropriados para a obtenção dos potenciais de redução de pico catódico e dos potenciais de meia onda de todos os derivados em meio aprótico e misto. Em meio aquoso, obtiveram-se os valores dos potenciais de redução de pico catódico apenas para os derivados tiofilidênicos. A partir dos resultados obtidos na análise conformacional, observou-se a quebra de efeito co-planar estrutural na região do anel benzênico substituído e porção hidrazídica molecular. Adicionalmente, nos mapas de densidade de potencial eletrostático, registrou-se a ocorrência de efeito mesomérico conjugado reduzido entre as porções moleculares citadas acima. De posse destes dados, em conjunto com as visualizações de sinais característicos para grupos cromóforos distintos em espectros UV/visível, sugere-se a ausência de efeito conjugado molecular e, como conseqüência, de influência dos grupos substituintes em posição para do anel benzênico sobre o grupo nitro ligado ao anel heterocíclico. Nos resultados obtidos a partir das análises de QSPR, observou-se que não ocorre correlação entre as propriedades físico-químicas, determinadas para os nitrocompostos, e os descritores eletroquímicos, obtidos em diferentes meios de reação. Diante deste fato, sugere-se que a quebra do efeito co-planar interfere diretamente na intensidade da influência das propriedades físico-químicas de grupos substituintes sobre o potencial de redução dos nitrocompostos. Observou-se, também, que a maioria dos compostos apresentou atividade antichagásica superior ao fármaco de referência, o benznidazol. Verificou-se que os compostos não substituído e cloro derivado da série dos 5-nitro-2-furfurilidênicos são mais ativos que os correspondentes análogos das 5-nitro-2-tiofilideno benzidrazidas. Em análise preliminar, observou-se que o potencial de redução e o potencial de meia onda não exercem influência sobre a atividade antichagásica dos nitrocompostos. / Some nitrocompounds have show activity against to Trypanosoma cruzi. Those biological activity are resulting from nitro group reduction with subsequent formation of radical nitro anion and other reaction\'s intermediates, which are toxic to the parasite. The structural difference among the 5-nitro-heterocyclic derivatives might exert influence on these compounds reduction and also on the antichagasic activity. In this way, the study of reduction process by voltammetric techniques, such as cyclic voltammetry and square wave voltammetry, is a mode of evaluate the biological action mechanism. Therefore, the evaluation of the electrodic process and the determination of nitrocompounds reduction potential provide information for elucidating the antitrypanosomal activity mechanism. In the present work, the procedures employed to determinate the cathodic reduction and reduction potentials were carried out on twenty three nitrocompounds (5-nitro-2-tiofilidenic benzhidrazides and 5-nitro-2-furfurilidenic benzhidrazides) with the aim using those data in QSPR and SAR analysis. Cyclic voltammetry and square wave voltammetry techniques were used to determined the reduction potential in electrochemical cell of 10 mL in the protic media (PIPES buffer and electrolyte of support NaNO3 0.1 mol L-1), mixed media of DMSO and PIPES buffer 50:50 v/v (electrolyte of support NaNO3 0.1 mol L-1), and aprotic media of DMSO (electrolyte of support Bu4NH4BF4 0.1 mol L-1). The reference and counter electrodes used were Ag/AgCl (with KCl saturated) and platinum, respectively. In these studies, the work electrodes were glassy carbon (aprotic and mixed media), carbon paste (mixed and protic media) and carbon paste modified with nitrocompounds (protic media). Molecular modeling studies and UV/visible spectrophotometry were investigated with the aim for determining the lowest conformational energy and the occurrence of co-planarity molecular structure. In this work were performed the geometry optimization (using the quantum chemistry AM1, ab initio HF3-21G* and HF6-31G* level of theory), conformational analysis(AM1 and ab initio HF6-31G*) and single point calculations (AM1). All the lowest energy conformers from conformational analysis were submitted to single point calculations. These models were used to determine the physicochemical properties (energy of formation in vacuum and solvated media, energy of HOMO, energy of LUMO, dipole moment, hardness, chemistry potential, electronic affinity, ionization potential and charges of electrostatic potential) and the molecular electrostatic potential, HOMO and LUMO maps, respectively. All calculations were performed using the computational software Spartan O2 for Linux, Spartan O4 for Windows, and the ClogP 4.0 (only for hydrophobicity). The cathodic reduction and half-wave potentials values and the set of physicochemical properties, which are obtained from quantum chemistry calculations, as well as the electronic effect (σp and σR derived of σ Hammett) and the hydrophobicity property (ClogP, and π of Hansch) were used in QSPR analysis applying Hansch analysis and PLS methodologies. Afterwards, the antichagasic activity of ten compounds (eight thiofilidenic and two furfurylidenic derivatives) was evaluated considering the influence of nitrocompounds reduction on biological activity. This assay allows to analyse the antiproliferative effect of nitrocompounds on the parasites growing, in twenty-four (24) hours, reading the number of trypanosomes in Haemocytometer. The voltammetric methodologies allowed to determinate the cathodic reduction and half-wave potential values in aprotic and mixed media to all nitrocompounds. In protic media, however, the cathodic reduction potential values were obtained only for the 5-nitro-2-tiofilidenic benzhidrazides. Those procedures seen to be appropriated for perform the electrochemistry reduction evaluation. The geometry optimization and conformational analysis allows determining a lack of structural planarity in all derivatives located at the bond between the carbonilic carbon and benzhoyl group. Additionally, the electrostatic potential maps presented a decreased on the electronic conjugate effect of the structures of nitrocompounds investigated. The conformational analysis allows us to determine the nitroheterocyclic biosostere conformations dependence on the reaction phase studied. The UV/visible spectra presented two waves, which are indicating responses related to different chromophores. These results suggest the absence of conjugate effect in nitrocompound structure, indicating that the presence of groups attached to benzene ring do not exert influence on the nitro group attached to heterocyclic ring. In the QSPR analysis was not detected any correlations between thel physicochemical properties and the cathodic reduction and reduction potentials, in all reaction media investigated. Those results might be occurred due to the lack of molecular co-planarity, which is reducing the influence of any property on the nitrocompounds reduction. Almost all nitrocompounds investigated show higher antichagasic activity than the reference drug, benznidazol. In a preliminary analysis it was verified that the cathodic reduction and half-wave potentials do not exert influence on the nitrocompounds antichagasic activity.

Este trabalho teve como objetivo a realização de estudos de Relações Quantitativas Tridimensionais Estrutura-Atividade, QSAR 3D, com identificação de alvos potenciais para compostos 5-nitro-heterocíclicos com estruturas azometínica ou oxadiazolínica com bioatividade dual, antifúngica e anti-T. cruzi, visando à identificação de novos compostos que possam ser aproveitados como possíveis candidatos a fármaco ou como compostos-líderes para novos estudos de modificação molecular. Os compostos estudados pertencem a quatro séries intimamente relacionadas, a saber: Série AzoO: 5-nitro-2-furfurilideno benzidrazidas 4-substituídas; Série AzoS: 5-nitro-2-tiofilideno benzidrazidas 4-substituídas; Série OxaO: 3-acetil-oxadiazolina 2,5-furfurilideno benzidrazidas 4-substituídas e Série OxaS: 3-acetil-oxadiazolina 2,5-tiofilideno benzidrazidas 4-substituídas. A utilização de forma integrada de ferramentas pertencentes às áreas de química farmacêutica, quimioinformática e bioinformática permite a caracterização de cavidades catalíticas de proteínas e a identificação das propriedades físico-químicas consideradas essenciais para a interação adequada entre ligante e biomacromolécula-alvo. Por outro lado, permitem também a identificação de alvos a partir da comparação de estruturas químicas, com as propriedades físico-químicas associadas, entre ligantes conhecidos e possíveis alvos; este procedimento de comparação de perfis moleculares, conhecido como virtual profiling, parte do princípio de que moléculas semelhantes a ligantes conhecidos de proteínas têm o potencial de interagir com essa mesma proteína, onde o grau de semelhança é determinado não somente pela estrutura química, mas também pela distribuição de características físico-químicas. As ferramentas utilizadas em estudos in silico incluem também análise de descritores topológicos que permitem a caracterização de propriedades físico-químicas considerando sua distribuição espacial em relação à estrutura química de uma dada molécula. O uso destes descritores permite a determinação do grau de semelhança entre moléculas ou partes de moléculas (fragmentos moleculares), e encontra-se na base das metodologias de triagem e de caracterização de sítios catalíticos de proteínas. Dentro da proposta de trabalho foram realizados estudos envolvendo virtual profiling, análise de fragmentos moleculares com base em descritores físico-químicos capazes de caracterizar superfícies de proteínas e ligantes, caracterização de cavidade catalítica de possível alvo e docking dos compostos a alvos putativos. Foi também realizada a determinação de atividade antifúngica e análise de resultados de QSAR 3D, levando finalmente à construção de uma hipótese quanto ao possível alvo biológico para os compostos azometínicos e oxadiazolínicos. O procedimento de virtual profiling apontou como possíveis alvos as enzimas CYP19A (aromatase), CYP3A4, CYP3A5 e CYP3A7. Embora estas enzimas não estejam diretamente envolvidas com as atividades biológicas testadas para os compostos até o momento, existem estudos que demonstram a interação de compostos azólicos de ação antifúngica, cujo alvo é CYP51 (14α-desmetilase), também com CYP19A e CYP3A4. Em conjunto com a análise da caracterização das propriedades físico-químicas com uso de descritores topológicos e a própria dualidade da atividade biológica, concluiu-se que seria possível a interação dos compostos das séries estudadas com CYP51, sendo esta enzima alvo não somente de antifúngicos azólicos, como também de compostos com ação anti-T. cruzi. A caracterização da cavidade catalítica de CYP51, tanto em termos de descritores de propriedades físico-químicas como de características estereoquímicas, associada ao estudos de QSAR 3D, confirmaram a possibilidade de interação da série oxadiazolínica com a CYP51, em orientação semelhante à dos antifúngicos azólicos. A série azometínica, que apresenta a mesma atividade dual da série oxadiazolínica, embora com potências diferentes, não possui conformação adequada para a interação da forma proposta. Existem, no entanto, dados que indicam a possibilidade de interação de compostos no sítio catalítico da enzima de forma diferente em relação a compostos oxadiazolínicos e antifúngicos azólicos. / This study had as objective the study of Tridimensional Quantitative Structure-Activity Relationships, 3D QSAR, and the identification of potential targets for 5-nitro-heterocyclic compounds with azomethynic or oxadiazolynic structures presenting dual antifungal and anti-T. cruzi bioactivity, aiming the discovery of new compounds to be used as possible drug candidates or lead compounds. The studied compounds belong to four closely related series: AzoO series: 4-substituted 5-nitro-2-furfurylidene benzhydrazides; AzoS Series: 4-substituted 5-nitro-2-thiophylidene benzhydrazides; OxaO Series: 4-substituted 3-acetyl-oxadiazolyne 2,5-furfurylidene benzhydrazides; and OxaS Series: 4-substituted 3-acetyl-oxadiazolyne 2,5-thiophilydene benzhydrazides. The integrated use of tools belonging to the areas of medicinal chemistry, chemoinformatics, and bioinformatics allow for the characterization of catalytic cavities of proteins and the identification of physicochemical properties considered essential for the adequate interaction between ligand and target biomacromolecule. In addition to this, they also make possible the identification of targets through the comparison of chemical structures, with their associated physicochemical properties, of known ligands and their possible targets; this approach, known as virtual profiling, is based on the principle that molecules similar to known ligands of proteins can potentially interact with this same protein, with the similarity degree being determined not only by chemical structure but also through the distribution of physicochemical characteristics. The tools used in studies in silico also include the analysis of topological descriptors that can be used to analyse physicochemical characteristics considering their spacial distribution in relation to the chemical structure of a given molecule. The use of these descriptors makes possible the determination of the similarity degree between molecules or part of molecules (molecular fragments), and is the basis for methodologies of screening and characterization of the catalytic sites or proteins. Considering the proposed objective, studies were carried involving virtual profiling, analysis of molecular fragments based on physicochemical descriptors able to characterize the surface of ligands and proteins, characterization of the catalytic site of a possible target, and docking of the compounds to putative targets. The antifungal activity of compounds was determined and 3D QSAR results analysed, leading to the formulation of a hypothesis for the possible biological target for the azomethynic and oxadiazolynic compounds. Virtual profiling results pointed as possible targets the P450 enzymes CYP19A (aromatase), CYP3A4, CYP3A5, and CYP3A7. Although these enzymes are not directly involved with the currently tested biological activities for these compounds, there are studies reporting the interaction of azole antifungal compounds, whose target is CYP51 (14α-demethylase), with CYP19A and CYP3A4. Taken together with the analysis of physicochemical characteristics based on topological descriptors, and considering the duality of determined biological activities, it was concluded that the interaction of the studied compounds with CYP51 was possible since this enzyme is the target nor only for azole antifungals, but also for anti-T. cruzi compounds. Characterization studies of CYP51 catalytic cavity considering not only physicochemical properties descriptors but also stereochemical characteristics, associated to the results of 3D QSAR, confirmed the possibility of interaction of compounds from the oxadiazolynic series with CYP51, in an orientation similar to azole antifungals. The azomethynic series, that also presents the same dual biological activity though with different potency, does not present an adequate conformation for the ineraction proposed for the oxadiazolynic series; however, there are reports indicating the possibility of interaction of compounds in the catalytic site of the enzyme in a different way from that of antifungal azoles and the proposed interaction of oxadiazolynic compounds.
