Impaired Cardiac cAMP-specific PDE4, β1-AR, and NE in an Ischemia-Reperfusion Rat Model

Ischemic injury in the heart is followed by an increase in SNS activity and the higher this activity, the poorer patient outcomes. An index of SNS activity in models of ischemia can be achieved by measuring NE, β-AR, and perhaps indirectly PDE4 to give an intracellular aspect on SNS signaling. A 20 minute ischemia-reperfusion was induced in a rat model with physiological measurements at 2-5 weeks post IR. At 3 weeks post IR, rats displayed increased PDE4 expression, decreased β 1-AR expression, increased plasma NE, decreased tissue NE storage, increased Doppler E/A ratio and unchanged LV ejection fraction. PET analysis with FDG revealed no infarct at 2 weeks, while analysis with [13N]NH3 displayed no resting flow defect but revealed trends in flow reserve impairment as early as 2.5 weeks with recovery at 5 weeks post-surgery. Applications of this model could be non-invasive imaging of PDE4 with (R)-[11C]Rolipram PET at early time points for development towards prognostic and therapy guidance in humans.

Rakouská teorie hospodářského cyklu - pohled současné makroekonomie

Tato diplomová práce analyzuje rakouskou teorii hospodářského cyklu. Představuje její základní stavební kameny. V této teorii neexistuje rozlišení mezi krátkým a dlouhým obdobím, proto je spojena analýza hospodářského cyklu a hospodářského růstu. Hlavním klíčem k jejímu pochopení je teorie kapitálu, jež se zrcadlí v tzv. intertemporální struktuře produkce. Hospodářský cyklus je způsoben monetárním šokem, který atakuje cenový systém ekonomiky, zejména pak úrokovou míru. Tyto disturbance narušují intertemporální strukturu kapitálu a zahajují hospodářský cyklus. Diplomová práce analyzuje ne-neutralitu peněz, a to i v případě změn poptávky po penězích. Dále se pokouší představit slabiny této teorie a nalézá možná řešení. Diplomová práce také porovnává rakouskou teorii s moderními teoriemi hospodářského cyklu a upozorňuje na některé vady těchto teorií. Na závěr jsou vysvětleny hlavní pohyby české ekonomiky pomocí rakouské teorie hospodářského cyklu.

The thesis deals with intelligence, as a socio-cultural concept that has its own history and development. It thus deconstructs the naturalized notions of a biological "essence" of intelligence, which has not been proved to this day. The scientific and philosophical modes of thinking, that participated in both "inventing" intelligence and its forming, are introduced. The analysis of the primary sources enables to compass the reflection of these dominant scientific approaches towards intelligence in the Czech expert discourses. The thesis also reflects on the important role intelligence has in the assessment of mental norm and disability, and it thus analyses the perception of disability during the defined historical period, including the discourse analysis of the different representations of disability. This diploma thesis focuses on the topic of the principles applied during proceedings regarding offences, mainly the principle of non bis in idem, which this thesis focuses on. The non bis in idem principle means that a matter cannot be judged twice, because it is generally accepted that two punishments for the same crime are unfair. This principle is applied in proceedings on criminal charges and because the concept of criminal charges is autonomous according to the European Court for Human rights, i.e. it has specific objective content, I chiefly discussed interpretation of this concept. According to the European Court for Human Rights, in order for the issue to be criminal charges within the meaning of Article 4(1) of the Protocol to the European Convention on Human Rights, the charges must fulfil the so-called Engel criteria, which I have endeavoured to explain to the reader. These criteria were created by the European Court for Human Rights in the case of Engel and Others v. the Netherlands (1976), whereas these criteria were developed before the same court in the case of Bendenoun v. France (1994). Presented thesis deals with conversations that arise within the online mentoring counseling center "S barvou ven". Mainly young people going through the process of coming out are the target group of counseling center. In the theoretical part of thesis, I describe the counseling center, the social atmosphere that prevails around the issue of people with minority sexual orientation, clarify what a mentoring relationship is and its importance in the lives of adolescents and describe the process of coming out. The empirical part deals with the qualitative analysis of nineteen conversations. The results of the analysis showed that clients entering the counseling center use the conversation with the mentor to meet various needs, from the need to share their feelings or experiences to the desire to find their "friend" or partner in the mentor. The results of the second part of the analysis provide an indicative overview of the client's needs, as well as when and why their needs could or could not be met. The results also show how mentors work with needs which clients can not consciously describe. Last but not least, the results overview shows that mentors use a wide range of questions, offer information or provide clients, for example, a sense of belonging or hope, or offer their own experience.

Volcanic stratigraphy and a kinematic analysis of NE-trending faults of Allens Ranch 7.5' quadrangle, Utah County, Utah

The mineral resources of the Tintic Mining District are influenced by three major events in its geologic history; the Mesozoic Sevier Orogeny, Paleogene volcanism and Late Neogene Basin and Range extension. In this paper a detailed analysis of each these geologic events is presented to help us understand the structural host, mineralization and exhumation of the Tintic Mining District ore. A kinematic analysis of the faults was completed to determine the origin of NE-trending faults, Sevier Orogeny or Basin and Range extension, in the northern part of the East Tintic Mountains in Allens Ranch 7.5' quadrangle, near the eastern margin of the Great Basin of central Utah. The structural history of the NE-trending faults found in the quadrangle was reconstructed to determine stress directions and fault kinematics. Maximum paleostress direction for the East Tintic fold and thrust system is between 80º–100º with fold axes oriented at ~350º. For example, the Gardison Ridge and Tintic Prince faults are NE-trending right-lateral transverse faults that formed at ~30º to paleostress directions similar to those of the Sevier Orogeny. The dominant NE-trending faults in the region are likely due to (1) differential shortening during progressive orocline development, (2) the pre-deformational Pennsylvanian-Permian Oquirrh basin geometry, and (3) the influence of the Leamington transverse zones of the Provo salient. Conversely, mixed paleostress directions for the north-trending Tintic Davis Canyon fault show it is a Basin and Range extension-related normal fault that may have originated as a Sevier related fault. Other N-trending faults within the quadrangle are only related to Basin and Range extension. However, large offset, range-bounding faults are buried by valley fill throughout the quadrangle and no young fault scarps are identified cutting Lake Bonneville deposits. An Oligocene to Miocene suite of extrusive volcanic units in the quadrangle correlates well with those of the East Tintic and Soldiers Pass volcanic fields. The Paleogene volcanic section is dominated by a suite of high-K calc-alkaline extrusive rocks (35 to 32 Ma). This intermediate to silicic sequence was followed by eruption of the mildly alkaline Mosida Basalt during the Miocene (19.5 Ma) marking the transition from subduction-related intermediate and silicic volcanism to extension-related mafic volcanism in the eastern Great Basin.
