Considerada multifatorial e progressiva, a RD afeta células neurais e gliais, e também elementos vasculares da retina. Sabe-se que diversas vias estão envolvidas na patogênese da RD, no entanto, os mecanismos que levam a exacerbação da inflamação e morte de células gliais/neurais o que caracteriza a neurodegeneração da retina, ainda permanecem desconhecidos. Diante disso, a redução desses fatores tem sido extensivamente estudada como alvo no combate a RD. As Sirtuínas, histonas desacetilases dependentes de nicotinamida adenina dinucleotídeo (NAD+), atuam em resposta a vários estresses e atualmente tem sido relacionadas às importantes funções moleculares na regulação de várias doenças. Considerado um redox sensível, a SIRT1 pode estar reduzida em condição de doença, o que agravar ainda mais a situação patológica. No entanto, não se sabe ao certo o mecanismo de modulação e/ou atuação da SIRT1 frete às doenças neurodegenerativas, tais como a RD. No Artigo I, foram avaliados os possíveis efeitos protetores do cacau rico em polifenóis na retina diabética. As células Müller da retina (rMC-1) foram expostos por 24h à glicose normal (NG), alta glicose (HG) ou peróxido de hidrogênio (H2O2) e submetidas ao tratamento com cacau na presença ou não de um inibidor da SIRT1 ou siRNA. O estudo animal foi desenvolvido em ratos experimentalmente diabéticos induzidos por estreptozotocina e randomizado para receber tratamentos com cacau com baixa, intermediária, ou elevada dose de polifenol (0,12 mg; 2,9 mg; 22,9 mg/kg/dia) por gavagem durante 16 semanas. As células expostas a H2O2 ou HG apresentaram aumento de proteína acídica fibrilar glial (GFAP) e acetil-RelA/p65 e diminuição da atividade/expressão da SIRT1. Estes efeitos foram anulados pelo cacau, que diminuiu a produção de espécies reativas de oxigênio e reduziu a ativação da poli(ADP-ribose) polimerase-1 (PARP-1); melhorou os níveis intracelulares de NAD+ e consequentemente aumentou da atividade da SIRT1. As retinas dos ratos diabéticos exibiram os primeiros marcadores de retinopatia acompanhada pela eletrorretinografia prejudicada. A presença de diabetes levou a ativação da PARP-1 e diminuição dos níveis de NAD+, resultando em comprometimento da SIRT1. O aumento na acetilação do RelA/p65 levou na hiperexpressão do GFAP. A administração oral de cacau polifenol restaurou as alterações acima referidas. Este estudo revelou que o cacau enriquecido com polifenóis teve efeito protetor da retina diabética restabelecendo a via da SIRT-1. No Artigo II, foi investigado o possível efeito terapêutico de células derivadas de animais saudáveis (Dock7 m +/+ Leprdb db/m) e diabéticos (BKS.Cg-Dock7 m +/+ Leprdb/J, db/db) na retinopatia diabética (RD). Os camundongos db/db (espontaneamente diabéticos) com 8 semanas de idade foram randomizados para receber uma única injeção intravenosa de PBS ou células early outgrowth (EOCs) de doadores db/m ou db/db. Quatro semanas mais tarde, os animais foram sacrificados e os olhos enucleados. Para estudo in vitro, o meio condicionado das EOCs (EOC-CM) foi gerado a partir do cultivo de EOCs de animais db/m e db/db. As células rMC-1 foram expostas por 24h a NG ou HG e submetidas ao tratamento com db/m ou db/db EOC-CM, em presença ou não de um inibidor farmacológico (EX527) ou gênico (siRNA) da SIRT1. Nos ratos diabéticos, houve um aumento de marcadores de RD e do dano oxidativo, acompanhado por uma diminuição da proteína SIRT1 e seguido pelo aumento da acetilação da lisina-310 do complexo p65-NFkB. A terapia celular com EOCs reduziu significativamente todas as alterações mencionadas acima. As rMC-1 expostas a HG apresentaram aumento da expressão de GFAP, fator de crescimento do endotélio vascular e Nox4, acompanhado pelo aumento dos níveis de espécies reativas de oxigênio e acetil-lisina-310-p65-NFkB. Além disso, a expressão/atividade da SIRT1 foram reduzidas em ambiente diabético. O tratamento com EOC-CM impediu todas estas alterações. Este estudo demonstra que a capacidade parácrina das EOCs, na secreção de fatores, é eficaz no restabelecimento da via de SIRT1 retina, e assim, proteger a retina dos insultos diabéticos. Em resumo, a presente tese fornece evidências que tanto a administração oral do cacau enriquecido com polifenóis quanto à terapia celular com EOCs, conferem neuroproteção da retina aos insultos do diabetes. Portanto, intervenções que modulem a atividade das sirtuínas são promissoras no tratamento farmacológico da retinopatia diabética / Abstract: The diabetic retinopathy (RD) is a devastating disease and the principal cause of blindness among people in adulthood worldwide. The RD is considered a multifactorial and progressive disease, affecting neuronal and glial cells, and also vascular elements of the retina. It is known that several pathways are involved in the pathogenesis of RD, however, the mechanisms that lead to exacerbation of inflammation and death of glial/neuronal cell, characterizing retinal neurodegeneration, remain unknown. Therefore, the reduction of these factors have been extensively studied as a therapeutic target against RD. Sirtuin 1 (SIRT1), a family of histone deacetylase enzyme, acts in response to various stresses and, currently, has been related to important molecular functions in the regulation of various diseases. Considered a redox-sensitive, SIRT1 may be reduced under disease condition, whereby aggravate the pathological situation. However, is not known the mechanism of modulation/activity of SIRT1 in neurodegenerative diseases, such as RD. In the article I, were studies the possible protective effects of cocoa in the diabetic retina were assessed. rMCs exposed to NG, HG or H2O2 were submitted to cocoa treatment in the presence or absence of SIRT-1 inhibitor and siRNA. The experimental animal study was conducted in streptozotocin-induced diabetic rats and randomized to receive low, intermediate, or high polyphenol cocoa treatments via daily gavage for 16 weeks (i.e., 0.12 mg/kg/day, 2.9 mg/kg/day, or 22.9 mg/kg/day of polyphenols). The rMCs exposed to HG or H2O2 exhibited increased GFAP and acetyl-RelA/p65 and decreased SIRT1 activity/expression. These effects were cancelled out by cocoa, which decreased ROS production and PARP-1 activity, augmented the intracellular pool of NAD+, and improved SIRT1 activity. The rat diabetic retinas displayed the early markers of retinopathy accompanied by markedly impaired electroretinogram. The presence of diabetes activated PARP-1 and lowered NAD+ levels, resulting in SIRT1 impairment. This augmented acetyl RelA/p65 had the effect of upregulated GFAP. Oral administration of polyphenol cocoa restored the above alterations in a dose-dependent manner. This study reveals that cocoa enriched with polyphenol improves the retinal SIRT-1 pathway, thereby protecting the retina from diabetic milieu insult. In the article II, were investigated the possible therapeutic effect of cells derived from control (db/m) and spontaneously diabetic (db/db) mice on diabetic retinopathy. The db/db mice with 8 weeks of age were randomized to receive a unique intravenous injection of PBS or 0,5x105 db/m EOCs or 0,5x105 db/db EOCs. Four weeks later, the animals were euthanized and the eyes enucleated. For in vitro study, EOC-CM was generated from db/m and db/db EOCs cultures. rMCs were exposed for 24h to NG or HG combined or not with db/m or db/db EOC-CMs. In diabetic rats, there was an increase of DR and oxidative damage markers, accompanied by decrease in SIRT1 protein followed by lysine-310-p65-NF?B acetylation. The treatment with cells from db/m significantly reduced all the above-mentioned, but interestingly the treatment with cells from db/db mice fully restored the above alterations to normal levels. rMCs exposed to HG displayed GFAP and VEGF expression up regulated, accompanied by increase in Nox4 expression and ROS levels, and acetyl-lysine-310-p65-NF?B. SIRT1 protein expression and activity were markedly reduced in diabetic milieu conditions. The treatment with both EOC-CMs prevented all these abnormalities, but db/db EOC-CM fully restored to NG conditions. This study demonstrates that endocrine capacity of EOCs is effective in improving retinal SIRT1 pathway thus protecting the retina from diabetic milieu insult. In summary, compelling novel evidence is provided herein that either through oral administration of polyphenol enriched cocoa or cell therapy with EOCs, conferred retinal neuroprotection against diabetic insults in animal models. The identification of SIRT-1 as a potential therapeutic target in the treatment of diabetic retinopathy may provide new perspective in the pharmacological treatment of this diabetic complication / Doutorado / Clinica Medica / Doutor em Clínica Médica

A Role for c-Jun Kinase (JNK) Signaling in Glial Engulfment of Degenerating Axons: A Dissertation

The central nervous system (CNS) is composed of two types of cells: neurons that send electrical signals to transmit information throughout the animal and glial cells. Glial cells were long thought to be merely support cells for the neurons; however, recent work has identified many critical roles for these cells during development and in the mature animal. In the CNS, glial cells act as the resident immune cell and they are responsible for the clearance of dead or dying material. After neuronal injury or death, glial cells become reactive, exhibiting dramatic changes in morphology and patterns of gene expression and ultimately engulfing neuronal debris. This rapid clearance of degenerating neuronal material is thought to be crucial for suppression of inflammation and promotion of functional recovery, but molecular pathways mediating these engulfment events remain poorly defined. Drosophila melanogaster is a genetically tractable model system in which to study glial biology. It has been shown that Drosophila glia rapidly respond to axonal injury both morphologically and molecularly and that they ultimately phagocytose the degenerating axonal debris. This glial response to axonal debris requires the engulfment receptor Draper and downstream signaling molecules dCed-6, Shark, and Rac1. However, much remains unknown about the molecular details of this response. In this thesis I show that Drosophila c-Jun kinase (dJNK) signaling is a critical in vivo mediator of glial engulfment activity. In response to axotomy, glial dJNK signals through a cascade involving the upstream MAPKKKs Slipper and TAK1, the MAPKK MKK4, and ultimately the Drosophila AP-1 transcriptional complex composed of JRA and Kayak to initiate glial phagocytosis of degenerating axons. Interestingly, loss of dJNK also blocked injury-induced up-regulation of Draper levels in glia and glial-specific over-expression of Draper was sufficient to rescue phenotypes associated with loss of dJNK signaling. I have identified the dJNK pathway as a novel mediator of glial engulfment activity and show that a primary role for the glial Slipper/Tak1→MKK4→dJNK→dAP-1 signaling cascade is activation of draper expression after axon injury.

Efekat akutnog izlaganja peroralno unetog akrilamida na histološke strukture želuca pacova soja Wistar / The effect of acute exposure to orally ingested acrylamide on histological structure of stomach in Wistar rats

<p>Akrilamid je toksična hemijska supstanca koja ima vrlo &scaron;iroku primenu u hemijskoj industriji, a 2002. godine otkriveno je njegovo prisustvo u namirnicima bogatim skrobom koje se pripremaju na visokim temperaturama. U poslednjh desetak godina primećen je veliki porast gastrointestinalnih tegoba u ljudskoj populaciji. Cilj istraživanja bio je ispitati patohistolo&scaron;ke promene u tkivu želuca pacova soja Wistar izazvanih peroralnim aplikovanjem akrilamida i na taj način povući paralelu sa mogućim gastrointestinalnim tegobama nastalim kao posledica konzumiranja hrane bogate akrilamidom. U istraživanju je ispitivano 6 grupa od po 5 eksperimentalnih životinja (pacovi soja Wistar). Dve kontrolne grupe kojima je peroralno aplikovana destilovana voda i koje su žrtvovane posle 24h i 72h; dve eksperimentalne kojima je peroralno aplikovan akrilamid u dnevnoj dozi od 25 mg/kg i koje su žrtvovane posle 24h i 72h; dve eksperimentalne grupe kojima je peroralno aplikovan akrilamid u dnevnoj dozi od 50 mg/kg i koje su žrtvovane posle 24h i 72h. Na histolo&scaron;kom materijalu tkiva želuca primenjena je kvalitativna histolo&scaron;ka analiza pod svetlosnim mikroskopom, semikvantitativna procena tipa mucina u epitelnim ćelijama sluznice želuca, prisustvo limfocita i granulocita u sluznici želuca, stereolo&scaron;ka merenja pojedinih kompartmana zida želuca, linearna merenja broja i veličine ganglijskih ćelija u Maissner-ovom i Auerbach-ovom nervnom pleksusu, kao i broj mastocita u lamini propriji sluznice i podsluznici želuca. Dobijene vrednosti merenih parametara su potom statistički obrađene. Nastale promene na tkivu želuca pacova soja Wistar se ogledaju u vidu blagog direktnog o&scaron;tećenja povr&scaron;nog epitela sa propratnom blagom inflamatornom reakcijom i blagom degranulacijom mastocita. U Maissner-ovom i Auerbach-ovom nervnom pleksusu su smanjene volumenske gustine nervnih vlakana i ganglijskih ćelija, kao i broj i veličina ganglijskih ćelija. Direktno toksično delovanje na epitel dovodi do posledične obnove epitela, te je potvrđeno prisustvo nezrelijih oblika mukoproduktivnih ćelija koje sadrže kisele, AB pozitivne mucine. Ispitani inflamatorni i degenerativni parametri pokazuju pozitivnu korelaciju u odnosu na dozu i/ili dužinu ekspozicije akrilamidu. Primena akrilamida peroralno pokazala je da postoje patohistolo&scaron;ke promene na tkivu želuca u vidu direktnog toksičnog o&scaron;tećenja epitela, inflamatorne reakcije i o&scaron;tećenja nervnih pleksusa. Poznavanjem mehanizma delovanja ove toksične materije moguće je primeniti adekvatnu prevenciju u ishrani i izvr&scaron;iti odgovarajući izbor terapijskih metoda.</p> / <p>Acrylamide is a toxic chemical substance with wide implementation in chemical industry. In 2002 it was discovered the presence of acrylamide in foods rich in starch which are prepared at high temperatures. In the last ten years there is a large increase in gastrointestinal illnesses in human population. The aim of this study was to investigate the histopathological changes in the gastric tissue in Wistar rats induced with injection of oral acrylamide and thus draw a parallel with possible gastrointestinal problems arising as a result of the consumption of foods rich in acrylamide. The research was carried out 6 groups of 5 experimental animals (Wistar rats). Two control groups that are orally concomitant application of distilled water and which were sacrificed after 24h and 72h; two experimental groups which are orally administrated acrylamide in a daily dose of 25 mg / kg and that were sacrificed after 24h and 72h; two experimental groups which were orally administrated acrylamide in a daily dose of 50 mg / kg and that were sacrificed after 24h and 72h. On histological gastric tissue material is applied qualitative histological analysis by light microscopy, semi-quantitative assessment of the type of mucin in epithelial cells of the stomach lining, the presence of lymphocytes and granulocytes in gastric mucosa, stereological measurements of individual compartments of the stomach wall, linear measuring the number and size of ganglion cells in the Maissner and Auerbach&#39;s nerve plexus, and the number of mast cells in the lamina propria of the mucosa and in the submucosis of the stomach. Obtained values of measured parameters were statistically processed. Histological changes in the stomach tissue of Wistar rats are seen as a direct slight damage of the surface epithelium, with accompanynig mild inflammatory reaction and the degranulation of mast cells. The Meissner&#39;s and Auerbach&#39;s nerve plexus decreased volume density of nerve fibers and ganglion cells, as well as the number and size of the ganglion cells. Directly toxic effect on epithelium leads to the result of the reconstruction of the epithelium, which is confirmed by the presence of immature form of mucoproductive cells which contain acid, AB positive mucins. Examined inflammatory and degenerative parameters show a positive correlation with respect to dose and / or a time of exposition to acrylamide. Acrylamide oral application revealed that there are histologic changes in the stomach tissue in the form of a direct toxic damage to the epithelium, inflammatory reaction and damage to the nerve plexus. Knowing the mechanism of action of these toxic substances allows to apply adequate prevention in nutrition and make an appropriate choice of therapeutic methods.</p>

Para avaliar a eficácia do método de pré-degeneração em nervos ciáticos de ratos durante diferentes intervalos de tempo, foram realizados estudos histomorfométricos de cortes laminares obtidos ao nível do enxerto e no segmento distal do nervo receptor. Os resultados foram comparados com a técnica convencional de enxertia nervosa. Verificou-se que o tempo de pré-degeneração interfere na regeneração de novos axônios e que o período mais adequado para sua utilização situou-se ao redor de 2 semanas no modelo experimental adotado / In order to evaluate the efficacy of a predegeneration method in rat sciatic nerves during different periods of time, histomorphometric studies were performed at the graft and distal segment sites of the recipient nerves. The results were compared with the conventional nerve grafting technique. It was shown that the period of predegeneration interfered in the regeneration of new axons and the most favorable time for its use is around 2 weeks, in this experimental model

Estudo experimental comparativo entre enxerto de nervo convencional e enxerto de nervo preservado a frio / Experimental comparative study between conventional nerve graft and cold preserved nerve graft

INTRODUÇÃO: A reparação das lesões de nervos periféricos com perda extensa de substância, onde a sutura direta não é viável, ainda apresenta nos dias atuais resultados variáveis e dependentes de diversos fatores. O tratamento mais comumente utilizado nestes casos é a auto-enxertia de nervos, com sacrifício de um nervo de outra região do corpo, procedimento que, entretanto, pode trazer algumas dificuldades e consequências. Desta forma, a busca por novas técnicas, como a possibilidade de utilização de nervos preservados em baixas temperaturas, representa um avanço inestimável no campo da reparação de lesões nervosas. OBJETIVO: O objetivo deste estudo foi realizar avaliações funcionais, eletrofisiológicas e histomorfométricas que permitam comparar a regeneração nervosa autógena em enxerto convencional versus enxerto preservado a frio, em modelo experimental de ratos, após denervação a fresco ou conservação de um segmento do nervo em baixa temperatura por 14 dias e por 50 dias. MÉTODOS: Foram utilizados 20 ratos Wistar de peso e idades aproximadamente iguais, divididos em quatro grupos de cinco animais. Os grupos 1 e 3 serviram de controle respectivamente para os grupos 2 e 4, utilizando enxertia de nervo convencional por 14 dias (grupo 1) e por 50 dias (grupo 3). O grupo 2 utilizou enxertia de nervo preservado a 4 graus Celsius em solução Celsior® por 14 dias, enquanto o grupo 4 foi submetido à preservação a frio na mesma solução por 50 dias. Foram realizadas análises funcionais da marcha, análises de potenciais evocados e análises histomorfométricas dos animais em diversos momentos. As análises funcionais utilizaram uma aparelhagem própria para estudo da marcha em pequenos animais de experimentação, denominada catWalk®, que fornece medidas estáticas e dinâmicas da marcha, com parâmetros como a pressão em relação à pata contralateral e a área máxima da impressão plantar do animal, tendo sido captados os dados antes do procedimento de retirada do enxerto e após a realização da enxertia, neste último caso com avaliações quinzenais até que tenham sido completados 60 dias de pós- operatório. As análises de potenciais evocados motores analisaram a latência e a amplitude dos estímulos nervosos e foram realizadas 60 dias após os procedimentos de enxertia. As análises microscópicas observaram a contagem de axônios mielinizados e a área destas fibras nervosas nas regiões proximal e distal aos reparos, aos 60 dias após os procedimentos, comparando também as relações entre a região distal e proximal de cada um destes parâmetros através dos índices de regeneração e mudança de área. RESULTADOS: A enxertia com nervo preservado a frio por 14 dias apresentou resultado funcional semelhante ao seu grupo controle na análise da área máxima de contato e da pressão máxima de contato da pata operada em todas as avaliações. Já a conservação do enxerto a frio por 50 dias resultou em superioridade funcional em todos as avaliações em relação a seu grupo controle. Os estudos eletrofisiológicos mostraram cada grupo de enxertia preservada a frio com resultados similares a seu grupo controle, tanto em relação à latência, quanto à amplitude nos dois músculos avaliados. As análises histomorfométricas resultaram em índices de regeneração e de mudança de área semelhantes na comparação entre os grupos 60 dias após os procedimentos de enxertia. CONCLUSÕES: A conservação a frio do enxerto de nervo durante 14 dias e durante 50 dias apresentou resultados funcionais da regeneração iguais ou superiores aos enxertos convencionais e resultados eletrofisiológicos e histológicos semelhantes aos respectivos grupos controle de enxertos convencionais, demonstrando um futuro promissor para a utilização clínica de enxertos preservados a frio em um \"banco de nervos\" / INTRODUCTION: The repair of peripheral nerve injuries with extensive loss of substance, where direct suture is not feasible, at the present time still has variable results and dependence on many factors. The treatment most commonly used in these cases is the nerve autograft, with sacrifice of a nerve from another region of the body. This procedure, however, can sometimes lead to some difficulties and consequences. Therefore, the search for new techniques such as the possibility of using cold preserved nerves, is a great advancement in the field of repairing nerve damage. OBJECTIVE: The purpose of this study was to perform functional, electrophysiological and histomorphometric evaluations to compare conventional autografts versus cold-preserved autografts of the sciatic nerves of rats, after fresh denervation or conservation of a nerve segment at low temperature for 14 days and 50 days. METHODS: 20 Wistar rats of approximately equal ages and weight were divided into 4 groups of 5 animals. Groups 1 and 3 were treated with a conventional nerve graft after denervation for 14 days and 50 days, respectively; they served as controls for groups 2 and 4, which were treated with cold-preserved nerve grafts immersed in a Celsior® solution at 4 degrees Celsius for 14 and 50 days, respectively. Functional gait analysis, evoked potential analysis and histomorphometric analysis of the animals were performed at different times. Functional analysis used equipment for gait study in small animal experiments, called catWalk®, which provides static and dynamic measurements, with parameters such as pressure relative to contralateral paw and the maximum area of the footprint of the animal, and these data were captured before the graft withdrawal procedure and after grafting, in this latter case the functional analysis was made every 15 days until they had been completed 60 days after surgery. The motor evoked potential analysis examined the latency and amplitude of nerve stimuli and was made 60 days after the grafting procedures. The microscopic analysis measured myelinated axons and the area of these nerve fibers in the proximal and distal regions to the repair sites at the end of 60 days after the procedures, also comparing the relationship between the distal and proximal regions of each of these parameters through the regeneration and area change rates. RESULTS: Cold preservation of nerve graft for 14 days showed functional results similar to those of its control group for the maximum contact area and for the maximum pressure intensity of the operated paw in all evaluations. Cold preservation of nerve graft for 50 days resulted in functional superiority in all assessments compared with its control group. Cold preservation of nerve graft for 14 days and 50 days showed electrophysiological results similar to those of their respective control groups, both in terms of latency, as to the amplitude in the two muscles evaluated. Histomorphometric analysis showed similar regeneration and area change rates for all the groups 60 days after the grafting procedures. CONCLUSIONS: The cold preservation of nerve grafts for 14 days and 50 days showed similar or superior functional results and similar electrophysiological and histological results compared with their respective conventional graft control groups, indicating a promising future for the clinical utilization of cold preserved grafts in a \"nerve bank\"

A study of the enteric nervous system and interstitial cells of Cajal in a mouse model of Alzheimer's disease.

Peristalsis is modulated by slow waves that originate from pacemaker cells called interstitial cell of Cajal (ICC). ICCs also modulate and transduce inputs from the enteric nervous system (ENS) to the smooth muscle. Recent studies in rodents and humans demonstrated that a decrease in ICC number and enteric neurodegeneration during ageing is associated with difficult bowel movements and constipation. By studying ICC and the ENS during normal aging and in a mouse model (Tg2576) of Alzheimer’s disease (AD) where cholinergic loss may be exaggerated, we may gain new perspectives on the treatment of degenerative diseases. The aim of the present study therefore, was to investigate the morphological and functional changes of the GI tract of mice during ageing and in an AD mouse model over-expressing amyloid precursor protein (APP) using an isolated tissue approach. / Whole mount immunohistochemistry of 6-month-old Tg2576 mice and their age-matched wild type (WT) controls revealed that there were significant losses of enteric neurons in the duodenum (P < 0.05) and ileum (P < 0.001), and of GFAP-positive enteric glial cells in the ileum (P < 0.001). There was also a loss of S100-positive glial cells in the antrum (pacemaker region in the stomach) (P < 0.05), ileum (P < 0.05) and colon (P < 0.05). These results indicated the alteration of the ENS during the early stages of AD. There were no differences in ICC arears of all GI regions between 6-month-old Tg2576 mice and their age-matched WT controls (P > 0.05), and there was no alteration of basal peristaltic rhythm during the early stages of AD. The non-significant increase of GFAP to S100 enteric glial cell ratio in the duodenum and colon might indicate an ongoing inflammatory process in these two GI regions during the early stages of AD. / The presence of insoluble amyloid plaques was studied using Aβ immunohistochemistry, Sirius red assay and Thioflavin-T assay on paraffin wax sections. The aggregation of amyloid plaques was observed in all the GI regions of 6-month-old Tg2576 mice and the levels of amyloid plaque varied in different regions. No amyloid plaques were found in the GI tract of 6-month-old WT animals excepting the colon. The increase in formation of amyloid plaques might be correlated to the losses of enteric neurons and enteric glial cells during the early stages of AD. / Western blot analysis was performed on frozen sections of tissues from the ileum and colon to investigate whether there were changes in choline acetyltransferase (ChAT, from excitatory neurons), neuronal nitric oxide synthase (nNOS, from inhibitory neurons), glial cell line-derived neurotrophic factor (GDNF, from enteric glia) and soluble Aβ oligomers between 6-month-old Tg2576 mice and WT controls. nNOS expression significantly increased in the ileum (P < 0.05) but not in the colon (P > 0.05) of Tg2576 mice compared with WT controls. There were no differences in the expressions of ChAT, GDNF and Aβ oligomers (docecamer, nonamer and hexamer) in the ileum and colon between Tg2576 mice and WT controls (P > 0.05). These results imply that there is an increase in the inhibitory signal in the GI tract during the early stages of AD but soluble Aβ oligomers might not be the cause of neuronal and glial losses in the GI tract. / Following histological and biochemical studies of different GI regions, slow wave signals from the antrum and ileum were measured using a microelectrode array (MEA) system. The dominant frequencies (DFs) and power distributions were measured and these served as parameters for measuring functional changes in the GI tract during ageing in ICR mice and the early stages of AD. In the presence of nifedipine, nicotine significantly stimulated the slow wave activities in the antrum and ileum of 3-month-old (P < 0.05) and 6-month-old (P < 0.05) ICR mice but failed to trigger the slow wave activities in 12-month-old (P > 0.05) ICR mice, suggesting the neurodegeneration might begin with the age between 6 and 12 months. With the addition of tetrodotoxin, nicotine failed to stimulate the slow wave activities in the antrum and ileum of three age groups (P > 0.05) and it showed that nicotine only acted on enteric neurons to trigger the ICC activities. There were no differences in the antral and ileal baseline recordings between 6-month-old Tg2576 mice and their age-matched WT controls (P > 0.05). However, nicotine significantly increased DFs and tachygastria ranges of the antrum and ileum in WT controls (P < 0.05) but failed to increase electrical activitiy of the antrum and ileum in Tg2576 mice (P > 0.05), thus suggesting a loss of neuronal and/or glial cells in the GI tract during the early stages of AD. / In conclusions, these findings suggest the mouse model for AD has morphological, biochemical and functional changes in the GI tract. The present studies provide a foundation for the investigation of degenerative diseases and support the hypothesis that the ENS may be the gateway for the early pathological changes in the central INTRODUÇÃO: A reparação das lesões de nervos periféricos com perda extensa de substância, onde a sutura direta não é viável, ainda apresenta nos dias atuais resultados variáveis e dependentes de diversos fatores. O tratamento mais comumente utilizado nestes casos é a auto-enxertia de nervos, com sacrifício de um nervo de outra região do corpo, procedimento que, entretanto, pode trazer algumas dificuldades e consequências. Desta forma, a busca por novas técnicas, como a possibilidade de utilização de nervos preservados em baixas temperaturas, representa um avanço inestimável no campo da reparação de lesões nervosas. OBJETIVO: O objetivo deste estudo foi realizar avaliações funcionais, eletrofisiológicas e histomorfométricas que permitam comparar a regeneração nervosa autógena em enxerto convencional versus enxerto preservado a frio, em modelo experimental de ratos, após denervação a fresco ou conservação de um segmento do nervo em baixa temperatura por 14 dias e por 50 dias. MÉTODOS: Foram utilizados 20 ratos Wistar de peso e idades aproximadamente iguais, divididos em quatro grupos de cinco animais. Os grupos 1 e 3 serviram de controle respectivamente para os grupos 2 e 4, utilizando enxertia de nervo convencional por 14 dias (grupo 1) e por 50 dias (grupo 3). O grupo 2 utilizou enxertia de nervo preservado a 4 graus Celsius em solução Celsior® por 14 dias, enquanto o grupo 4 foi submetido à preservação a frio na mesma solução por 50 dias. Foram realizadas análises funcionais da marcha, análises de potenciais evocados e análises histomorfométricas dos animais em diversos momentos. As análises funcionais utilizaram uma aparelhagem própria para estudo da marcha em pequenos animais de experimentação, denominada catWalk®, que fornece medidas estáticas e dinâmicas da marcha, com parâmetros como a pressão em relação à pata contralateral e a área máxima da impressão plantar do animal, tendo sido captados os dados antes do procedimento de retirada do enxerto e após a realização da enxertia, neste último caso com avaliações quinzenais até que tenham sido completados 60 dias de pós- operatório. As análises de potenciais evocados motores analisaram a latência e a amplitude dos estímulos nervosos e foram realizadas 60 dias após os procedimentos de enxertia. As análises microscópicas observaram a contagem de axônios mielinizados e a área destas fibras nervosas nas regiões proximal e distal aos reparos, aos 60 dias após os procedimentos, comparando também as relações entre a região distal e proximal de cada um destes parâmetros através dos índices de regeneração e mudança de área. RESULTADOS: A enxertia com nervo preservado a frio por 14 dias apresentou resultado funcional semelhante ao seu grupo controle na análise da área máxima de contato e da pressão máxima de contato da pata operada em todas as avaliações. Já a conservação do enxerto a frio por 50 dias resultou em superioridade funcional em todos as avaliações em relação a seu grupo controle. Os estudos eletrofisiológicos mostraram cada grupo de enxertia preservada a frio com resultados similares a seu grupo controle, tanto em relação à latência, quanto à amplitude nos dois músculos avaliados. As análises histomorfométricas resultaram em índices de regeneração e de mudança de área semelhantes na comparação entre os grupos 60 dias após os procedimentos de enxertia. CONCLUSÕES: A conservação a frio do enxerto de nervo durante 14 dias e durante 50 dias apresentou resultados funcionais da regeneração iguais ou superiores aos enxertos convencionais e resultados eletrofisiológicos e histológicos semelhantes aos respectivos grupos controle de enxertos convencionais, demonstrando um futuro promissor para a utilização clínica de enxertos preservados a frio em um \"banco de nervos\" / INTRODUCTION: The repair of peripheral nerve injuries with extensive loss of substance, where direct suture is not feasible, at the present time still has variable results and dependence on many factors. The treatment most commonly used in these cases is the nerve autograft, with sacrifice of a nerve from another region of the body. This procedure, however, can sometimes lead to some difficulties and consequences. Therefore, the search for new techniques such as the possibility of using cold preserved nerves, is a great advancement in the field of repairing nerve damage. OBJECTIVE: The purpose of this study was to perform functional, electrophysiological and histomorphometric evaluations to compare conventional autografts versus cold-preserved autografts of the sciatic nerves of rats, after fresh denervation or conservation of a nerve segment at low temperature for 14 days and 50 days. METHODS: 20 Wistar rats of approximately equal ages and weight were divided into 4 groups of 5 animals. Groups 1 and 3 were treated with a conventional nerve graft after denervation for 14 days and 50 days, respectively; they served as controls for groups 2 and 4, which were treated with cold-preserved nerve grafts immersed in a Celsior® solution at 4 degrees Celsius for 14 and 50 days, respectively. Functional gait analysis, evoked potential analysis and histomorphometric analysis of the animals were performed at different times. Functional analysis used equipment for gait study in small animal experiments, called catWalk®, which provides static and dynamic measurements, with parameters such as pressure relative to contralateral paw and the maximum area of the footprint of the animal, and these data were captured before the graft withdrawal procedure and after grafting, in this latter case the functional analysis was made every 15 days until they had been completed 60 days after surgery. The motor evoked potential analysis examined the latency and amplitude of nerve stimuli and was made 60 days after the grafting procedures. The microscopic analysis measured myelinated axons and the area of these nerve fibers in the proximal and distal regions to the repair sites at the end of 60 days after the procedures, also comparing the relationship between the distal and proximal regions of each of these parameters through the regeneration and area change rates. RESULTS: Cold preservation of nerve graft for 14 days showed functional results similar to those of its control group for the maximum contact area and for the maximum pressure intensity of the operated paw in all evaluations. Cold preservation of nerve graft for 50 days resulted in functional superiority in all assessments compared with its control group. Cold preservation of nerve graft for 14 days and 50 days showed electrophysiological results similar to those of their respective control groups, both in terms of latency, as to the amplitude in the two muscles evaluated. Histomorphometric analysis showed similar regeneration and area change rates for all the groups 60 days after the grafting procedures. CONCLUSIONS: The cold preservation of nerve grafts for 14 days and 50 days showed similar or superior functional results and similar electrophysiological and histological results compared with their respective conventional graft control groups, indicating a promising future for the clinical utilization of cold preserved grafts in a \"nerve bank\"

Para avaliar a eficácia do método de pré-degeneração em nervos ciáticos de ratos durante diferentes intervalos de tempo, foram realizados estudos histomorfométricos de cortes laminares obtidos ao nível do enxerto e no segmento distal do nervo receptor. Os resultados foram comparados com a técnica convencional de enxertia nervosa. Verificou-se que o tempo de pré-degeneração interfere na regeneração de novos axônios e que o período mais adequado para sua utilização situou-se ao redor de 2 semanas no modelo experimental adotado / In order to evaluate the efficacy of a predegeneration method in rat sciatic nerves during different periods of time, histomorphometric studies were performed at the graft and distal segment sites of the recipient nerves. No / Microglial activation and behavioral abnormalities occur before neuronal loss in experimental murine prion disease; the behavioral changes coincide with a reduction in synaptic plasticity. Because synaptic plasticity depends on an intact perineuronal net (PN), a specialized extracellular matrix that surrounds parvalbumin (PV)-positive GABAergic (gamma-aminobutyric acid [GABA]) inhibitory interneurons, we investigated the temporal relationships between microglial activation and loss of PN and PV-positive neurons in ME7 murine prion disease. Anesthetized C57Bl/6J mice received bilateral intracerebral microinjections of ME7-infected or normal brain homogenate into the dorsal hippocampus. Microglial activation, PrP accumulation, the number of PV-positive interneurons, and Wisteria floribunda agglutinin-positive neurons (i.e. those with an intact PN) were assessed in the ventral CA1 and subiculum at 4, 8, 12, 16, and 20 weeks postinjection. Hippocampal areas and total neuron numbers in the ventral CA1 and subiculum were also determined. Loss of PN coincided with early microglial activation and with a reduction in synaptic plasticity. No significant loss of PV-positive interneurons was observed. Our findings suggest that the substrate of the earliest synaptic and behavioral abnormalities in murine prion disease may be inflammatory microglia-mediated degradation of the PN.
