Herrera Quiero, Mauricio January 2003 (has links)

Maturana Ramírez, Andrea January 2007 (has links)

Attityder till NO

Lindström, Madelen January 2008 (has links)

Biological effects of dietary bleached kraft pulp mill effluent on mink (Mustela vison)

Smits, Judit Emmy Geraldine 01 January 1996 (has links)
Semi-aquatic predators such as mink are exposed to anthropogenic contaminants directly through the water and through bioaccumulation in the food chain. The biological impact of dietary bleached kraft pulp mill effluent (BKME) on mink (Mustela vison) was investigated. In a pilot study and two subchronic studies of 8 and 7 month duration, mink were fed diets containing 75% (year 1) and 45% (year 2) fish caught downstream of a BKME discharge point, and drinking water contained 25% BKME. In year two, the 45% fish diet had 15% soft-wood run BKME incorporated into the feed. The investigation was tiered. In the pilot study, behavioural, clinical, biochemical, hematological, and pathological effects were investigated. Repeating these variables, reproductive factors were added in Year one, while in Year two, hepatic enzyme (ethoxyresorufin-O-deethylase (EROD)) induction, cell mediated and humoral immune function, and hepatic vitamin A levels were evaluated. In vivo and in vitro immunotoxicity assays were developed for mink. Peripheral blood mononuclear cell (PBMC) proliferation was measured in response to mitogens in vitro. An enzyme-linked immunosorbent assay for antibody detection was developed for mink. Delayed type hypersensitivity (DTH) tests and antibody production responses were used to measure cell-mediated and humoral immunity in vivo in the experimental mink. No adverse effects were found on behavioural, gross pathological, histopathological, hematological or biochemical variables, on gestation, kit birth weight, kit survival, libido, estrus, sperm quality or hormone levels. In the Year two subchronic study, the relative liver size was increased in BKME-exposed males. Hepatic EROD activity was 1.8 times greater in exposed females (p = 0.0001) and 2.0 times greater in exposed males (p = 0.0004) relative to control mink. No difference in PBMC proliferation was seen between the control and exposed mink with any of the mitogens used. The DTH response was impaired (p = 0.014), while the antibody response was enhanced (p = 0.029) in the BKME-exposed mink. Hepatic vitamin A levels were not different in the females (mid-lactation), but were significantly decreased in the BKME-exposed males (post-breeding) (p = 0.0002). These changes represent a primary effect of bleached pulp mill effluent on the immune system, hepatic vitamin A stores, and hepatic detoxification enzyme system in exposed mink. Hepatic EROD activity provides a useful indicator for evidence of exposure to environmental toxicants in mink. The change in the immune response is occurring at the level of T lymphocyte differentiation, and therefore, affects the relative proportions of T lymphocyte subpopulations which are dedicated to cell mediated, or, T lymphocyte dependent, antibody mediated immunity. Immune deviation seen in the female mink is not associated with changes in hepatic vitamin A stores, while the decreased vitamin A in the males has an unknown effect on their immune response. The biological impact of bleached kraft pulp mill effluent does not cause dramatic or subclinical signs of toxicity in exposed mink. However, the interference with hepatic vitamin A storage, and changes to the immune response, present concerns regarding long term effects on health, reproduction and longevity in exposed mink.

N2s2 chelating agents as cys-x-cys biomimics for fe(no) and fe(no)2 complexes

Chiang, Chao-Yi 16 August 2006 (has links)
Nitric oxide plays an important role in many biological functions. A metallo derivative in biological systems is a protein-bound dinitrosyl iron complex (DNIC), which results from iron-sulfur cluster degradation in the presence of excess NO. Through model complexes I have examined the fundamental properties of a dithiolato-Fe(NO)2 complex, bismercaptoethandiazacyclooctane iron dinitrosyl or (H+bme-daco)Fe(NO)2 as a biomimic of dicysteinate coordination of [Fe(NO)2]. This complex was prepared and fully characterized in my studies. The DNIC moiety is in its oxidized state, {Fe(NO)2}9. Through reaction studies, monitored by IR spectroscopy (H+N2S2)Fe(NO)2 (N2S2 = bme-dach. Bme-pda) has been shown to transfer NO to FeIII in (TPP)FeCl (TPP = meso-tetraphenylporphyrin) as NO-. The remaining mononitrosyl converts into complex (N2S2)Fe(NO). The (N2S2)Fe(NO) complexes (N2S2 = bme-daco, bme*-daco, bme-dach) were prepared by direct reaction of dimeric [(N2S2)Fe]2 and NO gas. The analogous (N2S2)Co(NO) complex (N2S2 = bme-dach) has also been prepared. The series of square pyramidal (N2S2)M(NO) have been studied by cyclic voltammetry and ν(NO) IR spectroscopy.

Beneficial effects of dietary L-arginine supplementation to diabetic rats

Kohli, Ripla 30 September 2004 (has links)
Diabetic rats exhibit decrease in plasma arginine, NO synthesis and tetrahydrobiopterin in endothelial cells (EC). Treatment with L-arginine may be beneficial for enhancing NO synthesis in diseases associated with endothelial dysfunction. However, little is known about the mechanism responsible for the stimulatory effect of arginine on endothelial NO synthesis. We hypothesized that dietary arginine supplementation increases BH4 for NO synthesis in EC of diabetic rats, thereby preventing endothelial dysfunction. In experiment I, streptozotocin (STZ) induced-diabetic male Sprague Dawley (SD) rats (a model of type-I diabetes) were individually pair-fed a casein-based diet on the basis of feed intake (per kg body weight) of non-diabetic SD rats. Addition of arginine-HCl or alanine to drinking water for the rats were adjusted daily to ensure isonitrogenous provision per kg body weight. In non-diabetic rats, arginine supplementation increased plasma arginine (144%), plasma insulin (44%), EC arginine (88%), EC BH4 (106%) and EC NO synthesis (80%), compared with alanine treatment. In diabetic rats, arginine supplementation reduced body weight loss (36%), and plasma glucose (54%), and increased plasma arginine (110%), plasma insulin (209%), EC arginine (173%), EC BH4 (128%) and EC NO synthesis (125%), compared with alanine treatment. In experiment II, male Zucker diabetic fatty (ZDF) rats (a model of type-II diabetes) were individually pair-fed a Purina 5008 diet on the basis of feed intake by alanine-treated diabetic rats (per kg body wt). Addition of arginine-HCl or alanine to drinking water for the rats was adjusted daily to ensure isonitrogenous provision per kg body weight. Arginine supplementation to ZDF rats did not affect plasma glucose and insulin, reduced epidididmal fat (30%), abdominal fat (43%) and body weight gain (18%), and increased plasma arginine (273%), EC arginine (197%), EC BH4 (120%) and EC NO synthesis (122%), compared with alanine-treated ZDF rats. These results show that dietary L-arginine supplementation increases BH4 and NO synthesis in EC of both STZ-diabetic and ZDF rats. Strikingly, arginine treatment prevented hyperglycemia in STZ-diabetic SD rats and reduced obesity in ZDF rats. Collectively, results demonstrate that oral administration of arginine is beneficial for both type-I and type-II diabetic rats.
