A new concept of regenerative proton exchange membrane fuel cell (R-­‐PEMFC) / Modélisation et simulation d’une pile à combustible réversible

Les travaux précédents trouvés dans la littérature ont mis l'importance sur la pile à combustible PEM ou électrolyseur PEM. Certains articles ont étudié également la pile à combustible réversible et le système d'alimentation en hydrogène par énergie solaire en intégrant à la fois la pile à combustible et électrolyseur. Contrairement à un « Unitised regenerative fuel cell (URFC)», notre conception a un compartiment individuel pour chaque système de PEM-Fuel Cell et d'electrolyseur-PEM et nommé Quasi - URFC. Grâce à ce nouveau concept, l'objectif principal est de réduire le coût de la pile à combustible régénératrice (RFC) en minimisant le rapport de surface superficielle géométrique du catalyseur de l'assemblage membrane électrodes (AME) des deux modes dans la cellule. D'ailleurs, nous visons également à construire un RFC plus compact, léger et portable par rapport à une pile à combustible ou l'électrolyseur classique. Ce travail de recherche est divisé en trois parties : la modélisation et simulation numérique, l'assemblage du prototype et le travail d'expérimentation. Quant à la partie de modélisation, un modèle physique multi-2D a été développé dans le but d'analyser les performances d'une pile à combustible à régénérée à trois-compartiments, qui se compose d'une piles à combustible et d'électrolyseur. Ce modèle numérique est basée sur la résolution des équations de conservation de masse, du momentum, des espèces et du courant électrique en utilisant une approche par éléments finis sur des grilles 2D . Les simulations permettent le calcul de la vitesse, de la concentration de gaz, la densité de courant et les distributions de potentiels en mode pile à combustible et en mode d'électrolyse, ainsi nous aider à prédire le comportement de quasi - RFC. En outre, l'assemblage du premier prototype du nouveau concept de pile à combustible à combustible régénérée a été achevée et testée au cours des trois années d'études dans le cadre d'une thèse. Les résultats expérimentaux de la 3 Compartiments R-PEMFC ont été prometteurs dans les deux modes, soit en mode piles à combustible et soit en mode d'électrolyseur. Ces résultats valideront ensuite les résultats de la simulation, obtenus auparavant par la modélisation. / The past works found in the literature have focused on either PEM fuel cell or electrolyzer-PEM. Some of the papers even studied the unitised reversible regenerative fuel cell (URFC) and the solar power hydrogen system by integrating both fuel cell and electrolyzer. Unlike the URFC, our design has an individual compartment for each PEMFC and E-PEM systems and named Quasi-URFC. With this new concept, the main objective is to reduce the cost of regenerative fuel cell (RFC) by minimizing the ratio of the catalyst’s geometric surface area of the membrane electrode assembly (MEA) of both cell modes. Apart from that, we also aim to build a compact, light and portable RFC.This research work is divided into three parts: the modeling, assembly of the prototype and the experimentation work. As for the modeling part, a 2D multi-physics model has been developed in order to analyze the performance of a three chamber-regenerative fuel cell, which consists of both fuel cell and electrolyzer systems. This numerical model is based on solving conservation equations of mass, momentum, species and electric current by using a finite-element approach on 2D grids. Simulations allow the calculation of velocity, gas concentration, current density and potential's distributions in fuel cell mode and electrolysis mode, thus help us to predict the behavior of Quasi-RFC. Besides that, the assembly of the first prototype of the new concept of regenerative fuel cell has been completed and tested during the three years of PhD studies. The experimental results of the Three-Chamber RFC are promising in both fuel cell and electrolyzer modes and validate the simulation results that previously obtained by modeling.

Propuesta de mejora del nivel de servicio de una vía urbana mediante el uso del carril reversible

El estudio evalúa el impacto de aplicar el carril reversible para reducir la congestión vehicular en una zona urbana. La evaluación se realizó usando simulación y desarrollando a su vez escenarios de ciclo de semáforo que se acople a su mejoría, de esta manera conocer los cambios de reducción de sus parámetros como demoras, tiempos de viaje y longitud de cola. El análisis parte de los datos recopilados del estado real del tráfico a través de la implementación y simulación microscópica con la aplicación del software VISSIM 9.0. Una vez calibrado y validado el modelo se implementa el carril reversible en la Av. José Carlos Mariátegui entre la av. Magnolias – av. 1ro de mayo. Los resultados de acuerdo a los parámetros se obtuvieron la mejora del nivel de servicio en las intersecciones 1 pasando del nivel F al nivel E y en la intersección 2 del nivel E al nivel D. También, mejoro el tiempo de viaje en la intersección 1 a un 42% y la intersección 2 en un 15%. En cuanto a la simulación de los escenarios del semáforo implementado con el carril reversible, se realizó dos escenarios 1 y 2, lo cual, se tomará en cuenta el escenario 2 del ciclo de semáforo, ya que en ella se ve una gran mejora en la longitud de cola en un 53%. / The study evaluates the impact of applying the reversible lane to reduce traffic congestion in an urban area. The evaluation was carried out using simulation and in turn developing traffic light cycle scenarios that are coupled to its improvement, in this way knowing the reduction changes of its parameters such as delays, travel times and queue length. The analysis is based on the data collected from the real state of the traffic through the implementation and microscopic simulation with the application of the VISSIM 9.0 software. Once the model has been calibrated and validated, the reversible lane is implemented on Av. José Carlos Mariátegui between Av. Magnolias - av. May 1st. The results according to the parameters were obtained from the improvement of the service level at intersections 1, going from level F to level E and at intersection 2 from level E to level D. Also, the travel time at intersection 1 was improved. 42% and intersection 2 15%. Regarding the simulation of the traffic light scenarios implemented with the reversible lane, two scenarios 1 and 2 were carried out, which will take into account scenario 2 of the traffic light cycle, since it shows a great improvement in the tail length by 53%. / Trabajo de investigación

La congestión vehicular se ha convertido día tras día en una constante en las grandes ciudades del mundo y Lima es una de ellas. Sin embargo, existen formas de mitigar el tiempo que se pierde al momento de circular por estas vías de alto tránsito vehicular, en especial durante las horas pico de la noche como es el caso de la Av. José Carlos Mariátegui entre la Av. Las Magnolias y la Av. 1ero de Mayo. Por tal motivo, el presente trabajo consiste en proponer una alternativa de solución al problema para reducir un 30 % la congestión del tráfico vehicular, empleando para ello el carril reversible. Para la determinación de la factibilidad del empleo de esta solución se realizó previamente la modelación mediante el software VISSIM 9.0 de acuerdo a parámetros de nivel microscópico, los cuales fueron utilizados en la calibración y validación del modelo haciéndolo más cercano a la realidad considerando las características de la zona de estudio. Por otro lado, se realizó el cálculo del nivel de servicio mediante el manual de HCM 2010, que proporciona a una serie de pasos sucesivos para finalmente poder obtener el nivel de servicio de la Avenida en estudio. A partir de lo cual se concluye que la aplicación del carril reversible cumple la función de reducción del nivel de servicio pasando de un nivel “F” a un nivel “E” en la primera intersección, y de un nivel “E” a un nivel “D” en la segunda. Lo que representa una reducción de las demoras en 30%, tiempo de viaje en 29% y longitud de cola en 53 %. Por último, para complementar la mejora del nivel de servicio del carril reversible se propuso dos escenarios de semaforización la cual, el escenario 2 resulto ser la más óptima con los intervalos propuestos en las fases 1 y 2 sin alterar los ciclos actuales. / Day after day Traffic Congestion has become a constant in the great cities of the world and Lima is one of them. However, there are ways to mitigate the time that it’s lost in the moment to drive these high traffic roads, especially during peak hours at night, such as Av. José Carlos Mariátegui between Av. Las Magnolias and Av. 1ero de Mayo. For this reason, the present work consists of proposing an alternative solution to the problem to reduce vehicular traffic congestion by 30%, using the reversible lane. To determine the feasibility of using this solution, modeling was previously carried out using the VISSIM 9.0 software according to microscopic level parameters, which were used in the calibration and validation of the model, making it closer to reality considering the characteristics of the study zone. On the other hand, the calculation of the level of service was performed using the HCM 2010 manual, which provides a series of successive steps to finally be able to obtain the level of service for the avenue under study. From which it is concluded that the application of the reversible lane fulfills the function of reducing the level of service, going from a level “F” to a level “E” at the first intersection, and from a level “E” to a level "D" in the second. This represents a reduction of delays by 30%, travel time by 29% and queue length by 53%. Finally, to complement the improvement of the service level of the reversible lane, two traffic light scenarios were proposed, which, scenario 2 turned out to be the most optimal with the intervals proposed in phases 1 and 2 without altering the current cycles. / Tesis

Identification of monoamine oxidase inhibitors using a molecular modelling approach / Anke Pienaar

Monoamine oxidase (MAO) is an enzyme located on the outer mitochondrial membrane and is considered to be a target for the treatment of diseases such as Parkinson’s disease and depression. MAO may be classified into two isoforms, MAO-A and MAO-B. Since MAO-A and MAO-B catalyzes the metabolism of serotonin and dopamine, respectively, MAO-A inhibitors are used in the therapy of depression while MAO-B inhibitors are useful in the treatment of Parkinson’s disease. The older nonselective and irreversible MAO inhibitors, however, are not frequently used because they may ellicit potentially dangerous side effects such as the “cheese reaction”. The cheese reaction occurs when irreversible MAO-A inhibitors block the metabolism of tyramine in the gastrointestinal tract. Excessive amounts of tyramine subsequently enter the systemic circulation and cause a hypertensive reaction. This problem may be overcome by the development of selective MAO-B inhibitors and reversible MAO-A inhibitors. Selective MAO-B inhibitors do not cause the cheese reaction, because tyramine is metabolized, in the intestines, by MAO-A. Tyramine also has the ability to displace reversible MAO-A inhibitors and can subsequently be normally metabolized, thus not causing the cheese reaction. Several reseach groups are therefore involved in the discovery of reversible MAO-A and MAO-B inhibitors. As mentioned above, such drugs may be used in the treatment of depression and Parkinson’s disease. One approach is the de novo design of novel molecules with affinities for MAO-A and MAO-B active sites. In a second approach, existing drugs may be reappropriated as MAO inhibitors. With this approach, approved drugs are screened for the possibility that they, in addition to their action at the indicated target, also act as inhibitors of MAO-A and/or MAO-B. Such drugs may then be applied as MAO inhibitors in the treatment of depression and Parkinson’s disease. From a toxicological point of view, it is also of importance to identify MAO-A inhibitory activities among existing drugs as this will alert to the occurance of potential side effects such as the cheese reaction. In this study the second approach will be followed. This study will screen a virtual library of approved drugs for inhibitory activity towards MAO-A and MAO-B. Molecular modeling may be used to screen virtual libraries of drugs as potential inhibitors of the MAO enzymes. This may conveniently be achieved by employing structure-based or ligand-based pharmacophore models. In this study a virtual library of approved drugs was screened for secondary inhibitory activities towards the MAO isoforms with the use of structure-based pharmacophore models. There are several advantages to this approach. Molecular modeling aims at reducing the overall cost associated with the discovery and development of a new drug by identifying the most promising candidates to focus the experimental efforts on. It aids in understanding how a ligand binds to the active site of an enzyme. It is relatively easier to re-register a drug for a second pharmacological activity. This approach may also lead to drugs with a multi-target mode of action. The structure-based pharmacophores were constructed using the known crystallographic structures of MAO-A and MAO-B with the inhibitors, harmine and safinamide, complexed in the active sites, respectively. Employing the MAO-A and MAO-B structure-based pharmacophore model in the virtual screening of a library of approved drugs, 45 compounds were found to map to the MAO-A and MAO-B pharmacophore models. Among the hits, 29 compounds were selected for in vitro evaluation as MAO-A and MAO-B inhibitors. The IC50 values for these compounds were determined. After in vitro evaluation, 13 compounds showed inhibitory activity towards MAO. Of the 13 compounds 3 showed interesting inhibitory activities. These compounds included caffeine (IC50 = 0.761 μM for MAO-A and 5.08 μM for MAO-B), esomeprazole (IC50 = 23.2 μM for MAO-A and 48.3 μM for MAO-B) and leflunomide (IC50 = 19.1μM for MAO-A and 13.7 μM for MAO-B). The MAO inhibitory properties of caffeine and esomeprazole were further investigated. The reversibility of MAO inhibition by caffeine and esomeprazole were determined by dialysis and dilution studies. Sets of Lineweaver-Burk plots were constructed to determine the modes of binding of these inhibitors to the MAO enzymes. Both caffeine and esomeprazole were found to be reversible and competitive inhibitors of MAO. Dialysis of mixtures of caffeine with MAO-A and MAO-B resulted in the recovery of enzyme activity to levels of 97% and 96%, respectively. Dialysis of mixtures of esomeprazole with MAO-A and MAO-B resulted in the recovery of enzyme activity to levels of 93% and 88%, respectively. Similarly, dilution of mixtures containing esomeprazole and MAO-A/MAO-B resulted in the recovery of enzyme activity to levels of 94% and 87%, respectively.For the inhibition of MAO-A and MAO-B by caffeine and esomeprazole, the Lineweaver-Burk plots were indicative of a competitive mode of inhibition. In an attempt to gain further insignt, caffeine, esomeprazole and leflunomide were docked into models of the active sites of MAO-A and MAO-B. An analysis of the interactions between the enzyme models and the ligands were carried out and the results are discussed in the dissertation The results of the present study show that screening of a virtual database of molecules with a pharmacophore model may be useful in identifying existing drugs with potential MAO inhibitory activities. The search for new reversible MAO inhibitors for the treatment of diseases, including Parkinson’s disease and depression, may be facilitated by employing a virtual screening approach. Such an approach also may be more costeffective than de novo inhibitor design. Synthesis and biological evaluation of 6-substituted coumaranone derivatives and related compounds as monoamine oxidase inhibitors / Adriaan Sarel van Dyk

Parkinson’s disease (PD) is an age related neurodegenerative disorder that presents with both motor and non-motor symptoms. The most common pathological characteristic of PD is the loss of the pigmented dopaminergic neurons of the substantia nigra pars compacta (SNpc), with the appearance of intracellular inclusions known as Lewy bodies in the affected neurons. The loss of the SNpc neurons results in a deficiency of dopamine in the nigrostriatal pathway of the brain, and it is this deficiency that is responsible for the motor symptoms of PD. Monoamine oxidase B (MAO-B) is predominantly found in the striatum and is responsible for the oxidative metabolism of dopamine. The first-line treatment of PD is dopamine replacement therapy with levodopa, the metabolic precursor of dopamine. Rapid metabolism of levodopa at central and peripheral level, however, hampers its therapeutic potential. MAO-B inhibition enhances striatal dopamine activity by means of inhibiting dopamine metabolism, and MAO-B inhibitors are thus used in the treatment of PD, particularly in combination with levodopa. The aim of this study was to design new potent, reversible MAO inhibitors with selectivity towards MAO-B for the symptomatic treatment of PD. Recent studies have shown that C5-substituted phthalide derivatives are highly potent inhibitors of human MAO-B. Phthalide derivatives were also found to be potent inhibitors of human MAO-A. The structural similarity between phthalide and 3-coumaranone suggests that 3-coumaranone may be a useful scaffold for the design of reversible MAO-B inhibitors. In the present study, 3-coumaranone derivatives were thus synthesised and evaluated as potential MAO-A and MAO-B inhibitors. By reacting 6-hydroxy-3-coumaranone with the appropriate alkylbromide in N,N-dimethylformamide in the presence of potassium carbonate, a series of twenty 3-coumaranone derivatives were synthesised. The structures of the compounds were verified with NMR spectroscopy and mass spectrometry. The purities of the compounds were determined by HPLC analyses. To determine the inhibition potencies, the recombinant human MAO-A and MAO-B enzymes were used, and the inhibition potencies were expressed as IC50 values. The results indicated that the 3-coumaranone derivatives are highly potent MAO-B inhibitors. For example, 9 of the 3-coumaranone derivatives inhibited MAO-B with IC50 values < 0.05 μM, with the most potent inhibitor exhibiting an IC50 value of 0.004 μM. Although the 3-coumaranone derivatives are selective MAO-B inhibitors, some compounds were also potent MAO-A inhibitors with the most potent inhibitor exhibiting an IC50 value of 0.586 μM. The reversibility of MAO-B inhibition by a representative inhibitor was examined by measuring the degree to which the enzyme activity recovers after dialysis of the enzyme-inhibitor complex. Since MAO-B activity was almost completely recovered after dialysis, it may be concluded that the 3-coumaranone derivatives bind reversibly to MAO-B. Lineweaver-Burk plots were constructed to show that the representative 3-coumaranone derivative is a competitive inhibitor of MAO-B. To conclude, the 3-coumaranone derivatives are potent, selective, reversible and competitive inhibitors of MAO-B. These compounds may find application in the treatment of neurodegenerative disorders such as PD. Potent MAO-A inhibitors were also discovered, which suggests that 3-coumaranone derivatives may serve as leads for the design of drugs for the treatment of depression. Monoamine oxidase B (MAO-B) is predominantly found in the striatum and is responsible for the oxidative metabolism of dopamine. The first-line treatment of PD is dopamine replacement therapy with levodopa, the metabolic precursor of dopamine. Rapid metabolism of levodopa at central and peripheral level, however, hampers its therapeutic potential. MAO-B inhibition enhances striatal dopamine activity by means of inhibiting dopamine metabolism, and MAO-B inhibitors are thus used in the treatment of PD, particularly in combination with levodopa. The aim of this study was to design new potent, reversible MAO inhibitors with selectivity towards MAO-B for the symptomatic treatment of PD. Recent studies have shown that C5-substituted phthalide derivatives are highly potent inhibitors of human MAO-B. Phthalide derivatives were also found to be potent inhibitors of human MAO-A. The structural similarity between phthalide and 3-coumaranone suggests that 3-coumaranone may be a useful scaffold for the design of reversible MAO-B inhibitors. In the present study, 3-coumaranone derivatives were thus synthesised and evaluated as potential MAO-A and MAO-B inhibitors. By reacting 6-hydroxy-3-coumaranone with the appropriate alkylbromide in N,N-dimethylformamide in the presence of potassium carbonate, a series of twenty 3-coumaranone derivatives were synthesised. The structures of the compounds were verified with NMR spectroscopy and mass spectrometry. The purities of the compounds were determined by HPLC analyses. To determine the inhibition potencies, the recombinant human MAO-A and MAO-B enzymes were used, and the inhibition potencies were expressed as IC50 values. The results indicated that the 3-coumaranone derivatives are highly potent MAO-B inhibitors. For example, 9 of the 3-coumaranone derivatives inhibited MAO-B with IC50 values < 0.05 μM, with the most potent inhibitor exhibiting an IC50 value of 0.004 μM. Although the 3-coumaranone derivatives are selective MAO-B inhibitors, some compounds were also potent MAO-A inhibitors with the most potent inhibitor exhibiting an IC50 value of 0.586 μM. The reversibility of MAO-B inhibition by a representative inhibitor was examined by measuring the degree to which the enzyme activity recovers after dialysis of the enzyme-inhibitor complex. Since MAO-B activity was almost completely recovered after dialysis, it may be concluded that the 3-coumaranone derivatives bind reversibly to MAO-B. Lineweaver-Burk plots were constructed to show that the representative 3-coumaranone derivative is a competitive inhibitor of MAO-B. To conclude, the 3-coumaranone derivatives are potent, selective, reversible and competitive inhibitors of MAO-B. These compounds may find application in the treatment of neurodegenerative disorders such as PD. Potent MAO-A inhibitors were also discovered, which suggests that 3-coumaranone derivatives may serve as leads for the design of drugs for the treatment of depression. Synthetic and kinetic investigations into living free-radical polymerisation used in the preparation of polymer therapeutics

The aim of this work was to successfully prepare polymers of N-(2-hydroxypropyl)methacrylamide, (PHPMA) using controlled/"living" free-radical polymerisation technique. For this purpose, atom transfer radical polymerisation (ATRP) and reversible addition-fragmentation (chain) transfer (RAFT) polymerisation were used in preparation of a number of base polymers with the intention of quantitatively converting them into PHPMA. Both methods were applied under varying polymerisation conditions, and the kinetics of the systems investigated. Various rate constants were measured, while computer modelling of the experimental data allowed estimation of other kinetic parameters of interest. Investigations into solvent and ligand effects on the kinetics of ATRP of the activated ester methacryloyloxy succinimide (MAOS) and one of the archetypal methacrylate monomers, methyl methacrylate (MMA) were carried out. The method of RAFT was also employed in polymerisation of MAOS and a number of other monomers in the hope of finding the best synthetic precursor of PHPMA. Polymers of methacryloyl chloride (MAC) and p-nitrophenyl methacrylate (NPMA) were prepared, as well as the polymers of HPMA itself and N-isopropyl methacrylamide. Polymerisation of MMA by RAFT was also attempted in view of adding to current knowledge on the monomer's behaviour and the kinetic characteristics of its RAFT polymerisation. Preparation of PHPMA from PMAOS, PMAC and PNPMA was attempted. Successful preparation of PHPMA from the polymer of the acid chloride was achieved under mild reaction conditions, while displacement of N-hydroxysuccinimide groups of PMAOS resulted in unexpected modification of the polymer under the conditions used. Conversion of PNPMA into PHPMA was not achieved. At this stage these results suggest inadequacy of both PMAOS and PNPMA as reactive polymeric precursors.

The antidepressant properties of selected methylene blue analogues / Anzelle Delport

The shortcomings of current antidepressant agents prompts the design of novel multimodal antidepressants and the identification of new antidepressant targets, especially those located at sub-cellular level. Such antidepressants should possess improved response rates as well as safety profiles. Methylene blue (MB) is reported to possess diverse pharmacological actions and is attracting increasing attention for the treatment of a variety of disorders including Alzheimer’s disease, bipolar disorder, anxiety and depression. MB acts on both monoamine oxidase (MAO) and the nitric oxide (NO)-cGMP pathway, and possesses antidepressant activity in rodents. The principal goal of this study was to design a close structural analogue of MB and to evaluate the effects of these structural changes on MAO inhibition, a well-known antidepressant target. Furthermore, MAO inhibition is also responsible for cardiovascular toxicity in clinically used MAOI inhibitors. For this purpose we investigated the antidepressant properties of the synthetic MB analogue (ethyl-thioniniumchloride; ETC) as well as azure B, the major metabolite of MB, in the forced swim test (FST). ETC was synthesized with a high degree of purity from diethyl-p-phenylenediamine with 6% yield. ETC was firstly evaluated as a potential inhibitor of recombinant human MAO-A and MAO-B. Azure B and ETC were evaluated over a dosage range of 4-30 mg/kg for antidepressant-like activity in the acute FST in rats, and the results were compared to those obtained with saline, imipramine (15 mg/kg) and MB (15 mg/kg) treated rats. Locomotor activity was evaluated to ensure that changes in swim motivation are based on antidepressant response and not due to an indirect effect of the drug on locomotor activity. The results document that ETC inhibits MAO-A and MAO-B with IC50 values of 0.51 μM and 0.592 μM, respectively. Furthermore, ETC inhibits MAO-A and MAO-B reversibly, while the mode of inhibition is most likely competitive. In the acute FST, azure B and ETC were more effective than imipramine and MB in reversing immobility, without inducing locomotor effects. Azure B and ETC increased swimming behaviour during acute treatment, which is indicative of enhanced serotonergic neurotransmission. Azure B and ETC did not affect noradrenergicmediated climbing behaviour. These results suggest that azure B may be a contributor to the antidepressant effect of MB, and acts via increasing serotonergic transmission. Exploration of Majority Logic Based Designs for Arithmetic Circuits

Since its inception, Moore's Law has been a reliable predictor of computational power. This steady increase in computational power has been due to the ability to fit increasing numbers of transistors in a single chip. A consequence of increasing the number of transistors is also increasing the power consumption. The physical properties of CMOS technologies will make this powerwall unavoidable and will result in severe restrictions to future progress and applications. A potential solution to the problem of rising power demands is to investigate alternative low power nanotechnologies for implementing logic circuits. The intrinsic properties of these emerging nanotechnologies result in them being low power in nature when compared to current CMOS technologies. This thesis specifically highlights quantum dot celluar automata (QCA) and nanomagnetic logic (NML) as just two possible technologies. Designs in NML and QCA are explored for simple arithmetic units such as full adders and subtractors. A new multilayer 5-input majority gate design is proposed for use in NML. Designs of reversible adders are proposed which are easily testable for unidirectional stuck at faults.
