Effects of the Influenza Vaccine on the Oral Cavity

According to existing literature, there is a suggested correlation between certain vaccinations and oral cavity symptoms. Studies have shown that the Diphtheria, Tetanus, Acellular Pertusis, as well as Polio Vaccinations have an association with presented symptoms of bleeding gums, sores, ulcers, white spots in the mouth or on lips, and unpleasant breath odor. Although these symptoms may not occur simultaneously or directly after administration of the immunization, there has been supported evidence of correlation. Given the relevance of an association between vaccinations and orally manifested side effects, an investigation on the presence of such association with the widely administered flu vaccine was conducted. Data for this work was collected from a population including undergraduate students studying at the University of Central Florida. A brief voluntary online survey requesting demographic information regarding administration of the vaccine as well as any experienced side effects was used. The study was cohort in nature as it tracked subjects with known exposure to the flu shot in the past six months to understand the outcome of interest. Results from the survey were used to determine that there is no correlation between orally manifested side effects and administration of the flu vaccine.

In my thesis I focused on the relationship between the influence of risk factors and the dental health status. The evaluation of the results was made on the basis of testing and verification of the set hypotheses, using my own graphs and statistical evaluation (in the program Statistica 6.0). For the research I used the data of 613 dental patients. I could draw these data from the provided file of their medical cards. The study is divided into the theoretical and practical part. In the theoretical part I focused on the anatomical structure of teeth, on dental caries, dental stone, and their creation and prevention. A large part of my work is dedicated to disease of the oral cavity and its hygiene. In this section I deal with various types of periodontal diseases (chronica of gingivitis, periodontitis) and their prevention. The practical part deals with the hypotheses themselves. I set seven hypotheses. The set hypotheses focus on risk factors, namely sex, residence, age, oral hygiene, frequency of preventive medical examinations, and illnesses which the patients suffer from. I evaluated the influence of these risk factors with regard to the cariosity and overall dental health of the dental patients. Um dos tipos de câncer mais frequentes no mundo é o carcinoma epidermoide de cabeça e pescoço. A dificuldade de resposta eficiente aos tratamentos está relacionada à complexidade molecular deste tumor. MicroRNAs regulam uma parte significativa dos genes humanos, dentre os quais genes responsáveis por controlar proliferação e apoptose, caracterizando seu papel na progressão tumoral e resistência a quimioterápicos. O entendimento sobre como moléculas com ação antineoplásica atuam sobre a regulação de microRNAs pode contribuir para a compreensão de resultados terapêuticos. Neste estudo avaliamos o efeito dos triterpenos quinonametídeos maitenina e 22-β-hidroximaitenina sobre a viabilidade celular e sobre a regulação de microRNAs associados com proliferação e apoptose. O efeito destas moléculas foi avaliado frente a linhagens tumorais e em queratinócitos orais livres de tumor. Os resultados indicam que os dois triterpenos avaliados reduzem eficazmente a viabilidade das células cancerígenas e a regulação de microRNAs faz parte dos mecanismos envolvidos neste efeito. / Head and neck squamous cell carcinoma is one of the most frequent cancer types in the world. The difficulty of effective response to treatment is related to the molecular complexity of this tumor. MicroRNAs, small non-coding RNA molecules, regulate a significant proportion of human genes, among which genes involved in cell growth, apoptosis and response to chemotherapy. The comprehension on how anticancer molecules interfere with microRNA expression should contribute to the understanding of treatment outcome. This study evaluated the effect of two quinonamethide triterpenoids, maytenin and 22-β-hidroxymaytenin, on cell viability and on the regulation of microRNAs associated with proliferation and apoptosis. The effect of these molecules was evaluated in tumor cells and oral keratinocytes. The results show that the two triterpenoids effectively reduced cell viability of cancer-derived cell lines and that microRNA regulation is one of the mechanisms involved in this effect.

Insight into a unique carbon resource partitioning mechanism in Aggregatibacter actinomycetemcomitans

Aggregatibacter actinomycetemcomitans is a Gram negative bacterium found exclusively in the mammalian oral cavity where it resides in the gingival crevice, the space between the tooth and gum tissue. Though it has historically been considered a common commensal organism, it is now appreciated that A. actinomycetemcomitans is an opportunistic pathogen associated with the diseases periodontitis and endocarditis. To cause infection, A. actinomycetemcomitans must interact and compete with neighboring bacteria for space and nutrients, though little is known about the physiology it employs within the gingival crevice. Using A. actinomycetemcomitans grown in a chemically defined medium containing carbon sources found in vivo, I use transcriptome analyses and growth studies to show that A. actinomycetemcomitans preferentially utilizes lactate over the phosphotransferase system (PTS) sugars glucose and fructose. Additionally, the presence of lactate or pyruvate inhibits the transport and metabolism of these sugars in a post-transcriptionally controlled process I have termed PTS substrate exclusion. Since lactate is an energetically inferior carbon source, PTS substrate exclusion appears to be a carbon resource partitioning mechanism that allows A. actinomycetemcomitans to avoid competition for energetically favorable sugars with other species, and I propose a model to describe this phenomenon. To begin to understand the mechanism of PTS substrate exclusion, I examine the first step of the proposed model by purifying and characterizing the L-lactate dehydrogenase (LctD) from A. actinomycetemcomitans. I demonstrate that, unlike other studied lactate dehydrogenases, the LctD from A. actinomycetemcomitans does not exhibit feedback inhibition in the presence of physiologically relevant concentrations of pyruvate, which supports my hypothesis that elevated intracellular pyruvate levels inhibit the PTS. The results of my studies provide insight into a new regulatory mechanism governing carbon utilization in this bacterium. / text

Nesse contexto, o objetivo desse estudo foi isolar e identificar Lactobacillus da cavidade bucal de indivíduos livres de cárie e avaliar seu potencial de inibição de C. albicans por meio de estudos in vitro e in vivo. Primeiramente, foram avaliados os efeitos de 30 isolados clínicos de Lactobacillus sobre o número de células viáveis (UFC) em biofilme de C. albicans e sobre a formação de hifas. Os isolados que obtiveram os maiores efeitos inibitórios sobre C. albicans foram selecionados para os testes de determinação da biomassa total dos biofilmes pela absorbância do cristal violeta, análise da arquitetura dos biofilmes por microscopia eletrônica de varredura (MEV) e quantificação da expressão de genes de C. albicans (ALS3, HWP1, EFG1 e CPH1) por qPCR. Esses isolados também foram submetidos a estudos in vivo usando os modelos de Galleria mellonella e Caenorhabditis elegans. Para o estudo em G. mellonella, a infecção experimental foi avaliada pela curva de sobrevivência, quantificação da carga fúngica na hemolinfa, densidade hemocitária, quantificação da expressão gênica de peptídeos antifúngicos (Gallerymicina e Galiomicina) e monitoramento da infecção de C. albicans por análise de bioluminescência. No modelo de C. elegans, a infecção foi avaliada por meio dos ensaios de curva de sobrevivência e estudo da filamentação de C. albicans. Os resultados dos ensaios in vitro demonstraram que L. paracasei 28.4, L. rhamnosus 5.2 e L. fermentum 20.4 foram as cepas com maior atividade antimicrobiana sobre os biofilmes de C. albicans. Nessas cepas, todos os genes analisados foram regulados negativamente na associação com Lactobacillus quando comparados com o grupo controle. No estudo in vivo, a injeção de L. paracasei 28.4 em G. mellonella infectadas com C. albicans aumentou a sobrevida das larvas, o número de hemócitos e a expressão de peptídeos antifúngicos, reduzindo assim a UFC de C. albicans. Em C. elegans, L. paracasei 28.4 também foi capaz de aumentar a sobrevida dos vermes infectados com C. albicans e reduzir a filamentação. Conclui-se que L. fermentum 20.4, L. paracasei 28.4 e L. rhamnosus 5.2 tem potencial para serem usados como probióticos na cavidade bucal devido sua ação anti-biofilme e sua regulação negativa dos genes de virulência de C. albicans. L. paracasei 28.4 foi capaz de prolongar a sobrevida nos dois modelos experimentais infectados com C. albicans por apresentarem ação antifúngica e imunomodulatória. / The study of the antifungal activity of Lactobacillus may contribute to the discovery of new therapeutic strategies for Candida infections. In this context, the objective of this study was to isolate and identify Lactobacillus from the oral cavity of caries-free subjects and to evaluate its effects through in vitro and in vivo studies. First, the effects of 30 clinical isolates of Lactobacillus on the number of viable cells (CFU) in biofilms of C. albicans and on hyphae formation were evaluated. The isolates that obtained the highest inhibitory effects on C. albicans were selected for biofilm biomass determination by violet crystal absorbance, analysis of biofilm architecture by scanning electron microscopy (SEM) and quantification of the expression of C. albicans (ALS3, HWP1, EFG1 and CPH1) by real time PCR. These isolates were also submitted to in vivo studies using the Galleria mellonella and Caenorhabditis elegans models. For the study in the model of Galleria mellonella, the experimental infection was evaluated by the survival curve, quantification of the fungal load in the hemolymph, hemocitary density, the gene expression of antifungal peptides (Gallerymicin and Galiomicin) and monitoring of C. albicans infection by bioluminescence analysis. In the Caenorhabditis elegans model, the infection was evaluated by the survival curve assays and the study of C. albicans filamentation. The results of in vitro tests demonstrated that L. paracasei 28.4, L. rhamnosus 5.2 and L. fermentum 20.4 were the strains with the highest antimicrobial activity on the biofilms of C. albicans. In these strains, all analyzed genes were negatively regulated in association with Lactobacillus when compared to the control group. In the in vivo study, the injection of L. paracasei 28.4 into the G. mellonella increased survival of the larvae, the number of hemocytes and the expression of antifungal peptides, thus reducing the CFU of C. albicans. In C. elegans, L. paracasei 28.4 was also able to increase the survival of worms infected with C. albicans and reduce the filamentation. We conclude that L. fermentum 20.4, L. paracasei 28.4 and L. rhamnosus 5.2 have potential to be used as probiotics in the oral cavity due to their anti-biofilm action and their negative regulation of virulence genes of C. albicans. The study sample was composed by 66 radiographs of white subjects, 26 male and 40 female, aged between 12 and 21 years, from schools in the region of the Great Paulista ABC. The criteria used for diagnosing normal occlusion was the Six keys to normal occlusion (Andrews, 1972) ; at least four out of the six keys should be present, necessarily the first one, which is the inter-arch relation. To take the X-rays, the patient was positioned in the natural head position, and they were taken immediately after the swallowing of barium sulphate. Posteriorly the cephalometric tracings were done and the following measurements were obtained: tongue length, tongue height, distance from the medium area of tongue dorsum to the hard palate, distance between tongue apex and incisal face of lower incisor. The results indicate that there is not a unique pattern for the tongue rest position in oral cavity, therefore presenting various possibilities. Convergence existed only in the fact that all of them touched the soft palate. There was no statistical significant relationship between the tongue rest position and the facial biotypes or the gender. / O propósito neste estudo foi determinar a posição de repouso da língua em indivíduos com oclusão dentária normal e respiração nasal, por meio de telerradiografias em norma lateral realizadas após a ingestão de bário. A amostra foi composta por 66 radiografias de indivíduos brancos com oclusão dentária normal, sendo 26 do sexo masculino e 40 do sexo feminino, na faixa etária de 12 a 21 anos de idade, procedentes de escolas da região do Grande ABC Paulista. O critério utilizado para diagnóstico da oclusão normal foi As Seis Chaves para a Oclusão Normal preconizadas por Andrews (1972), devendo estar presentes no mínimo quatro das seis chaves, sendo obrigatória a presença da primeira chave de oclusão que é a da relação interarcos. As radiografias foram obtidas com o indivíduo em posição natural da cabeça após a ingestão de contraste de sulfato de bário para evidenciar o controle da língua. Posteriormente foi feito o desenho anatômico das estruturas pesquisadas, marcados os pontos cefalométricos, traçadas as linhas e os planos, e por último obtidas as seguintes medidas lineares: comprimento e altura da língua, distância do dorso da língua na sua porção média até o palato duro e a distância entre a ponta da língua e a incisal do incisivo inferior. Por meio dos resultados encontrados verificou-se que não existe um padrão único de posicionamento de repouso da língua dentro da cavidade oral, em pacientes respiradores nasais, variando muito sua distância até a incisal dos incisivos inferiores, bem como até o palato duro, havendo uniformidade apenas no fato da língua tocar o palato mole em todos os indivíduos da amostra. Não houve relação estatisticamente significante entre a posição de repouso da língua e os biotipos faciais e nem dimorfismo sexual.
