Bioaccumulation of organochlorine pesticides and biomarker responses in Hydrocynus vittatus and Synodontis zambezensis from the Lower Phongolo River and Floodplain, KwaZulu-Natal, South Africa01 July 2015 (has links)
M.Sc. (Zoology) / Concern has been shown by Ezemvelo KZN Wildlife for the state of the Lower Pongolo River and Floodplain due to the ever increasing urban and industrial development in the area. This increase in development poses a threat to the fish populations within the system and their ability to support the surrounding communities. Continued spraying of pesticides e.g. DDT within the area are adding to stresses placed on the fish populations through decreasing water quality. The effectiveness of environmental water flows suggested by White et al. (1984), have never been assessed by conservation authorities. This means that the authorities have been unable to fulfil their legislative obligations to establish management and conservation plans (Hughes et al., 2001). Work on the biological responses of fish to anthropogenic stressors such as organic pollutants and metals; have been conducted by a group of experts in the field of ecological health. This section of the project focusses on two fish species, Hydrocynus vittatus and Synodontis zambezensis, sampled within the study area and used for biomarkers and tissue chemical level determination. Standard techniques were applied for Organochlorine Pesticide (OCP) analysis according to Yohannes et al. (2013) and these results have been compared to biomarker responses which have also been completed following standard protocols. Results found within both fish species, show the relationship between accumulation of toxicants and biomarker responses. The highest levels of ΣOCPs where found within H. vittatus at (Mean ± SE) 1101.61 ± 610.97 ng/g lipid weight, the highest constituent being technical grade Dichlorodiphenyltrichloroethane (DDT). High levels of ΣDDT were also found in S. zambezensis, but were lower than levels in H. vittatus and these levels correspond with positioning of each species within the food web. H. vittatus occupies a much higher level in the food web as they are top predators and S. zambezensis falls lower down on the food web due to their reliance on detritus and only small invertebrate species for food. Biomarker responses are clearly affected by bioaccumulation levels and this is especially evident within H. vittatus as Achetylcholinesterase (AChE) is greatly inhibited by high levels of ΣDDT. Changes in the level of biomarker responses are not as apparent in S. zambezensis and this is believed to be food web linked, due to differences in bioconcentration. Data collected during this study will contribute to baseline data on S. zambezensis and add to already accumulated data on H. vittatus. This new data will also help with the improvement of or alteration to already present monitoring programmes in the study area.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Poluições da água e do solo são duas das cinco categorias básicas de poluição ambiental. As outras três são do ar, ruído e luz. Poluição ocorre quando um material/produto químico é adicionado ao meio ambiente afetando-o adversamente. Uma vez que a poluição existe, retornar a matriz impactada para o seu estado anteriormente não poluído torna-se muito difícil ou até mesmo impossível em alguns casos. Os compostos organoclorados (OC) representam um importante grupo de poluentes orgânicos persistentes (POP), devido à sua toxicidade e elevada persistência no ambiente. Diferentes técnicas de remediação de solo e água subterrânea têm sido desenvolvidas com o objetivo de reparar as áreas contaminadas em menor tempo e custo possíveis e, assim, a oxidação química in situ tem se mostrado uma técnica cada vez mais atraente devido sua simplicidade e baixo custo. A degradação de dois compostos organoclorados via oxidação química por reagente de Fenton em solução aquosa é o objeto do presente trabalho. Para tanto, foram selecionados dois compostos, sendo um alifático e outro aromático. Assim, foram avaliadas as condições de degradação dos compostos organoclorados em relação às concentrações do oxidante em diferentes tempos reacionais, diferentes fontes de catalisador e, de forma qualitativa, quais foram os principais subprodutos formados. Os ensaios oxidativos foram realizados em meio aquoso com temperatura controlada a 25 C e em 3 configurações de [Fe2+] e [H2O2]. A extração dos compostos de interesse para a fase orgânica também foi estudada. A análise dos extratos revelou que o acetato de etila foi o solvente que melhor extraiu os compostos de interesse da fase aquosa. Além destes ensaios, foram avaliadas outras duas fontes de Fe(II), sendo 11 óxidos de manganês dopados com ferro e a classe de solo predominante no território nacional, que é caracterizada pela sua alta concentração de ferro endógeno (latossolo). O ensaio entre as fontes de ferro revelou que tanto o óxido de Mn quanto o latossolo produziram menos subprodutos em relação à reação com a solução de sulfato Fe(II). Entretanto, em termos quantitativos, a redução na concentração de clorobenzeno foi mais efetiva na reação com a solução de Fe(II) (cerca de 63%). A identificação dos sobprodutos foi realizada a partir da análise em cromatógrafo a gás acoplado a espectrômetro de massas e em análise comparativa dos espectros com a biblioteca NIST. Os ensaios oxidativos revelaram que em baixas concentrações de oxidante, há a possibilidade de geração de subprodutos mais tóxicos que os reagentes, sendo produzido bifenilas policloradas e hexaclorociclohexano nas reações com clorobenzeno e 1,2-dicloroetano, respectivamente

L'hexaclorobenzè (HCB) és un compost organoclorat àmpliament distribuït per tot el planeta, altament lipofílic que s'acumula als sistemes biològics. Els nounats s'exposen a aquests compostos organoclorats (OCs) a través de la placenta i de la lactància materna. Tot i que l'HCB és un dels OCs més comuns, la seva transferència a través de la placenta de la mare al fetus durant la gestació i a través de l'alletament matern està poc documentada. El coneixement sobre els seus possibles efectes sobre la salut infantil és també bastant limitat. A Flix, un poble de la Ribera d'Ebre, es van detectar nivells molt elevats d'HCB a l'atmosfera degut a la seva proximitat a una empresa electroquímica. Els objectius del present treball són els d'avaluar l'exposició a OCs a través de la placenta i de la lactància materna i els seus efectes sobre la salut infantil durant el primer any de vida en el conjunt de nounats d'aquesta població. / Hexachlorobenzene (HCB) is an organochlorine compound widely spread in the environment, highly lipophyilic which accumulates in all biological systems. Newborns are exposed to organochlorine compounds (OCs) through placenta and via breastfeeding. Although HCB is one of the most common OCs, its transference from the mother to the child through placenta and via breastfeeding is poorly documented. The knowledgement of the possible adverse effects on infant's health is also limited. In Flix, a small village in the Ribera d'Ebre area, high levels of HCB were detected in the atmosphere of the village due to the vicinity of an electrochemical factory. The objectives of the present study are to evaluate OCS exposition through placenta and breastfeeding and its effects on infant's health during the first year of life.

Investigation of selected organic compounds on water quality along the Olifants river catchmentMulanga, Tshimanga Christelle 06 1900 (has links)
Water is a crucial natural resource, indispensable to food production, life, the environment, power generation, industry, sanitation and hygiene. The presence of Organochlorine Pesticides (OCPs) in the environment is not wanted due to their negative effects on human beings and animals. As a result, there is a need to continuously monitor their presence in the environment. In this study, surface water samples were collected once a month during the dry season and during wet season from the selected five points along the Olifants River and stored at a temperature of < 50C before analysis. The OCPs were extracted with dichloromethane (DCM) using the Liquid-Liquid Extraction (LLE) method. After undertaking the sample through the clean- up process, the crude extracts obtained were put into the column chromatography and eluted with hexane, about 1.5 μL of the purified extracts were analysed by the Gas Chromatographic- Mass Spectrophotometer (GC/MS). The percentage recoveries, varied from 32- 116 % for p, p’-DDT and 4,4'-DDD respectively in triply spiked water samples. The standard deviation for most of the compounds is less than±0.04, with the exclusion of Heptachlor (±0.14). The seasonal variability of OCPs in water samples along Olifants River results show that in dry season, the Olifants River is mostly polluted at the Oxford site with (BHC-beta, Aldrin, Heptachlor-epoxide, Endosulfan-alpha and Endrin), at the Ga- Selati site with (Heptachlor-epoxide and Endrin) and at the Wolvekrans site with (Endosulfan-alpha), with Aldrin up to 834.20 ng/ L indicating the highest hazard toward the aquatic environment while in summer the Olifants River is mostly polluted at the Ga- selati site with BHC-beta and at the Waterval site with (Heptachlor and BHC-gamma) with BHC- gamma up to 560 ng/ L indicating the highest hazard toward the aquatic environment. The levels reached from the Olifants River catchment were meaningfully above the drinking water quality guidelines for organic chemical recommended by WHO, 2006 i.e. (BHC-gamma,DDT-44, Aldrin, dieldrin and Endrin are (2.0,1.0,0.03,0.03 and 0.6) respectively for the protection of the domestic use, aquatic ecology and agricultural use (irrigation and livestock watering) for compounds with local guideline values; while, the international water quality guidelines to protect the aquatic ecosystems are 0.00083 ng/mℓ (4,4’DDD), 0.00059 ng/mL (4,4’DDE), (4,4’DDT), 0.00021 ng/mL (heptachlor), 0.0092 ng/mℓ (α-HCH), 0.0186 ng/mL (γ- HCH), and chronic values are 0.056 ng/mL (ENDO I and II) and 0.0023 ng/mL endrin) (USEPA, 2002). Levels detected were significantly higher than some research studies conducted up to now in South African aquatic environments. Assessment of organochlorine pesticide residues in fish samples from the Okavango Delta, BotswanaMpofu, Christopher 28 February 2011 (has links)
This thesis presents an evaluation of the dispersive solid-phase extraction (d-SPE) method referred to as the quick, easy, cheap, effective, rugged and safe (QuEChERS) method for the determination of four organochlorine pesticide residues in fish samples. The pesticides investigated in this study were o, p′-DDT, p, p′-DDE, aldrin and dieldrin. The combined use of Gas Chromatography with an Electron Capture Detector (GC-ECD) and sensitive Time of Flight (TOF) mass detector facilitated the identification of the target analytes. In the absence of certified reference material, the overall analytical procedure was validated by systematic recovery experiments on spiked samples at three levels of 2, 5 and 10 ng/g. The targeted compounds were successfully extracted and their recovery ranged from 76 to 96% with relative standard deviations of less than 13%. The optimum QuEChERS conditions were 2g of fish powder, 10 ml acetonitrile and 1 min shaking time. The optimal conditions were applied to assess the levels of chlorinated pesticides in blunt-tooth catfish (Clarias ngamensis), tigerfish (Hydrocynus vittatus), Oreochromis andersonii and red-breasted tilapia (Tilapia rendalli) from the Okavango Delta, Botswana. Dieldrin, p, p′-DDE and aldrin were detected in all the analysed samples with a concentration range of 0.04 – 0.29, 0.07 – 0.33, 0.04 – 0.28 and 0.03 – 0.24 ng/g per dry weight in O. andersonii, C. ngamensis, T. rendalli and H. vittatus respectively. These concentrations were below the US-EPA 0.1 μg/g allowable limit in edible fish and the Australian Maximum Residue Limit (MRL) of 50 - 1000 ng/g in fresh fish. DDT was not detected in all the fish species investigated. The mean lipid content recorded in the fish samples were 1.24, 2.16, 2.18 and 4.21% for H. vittatus, T. rendalli, O. andersonni and C. ngamensis respectively. No systematic trend was observed between fish age and pesticide levels in fish. Acetylcholinesterase (AChE) activity assays were performed to assess the effects of organochlorine pesticides in Clarias ngamensis. The enzyme activity recorded in Clarias ngamensis from the Okavango Delta and the reference site was 12.31 μmol of acetylcholine iodide hydrolysed/min/g brain tissue. The enzyme activity remained the same indicating no enzyme inhibition. The conclusions drawn from this study are that the QuEChERS method is applicable for the determination of organochlorine pesticide residues in fish matrices. The fish from the Okavango Delta are safe for human consumption.

Le cancer du sein est le plus fréquent des cancers de la femme. Son incidence n’est expliquée que partiellement par les facteurs de risque bien établis comme les facteurs reproductifs, hormonaux ou génétiques. Plusieurs éléments suggèrent l’existence de facteurs de risque environnementaux, tels que les polluants organochlorés persistants connus comme perturbateurs endocriniens. Les objectifs de ce travail sont : d’étudier les niveaux d’exposition, et leurs déterminants, au DDE (principal métabolite de l’insecticide DDT) et aux polychlorobiphényles (PCB) ; d’estimer par une modélisation pharmacocinétique à base physiologique (PBPK) les niveaux d’exposition au PCB-153 à la période péri-pubertaire qui constitue une fenêtre de susceptibilité accrue de la glande mammaire aux agents environnementaux ; d’étudier le rôle du DDE et du PCB-153 dans le risque de cancer du sein, à la fois à partir de prélèvements sanguins actuels et, pour le PCB-153, à partir de niveaux estimés à la période péri-pubertaire.Ce travail a été effectué à partir de l’étude CECILE, étude cas-témoins en population générale incluant 1055 témoins et 1080 cas de cancer du sein diagnostiqués de février 2005 à mars 2008 dans les départements d’Ille-et-Vilaine et de Côte d’Or chez lesquels un prélèvement sanguin a permis d’effectuer des dosages de p,p’-DDE et de PCB-153. La modélisation PBPK mise en place en coopération avec une équipe de toxicologues canadiens a permis de simuler des profils toxicocinétiques vie entière de PCB-153 et d’estimer les niveaux d’exposition au cours de la puberté. Cette modélisation est basée sur les niveaux mesurés au moment de l’enquête, les facteurs individuels de variation (poids, taille, grossesses, allaitement) connus d’après le questionnaire et la courbe de contamination environnementale au cours du temps.Les niveaux de contamination biologique observés sont plus faibles que ceux rapportés précédemment dans les populations occidentales. Nous montrons également que les principaux facteurs prédictifs des niveaux sériques de p,p’-DDE et de PCB-153 chez les femmes incluent l’âge, les habitudes alimentaires, le poids et ses variations au cours du temps, les grossesses et l’allaitement, et le lieu de résidence. Nous n’observons pas d’association entre les niveaux de p,p’-DDE et le risque de cancer du sein, mais une association inverse apparaît pour les cancers du sein de type négatifs pour les récepteurs hormonaux. Nous observons également une association inverse avec les niveaux de PCB-153 chez les femmes de 50 ans et plus. L’association inverse est renforcée lorsque les niveaux sont estimés pour la période péri-pubertaire à l’aide des modèles PBPK chez les femmes de la génération 1951-1960, correspondant à la cohorte de naissance des femmes de plus de 50 ans ayant connu les expositions aux PCB les plus élevées durant leur puberté.Plusieurs mécanismes biologiques pourraient expliquer l’association inverse observée entre le cancer du sein et l’exposition aux PCB. Ces résultats doivent être confirmés par des études tenant compte des fenêtres de susceptibilité de la glande mammaire et constituent une piste intéressante à approfondir. / Breast cancer is the most frequent cancer in women. Its incidence is only partially explained by well-established risk factors as reproductive, hormonal or genetic factors. It has been suggested that endocrine disrupting chemicals such as persistent organochlorine pollutants also play a role in breast cancer. The objectives of this study were: to assess exposure levels to DDE (main metabolite of DDT) and to polychlorobiphenyls (PCBs), and to explore their determinants; to estimate PCB-153 levels during the puberty using physiologically-based pharmacokinetic (PBPK) models, as this period of life constitutes a critical exposure window with increased susceptibility to carcinogens; to study the associations between breast cancer risk and separately DDE, PCB-153 measured at the time of diagnosis, and PCB-153 levels estimated during puberty.The analyses were based on data from the CECILE study, a population-based case-control study including 1055 controls and 1080 breast cancer cases who gave a blood specimen. The cases were diagnosed from February 2005 to March 2008 in two French administrative areas (Ille-et-Vilaine and Côte d’Or). p,p’-DDE and PCB-153 serum levels were measured. PBPK modeling allowed simulating PCB-153 lifetime toxicokinetic profiles and estimating exposure levels during puberty. These PBPK models were based on blood measurements at the time of recruitment in the study, on individual characteristics elicited from the questionnaire (weight, height, pregnancies, breastfeeding) and on the curve of environmental contamination by PCBs estimated since the 1930’s. The levels of biological contamination were lower than those previouly reported in Western populations in earlier periods. The main predictive factors of p,p’-DDE and PCB-153 serum levels included age; dietary habits; weight and its variations; pregnancies and breastfeeding; and residence area. We did not observed any association between p,p’-DDE levels and the risk of breast cancer, but an inverse association was observed for estrogen receptor-negative breast cancers. We also observed an inverse association of breast cancer with PCB-153 among women above 50 years. This inverse association was reinforced when considering PCB-153 levels estimated during peripubertal period from PBPK models, particularly among women born in 1951-1960, a birth cohort with peak exposure to PCB-153 during puberty. Biological mechanisms explaining the observed inverse association between breast cancer and PCB exposure are considered. These findings should be confirmed by further studies.
