Fitocistatinas em arroz (Oryza Sativa) e caracterização funcional da fitocistatina XII

Cistatinas são inibidores competitivos de proteases cisteínicas. Em plantas, as fitocistatinas, constituem uma família independente, caracterizada pela presença do motivo conservado LARFAV. Elas podem estar envolvidas em uma grande variedade de processos fisiológicos, desde o controle da proteólise endógena em órgãos reprodutivos e vegetativos, até a inibição de proteases extracelulares de herbívoros, parasitas e patógenos. Algumas fitocistatinas possuem uma extensão carboxi-terminal em sua sequência, com um motivo consenso SNSL importante para a inibição de legumaínas. Em arroz, existem doze genes distintos de fitocistatinas, sendo a fitocistatina XII (OcXII) a única com um domínio extra carboxi-terminal. Considera-se esta extensão extremamente importante, pois permitiria a atuação bifuncional deste inibidor de proteinase, simultaneamente inibindo dois tipos de proteases, papaínas e legumaínas. O objetivo principal que norteou o desenvolvimento deste trabalho foi identificar as doze fitocistatinas de arroz (Oryza sativa), caracterizando, principalmente, o gene da OcXII, de forma a analisar as prováveis implicações funcionais deste gene na planta. A caracterização da expressão dos doze genes codificadores das fitocistatinas revelou que os genes comumente mais expressos são OcI, OcIII e OcXII, sendo que OcXII destaca-se nas plantas em estádio final de maturação das sementes, em tecidos específicos de endosperma e células em cultura. Para germinação e crescimento de plântulas, é necessária uma redução da expressão de OcXII. O padrão evolutivo das fitocistatinas, em diversas plantas, sugere uma origem comum para estes genes, seguida de divergências e duplicações específicas em cada espécie. A OcXII é mantida em um grupo monofilético com as demais fitocistatinas com extensão C-terminal. Nos estudos funcionais sobre a fitocistatina XII de arroz, nove linhagens de plantas RNAi para o gene OcXII foram obtidas via transformação de calos de arroz (ssp. japonica, cv. Nipponbare). Quatro destas linhagens apresentaram nível de pelo menos 90% de silenciamento de OcXII, sem alteração na expressão gênica das demais fitocistatinas e legumaínas. Duas destas linhagens tiveram alterações fenotípicas com redução da viabilidade das sementes geradas. O silenciamento de OcXII e expressão de demais fitocistatinas manteve-se inalterado na geração T1, enquanto as legumaínas 1 e 3 sofreram alterações em pelo menos uma das linhagens. A transformação de plantas para estudos de localização celular com o promotor do gene OcXII, resultou em quatro linhagens transformadas, sendo que em uma delas a expressão do gene repórter gus, controlado pelo promotor OcXII, foi significativamente detectada nos calos em vias de regeneração de plantas. Também, pode ser verificado que a OcXII responde ao tratamento com ABA (ácido abscísico), tendo um aumento de expressão significativo após 4 horas, tanto em tecidos de folha quanto de raiz. Desta forma, a fitocistatina XII é um dos genes de fitocistatinas mais expressos durante todo o desenvolvimento das plantas de arroz, sendo o único gene com provável capacidade de inibir proteases cisteínicas do tipo legumaínas. / Cystatins are cysteine proteases competitive inhibitors. The phytocystatins evolution patterns in various plants, suggests a common origin for these genes, followed by divergences and duplications, specific to each species. The OcXII, is kept in a monophyletic group with all the other phytocystatins with C-terminal extension. In the functional studies on phytocystatin XII from rice, nine lines of RNAi plants were obtained by transformation of rice’s callus (japonica ssp. cv. Nipponbare). Four of these strains had low levels of OcXII mRNA, revealing that this gene were at least 90% silenced, any changes in gene expression for other phytocystatins or legumains were detected. Two of these transgenic lines showed phenotypic changes with reduced seed viability. The OcXII gene silencing and other phytocystatins expression remained unchanged in the T1 generation, while the first and third legumains varied in at least one of the strains. The plants transformation, for cellular localization studies with OcXII gene promoter resulted in four transgenic strains, and in one of them expression of the gus reporter gene, controlled by the promoter OcXII, was significantly detected in the calli undergoing the plant regeneration process. Also, it can be noticed that the OcXII gene expression responds to the treatment with ABA (abscisic acid), with a significant increase, in both tissues as leaf root, after 4 hours of treatment. Thus, phytocystatin XII is one of the most expressed genes troughout the development of rice plants, being the only gene with the probable ability to inhibit cysteine proteases of legumain type, in addition to papain proteases.

Caracterização funcional dos genes ASR na resposta ao alumínio em arroz (ORYZA SATIVA)

O arroz é considerado o cereal mais tolerante ao alumínio (Al). Entretanto, a variabilidade entre os genótipos leva a uma considerável diferença quanto ao grau de tolerância para cultivares distintas. Diversos estudos mostram que plantas de arroz toleram o Al por intermédio de mecanismos internos e externos. Neste trabalho foi analisada a modulação da expressão da família gênica ASR (Aba, Stress and Ripening) em função do tratamento com Al. O gene ASR5 foi diferencialmente expresso em raízes de arroz tolerante ao Al (ssp Japonica cv Nipponbare). Entretanto, ASR5 não respondeu a exposição ao Al em raízes de arroz sensíveis ao Al (ssp Indica cv Taim). Plantas transgênicas silenciadas para os genes ASR apresentaram um aumento da sensibilidade ao Al. Calos embriogênicos de arroz transformados com a fusão ASR5-GFP revelaram que a proteína ASR5 localiza-se no núcelo e no citoplasma. Em protoplastos transformados, ASR5 localizou-se nos cloroplastos. Usando uma abordagem proteômica, comparando plantas não-transformadas e plantas ASR5_RNAi, um total de 41 proteínas com padrões contrastantes foi identificado. No intuito de identificar genes com expressão alterada pelo Al em arroz, e buscar genes afetados pelo silenciamento de ASR5, foi realizado o sequenciamento total dos transcritos utilizando plantas de arroz não transformadas e ASR5_RNAi em duas condições: controle e tratamento com Al. Essas análises transcriptômicas revelaram que 961 genes responderam ao Al em raízes de plantas de arroz não transgênicas submetidas ao Al. Nas plantas ASR5_RNAi, apenas 309 genes responderam ao tratamento com Al. Entretanto, apenas 52 desses genes se sobrepuseram quando comparados ao grupo de genes modulados em plantas não transformadas, sugerindo que a planta ASR5_RNAi perdeu a capacidade de regular um conjunto de genes. Além disso, análises de imunoprecipitação da cromatina seguida de sequenciamento em massa, revelaram que ASR5 liga-se ao promotor do gene STAR1, entre outros, regulando sua expressão em resposta ao Al. Os resultados mostram pela primeira vez que ASR5 atua como fator de transcrição em arroz e que está envolvido na regulação de genes responsivos ao Al conferindo tolerância frente à toxicidade do Al. / Among cereal crops, rice is considered the most aluminium (Al) tolerant species. However, variability among rice genotypes leads to remarkable differences in the degree of Al tolerance for distinct cultivars. A number of studies have demonstrated that rice plants achieve Al tolerance through internal and external mechanisms. We have analyzed expression changes of the rice ASR (Aba, Stress and Ripening) gene family as a function of Al treatment. The gene ASR5 was differentially regulated in the Al-tolerant rice ssp Japonica cv Nipponbare. However, ASR5 expression did not respond to Al exposure in Indica cv Taim rice roots, which are highly Al-sensitive. Transgenic plants carrying RNAi constructs that targeted the ASR genes displayed increased Al susceptibility in T1 plants. Rice embryogenic calli expressing an ASR5-GFP fusion revealed that ASR5 was localized in both the nucleus and cytoplasm. In transformed protoplasts, ASR5-GFP appears in chloroplast cells. Using a proteomic approach to compare non-transformed and ASR5_RNAi plants, a total of 41 proteins with contrasting expression patterns were identified. In order to identify genes with differential modulation under Al stress in rice, and search for genes affected by ASR5 silencing, RNA-seq was made in non-transformed plants and ASR5_RNAi plants in control conditions and after Al treatment. These analyses revealed that 961 genes responded to Al in non-transformed rice roots under Al stress. A total of 309 genes responded to Al in ASR5_RNAi plants. Only 52 genes overlapped between non transformed and ASR5_RNAi plants when comparing genes modulated by Al, showing that ASR-silenced plants lost the ability to modulate a set of genes in response to Al treatment. Furthermore, ChIP-Seq analysis revealed that ASR5 can bind to the promoter of STAR1, among other genes, regulating its expression under Al stress. These results show for the first time that ASR5 act as a transcription factor in rice and that it regulates Al-responsive genes conferring tolerance in rice against Al toxicity.

Identificação e caracterização do gene OsPMBP1 (Prenylated Metal Binding Protein 1) de arroz (Oryza sativa)

O número de genes atualmente identificados no genoma do arroz (Oryza sativa ssp japonica) é de cerca de 32.000. Graças aos esforços de diferentes grupos ao redor do globo, o papel de muitos desses genes é melhor compreendido no momento. A função de um gene pode ser inferida principalmente pela análise de seu padrão de expressão, da estrutura da proteína codificada e pela análise de mutantes nos quais o gene se encontre interrompido, silenciado ou super-expresso. Para esse propósito, nosso grupo de pesquisa vem desenvolvendo linhagens transgênicas pela inserção de uma construção de T-DNA baseada no sistema de transposons de dois componentes iAc/Ds. Uma dessas linhagens (Ds72) apresenta estatura reduzida e sua inserção de T-DNA interrompe um locus, cuja função gênica ainda é desconhecida. O objetivo desse trabalho é a identificação e caracterização desse gene, que denominamos de OsPMBP1 (Prenylated Metal Binding Protein 1). Para tal, analisamos a linhagem mutante, a estrutura da proteína prevista, as possíveis relações filogenéticas com outros genes e o padrão de expressão desse gene. Em paralelo, induzimos a mobilização do elemento Ds da sua inserção original no T-DNA da linhagem original para gerar novas linhagens mutantes. A proteína codificada por esse gene contém um motivo de associação a metais pesados e um sítio de prenilação. Identificamos esse gene como um membro de um ramo até agora desconhecido da família das Proteinas Farnesiladas (FP), proteínas preniladas que se ligam a íons metálicos. Nossas análises mostram que a expressão de OsPMBP1 é semelhante em raízes e na porção aérea de plântulas, porém, em condições de excesso de alumínio, a expressão nesses órgãos se altera, sendo induzida em folhas e caules enquanto que em raízes é reprimida. Outros genes da família FP também respondem à estresses por excesso de íons metálicos, e foram caracterizados como diretamente envolvidos no transporte e/ou detoxicação de metais pesados. Em condições normais, esse gene parece ser regulado pelo relógio circadiano, sendo um gene de expressão tardia, com pico de expressão próximo ao entardecer. A análise detalhada da linhagem Ds72 indicou que não há ligação entre o fenótipo observado e a inserção de T-DNA. Os dados obtidos demonstram que OsPMBP1 pode estar envolvido na resposta a estresse por metal pesado ou no transporte desses íons, porém ainda são necessários mais estudos para melhor entender a função desse gene na planta. / The current number of identified genes in the rice genome (Oryza sativa ssp japonica) is about 32,000. Thanks to the efforts of different researchers around the world, the role of many of these genes is currently better understood. The gene function can be inferred mainly by the analysis of its expression pattern, the structure of the encoded protein and by the analysis of mutants where the gene is interrupted, silenced or over-expressed. For that purpose, our research group has developed transgenic lines by the insertion of a T-DNA construction that is based on the two-component iAc/Ds system of transposons. Ours analyses showed that the OsPMBP1 expression is equivalent in seedling roots and shoots, but, under aluminum excess, its expression change in these organs, it was inducted in the shoots and repressed in the roots. Others genes of the FP family also respond to metallic ion stresses and have been characterized as directly involved in heavy metal transport and detoxification. In normal conditions, this gene is apparently regulated by the circadian clock, with expression apices close to the evening/dusk interface. The detailed analysis of the Ds72 line indicated that there isn’t a relationship between the observed phenotype and the T-DNA insertion. The data showed that OsPMBP1 may be involved in the response to heavy metal stress or in its transport, but more analyses are required to a better understanding of the role of this gene in the plant.

Aspectos funcionais e evolutivos da família WAK em Oryza Sativa

O ambiente é um sistema dinâmico que está sempre mudando e a capacidade de reconhecimento dessas modificações é uma característica crucial para a vida. A família gênica RLK (do inglês Receptor-Like Kinase) engloba as proteínas receptoras do tipo quinase que estão aptas a reconhecer sinais ambientais, através de seu domínio extracelular, e ativar uma cascata de sinalização por modificações pós-traducionais em outras proteínas utilizando a atividade de fosforilação de seu domínio quinase. A subfamília WAK (do inglês The Wall-Associated Kinase) pertence a família gênica RLK, sendo que algumas proteínas desta subfamília foram identificados associados fisicamente à parede celular, sugerindo estes genes como fortes candidatos para agir como sensores, ligando diretamente o ambiente extracelular com o citoplasma e desencadeando sinais intracelulares. Os genes WAK formam uma grande subfamília em arroz, com 130 genes descritos para a subespécie japonica, em comparação aos 27 genes WAK descritos para arabidopsis. A ausência de dados sobre a expansão desta subfamília e suas implicações nas plantas justifica uma análise evolutiva e estrutural entre os membros da subfamília WAK de arabidopsis e arroz, com uma comparação mais extensiva entre os genomas das subespécies de arroz indica e japonica. Quando a organização e os resíduos conservados do domínio quinase das WAKs foram comparados com a superfamília de proteínas quinases de eucariotos, a identificação de dois grupos distintos de WAKs tornou- se evidente em arabidopsis e arroz. Um grupo é formado somente por OsWAKs que provavelmente se expandiu depois da separação entre monocotiledônea- dicotiledônea, os quais derivaram para a classe de quinases não-RD. O outro grupo corresponde à classe RD-quinase, sendo esta a classe de quinase mais frequente entre os eucariotos, formado por ambos os genes AtWAK e OsWAK. Além disso, com os resultados de comparação entre os genomas das subespécies indica e japonica foi possível identificar uma grande variação com relação aos domínios das proteínas e ao padrão de expressão entre os genes OsWAK dessas subespécies. O conjunto de resultados sugere que as WAKs constituem duas subfamílias evolutivamente relacionadas, mas independentes: OsWAK-RD e WAK-nonRD. O reconhecimento desta divisão poderá contribuir para a compreensão das funções e regulação das WAKs. / The environment is a dynamic system, which is always changing and the recognition of its modifications is a crucial feature for life. The RLK is a gene family of receptor quinase in plants that are able to recognize the environment signals, through its extracellular domain, and activating a signal cascade through the post-translational modifications of others protein using the phosphorylation activity from the quinase domain. The Wall-Associated Kinase (WAK) is a subfamily from RLK, with some members identified as associated to the cell wall, suggesting these genes are strong candidates to act as sensors directly linking the extracellular environment to the citoplasm and triggering intracellular signals. The WAK is a large subfamily in rice genome, with 130 genes being described in japonica, compared to the twenty-seven in A. thaliana. The absence of data about this gene subfamily expansion and their implications for plants, justify an evolutionary and structural analysis of A. thaliana and O. sativa members of the WAK subfamily, with a more extensive comparison between genomes of japonica and indica rice subspecies. When the organization of plant WAK domains and conserved residues are compared with those of other eukaryotes protein kinase superfamilies, the identification of two distinct groups of WAKs become evident in Arabidopsis and Rice. One group corresponds to a cluster containing just OsWAK that should have expanded after the monocot-dicot separation, which evolved to form a non-RD kinase class. The other group corresponds to the classical RD-kinases with both AtWAK and OsWAK representatives. Moreover, by comparing OsWAK from indica and japonica subspecies was possible to identify a large divergence in the protein domain features and gene pattern expression. We propose that plant WAKs constitutes two evolutionary related but independent subfamilies: the WAK-RD and the WAK- nonRD. The recognition of this division would contribute to the understanding of WAK function and regulation.

Identificação e caracterização do gene OsPMBP1 (Prenylated Metal Binding Protein 1) de arroz (Oryza sativa)

O número de genes atualmente identificados no genoma do arroz (Oryza sativa ssp japonica) é de cerca de 32.000. Graças aos esforços de diferentes grupos ao redor do globo, o papel de muitos desses genes é melhor compreendido no momento. A função de um gene pode ser inferida principalmente pela análise de seu padrão de expressão, da estrutura da proteína codificada e pela análise de mutantes nos quais o gene se encontre interrompido, silenciado ou super-expresso. Para esse propósito, nosso grupo de pesquisa vem desenvolvendo linhagens transgênicas pela inserção de uma construção de T-DNA baseada no sistema de transposons de dois componentes iAc/Ds. Uma dessas linhagens (Ds72) apresenta estatura reduzida e sua inserção de T-DNA interrompe um locus, cuja função gênica ainda é desconhecida. O objetivo desse trabalho é a identificação e caracterização desse gene, que denominamos de OsPMBP1 (Prenylated Metal Binding Protein 1). Para tal, analisamos a linhagem mutante, a estrutura da proteína prevista, as possíveis relações filogenéticas com outros genes e o padrão de expressão desse gene. Em paralelo, induzimos a mobilização do elemento Ds da sua inserção original no T-DNA da linhagem original para gerar novas linhagens mutantes. A proteína codificada por esse gene contém um motivo de associação a metais pesados e um sítio de prenilação. Identificamos esse gene como um membro de um ramo até agora desconhecido da família das Proteinas Farnesiladas (FP), proteínas preniladas que se ligam a íons metálicos. Nossas análises mostram que a expressão de OsPMBP1 é semelhante em raízes e na porção aérea de plântulas, porém, em condições de excesso de alumínio, a expressão nesses órgãos se altera, sendo induzida em folhas e caules enquanto que em raízes é reprimida. Outros genes da família FP também respondem à estresses por excesso de íons metálicos, e foram caracterizados como diretamente envolvidos no transporte e/ou detoxicação de metais pesados. Em condições normais, esse gene parece ser regulado pelo relógio circadiano, sendo um gene de expressão tardia, com pico de expressão próximo ao entardecer. A análise detalhada da linhagem Ds72 indicou que não há ligação entre o fenótipo observado e a inserção de T-DNA. Os dados obtidos demonstram que OsPMBP1 pode estar envolvido na resposta a estresse por metal pesado ou no transporte desses íons, porém ainda são necessários mais estudos para melhor entender a função desse gene na planta. / The current number of identified genes in the rice genome (Oryza sativa ssp japonica) is about 32,000. Thanks to the efforts of different researchers around the world, the role of many of these genes is currently better understood. The gene function can be inferred mainly by the analysis of its expression pattern, the structure of the encoded protein and by the analysis of mutants where the gene is interrupted, silenced or over-expressed. For that purpose, our research group has developed transgenic lines by the insertion of a T-DNA construction that is based on the two-component iAc/Ds system of transposons. MicroRNAs como reguladores do desenvolvimento em plantas e o papel regulatório em raízes de arroz (Oryza sativa L.) na resposta ao alumínio

Diversos trabalhos demonstram que a resposta das plantas ao alumínio é complexa. Porém, nenhum trabalho caracterizou o envolvimento de microRNAs nesta resposta. Parte deste trabalho visou caracterizar o perfil de expressão de diferentes famílias de microRNAs em resposta ao alumínio comparando-se as raízes de plantas de arroz japonica e indica. De um total de dezesseis microRNAs diferencialmente expressos, treze microRNAs tiveram a expressão reduzida e outros seis tiveram a expressão aumentada nas raízes de plantas de arroz japonica tratadas com 450 M de AlCl3 após 8h. Nas plantas de arroz indica tratadas nas mesmas condições, nove microRNAs foram detectados como diferencialmente expressos. Destes, quatro tiveram a expressão aumentada e os outros cinco tiveram a expressão reduzida. Por RT-qPCR foram confirmados dois alvos do miR528. Um dos alvos, o gene L-Ascorbato Oxidase está relacionado com a regulação da divisão celular. Sugere-se que a regulação pelo miR528 pode estar de acordo com a inibição do desenvolvimento das raízes em resposta ao alumínio em plantas sensíveis. Estes resultados demonstram que a resposta dos microRNAs ao tratamento com alumínio também é complexa, pois vários microRNAs, que provavelmente regulam alvos distintos tiveram sua expressão modulada após o tratamento das plantas. Já foi demonstrado que o desenvolvimento de raízes laterais sofre regulação por microRNAs em Arabidopsis. A outra parte deste trabalho visou caracterizar funcionalmente o miR164 e a expressão espacial de diferentes membros da família miR164 em raízes de plantas de arroz. Plantas superexpressando o miR164 apresentaram as raízes laterais reduzidas em comparação com as plantas não transformadas. Interessantemente, a análise por RT-qPCR de dois alvos do miR164 revelaram resultados inversos. A análise das plantas contendo os promotores de três genes da família miR164 fusionados ao gene repórter GUS revelou uma sobreposição da expressão espacial no órgão. Os microRNAs miR164a e miR164d localizam-se nas raízes laterais, e o miR164f está localizado nas raízes laterais e também na raiz primária. Porém, cortes transversais das raízes demonstraram que os miR164a e o miR164f localizam-se na endoderme e no estelo, respectivamente. Uma análise in silico das seqüências dos promotores dos seis membros da família miR164 e de seus respectivos alvos revelou a presença de motivos de DNA provavelmente responsivos a fatores de transcrição envolvidos no desenvolvimento de meristemas primários. Com base nestes resultados, sugere-se que os microRNAs miR164a e miR164f devem estar regulando seus alvos em diferentes tecidos da raiz. Este resultado está de acordo com o desenvolvimento inicial das raízes laterais, pois estas se originam do periciclo componente do estelo. Os microRNAs têm função crucial ao longo do desenvolvimento e em resposta a estresses abióticos. O papel regulatório dos microRNAs reside na complexidade e diversidade das respostas a estresses abióticos e ao desenvolvimento, visto que diversos microRNAs, que provavelmente regulam distintos alvos, estão envolvidos em redes complexas na regulação da expressão gênica. / Previous works showed that the response to aluminum (Al) in plants is complex. However, at present, there is no data regarding microRNA expression in this response. Part of this work aimed to characterize the expression profile of different families of microRNAs in response to Al comparing roots of japonica and indica roots. From sixteen microRNAs differentially expressed, thirteen were down-regulated and six were upregulated in japonica rice roots treated with 450 M of AlCl3 after 8h. For the indica rice roots under the same conditions, nine microRNAs were differentially expressed. From these microRNAs, four were up-regulated and five were down-regulated. Two miR528 targets were confirmed by RT-qPCR. One of the targets is the L-AScorbate oxidase gene, which regulate cell divisions. These results help explaining rice root inhibition under Al teatment in sensitive plants. Our results suggest that microRNA response to Al is also complex, because several microRNAs that had their expression modulated probably regulate distinct target genes. It is known that lateral root development is regulated by microRNAs in Arabidopsis. The other part of this work was to characterize the miR164 function and the spatial expression of different members of the miR164 family in rice roots. Plantas overexpressing the miR164 had less lateral roots when compared to the non-transformed plants. Interestingly, the expression by RT-qPCR of two miR164 target was inverse in the miR164 overexpressing plants. The spatial expression analysis of different members of the miR164 family revealed overlapping domains. MicroRNAs miR164a and miR164d localized in the lateral roots, and miR164 is localized in lateral roots and also in primary roots. However, hand-sectioning of the miR164a and miR164f GUS fusion plants demonstrate that miR164a is expressed in the endodermis and miR164f is expressed in the stele. An in silico analysis of promoter sequences of all miR164 genes revealed the presence of DNA motifs probably responsive to transcription factors involved in the development of primary meristems. Base on these results, we suggest that different members of the miR164 family are regulating their targets in different tissues of rice roots. These results are in agreement with the initial development of lateral roots originating from the pericycle cells. The key of the regulatory role of microRNAs in response to abiotic stresses and during development brought complexity to the regulatory networks of gene expression.

Análise funcional das isoformas citosólicas e peroxissomais de ascorbato peroxidade em arroz (Oryza sativa L)

As espécies reativas de oxigênio (ERO) são produzidas constantemente pelo metabolismo aeróbico. Em situações de estresse biótico ou abiótico, a produção é aumentada e a toxicidade das ERO pode conduzir a diversos danos celulares. As ERO atuam também como moléculas sinalizadoras regulando a expressão de genes de defesa a estresses, senescência, morte programada da célula, crescimento e desenvolvimento da planta, entre outros processos. Uma vez que as ERO são tóxicas e também participam de eventos de sinalização, as células vegetais requerem mecanismos que regulem finamente a concentração intracelular dessas moléculas. A ascorbato peroxidase (APx) é uma enzima fundamental do metabolismo antioxidante, catalisando a decomposição do peróxido de hidrogênio. Em arroz, a família APx é codificada por oito genes, cujas isoformas são caracterizadas por sua localização subcelular: citosólica, peroxissomal, mitocondrial ou cloroplastidial. Este trabalho teve como objetivo caracterizar funcionalmente as ascorbato peroxidases citosólicas (APx1 e APx2) e peroxissomais (APx3 e APx4) de arroz, estudando o papel destas isoformas nos mecanismos de defesa das plantas. A estratégia utilizada foi a obtenção e caracterização de plantas silenciadas para diferentes genes por RNA de interferência (RNAi). O padrão de expressão global da planta silenciada para os genes de APx citosólicas, em comparação com a planta não-transformada, foi avaliado através de análises de microarranjo e proteômica. A análise das plantas silenciadas APx1/2s revelou a existência de um mecanismo de compensação, com alteração de parâmetros fotossintéticos, ativação de outras enzimas antioxidantes e aclimatação, possivelmente sinalizados pelos níveis mais elevados de peróxido de hidrogênio. As plantas silenciadas para os genes de APx peroxissomais apresentaram atraso no desenvolvimento da panícula e maior susceptibilidade à senescência. Os genes OsAPx3 e OsAPx4 são mais expressos em folhas. A isoforma APx4 apresentou expressão em folhas, raízes e panícula de arroz, principalmente nas regiões do sistema vascular. Estas análises mostram que as isoformas de APx de arroz possuem funções diferentes, relacionadas com o compartimento celular em que estão localizadas. As enzimas APx fazem parte do complexo sistema antioxidante vegetal e estes resultados visam contribuir para um melhor entendimento do papel de APx no metabolismo celular e na defesa da planta. / The reactive oxygen species (ROS) are produced constantly by aerobic metabolism. In abiotic and biotic stress, the production is increased and ROS toxicity can cause several cellular damages. ROS also act as signaling molecules in the regulation of defense gene expression, senescence, programmed cell death, plant growth and development and other processes. Since ROS are toxic and also participate of signaling events, plant cells require mechanisms that tightly regulate the intracellular concentration of these molecules. Ascorbate peroxidase (APx) is an antioxidant metabolism enzyme, which catalyzes the hydrogen peroxide scavenging. In rice, the APx family is formed by eight genes, which isoforms are characterized by their subcellular localization: cytosol, peroxisome, mitochondria and chloroplast. This work aimed to characterize at functional level the cytosolic (OsAPx1 and OsAPx2) and the peroxisomal (OsAPx3 and OsAPx4) ascorbate peroxidase encoding genes in rice, studying the role of their products in plant defense mechanisms. The general strategy used was to produce and to characterize plants silenced for different APx genes by RNA interference (RNAi). The global expression pattern of silenced plants for cytosolic APx, compared with the non-transformed plant, was analyzed by microarray and proteomics experiments. These revealed the existence of a compensatory mechanism, which include alterations in photosynthetic parameters, activation of other antioxidant enzymes and acclimation, possibly induced by higher levels of hydrogen peroxide. Plants silenced for peroxisomal APx genes showed a delay in panicle development and were more susceptible to senescence. The expression analyses revealed that OsAPx3 and OsAPx4 genes present higher expression in leaves. The APx4 isoform is expressed in leaves, roots and panicles of rice, mainly in the vascular system. These analyses showed that rice APx isoforms may play different functions related to the cellular compartment in which they are located. APx enzymes are part of the complex plant antioxidant system and these results may contribute to a better understanding of the APx role in the cellular metabolism and plant defense.

Caracterização funcional dos genes ASR na resposta ao alumínio em arroz (ORYZA SATIVA)

O arroz é considerado o cereal mais tolerante ao alumínio (Al). Entretanto, a variabilidade entre os genótipos leva a uma considerável diferença quanto ao grau de tolerância para cultivares distintas. Diversos estudos mostram que plantas de arroz toleram o Al por intermédio de mecanismos internos e externos. Neste trabalho foi analisada a modulação da expressão da família gênica ASR (Aba, Stress and Ripening) em função do tratamento com Al. O gene ASR5 foi diferencialmente expresso em raízes de arroz tolerante ao Al (ssp Japonica cv Nipponbare). Entretanto, ASR5 não respondeu a exposição ao Al em raízes de arroz sensíveis ao Al (ssp Indica cv Taim). Plantas transgênicas silenciadas para os genes ASR apresentaram um aumento da sensibilidade ao Al. Calos embriogênicos de arroz transformados com a fusão ASR5-GFP revelaram que a proteína ASR5 localiza-se no núcelo e no citoplasma. Em protoplastos transformados, ASR5 localizou-se nos cloroplastos. Usando uma abordagem proteômica, comparando plantas não-transformadas e plantas ASR5_RNAi, um total de 41 proteínas com padrões contrastantes foi identificado. No intuito de identificar genes com expressão alterada pelo Al em arroz, e buscar genes afetados pelo silenciamento de ASR5, foi realizado o sequenciamento total dos transcritos utilizando plantas de arroz não transformadas e ASR5_RNAi em duas condições: controle e tratamento com Al. Essas análises transcriptômicas revelaram que 961 genes responderam ao Al em raízes de plantas de arroz não transgênicas submetidas ao Al. Nas plantas ASR5_RNAi, apenas 309 genes responderam ao tratamento com Al. Entretanto, apenas 52 desses genes se sobrepuseram quando comparados ao grupo de genes modulados em plantas não transformadas, sugerindo que a planta ASR5_RNAi perdeu a capacidade de regular um conjunto de genes. Além disso, análises de imunoprecipitação da cromatina seguida de sequenciamento em massa, revelaram que ASR5 liga-se ao promotor do gene STAR1, entre outros, regulando sua expressão em resposta ao Al. Os resultados mostram pela primeira vez que ASR5 atua como fator de transcrição em arroz e que está envolvido na regulação de genes responsivos ao Al conferindo tolerância frente à toxicidade do Al. / Among cereal crops, rice is considered the most aluminium (Al) tolerant species. However, variability among rice genotypes leads to remarkable differences in the degree of Al tolerance for distinct cultivars. A number of studies have demonstrated that rice plants achieve Al tolerance through internal and external mechanisms. We have analyzed expression changes of the rice ASR (Aba, Stress and Ripening) gene family as a function of Al treatment. The gene ASR5 was differentially regulated in the Al-tolerant rice ssp Japonica cv Nipponbare. However, ASR5 expression did not respond to Al exposure in Indica cv Taim rice roots, which are highly Al-sensitive. Transgenic plants carrying RNAi constructs that targeted the ASR genes displayed increased Al susceptibility in T1 plants. Rice embryogenic calli expressing an ASR5-GFP fusion revealed that ASR5 was localized in both the nucleus and cytoplasm. In transformed protoplasts, ASR5-GFP appears in chloroplast cells. Using a proteomic approach to compare non-transformed and ASR5_RNAi plants, a total of 41 proteins with contrasting expression patterns were identified. In order to identify genes with differential modulation under Al stress in rice, and search for genes affected by ASR5 silencing, RNA-seq was made in non-transformed plants and ASR5_RNAi plants in control conditions and after Al treatment. These analyses revealed that 961 genes responded to Al in non-transformed rice roots under Al stress. A total of 309 genes responded to Al in ASR5_RNAi plants. Only 52 genes overlapped between non transformed and ASR5_RNAi plants when comparing genes modulated by Al, showing that ASR-silenced plants lost the ability to modulate a set of genes in response to Al treatment. Furthermore, ChIP-Seq analysis revealed that ASR5 can bind to the promoter of STAR1, among other genes, regulating its expression under Al stress. These results show for the first time that ASR5 act as a transcription factor in rice and that it regulates Al-responsive genes conferring tolerance in rice against Al toxicity.
