Transferência de oxigênio e condições de cisalhamento em biorreator convencional com impelidores orelha de elefante

However, this impeller presents disadvantages such as high power consumption, and generates higher shear in the medium, which can cause irreversible morphological changes in microorganisms, damaging the culture performance. In literature, impellers called "Elephant Ear" (EE), presented in down-flow (down-pumping) (EEDP) and up-flow (up-pumping) (EEUP) configurations, are indicated as suitable for cultivation of shear sensitive cells. However, this characteristic has never been demonstrated quantitatively. In the present study, the oxygen transfer and hydrodynamics parameters were evaluated and compared in a conventional aerated stirredtank bioreactor equipped with impellers as the classical impeller Rushton turbine (RT) and with EE in EEDP and EEUP modes. Firstly, prediction equations were obtained for quantifying the average shear rate (Ýav) in the different systems, as function of the operating conditions (600≤N≤1000 rpm and 0.40≤Ýavar≤1.2 vvm) and rheological properties of the pseudoplastic fluids (K and n), based on a methodology that uses the volumetric oxygen transfer coefficient as characteristic parameter. The values of Ýav obtained from the correlations for the RT impeller were within a range of values estimated from correlations of the literature, showing that the methodology utilized is valid. Based on correlations of Ýav and on results of power consumption, the energy dissipation and Kolmogorov micro-scale (micro eddy sizes) could be estimated. For non-Newtonian fluids, the RT and EEUP impellers generated micro-eddies with smaller sizes in the range from 45.0-127.0 and 80.6-123.7 μm, respectively, showing that the impellers RT and EEUP are more shear than EEDP impeller. Using the prediction equations of Ýav the effect of shear conditions in cultures of Streptomyces clavuligerus were evaluated in conventional bioreactor equipped with RT, EEDP and EEUP impellers at 800 rpm, 0.50 vvm and 30C. Profiles of the volumetric oxygen transfer coefficient and shear rate were obtained during the cultivation. The morphological changes resulted from shear conditions imposed for the different impellers were analyzed using image analysis technique. The RT and EEDP impellers imposed the highest oxygen transfer and smaller shear to the broth, respectively. The flow pattern generated by EE impellers was crucial at the beginning of the cultivation, and is highly beneficial when EEUP impeller were used, since it presented a controlled shear throughout the cultivation. The best cultivation condition for the clavulanic acid production was obtained using the EEUP impeller, which resulted in a maximum concentration of 453 mg/L and a consistency index K= 2.5 Pa.s0.27. The results suggested that the isolated hyphae are directly related to the K of the broth and clavulanic acid production. According to the rapid fragmentation of clumps and emergence of isolated hyphae, it was concluded that the RT impeller presents shear rates higher than the EEs impellers, since the size of branched and non-branched hyphae were smaller and in greater quantities. This fact shows an advantage in the EEUP impeller use in shear sensitive microorganism cultures that requiring high levels of oxygen, without affecting the process performance. This advantage is associated with the flow pattern generated for the EEUP impeller. / Devido à natureza dos caldos de cultivo envolvendo microrganismos filamentosos aeróbios, de modo de obter adequada transferência de oxigênio e homogeneização do meio são tradicionalmente empregados biorreatores convencionais tipo tanque agitado e aerado utilizando o impelidor turbina de Rushton. Porém, esse impelidor apresenta desvantagens tais como o alto consumo de energia e elevado cisalhamento imposto ao meio, que pode provocar mudanças morfológicas irreversíveis aos microrganismos, prejudicando o rendimento dos cultivos. Na literatura especializada são indicados os impelidores denominados orelha de elefante ou Elephant Ear (EE), nas configurações de escoamento descendente (down-pumping) (EEDP) e de escoamento ascendente (up-pumping) (EEUP), como adequados para cultivos de células sensíveis ao cisalhamento. No entanto, esta caraterística nunca foi demostrada quantitativamente. No presente trabalho foram avaliados e comparados parâmetros de transferência de oxigênio e hidrodinâmicos em biorreator convencional tipo tanque agitado e aerado, equipado com impelidores como a clássica turbina de Rushton (TR) e com EE nas configurações EEDP e EEUP. Inicialmente foram obtidas equações de previsão para a quantificação da velocidade de cisalhamento média (Ýav Ýav) nos diferentes sistemas em função da frequência de agitação do impelidor e vazão especifica de ar e das propriedades reológicas de fluidos pseudoplásticos, com base em metodologia que utiliza o coeficiente volumétrico de transferência de oxigênio como parâmetro característico. Valores obtidos de Ýav para o impelidor TR pelas correlações obtidas ficaram dentro de uma faixa de valores estimados por correlações de literatura validando a metodologia empregada. Com base nas correlações de Ýav e nos resultados de consumo de potência, pode-se quantificar a dissipação de energia e a microescala de Kolmogorov (tamanho de micro-turbilhões). Para fluidos não-Newtonianos, os impelidores TR e EEUP geraram os menores micro-turbilhões com tamanhos nas faixas de 45,0-127,0 μm e de 80,6-123,7 μm, respectivamente, mostrando serem esses impelidores mais cisalhantes que o impelidor EEDP. Também com base nas equações de previsão de Ýav, foram avaliadas as influências das condições de transferência de oxigênio e de cisalhamento em cultivos de Streptomyces clavuligerus visando a produção de ácido clavulânico em biorreator convencional com os impelidores TR, EEDP e EEUP a 800 rpm, 0,50 vvm e 30C. A partir das amostras coletadas ao longo dos cultivos, foram obtidos perfis de kLa e de Ýav. Foram acompanhadas as mudanças morfológicas decorrentes das condições de cisalhamento impostas pelos diferentes impelidores mediante a técnica de análise de imagens. Os impelidores TR e EEDP impuseram, respectivamente, maiores e menores transferências de oxigênio e condições de cisalhamento ao caldo. A eficiência do padrão de escoamento gerado pelos impelidores EEs foi crucial nas primeiras horas do cultivo, sendo altamente benéfico quando utilizado o impelidor EEUP, o qual apresenta um cisalhamento controlado ao longo do cultivo. A melhor condição de cultivo em função da produção de ácido clavulânico foi obtida utilizando o impelidor EEUP obtendo uma produção máxima de 453 mg/L e índice de consistência K=2,5 Pa.s0,27. Os resultados sugerem que a quantidade de hifas isoladas está diretamente relacionada com o K do caldo e a produção de ácido clavulânico. De acordo com a rápida fragmentação de clumps e surgimento de hifas isoladas, conclui-se que o impelidor TR cisalha mais que os impelidores EE, isto é, pelos tamanhos das hifas ramificadas e não ramificadas que foram geradas em maiores quantidades e em menores dimensões. Tal fato mostra uma vantagem no uso do impelidor EEUP em cultivos de microrganismos sensíveis ao cisalhamento, e que necessitam de níveis superiores de oxigenação, não prejudicando o desempenho do processo. Estes resultados sugerem essa vantagem está associada ao padrão de escoamento imposto pelo impelidor EEUP.

Produção e otimização do processo de obtenção de ácido gama-poliglutâmico através do cultivo de Bacillus subtilis BL 53

O ácido y-poliglutâmico (y-PGA) é um biopolímero solúvel em água, aniônico, atóxico, biodegradável e biocompatível, produzido por Bacillus e que possui aplicações nas áreas química, médica, ambiental e de alimentos. Este trabalho tem como objetivo estudar o processo de produção do y-PGA a partir do cultivo de uma nova linhagem de Bacillus isolada de ambiente amazônico, o Bacillus subtilis BL53. Este estudo inclui a seleção de culturas, a otimização de condições de cultivo e a prospecção de substratos de produção alternativos. A habilidade de produção de y-PGA por linhagens de Bacillus isoladas na região amazônica foi investigada. A linhagem BL53 foi a linhagem selecionada, sendo identificada como Bacillus subtilis, através da análise da seqüência do gene 16S DNAr e de suas características bioquímicas. A avaliação das condições de cultivo submerso foi conduzida em agitador orbital mediante delineamento composto central rotacional (DCCR) que apontou como ponto ótimo de produção de y-PGA, a temperatura de 37 oC, o pH inicial de 6,9 e a concentração de 1,22 mM de Zn2+ suplementada ao Caldo E. Nas condições otimizadas, a produção de y-PGA foi igual a 10,4 g/L, cerca de três vezes maior que a obtida em condições convencionais de cultivo em Caldo E, utilizando a mesma linhagem. A influência da disponibilidade de oxigênio sobre a produção de y-PGA por B. subtilis BL53 foi avaliada em biorreator agitado de 5 L, com o emprego das velocidades de agitação de 500, 750 e 1000 rpm, sendo mantida fixa a taxa de aeração de 2 vvm. A produção de y-PGA mostrou-se altamente dependente da transferência de oxigênio, sendo que o teor de oxigênio dissolvido decaiu rapidamente nas primeiras 15 horas de cultivo, como resultado da elevada demanda de oxigênio pelas bactérias. O aumento na velocidade de agitação no biorreator possibilitou uma maior transferência de oxigênio e induziu o aumento na taxa de consumo de oxigênio pelas bactérias, conduzindo a maior produção e maior produtividade de y-PGA. A intensificação da agitação também influenciou os parâmetros cinéticos de crescimento do Bacillus subtilis BL53, provocando um aumento na velocidade específica de crescimento na fase logarítmica (max) sem provocar a perda da viabilidade celular. Com a utilização da velocidade de agitação de 1000 rpm em biorreator, o tempo de cultivo pode ser reduzido para menos de 48 h, cerca de 50% do tempo necessário para operação a 500 rpm. A investigação de substratos alternativos para produção de y-PGA mostrou o glicerol residual de biodiesel e o resíduo fibroso de soja como substratos promissores, apontando para a possibilidade de investigação em trabalhos futuros. / The poly-gamma-glutamic acid (y-PGA) is a water-soluble biopolymer, anionic, non toxic, biodegradable and biocompatible, it is produced by Bacillus and it has applications in chemical, medical, environmental and food industries. This work aims to study the process of production of y-PGA through cultivation of a new strain of Bacillus isolated from the Amazonian environment, Bacillus subtilis BL53. This study includes the screening of strains, the optimization of culture conditions and the investigation of alternative substrates. The ability of y-PGA production by Bacillus strains isolated from Amazonian environment was investigated. The BL53 strain was selected and identified as Bacillus subtilis, through analysis of 16S rDNA gene sequence and its biochemical characteristics. Evaluation of culture conditions in submerged cultivation was conducted in shaker using central composite design (CCD), which showed the temperature of 37 oC, the initial pH 6.9 and concentration of 1.22 mM Zn2+ in Medium E as the optimal conditions to y-PGA production. Under optimized conditions, the production of y-PGA was 10.4 g/L, about threefold what was obtained using this strain under conventional cultivation in Medium E. The influence of oxygen availability on the production of y-PGA by B. subitlis BL53 was evaluated in a 5 l stirred bioreactor with the use of stirring rates of 500, 750 and 1000 rpm, using a fixed aeration rate of 2 vvm. Production of y-PGA was highly dependent on oxygen transfer, and the dissolved oxygen content decreased rapidly in the first 15 hours of culture as a result of high oxygen demand by bacteria. The increase in stirring rate in the bioreactor allowed a better oxygen transfer and induced a rise in the oxygen uptake rate by bacteria, leading to higher production and higher productivity of y-PGA. The intensification of the stirring also influenced the kinetics growth parameters of Bacillus subtilis BL53, producing an increase in specific growth rate in the logarithmic phase (max) without causing reduction of cell viability. Using the stirring rate of 1000 rpm in a bioreactor, the cultivation time can be reduced to less than 48 h, about 50% of the time required for operation at 500 rpm. The investigation of alternative substrates for the production of y-PGA showed crude glycerol from biodiesel and soybean industrial fibrous residue as promising ones, pointing to research possibilities in future work.

Uso de carreadores de oxigênio na produção de ácido-poliglutâmico através do cultivo de bacillus subtilis bl53 e caracterização do biopolímero

O ácido ƴ-poliglutâmico (ƴ-PGA) é uma homopoliamida aniônica, biodegradável, comestível e atóxica, sintetizada por bactérias do gênero Bacillus, podendo ser utilizado nas indústrias alimentícia e de cosméticos, na medicina e no tratamento de águas residuais. Este trabalho teve como objetivo caracterizar e identificar potenciais aplicações para o ƴ-PGA obtido através do cultivo submerso de Bacillus subtilis BL53, conduzido sob condições otimizadas em trabalhos anteriores. Além disso, foi avaliado o efeito de diferentes inóculos e da adição de precursores da rota metabólica na produção do biopolímero. A melhor condição obtida foi testada em biorreatores com adição de polidimetilsiloxano (PDMS) como carreador de oxigênio, com o objetivo de aumentar a produtividade do biopolímrero. A massa molar média (Mw), obtida através de espalhamento de luz estático, na ordem de 106 g mol-1 não apresentou diferenças significativas para o biopolímero obtido após 48 e 96 h de cultivo. As análises reológicas conduzidas em viscosímetro rotacional indicaram que os polímeros obtidos após 48 e 96 horas apresentaram comportamento Newtoniano, sendo que após 96 horas a viscosidade absoluta foi maior. As análises térmicas (calorimetria diferencial exploratória e análise termogravimétrica) indicaram a temperatura de fusão (Tm) de 134 ºC e 128 ºC e o intervalo de degradação (Td) entre 120 ºC - 190 ºC e 120 ºC - 215 ºC, para os biopolímeros obtidos após 48 e 96 horas de cultivo respectivamente. O caldo LB apresentou-se como o melhor inóculo para a produção de ƴ-PGA. A adição dos precursores L-glutamina e ácido -cetoglutárico aumentou em 20 % a produção do biopolímero. A adição de 10 % de PDMS nos cultivos em biorreatores aumentou o coeficiente volumétrico de transferência de massa (KLa) e a produção e produtividade do ƴ-PGA, sendo produzidos 23.5 g L-1 do biopolímero em 24 horas de cultivo, uma produtividade aproximadamente 40 % superior às obtidas por outros autores utilizando o mesmo microrganismo. / Poly-ƴ-glutamicacid (ƴ-PGA) is an anionic, biodegradable, non-toxic and edible homopolyamide, synthesized by bacteria of the genus Bacillus, being used in food, cosmetics, medicine and waste water treatment. The aim of this study is to characterize and indentify potencial applicatiions for the ƴ-PGA obtained by submerged cultivation of Bacillus subtilis BL53, conducted under optimized conditions in previous studies. We also evaluated the effect of different inoculants and addition of precursors in the metabolic pathway of production of the biopolymer. The best condition obtained yet been tested in bioreactors with addition of polydimethylsiloxane (PDMS) as a carrier of oxygen in order to further increase the productivity of biopolymer. The average molecular weight (Mw) obtained by static light scattering, on the order of 106 g mol-1, showed no significant differences for biopolymer obtained after 48 and 96 h of cultivation. Analyses conducted in rotational viscometer indicated that biopolymers after 48 and 96 h have a Newtonian behavior, and the 96 hours had higher absolute viscosity. The thermal analysis (differential scanning calorimetry and thermo gravimetric analysis) indicated the melting temperature (Tm) as 134 ºC and 128 ºC and degradation temperature range (Td) of 120 ºC - 190 ºC and 120 ºC - 215 ºC, after 48 and 96 hours respectively. It was found that the best inoculum medium for biopolymer production was the LB broth. The addition of the precursors L-glutamine and -ketoglutaric acid increased in 20% the ƴ-PGA production. The addition of 10% of PDMS in bioreactors cultures increased the mass transfer volumetric coefficient (KLa) and the production and productivity of ƴ-PGA, being produced 23.5 g l-1 of the biopolymer in 24 hours of cultivation, a productivity about 40 % higher than those obtained by other authors using the same microorganism.

Plants produce a variety of secondary metabolites, e.g. to defend themselves against herbivores or to attract pollinating insects. Plant cell biotechnology offers excellent opportunities in order to use such secondary plant metabolites to produce goods with consistent quality and quantity throughout the year, and therefore to act independently from biotic and abiotic environmental factors. This article presents results of an extensive study of plant cell in vitro cultivation in a modern shake flask system with non-invasive online respiration activity monitoring unit. Comprehensive screening experiments confirm the successful transfer of a model culture (sunflower suspension) into the shake flask monitoring device and the suitability of this respiration activity monitoring unit as qualified tool for screening of plant in vitro cultures (sunflower and sage suspension). The authors demonstrate deviations between online and offline data due to varying water evaporation from different culture flask types. The influence of evaporation on growth-specific parameters thereby rises with increasing cultivation time. Furthermore, possibilities to minimize the impact of evaporation, either by adjusting the inlet air moisture or by measuring the evaporation in combination with an appropriate correction of the measured growth values, are shown.

Phototrophic growth of Arthrospira platensis in a respiration activity monitoring system for shake flasks (RAMOS)

Optimising illumination is essential for optimising the growth of phototrophic cells and their production of desired metabolites and/or biomass. This requires appropriate modulation of light and other key inputs and continuous online monitoring of their metabolic activities. Powerful non-invasive systems for cultivating heterotrophic organisms include shake flasks in online monitoring units, but they are rarely used for phototrophs because they lack the appropriate illumination design and necessary illuminatory power. This study presents the design and characterisation of a photosynthetic shake flask unit, illuminated from below by warm white light-emitting diodes with variable light intensities up to 2300 μmol m-2 s-1. The photosynthetic unit was successfully used, in combination with online monitoring of oxygen production, to cultivate Arthrospira platensis. In phototrophic growth under continuous light and a 16 h light/8 h dark cycle (light intensity: 180 μmol m-2 s-1), the oxygen transfer rate and biomass-related oxygen production were - 1.5 mmol L-1 h-1 and 0.18 mmol O2 gx-1 h-1, respectively. The maximum specific growth rate was 0.058 h-1, during the exponential growth phase, after which the biomass concentration reached 0.75 g L-1.

Réponse biologique de cellules animales à des contraintes hydrodynamiques : simulation numérique, expérimentation et modélisation en bioréacteurs de laboratoire / Biological response of animal cell to hydrodynamic stresses : numerical simulation, experimentation and modelling in bench-scale bioreactors

La réponse globale de cellules animales à des contraintes hydrodynamiques lors de leur culture en suspension dans des réacteurs agités a été étudiée grâce à une approche intégrative couplant les outils du génie biochimique à ceux de la mécanique des fluides numérique. En premier lieu, la description de l’hydrodynamique moyenne et locale de deux systèmes de culture agités de laboratoire, spinner et bioréacteur, a été réalisée. Puis, l'étude des cinétiques macroscopiques de cellules CHO cultivées en suspension, en milieu sans sérum et sans protéine, a été réalisée avec différentes vitesses d’agitation, pour évaluer l'impact de l'agitation sur les vitesses de croissance et de mort cellulaires, ainsi que de consommation des substrats et de production des métabolites et de l'interféron-gamma recombinant. Des caractérisations supplémentaires des cellules (apoptose, protéines intracellulaires) et de l'interféron ont également été réalisées. Les effets de l'intensification de l'agitation ont été représentés avec plusieurs corrélations globales reliant : (i) en milieu contenant du pluronic, l'intégrale des cellules viables au nombre de Reynolds, et la proportion de cellules lysées à la valeur moyenne de l'énergie de dissipation, <[epsilon]? (ii) en milieu sans pluronic, les vitesses spécifiques de croissance et de mort cellulaires à <[epsilon]. De plus, l'analyse par CFD de la distribution spatio-temporelle des contraintes indique que la lyse cellulaire, observée dans le réacteur aux conditions extrêmes d'agitation, serait plutôt liée à des valeurs locales très élevées de [epsilon], ainsi qu’à la fréquence d'exposition des cellules dans ces zones énergétiques. Un modèle hydro-cinétique original, couplant l’hydrodynamique locale aux cinétiques cellulaires de croissance et de mort, et basé sur l’intermittence de la turbulence permet la prédiction de la lyse massive observée en réacteur sous certaines conditions. Pour confirmer le fait que les effets liés à l'intensification de l'agitation sont bien le résultat d'une augmentation des contraintes hydrodynamiques, et non d'une amélioration du transfert d'oxygène, ce dernier a été mesuré et modélisé par couplage avec une simulation numérique de type Volume Of Fluid , concluant en une absence de limitation d'oxygène. Enfin, la conception, le dimensionnement et la caractérisation hydrodynamique d'un réacteur innovant de type Couette-Taylor, sont proposées pour la mise en œuvre de cultures perfusées dans un environnement hydrodynamique mieux contrôlé / The global response of animal cells to hydrodynamic stress when cultivated in suspension in stirred tank reactors was studied. To do this, an integrative approach coupling biochemical engineering and fluid mechanics tools were used. First, the description of the global and local hydrodynamics of two bench-scale agitated reactors, a spinner flask and a bioreactor, was carried out. Then, macroscopic kinetics of CHO cells cultivated in a serum and protein-free medium were obtained at various agitation rates, in order to evaluate the impact of agitation on cellular growth and death, as well as substrates consumption and metabolites and recombining IFN-[gamma] production. IFN-[gamma] and cells physiological state were more precisely characterised by glycosylation, apoptosis state and intracellular proteins measurements. The effects of the agitation increase were represented by several global correlations that related: (i) in a medium containing Pluronic F68, the Integral of the Viable Cells Density to the Reynolds number, and the proportion of lysed cells with the average value of energy dissipation rate <[epsilon]? (ii) in a medium without pluronic, specific cell growth and death rates to <[epsilon]. Moreover, CFD analysis of the stress distribution indicated that the cellular lysis observed in the bioreactor at the highest agitation rate, would be related to very high local values of [epsilon], and to the exposure frequency of the cells in these energetic zones. An original hydro-kinetic model based on the intermittency of turbulence and coupling the local hydrodynamics with cell growth and death kinetics, allowed the prediction of the massive cell lysis observed in the bioreactor under some mixing conditions. To decouple shear stress effects from oxygen transfer improvement, the oxygen transfer coefficient was experimentally measured and modelled using a Volume Of Fluid numerical simulation. Our results indicated the absence of an oxygen limitation, which confirmed that this cell response resulted from the hydrodynamic stress increase alone. Lastly, an innovative continuous and perfused Couette-Taylor reactor, allowing a better-controlled hydrodynamic environment was designed and sized. A atividade catalítica de clusters trigonais de acetato de rutênio e porfirinas supramoleculares contendo quatro unidades de clusters periféricos ou complexos de bis(bipiridina)rutênio, na oxidação de substratos orgânicos por iodosil-benzeno ou terc-butil-hidroperóxido, é abordada nesta tese. Foram feitos estudos cinéticos para elucidar o mecanismo de catálise, com especial destaque para duas espécies supramoleculares isoméricas representadas por Mn(3-TRPyP) ou Mn(4-TRPyP), nos quais os complexos de bis(bipiridina)(cloro)rutênio se coordenam à tetrapiridilporfirina através das posições meta ou para da ponte piridínica, respectivamente. Além da maior seletividade proporcionada pelos catalisadores supramoleculares, a substituição do íon cloreto pela água nos complexos periféricos de rutênio, intensificou a atividade catalítica, gerando novos sítios ativos, do tipo Ru(IV)=O. Também foram estudados clusters trinucleares de rutênio, altamente reativos, no estado Ru(III)Ru(IV)Ru(IV)=O. Estes foram gerados eletroquimicamente em solução aquosa, apresentando pronunciada atividade catalítica na oxidação do alcool benzílico. Finalmente, uma espécie dimérica de cluster, ainda inédita, com ponte oxo, foi caracterizada e investigada do ponto de vista catalítico. / The catalytic activity of trinuclear ruthenium clusters and supramolecular tetrapyridylporphyrins containing four peripheral cluster units or bis(bipyridine)ruthenium complexes, in the oxidation of organic substrates by iodosylbenzene or tert-butil-hydroperoxide, is dealt with in this thesis. Kinetic studies were performed in order to elucidate the mechanisms of catalysis involving two isomeric species denoted Mn(3-TRPyP) or Mn(4-TRPyP), in which the bis(bipyridine)(chloro)ruthenium complexes are bound to the tetrapyridylporphyrin center, at the meta or para positions of the pyridine bridge, respectively. In addition to the improved selectivity by such supramolecular catalysts, the substitution of the chloride ion by water in the peripheral ruthenium complexes, greatly enhanced their catalytic activity by generating new Ru(IV)=O reactive sites. Highly reactive trinuclear ruthenium clusters of the type Ru(III)Ru(IV)Ru(IV)=O were also generated electrochemically in aqueous solution, exhibiting pronounced catalytic activity in the oxidation of benzyl alcohol. Finally, a novel dimeric oxo-bridged cluster species was characterized, and its role in catalysis investigated.
