Optical Scattering Properties of Fat Emulsions Determined by Diffuse Reflectance Spectroscopy and Monte Carlo Simulations

To estimate the propagation of light in tissue-like optical phantoms (fat emulsions), this thesis utilized the diffuse reflectance spectroscopy in combination with Monte Carlo simulations. A method for determining the two-parametric Gegenbauer-kernal phase function was utilized in order to accurately describe the diffuse reflectance from poly-dispersive scattering optical phantoms with small source-detector separations. The method includes the spectral collimated transmission, spatially resolved diffuse reflectance spectra (SRDR) and the inverse technique of matching spectra from Monte Carlo simulations to those measured. An absolute calibration method using polystyrene micro-spheres was utilized to estimate the relation between simulated and measured SRDR intensities. The phase function parameters were comparable with previous studies and were able to model measured spectra with good accuracy. Significant differences between the phase functions for homogenized milk and the nutritive fat emulsions were found.

Optical coherence tomography is an imaging modality with an outstanding resolution. During the project, a time domain OCT system based on a Michelson fibre interferometer was implemented and put into operation. A super-luminescent diode with a centre wavelength of 1295nm and a bandwidth of 45nm was selected as light source and a linear variable delay line as reference. Basic tests were made on phantoms constructed of filter foils and on gel-like agar slices with optical properties similar to human tissue. It was shown that the achievable resolution was at least 36um and can be increased. The system can easily be enhanced to create two-dimensional images. / Optische Kohärenztomographie ist ein bildgebendes Verfahren mit einer hervorragenden räumlichen Auflösung. Im Laufe des Projekts wurde ein OCT-System basierend auf einem faseroptischen Michelson-Interferometer implementiert und in Betrieb genommen. Als Lichtquelle wurde eine Superlumineszenzdiode mit einer Mittenwellenlänge von 1295nm und einer Bandbreite von 45nm gewählt. Eine variable optische Verzögerungsleitung diente als Referenz. Erste Messungen an Filterfolien und gelähnlichen Agarphantomen, die die optischen Eigenschaften von menschlichem Gewebe nachbildeten, lieferten eine räumliche Auflösung von mindestens 36um. Durch die modulare Bauweise ist das System leicht für zweidimensionale Aufnahmen erweiterbar.

Ultrasound Assisted Optical Elastography For Measurement Of Mechanical Properties Of Soft Tissue Mimicking Phantoms

This work describes the development of an optical probe for measuring movement of tissue particles deep inside which are loaded by an ultrasound remote palpation device. The principle of the method is that ultrasound force which can be applied inside the tissue makes the tissue particles vibrate and this vibration phase-modulates the light intercepting the insoniified region which results in a modulated speckle intensity on detection outside the object. This speckle intensity modulation detected through the measured intensity autocorrelation is a measure of the vibration amplitude. Since the vibration amplitude is related to the local elastic properties of the medium, the measured modulation depth in intensity autocorrelation can be used to map the elastic property in the insonified region. In this work, first the ultrasound induced force is calculated for both plane and focused ultrasound beams, and converted to amplitude of vibration and refractive index modulation, solving the forward elastography equation. Light propagation inside an insonified object is modelled using Monte Carlo simulation and the amplitude and intensity correlations are computed. The modulation depth on the autocorrelation is estimated and shown that it is inversely correlated to the local elastic modulus and optical absorption coefficient. It is further shown that whereas the variation in modulation depth is linear with respect to absorption coefficient, the same variation with elastic property is nonlinear. These results are verified experimentally in a tissue mimicking phantom. The phantom was constructed out of poly vinyl alcohol(PVA) whose optical, mechanical and acoustic properties are independently controlled. It is also shown that for loading with focused ultrasound beam the displacement is almost along the ultrasound transducer axis and therefore the contribution from refractive index modulation alone can be ascertained by probing the insonified perpendicular to the transducer axis. This helps one to find the contribution to the modulation depth from the ultrasound-induced vibration, which can be used to compute a quantitative estimate of the elastic modulus from the modulation depth.

MODELING AND DEVELOPMENT OF THREE-DIMENSIONAL GEL DOSIMETERS

A dynamic mathematical model was developed to simulate the response of polyacrylamide gel (PAG) dosimeters to a single spherical radioactive brachytherapy seed. Simulations were conducted for a high dose-rate (HDR) seed using 192Ir and a low dose-rate (LDR) seed using 125I. The model is able to predict the amount of polymer formed, the crosslink density, and the volume fraction of aqueous phase as a function of radial distance and time. Results show that PAG dosimeters can provide accurate HDR brachytherapy dosimetry at distances larger than 4 mm from the centre of the seed but will give poor results for LDR due to monomer diffusion. Experiments were conducted to evaluate the potential for using pentacosa-10,12-diynoic acid (PCDA) as the reporter molecule in micelle gel dosimeters for optical computed tomography (CT) readout. Several gels containing PCDA that was solubilized using sodium dodecyl sulfate (SDS) responded to radiation by changing from colourless to blue. Unfortunately, all phantoms that showed colour changes were turbid, making them unsuitable for optical CT scanning. Several techniques were used to produce transparent gels containing PCDA but none of these gels responded noticeably to radiation. Only turbid gels with precipitated PCDA responded, indicating that the colour change was due to oligomerization within PCDA crystals and that PCDA molecules solubilized in micelles did not undergo oligomerization. As a result, PCDA is not suitable for use in radiochromic micelle gel dosimeters. A new recipe for a radiochromic leuco crystal violet (LCV) micelle gel dosimeters with enhanced dose sensitivity was developed for optical CT readout. The recipe contains LCV, trichloro acetic acid (TCAA), Cetyl Trimethyl Ammonium Bromide (CTAB), 2,2,2-Trichloroethanol (TCE), and gelatin. Experiments were conducted to improve understanding about interactions between the different components of LCV micelle gel, highlighting the importance of pH on dose sensitivity and transparency. Results also showed the effectiveness of chlorinated compounds in improving dose sensitivity. Statistical techniques were used to build empirical models that were used to optimize the gel recipe. Este trabalho apresenta o desenvolvimento de um estimulador galvânico vestibular para ser usado em experimentos de imageamento por ressonância magnética funcional (fMRI). Em experiências de fMRI é necessário a produção de estímulos somatossensoriais no paciente. Os estímulos devem ser sincronizados com a sequência de pulsos da fMRI. O estimulador foi dividido em circuitos analógicos (colocados dentro da sala do magneto) e circuitos digitais (sala de comando do sistema de MRI). A comunicação entre os circuitos é feita através de fibra óptica. Experimentos de fMRI realizados com voluntários demonstraram que o estimulador proposto é capaz de manter a sincronização com sistema de fMRI e pode ser usado para localizar as áreas do cérebro que são ativados pelo sistema vestibular. / This work presents the development of a galvanic vestibular stimulator to be used in functional magnetic resonance imaging experiments (fMRI). In fMRI experiments it is required the production of somatosensory stimuli in the patient must be sincronized with the fMRI pulse sequence. The stimulator circuits were divided in analog circuits (placed within the magnet room) and digital circuits (placed in the MRI command room). The communication between the circuits is made through optical fiber. fMRI experiments performed with volunteers demonstrated that the proposed stimulator is able to keep the sincronization with the MRI system and can be used to locate the brain areas that are activated by the vestibular system.
