Isolation, characterisation and molecular typing of feline mycoplasma speciesRobinson, Sally Rae January 2009 (has links)
The exact role of mycoplasma in feline ocular and respiratory disease is not yet understood. The results of previous studies are contradictory in this regard. There is some evidence to suggest that M. felis has a pathogenic role in such diseases, but it is inconclusive. / The aim of this study was to investigate the prevalence and anatomical distribution of mycoplasmas in a population of shelter cats, to determine which species were present, and establish the association of their presence with ocular or respiratory disease. / The prevalence of mycoplasma in the 110 cats examined was 71.8%, as determined by in vitro culture. Mycoplasma was most commonly isolated from the pharynx, followed by the bronchus and conjunctiva. In infected cats, mycoplasmas were likely to be isolated from multiple anatomical sites. / The polymerase chain reaction (PCR) was used to amplify part of the 16S rRNA gene, and the mutation scanning technique non-isotopic single-strand conformation polymorphism (SSCP) was utilised to delineate mycoplasma isolates based on nucleotide sequence variation. PCR-SSCP proved to be a useful method to screen large numbers of samples for variation and to group them according to species. / The species of mycoplasma identified by nucleotide sequencing were M. felis and M. gateae. It was not determined whether it was possible to differentiate between M. gateae and M. arginini based on SSCP profile results with the target DNA region used due to their almost identical nucleotide sequence. This group of M. gateae/M. arginini served as a useful non-pathogenic comparison group to M. felis. / There was no statistically significant difference between M. felis and the M. gateae/M. arginini group with respect to prevalence or anatomic distribution. There was no evidence of any association of mycoplasma with disease linked to any of the anatomic locations studied. / Mycoplasmas were isolated from the lower respiratory tract in 42.7% of cats. The isolation of mycoplasmas from the lower respiratory tract of healthy cats has been reported once, but this is the first report of M. felis being isolated from this location in healthy cats. This finding indicates that the isolation of mycoplasmas from the lower respiratory tract is not sufficient evidence to implicate a role in respiratory disease. / Mycoplasmas were not significantly involved in ocular or respiratory disease in the population of cats studied. More likely, they are commensal organisms in the conjunctiva, pharynx and bronchus. Whether they are capable of playing an opportunistic role in disease, or what conditions may facilitate such a role remains to be determined.

Endoparazité koní / Endoparasites of horsesScháňková, Štěpánka January 2015 (has links)
Nine ponies were randomly allocated to one of three groups (A, B, C). Gradually, these groups of ponies were experimentally infected with third stage cyathostome larvae. Each pony was infected with 30,000 infective cyathostome larvae stored 4 weeks under the natural environmental conditions of the Czech Republic. These animals were examined post--mortem for small strongyle infections from October 2011 to January 2012. Seventeen species of the following Cyathostominae genera were identified: Coronocyclus, Cyathostomum,Cylicocyclus, Cylicostephanus, Gyalocephalus and Petrovinema. Of the 17 species of Cyathostominae recovered, Cyathostomum catinatum, Cyathostomum alveatum and Cylicocyclus nassatus were the most prevalent (100 %). From the entire population of Cyathostominae, 65,6 % were located in the caecum and only 34,4 % in the colon. Only 1,736 (24,6 %) females and 5,329 (75,4 %) males were observed. In the first and second phase (group A, B) of the experiment, 98.57 % and 95.48 % of adult nematodes were found, respectively. However, only 29,07% of adults were found in the third phase (group C) of experiment, and the proportion of arrested larvae (EL3) was 67.73%. All ponies were treated orally with ivermectin at a dosage of 400 ug /kg. However, adult Oxyuris equi were present in six horses. This is the first European study to demonstrate anthelmintic resistence in Oxyuris equi to macrocyclic lactones.

Diverzita kryptosporidií infikujících hlodavce podčeledi Arvicolinae v České republice / Diversity of Cryptosporidium spp. infecting rodents from subfamily Arvicolinae in the Czech RepublicHÁJKOVÁ, Ivana January 2013 (has links)
Abstract In order to examine the prevalence of Cryptosporidium in wild Arvicolinae in the Czech Republic and understand the role that wild rodents play in the transmission of this parasite to humans and livestock, 152 faecal samples from 129 common voles (Microtus arvalis) and 23 bank voles (Clethrionomys glareolus) were collected on 9 localities in 2012. All samples were examined for presence of Cryptosporidium sp. using both the aniline-carbol-methyl violet staining method and molecular tools. The age, sex and faecal consistency were noted at the time of sampling. Microscopical examination revealed the presence Cryptosporidium sp. oocysts in 2 samples originated from common voles and 2 samples from bank voles. Genotyping was done through PCR amplification and characterization of the SSU rRNA and actin loci. Cryptosporidium specific DNA was detected in 10 samples (4 from common voles and 6 from bank voles) including those microscopically positive. Cryptosporidium infection was not linked to diarrhoea. Sequence and following phylogeny analyses revealed two new Cryptosporidium genotypes originated from bank voles and two new genotype from common vole, phylogeneticaly distinct from known species and genotypes. The host specificity needs to be verified by experimental infection in the future.

Endoparazitární infekce ovcí a koz v různých podmínkách chovu / Endoparasite infections in sheep and goats in diverse farming systemsUHLÍŘOVÁ, Barbora Isatou January 2013 (has links)
Samples of excrements for parasitological investigation were obtained in two year period 2011 and 2012 on three sheep farms and two goat farms in South Bohemia (a farm conventional, an organic and converting to an organic farming system). A total of 400 samples were examined using four parasitological methods. Thirteen species of gastrointestinal nematodes and one species of Protozoa were recovered. In farms Coccidia Eimeria spp. was the most diagnosed. Results revealed that goats were 1,8 times more often infected with parasites than sheep (x2 = 6,274; d. f. = 1; p = 0, 00576; OR = 1,82). Animals younger 6 months were 3 times more often infected with Eimeria than older ones (x2 = 17,174; d. f. = 1; p = 0,00003; OR = 3,16). Goats were 11 times more often infected with parasites in winter than sheep (x2 = 5,8174; d. f. = 1; p = 0,0050; OR = 10,78). Goats were 2 times more often infected with lungworm infection than sheep (x2 = 8,407; d. f. = 1; p = 0,0019; OR = 1,99). A statistically significant difference in infectious contamination between conventional and an organic system bred animals was not discovered (x2 = 0,0145; d. f. = 1; p = 0,452; OR = 1,05).

Střevní paraziti koz ve vybraném chovu / Gut parasites of goats on a chosen farmVANĚČKOVÁ, Michala January 2011 (has links)
The master thesis is aimed to evaluate intestinal parasites occurrence in a goat breeding. A herd of goats chosen for this survey was kept by a private enterprise focused on milk production. The practical research was conducted during two years, 2009 and 2010. Samples of feaces were collected during that period and analyzed for parasites occurrence. The analyses proved presence of the following parasites: Trichuris ovis, parasites from the order Strongylida, Fasciola hepatica, Dicrocellium lanceatum and coccidia. Total prevalence was significant; 85% of the samples were found to be positive. Species from the order Strongylida were identified as the most often 41,46 %, which meant 199 cases from the total of 509. During the examined period, the highest prevalence of parasites was detected in summer 2009, the second highest then in spring 2010.

InfecÃÃes relacionadas à assistÃncia à saÃde e fatores associados em pacientes transplantados renais em Fortaleza â CE. / Healthcare-related infections and associated factors in renal transplant recipients in Fortaleza-CE.Regina Kelly GuimarÃes Gomes 17 July 2014 (has links)
The healthcare-related infections are among the major complications in patients who undergo kidney transplant, by virtue not only of immunosuppression schemes to which they are subjected, as the care carried out by health teams. The aim of this study was to analyze healthcare-related infections in renal transplant recipients in 2012, in the city of Fortaleza, which has consolidated renal transplant services. A cross-sectional study where outpatient records were analyzed, charts and tokens for notification and investigation of patients who performed CCIH kidney transplant in 2012, in HUWC and HGF, institutions with successive records in this type of procedure. A total of 237 attendees, being, 101 (average age: 43.2 years) belonging to the HUWC and 136 (average age: 45.4 years) to the HGF, was included in the study. In both institutions, most people were male, married and resided in the Capital of CearÃ. Most of them also had the normal BMI was hypertensive, had as main causes of IRC: inderterminada, SAH and lupus nephritis. The proportion of invasive procedures performed were: graft biopsy (HUWC: 45.54%; HGF: 26.47%), central venous catheter puncture (HUWC: 98.01%; HGF: 97.06%), FAV (HUWC: 66.33%; HGF: 94.11%), and passage of double-J catheter (HUWC: 39.6%; HGF: 22.06%). The estimated prevalence of IRAS in 101 renal transplant recipients at HUWC was 50 (49.05%), and HGF, 31 (22.79%). The most common IRAS, both in HUWC, as in HGF, were urinary tract infections, and the main etiological agents isolated Klebsiella pneumoniae and Escherichia coli. In HUWC, the socio-demographic factors, clinicians and epidemiologists who presented statistically significant association with involvement by IRAS were: the classification of BMI (p<0.03) time on dialysis before transplantation (p<0.05), the total length of stay (p<0.0001), the time of surgery (p<0.001), cold ischemia time (p<0.01), the passage of double-J catheter (p<0.003), the speaking time of TOT (p<0.03) and time of use of the SVD (p<0.04) in HUWC; on HGF, classification of BMI (p<0.04), LES as a cause of IRC (p<0.01), blood transfusion before transplantation (p<0.02), the total length of stay (p<0.001) and time of use of the CVC (p<0.04). Therefore, throughout the perioperative period, there is a need for development of cautious actions throughout the health team, in order to prevent infections and unnecessary government spending with hospitalizations and prolonged treatments, supporting the growth of renal transplantation technique in the State in recent years.

O PCR, nested-PCR e seqüenciamento do gene recA apresentaram boa especificidade na identificação do CBc e dos genomovares, entretanto, ainda ocorrem algumas limitações, decorrentes da variedade genotípica, sendo necessária a utilização de métodos mais abrangentes, como o MSLT / Abstract: Objectives: Determine the Burkholderia cepacia complex (BCC) colonization/infection and genomovar prevalence in cystic fibrosis patients and compare the clinical, tomographic and functional severity indicators of lung injury in two groups of patients: GI - patients colonized or infected with BCC and GII - patients without BCC colonization or infection, with the aim of assessing the impact of this microorganism in pulmonary deterioration. Methods: A clinical-laboratory and prospective study was conducted with 222 CF patients seen in the CF outpatient (pediatrics and adults) in UNICAMP. Patients were divided into two groups: GI (n = 50) and GII (n = 134). Microorganisms were identified by conventional biochemical tests, Vitek2®Compact, recA-PCR and recA-nested-PCR with species-specific primers. The recA gene sequencing was performed for strains with inconclusive PCR reactions. Patients were classified by Schwachman's clinical score and measures of weight/age, height/age and BMI/age were used to assess nutritional status. The tomography changes were classified by Bhalla's score and lung function was assessed by spirometry. Data were compared between groups with the aim of assessing the CBC impact in lung injury. Results: The BCC prevalence was 22.5% and the most prevalent genomovars was: Bukholderia multivorans (30.0%), followed by Burkholderia cepacia (24.0%), Burkholderia cenocepacia IIIA (10.0%), Burkholderia cenocepacia IIIB (2%) and Burkholderia vietnamiensis (2.0%). In group I, 26.0% of patients were infected, 18.0% had transient colonization and 56.0% intermittent colonization by BCC. There was no statistical difference in the average point of Schwachman's score (p = 0.07) or in the severity classification (p = 0.611) but had differences in variables: nutritional status (p = 0.020) and general activity (p = 0.026). In the average point Bhalla's score was statistical difference (p = 0.04) and between the parameters: severity of bronchiectasis (p = 0.007), peribronchial thickening (p = 0.013), extent of bronchiectasis (p = 0.010), general the bronchial tree (p = 0.020). The pulmonary function test showed: FVC (%) (p = 0.076), FEV1 (%) (p = 0.066), FEF25-75% (p = 0.312) and FEV1/FVC (p = 0.312). Respiratory disorders classification: DVO in GI and GII = 4.8% = 23.8%, DVR in GI and GII = 9.5%, DVM in GI and GII = 19.0% = 1.6% and with DVM FVC decreased in GI and GII = 47.6% = 30.2%. Nutritional assessment was significant difference between the mean weight (kg) and height (cm) (p = 0.02.) CCoonncclluussiioonnss:: The BCC prevalence in our center is significantly higher than the dates provided by national and international literature. The increased Burkholderia multivorans prevalence differs from the most studies. It is suspected that may have been cross-infection between patients or is a regional characteristic. The Shwachman's and Bhalla score, spirometry and nutritional data showed that the BCC has serious impact on the deterioration and worsening pulmonary clinic. The phenotypic methods are useful for the presumptive BCC identification. The Vitek2®Compact showed good accuracy in BCC identification of, but with some errors due to limitations of the method/equipment. PCR, nested-PCR and recA sequencing showed good specificity in BCC genomovars identifying, however, there are still some limitations, stemming from different genotype, being necessary to use more comprehensive methods, such as the MSLT / Mestrado / Saude da Criança e do Adolescente / Mestre em Saude da Criança e do Adolescente

The considered objective was to evaluate the relationship between dental caries prevalence, socioeconomic variables and clinical (gingivitis and initial caries lesion) in 5-year-old preschool children in Piracicaba-SP, Brazil, in 2005. The used methodology was consisted of 728 preschool children attending 22 public (n=428) and 18 private (n=300) preschools. A calibrated examiner performed the clinical examination outdoors, under natural light, using dental mirror, CPI probe and air-drying. Dental caries and gingival status were measured using the dmft/dmfs and the alteration gingival indexes, respectively. Initial caries lesion (IL) was also recorded. The socioeconomic variables (type of school, monthly family income, number of people living in the household, parents¿ educational level and home ownership) were collected by means of a parental semi structured questionnaire. The work presented the following results: the mean values for dmft and dmfs indexes were 1.30 (SD=2.47) and 3.08 (SD=7.55) respectively; and children caries-free were 62.2%. The univariate analyses showed a significant association between dental caries and: monthly family income (p<0.0001), father¿s education (p<0.0001), mother¿s education (p=0.0012), type of school (p=0.0002), gingival status (p<0.0001), fluorosis (p<0.0001), dental crowding (p=0.0300), car own (p=0.0143). By means of multiple logistic regression analyses, the risk indicators of disease was gengivite (p=0.0002; OR=1.870); and monthly family income (p<0.0001; OR=0.494) and fluorosis (p=0.0004; OR=0.408) was protector indicators. The univariate analyses showed a significant association between dental caries + Li and monthly family income (p<0.0001), father¿s education (p=0.0002), mother¿s education (p=0.0053), type of school (p=0.0008), gingival status (p<0.0001), fluorosis (p<0.0001). By means of multiple logistic regression analyses, the risk indicators of disease was gengival status (p=0,0416; OR=1,806), and monthly family income (p<0.0001; OR=0,528) and fluorosis (p<0.0001; OR=0.393) was protector indicators. Mouth breathing (p= 0.0416; OR=1.378) and dental crowing(p=0.0255; OR=2.631) was the risk indicators of disease. It was possible to conclude that the prevalence of dental caries in 5-year-old preschool children in Piracicaba was low , but including IL its became moderate. There is a significant association between dental caries and IL and monthly family income. The risk indicators of disease was gengival status and fluorosis was protector indicator / Mestrado / Saude Coletiva / Mestre em Odontologia
