Caracterização microbiológica de cultura desnitrificante de reator anaeróbio horizontal de leito fixo utilizado na remoção de BTEX / Microbiologic characterization of anaerobic packed-bed reactor denitrifying culture used for BTEX removal

Hidrocarbonetos aromáticos conhecidos como BTEX (benzeno, tolueno, etilbenzeno e xilenos) estão entre os maiores problemas de poluição ambiental. Estes compostos são reconhecidos por sua recalcitrância em ambientes anóxicos, e a remoção destes hidrocarbonetos, de sítios anaeróbios contaminados é dependente da atividade de uma população de microrganismos adaptados capazes de promover a biodegradação destes compostos, nestas condições. Neste sentido, o presente trabalho buscou purificar, caracterizar e utilizar cultura desnitrificante, como inóculo para desenvolvimento de biofilme, em reator anaeróbio de leito fixo preenchido com espumas de poliuretano, além de avaliar o potencial deste biofilme em promover a biodegradação dos compostos de BTEX. Células da cultura apresentaram coloração Gram negativa, com morfologia de cocos (diplococos e cocobacilos). A cultura foi capaz de crescer, sob condições desnitrificantes, utilizando diferentes substratos. A cultura não apresentou capacidade de crescer sob condições sulfetogênicas. Velocidades de crescimento ('mü') de 0,046/h e 0,050/h e tempos de geração (Tg) de 15,1 horas e 13,9 horas foram obtidos para as células crescidas em benzeno e etilbenzeno, respectivamente. A cultura purificada foi utilizada para formar biofilme em RAHLF. Em todas as condições de alimentação as quais os reatores foram submetidos houve remoção da matéria orgânica, nitrato e dos hidrocarbonetos. A menor eficiência de remoção de hidrocarbonetos foi de 89,4%, obtida durante a alimentação dos reatores com benzeno. A matéria orgânica (DQO) foi removida com eficiência média de 87,1%. A eficiência média de remoção de nitrato foi de 94%. O sequenciamento das amostras revelou que o biofilme do RAHLF1 foi formado, principalmente, por espécies de Paracoccus, Pseudomonas e Bacteroides, enquanto que no RAHLF2, alimentado com etilbenzeno, observou-se espécies dos gêneros: Paracoccus, Pseudomonas, Xanthomonas e Variovorax / The monoaromatic hydrocarbons known as BTEX (benzene, toluene, ethylbenzene, and xylene) are among the greatest environmental pollution problems. In this study, a well biofilm-forming bacterial strain was isolated from oil contaminated water and sediment samples collected in petroleum storage Duc Giang, Hanoi. It was identified as Paracoccus sp. DG25 and registered in the GenBank database with the accession numbers KJ608354. Several biophysical and bio-chemical conditions for the biofilm formation of the strain were estimated such as pH, temperature, carbon sources and nitrogen sources. As the results the biofilm forming capacity was highest at pH 7, 37 oC, on maltose and supplemented with KNO3. Using these optimal conditions, the formed biofilm degraded 76.07 % of pyrene after 7 day-incubation, with the initial concentration of 300 ppm by high-performance liquid chromatography (HPLC) analysis. To our knowledge, there is rare publication on pyrene degradation by biofilm-forming bacteria. Therefore, the obtained results show that biofilm formed the strain Paracoccus sp. DG25 may considerably increase the degrading efficiency of pyrene and may lead to a new approach to treat polycyclic aromatic hydrocarbons containing in petroleum oil contaminated water in Vietnam. / Trong nghiên cứu này, từ các mẫu đất và nước nhiễm dầu lấy tại kho xăng Đức Giang, Hà Nội, chúng tôi đã phân lập được chủng vi khuẩn có khả năng tạo màng sinh học tốt. Chủng vi khuẩn này đã được phân loại và định tên là Paracoccus sp. DG25 với số đăng ký trên ngân hàng Gen là KJ608354. Chúng tôi cũng đã nghiên cứu một số điều kiện hóa lý ảnh hưởng tới khả năng hình thành màng sinh học như pH, nhiệt độ, nguồn Carbon và nguồn Nitơ. Kết quả cho thấy, chủng DG25 có khả năng tạo màng tốt nhất ở các điều kiện pH 7, 37 oC, nguồn Carbon là maltose và nguồn Nitơ là KNO3. Sử dụng các điều kiện tối ưu này để tạo màng và đánh giá khả năng phân hủy pyrene của màng tạo thành. Bằng phương pháp sắc ký lỏng cao áp, chúng tôi đã đánh giá được hàm lượng pyrene bị phân hủy sau 7 ngày nuôi tĩnh bởi màng sinh học của chủng DG25 lên tới 76,07 % với nồng độ ban đầu là 300 ppm. Cho tới nay, chưa có nhiều công bố về hiệu quả phân hủy pyrene của các chủng vi khuẩn tạo màng sinh học. Do vậy, kết quả đạt được này mở ra khả năng sử dụng màng tạo thành bởi chủng DG25 để nâng cao hiệu quả phân hủy pyren và có thể mở ra phương pháp mới nhằm xử lý các hợp chất hydrocarbon thơm có trong nước ô nhiễm dầu ở Việt Nam.

Biology of the oleander mealybug, Paracoccus burnerae (Brain) (Hemiptera: Pseudococcidae)

Thesis (MSc (Botany and Zoology))--University of Stellenbosch, 2010. / ENGLISH ABSTRACT:Chapter 1 - Mealybugs are tiny, soft-bodied insects which constitute the second largest scale insect family Pseudococcidae (Downie & Gullan 2004). The family comprises approximately 2000 species in 300 genera (Ben-Dov 1994), of which 20 species are pests of cultivated plants in South Africa (Annecke & Moran 1982). In South Africa, approximately 109 species of mealybugs have been recorded from 50 genera (Millar 2002). Chapter 2 - The effect of constant temperatures on the development, survival and fecundity of the oleander mealybug, Paracoccus burnerae on citrus was determined. Developmental time, rate of development, fecundity and survival were investigated at five constant temperatures and a 16L: 8D light: darkness regime. The rate of development increased linearly with an increase in temperature for the egg, 1st nymphal and pupal stages as well as the entire biological cycle (egg – adult), but was nonlinear for the 2nd and 3rd nymphal stages. Survival decreased with an increase in temperature. P. burnerae required 666.7 degree-days above a lower threshold of 8.7°C to complete one generation. The highest mean number of 68 eggs per female was reached at 22°C. A sex ratio of 0.52:0.48 (male:female) was obtained from the life table. The net reproductive rate (Ro) was >1 at all five temperatures, an indication that it is capable of increasing its population numbers despite the high mortality experienced in the 1st and 2nd nymphal stages. Chapter 3 - The oleander mealybug, Paracoccus burnerae (Brain) is a pest of citrus in South Africa. This study was carried out to determine the effect of temperature on development rate of P. burnerae and to investigate whether development rate is the reason why P. burnerae is out competing the citrus mealybug, Planococcus citri (Risso), in the Eastern and Western Cape Provinces of South Africa. The influence of temperature on life history traits of P. burnerae was determined at 20, 22, 25 and 27°C and compared with corresponding data for P. citri. The rate of development increased linearly with an increase in rearing temperature in the embryonic, first nymphal and pupal stages but reached a climax at 26.13 and 28.6°C in the second nymphal stage of both species, respectively. P. citri exhibited lower developmental thresholds except in first instar, shorter degree-days and higher developmental rates than P. burnerae. Results of the current study indicated that the dominance of oleander mealybug over the citrus mealybug is neither linked to developmental rates nor sum of effective temperatures. Chapter 4 - The importance of Paracoccus burnerae has risen over the years to an extent where it is now regarded as a quarantine pest for citrus fruit from South Africa. The field biology of P. burnerae on citrus in the Western Cape Province of South Africa was studied through periodic sampling of leaves from twigs enclosed in sleeve cages. The species composition and abundance of natural enemies was investigated. Both adult and immature stages attained maximum population peaks in March and P. burnerae had four generations. The highest level of mortality was experienced in the immature stages. Climate and an unidentified fungus were the key mortality factors. The level of abundance of the two observed predators, the harlequin beetle, Harmonia axyridis and the green lacewing, Chrysoperla sp. was relatively low. Although parasitism occurred in some cages, the level was low ranging between 1.62 to 9.43%. If biocontrol is the preferred method of controlling P. burnerae, suitable candidate parasitoids for inoculative biocontrol are Acerophagus sp., Leptomastix sp. and Microterys nietneri. The oleander mealybug does not share the same parasitoids with Planococcus citri, Pseudococcus calceolariae and Pseudococcus longispinus except the parasitoid Coccophagus sp. The most popular species of parasitoids used in the biolological control of mealybugs, Anagyrus sp. and Coccixenoides sp. were insignificant in the case of P. burnerae. Chapetr 5 - Biological control programs of mealybug species have relied on sprouting potatoes, pumpkins and butternut for rearing of both mealybugs and their natural enemies. In this study, the suitability of sprouting potatoes, butternuts and citrus as mass rearing substrates for the oleander mealybug, Paracoccus burnerae was investigated. Developmental times, rate and fecundity on each substrate were determined and compared at three different temperatures. The developmental time on sprouting potatoes was shorter than on citrus. P. burnerae was unable to complete its life cycle on butternut. The rate of development increased linearly with an increase in temperature on both sprouting potatoes and citrus. P. burnerae required 666.7 degree-days on citrus and 434.8 degree-days on sprouting potatoes above lower developmental thresholds of 7.6°C and 10.4°C respectively to complete one generation. The mean number of eggs per female was higher on sprouting potatoes (121.3) than on citrus (68), but declined with an increase in temperature from 22 to 27°C. Despite the shorter shelf life, sprouting potatoes are the preferred host for mass rearing of the oleander mealybug. Chapter 6 - general conclusions Chapter 7 - Researchers often present impressive results of their studies on the biology of the Coccoidea without mentioning the problems they came across and had to solve. Occurrence, Localization, and Possible Significance of an Ornithine-Containing Lipid in Paracoccus denitrificans

A ninhydrin-positive, phosphorus-negative lipid from Paracoccus denitrificans ATCC 13543 has been isolated and purified by mild alkaline methanolysis followed by silicic acid column chromatography and preparative thin-layer chromatography. The lipid was identified as an ornithine-containing lipid. The major ester-linked fatty acid was cis vaccenic acid. Major amide-linked fatty acids were 3-OH-20:1 and 3-OH-18:0. Ornithine-containing lipid was a major lipid component of P. denitrificans. Phospholipids made up about 57% and ornithine-containing lipid about 14% of the weight of the total lipid of the organism. The ratios of lipid ornithine: lipid phosphorus were 0.23, 0.65 and 0.58 in cytoplasmic membrane, outer membrane, and an NaCl extract, which is thought to represent chiefly outer membrane, respectively. Thus ornithine-containing lipid appears to be present in larger amounts in outer membrane than cytoplasmic membrane. No substantial variations in lipid ornithine levels were noted in stationary phase versus exposnential phase organisms, organisms grown in complex medium versus organisms grown in minimal medium with and without amino acid supplements, or in organisms grown in low phosphate-containing medium.

Étude et valorisation de composés naturels ou d’analogues de synthèse contrôlant l’adhésion de salissures marines / Study of natural or derivatives compounds which have an impact on biofouling

Le développement de salissures marines (ou biofouling) est à l’origine de nombreux problèmes économiques et écologiques. Ces salissures marines sont constituées de microorganismes (bactéries, microalgues…) formant le microfouling sur lequel va se développer le macrofouling constitué de macroorganismes tels que les algues, coquillages et éponges. La formation de ces salissures va induire un ralentissement des navires provoquant une surconsommation de carburant. De plus, l’utilisation de revêtements antisalissures ou peintures antifouling à base de métaux lourds et de biocides dans le passé a conduit à des problèmes environnementaux. L’objectif de la thèse est de rechercher une alternative aux composés actuellement utilisés (cuivre) en respectant le milieu marin. L’environnement est une source d’inspiration, une approche biomimétique pourrait être une stratégie de lutte efficace contre le biofouling. Dans une première partie, un criblage d’une centaine de composés est réalisé contre des souches bactériennes. Huit composés issus des deux familles : les batatasins et les hemibastadins sont étudiées en vue de comprendre leurs modes d’action. Parmi les composés sélectionnés, la famille des hemibastadins comprenant le DiBromoHemiBastadin-1 (DBHB) a montré des propriétés antifouling prometteuses. En effet, cette molécule est capable d’inhiber 50 % la formation du biofilm avec une IC50= 6,44 µg/mL pour la bactérie Pseudomonas aeruginosa PAO1 et une IC50 = 12,8 µg/mL pour la bactérie marine Paracoccus sp. 4M6. Afin de comprendre le mode d’action de cette molécule, son impact sur la communication bactérienne, le quorum sensing est étudié et le composé DBHB est capable de l’inhiber. Dans une seconde partie, un autre groupe d’organismes participant au microfouling est étudié : les microalgues. Afin d’évaluer l’impact de composés de la famille des hemibastadins et notamment du DBHB, l’adhésion et la formation de biofilm de microalgues sont étudiées. Le DBHB montre des inhibitions de l’adhésion et de la formation du biofilm uniquement envers la diatomée Cylindrotheca closterium. Une dernière partie, s’est intéressée à l’évaluation de revêtements contenant six composés de la famille des batatasins et des hemibastadins sur le microfouling naturel. Une méthode d’immersion de revêtements en conditions contrôlées (photobioréacteur) est mise au point afin de pallier aux contraintes environnementales. L’ensemble de cette étude a permis de mettre en évidence les propriétés antifouling du DBHB et de caractériser son mode d’action. Ce composé offre d’intéressantes voies d’étude dans la lutte contre le biofouling. De plus, l’approfondissement des connaissances sur les procédés d’adhésion et de formation de biofilm de microalgues permet de définir de nouvelles stratégies de lutte. / Biofouling induces important economic and ecological problems. This phenomenon includes microorganisms (bacteria, microalgae…) giving the microfouling which allows the macrofouling development with algae, invertebrates and sponges. These organisms colonize every immersed surface as boat hull. The colonization induces reduced speed of ships and fuel overconsumption. In the past, the utilization of AF coatings with heavy metals or pesticides caused environmental problems. The purpose of the study is to find an alternative to AF compounds (copper) respecting the marine medium. The marine environment is an inspiration; a biomimetic approach could be an interesting strategy to inhibit biofouling. In a first part, a screening of one hundred compounds is realized against marine and terrestrial bacteria. Eight molecules from two families (batatasins and hemibastadins) are studied to understand the way of action. Among selected compounds, Dibromohemibastadin-1 (DBHB) from hemibastadin family shows promising AF activities. This molecule is able to inhibit the biofilm formation with an IC50 of 6,44 µg/mL against the bacterium Pseudomonas aeruginosa PAO1 and 12,8 µg/mL for the marine bacterium Paracoccus sp. 4M6. To identify the way of action of DBHB, the impact on the bacterial communication named quorum sensing is investigated. The molecule shows an anti-quorum sensing property. In a second part, another group participating at microfouling is studied: microalgae. In order to evaluate the impact of hemibastadin family in particular DBHB, microalgae adhesion and biofilm formation are characterized. DBHB induces inhibition only on the adhesion and the biofilm formation of the diatom Cylindrotheca closterium. The last part presents the formulation of coatings containing six compounds from the batatasin and hemibastadin families. These coatings have been immersed in a harbor to evaluate the impact on natural microfouling. Furthermore, a new method for the evaluation of AF coatings is developed in controlled conditions, in a photobioreactor. This method allows the evaluation of coatings on the formation of a mixed biofilm (bacteria and microalgae). This method has been established to avoid environmental constraints by immersion in natural condition. This study allows the characterization of the AF property of DBHB. This compound provides promising research path to limit biofouling. Moreover, the development of a test allowing adhesion and microalgae biofilm formation in dynamic condition improves the characterization of compounds activities.

Anwendung kalorimetrischer Methoden zur Untersuchung des Alkanabbaus durch den Mikroorganismus Rhodococcus opacus 1CP

Ziel der vorliegenden Arbeit war die Charakterisierung des Wachstumsverhaltens von Mikroorganismen auf Alkanen am Beispiel des Bodenbakteriums Rhodococcus opacus 1CP. Unter Einsatz kalorimetrischer Methoden konnte das Wachstumsverhalten von 1CP auf Glucose und Alkanen mit sehr guter Reproduzierbarkeit aufgezeichnet werden. In Abhängigkeit der Substrateigenschaften wurden Änderungen der Wachstumskinetik registriert, welche auf die Aggregation der Zellkultur zurückgeführt werden konnten. Die kalorimetrischen Befunde zum Aggregationsverhalten wurden mittels Gaschromatographie und Partikelgrößenbestimmung bestätigt. Durch Biotensidquantifizierung in kalorimetrisch aufgezeichneten Kultivierungen von 1CP auf Alkanen konnte eine wachstumsassoziierte Biotensidbildung nachgewiesen werden. Elementaranalytische Untersuchungen ergaben eine Variation der Biomassezusammensetzung von 1CP in Abhängigkeit der C-Quelle. Unter Berücksichtigung von Biomasse- und Biotensidbildung wurden Summenreaktionen für die Wachstumsprozesse formuliert und durch Verifikation mit kalorimetrisch ermittelten Enthalpien des Substratumsatzes bestätigt.

In Abhängigkeit der Substrateigenschaften wurden Änderungen der Wachstumskinetik registriert, welche auf die Aggregation der Zellkultur zurückgeführt werden konnten. Die kalorimetrischen Befunde zum Aggregationsverhalten wurden mittels Gaschromatographie und Partikelgrößenbestimmung bestätigt. Durch Biotensidquantifizierung in kalorimetrisch aufgezeichneten Kultivierungen von 1CP auf Alkanen konnte eine wachstumsassoziierte Biotensidbildung nachgewiesen werden. Elementaranalytische Untersuchungen ergaben eine Variation der Biomassezusammensetzung von 1CP in Abhängigkeit der C-Quelle. Unter Berücksichtigung von Biomasse- und Biotensidbildung wurden Summenreaktionen für die Wachstumsprozesse formuliert und durch Verifikation mit kalorimetrisch ermittelten Enthalpien des Substratumsatzes bestätigt.
