<b>Effects of Natural Variation on Pollen Tube Sensitivity to Synergid Signals</b>Iyanu Adedeji (18410463), Sharon Kessler (18413778) 20 April 2024 (has links)
<p dir="ltr">Communication between the male gametophyte (the tip-growing pollen tube) and the female gametophyte (the synergid cells) is crucial for sexual reproduction in flowering plants. The reception of the pollen tube (PT) depends on its recognition and sensitivity to the signals from the synergid cells for its rupture, sperm release, and double fertilization. Mutations in genes regulating communication between the synergid cells and pollen tube lead to PT overgrowth. Despite significant advances in understanding the molecular mechanism of pollen tube reception in Arabidopsis, there remains a need for more comprehensive information on the impact of natural variations on physiological traits related to pollen tube and synergid signals in pollen tube reception. This research investigates the effects of natural variation on pollen tube sensitivity to synergid signals mediated by NORTIA. In <i>nortia</i> homozygous mutants, PT-synergid communication is disrupted due to lower levels of calcium signals in the synergid cells, resulting in PT overgrowth. Using the Aniline blue staining procedure, this study identified twelve ecotypes of Arabidopsis thaliana out of the twenty-four ecotypes that could suppress the PT overgrowth phenotype of <i>nta-1</i>. I observed that the suppressor ecotypes exhibit characteristics like the Columbia (Col-0) ecotype, while non-suppressor ecotypes resemble the Ws-2 ecotype. Comparing the impact of the Columbia (Col-0) and Wassilewskija (Ws-2) ecotypes on PT overgrowth in the <i>nta-2</i> mutant revealed that Columbia (Col-0) effectively suppressed PT overgrowth compared to Wassilewskija (Ws-2). We investigated the sensitivity of PT integrity mutants to synergid signals. Our results showed that compromised PT integrity mutant genes (<i>mlo1mlo15</i>) partially suppressed PT overgrowth in <i>nta-1</i>. I propose that suppressor ecotypes and mutants may exhibit a heightened sensitivity to synergid signals, that help them to regulate their response to synergid signals finely.</p>

Performance evaluation of low-cost GPS machine guidance systems for livestock production forage and pasture management applicationsWright, Conner Childress 10 May 2024 (has links) (PDF)
Global Positioning System (GPS) machine guidance aids in the precise navigation and piloting of farm machinery and serves to boost productivity and optimize farm operations. Characterization of low-cost GPS guidance systems for livestock production operations is necessary for return on investment (ROI) decision making. The objective of this research is to characterize the accuracy of four commercially available low-cost ($985 - $4,200) wide area augmentation system (WAAS) corrected GPS guidance systems. Testing procedures were designed in accordance with the ASABE/ISO standard 12188-2 for testing of satellite-based guidance systems during straight line travel. The vehicles lateral deviation of a planned path, characterized as cross-track error (XTE), was evaluated for short-term and long-term guidance for each system at speeds of 4.8, 9.6, and 14.5 kph (3, 6, and 9 mph).

Water use efficiency of six dryland pastures in CanterburyTonmukayakul, Nop January 2009 (has links)
The annual and seasonal water use efficiency of six pasture combinations were calculated from the ‘MaxClover’ Grazing Experiment at Lincoln University. Pastures have been established for six years and are grazed by best management practices for each combination. Measurements for this study are from individual plots of four replicates of ryegrass (RG)/white clover (Wc), cocksfoot (CF)/Wc; CF/balansa (Bal) clover; CF/Caucasian (Cc) clover; CF/subterranean (Sub) clover or lucerne. Water extraction measurements showed soils for all dryland pastures had a similar plant available water content of 280±19.8 mm. Dry matter measurements of yield, botanical composition and herbage quality were assessed from 1 July 2008 until 30 June 2009. Lucerne had the highest annual yield of 14260 kg DM/ha/y followed by the CF/Sub at 9390 kg DM/ha/y and the other grass based pastures at ≤ 6900 kg DM/ha/y. All pastures used about 670±24.4 mm/y of water for growth. Lucerne had the highest annual water use efficiency (WUE) of 21 kg DM/ha/mm/y of water used (total yield/total WU). The WUE of CF/Sub was the second highest at 15 kg DM/ha/mm/y, and the lowest was CF/Wc at 9 kg DM/ha/mm/y. The CF/Sub pastures had the highest total legume content of all grass based pastures at 21% and as a consequence had the highest annual nitrogen yield of 190 kg N/ha. This was lower than the monoculture of lucerne (470 kg N/ha). Ryegrass/white clover had the highest total weed component in all pastures of 61%. For dryland farmers spring is vital for animal production when soil temperatures are rising and moisture levels are high. The water use efficiency at this time is important to maximize pasture production. In spring lucerne produced 8730 kg DM/ha, which was the highest dry matter yield of all pastures. The CF/Sub produced the second highest yield of 6100 kg/DM/ha. When calculated against thermal time, CF/Sub grew 5.9 kg DM/ºCd compared with lucerne at 4.9 kg DM/ºCd. The higher DM yield from lucerne was from an extra 400 ºCd of growth. The highest seasonal WUE of all pastures occurred in the spring growing period. Linear regressions forced through the origin, showed lucerne (1/7/08-4/12/08) had a WUE of 30 kg DM/ha/mm (R2=0.98). Of the grass based pastures, CF/Sub produced 18 kg DM/ha/mm (R2=0.98) from 1/7 to 10/11/08 from 270 mm of water used. The lowest spring WUE was 13.5 kg DM/ha/mm by CF/Bal pastures which was comparable to the 14.3±1.42 kg DM/ha/mm WUE of CF/Wc, CF/Cc and RG/Wc pastures. During the spring, CF/Sub clover had the highest spring legume component of the grass based pastures at 42% and produced 120 kg N/ha. This was lower than the 288 kg N/ha from the monoculture of lucerne. Sub clover was the most successful clover which persisted with the cocksfoot. Based on the results from this study dryland farmers should be encouraged to maximize the potential of lucerne on farm, use cocksfoot as the main grass species for persistence, rather than perennial ryegrass, and use subterranean clover as the main legume species in cocksfoot based pastures. By increasing the proportion of legume grown the water use efficiency of a pasture can be improved. When pastures are nitrogen deficient the use of inorganic nitrogen may also improve pasture yields particularly in spring.

The effect of concentrate feeding strategies on rumen parameters, milk production and milk composition of Jersey cows grazing ryegrass (Lolium multiflorum) or kikuyu (Pennisetum clandestinum) pastureJoubert, Matthys Du Toit 03 1900 (has links)
Thesis (MScAgric)--Stellenbosch University, 2012 / ENGLISH ABSTRACT: Ruminal pH is the rumen condition that varies most. Studying the cause and effect of diurnal variation offers a means of improving ruminal conditions for rumen micro-organisms with subsequent improvements in fibre degradation, milk yields and milk composition. A research project was conducted to test the following hypothesis: feeding 6 kg (as is) concentrate per cow/day in different weight ratios, divided in a morning and an afternoon allocation, will result in an improved ruminal pH profile and a concurrent increase in fibre fermentation, milk yield and improved milk composition. The project was conducted at the Outeniqua Research Farm in the Southern Cape of South Africa. A randomized block design was used and two phases, viz. ryegrass phase and a kikuyu phase, were applied. Each phase was conducted during the growing season of the grass specie used as pasture. A rumen study and a production study were done in each phase. The rumen study used six cannulated multiparous cows per treatment, testing rumen pH, volatile fatty acid (VFA) concentration, in sacco degradation of neutral detergent fibre (NDF) and dry matter (DM). The production study used 42 multiparous cows, blocked according to days in milk (DIM), parity and milk yield, and cows were randomly divided into three treatment groups. The study tested milk yield, milk composition and changes in body weight (BW) and body condition score (BCS). Treatments consisted of a maize based concentrate supplement (6 kg per cow/day, as is) being fed to cows on pasture in different weight ratios between morning and afternoon allocation. Treatments used in the production study were:
Concentrate fed: 5 kg during morning; 1 kg during afternoon
• Concentrate fed: 4 kg during morning; 2 kg during afternoon
• Concentrate fed: 3 kg during morning; 3 kg during afternoon
The first and the last treatments mentioned above were used in the rumen studies. Results of the rumen study of the ryegrass phase showed no differences in ruminal pH between treatment means. The time (h) spent below the critical ruminal pH, i.e. 5.8, was of short duration (2.92 to 3.42 hours). The concentration of certain VFA’s differed at times, but the differences were not reflected in graphs and data of the ruminal pH. No differences were observed in in sacco NDF and DM degradation between treatment means. Similar results were mostly obtained in the kikuyu phase. No differences were found in either daily maximum, minimum or mean ruminal pH between treatment means. Though time (h) spent below the critical ruminal pH was of a long duration (7.1 hours) it did not differ between treatments. No differences were observed between treatment means for in sacco NDF and DM degradation.
Results of the production study of the ryegrass phase showed no differences in milk yield or milk composition between treatment means. The same was observed in the kikuyu phase except that milk fat was higher in the treatment group that received the 3:3 kg ratio treatment. The fact that treatments did not differ in terms of milk yield and milk composition in the production studies can be attributed to no differences between treatments in terms of rumen parameters observed in the rumen studies.
Thus, feeding a higher proportion of concentrate in the morning relative to the afternoon for a total of 6 kg per cow/day does not affect ruminal pH, fibre digestion or milk production. Based on the results of both phases it can be concluded that the null hypothesis should be rejected.

Substitution of maize with high fibre by-products in concentrates supplemented to dairy cows grazing kikuyu/ryegrass pasture during springLingnau, Werner August Leonhard 03 1900 (has links)
Thesis (MScAgric (Animal Sciences))--University of Stellenbosch, 2011. / ENGLISH ABSTRACT: Kikuyu over-sown with ryegrass forms the basis of pasture based systems in the Southern Cape. During
early spring, energy is the first limiting nutrient in kikuyu/ryegrass pasture, supplementation is thus
essential. Supplementation consists mainly of high starch concentrates (high maize inclusion), which is
expensive and could negatively affect rumen parameters. The objective of this study was to determine if
milk production could be improved or maintained, and if the rumen environment would be improved, by
replacing high starch concentrates with low starch (high by-product) concentrates for dairy cows on
kikuyu/ryegrass pasture.
Forty-five multiparous, high producing, lactating Jersey cows [body weight, 340 ± 34.7 kg; milk yield (MY),
19.6 ± 2.23kg/d; days in milk (DIM), 153 ± 33.5; lactation number, 3.6 ± 1.85; (mean ± SD)] were used in
the production study. A randomised block design was used. The forty-five cows were allocated to fifteen
groups of three each (blocking) on the basis of MY, DIM, and lactation number. Cows from each group
were randomly allocated to one of three treatment groups (high starch, medium starch and low starch
concentrate supplementation). Cows were fed 6kg (3kg during each milking) concentrate per day and
were allocated fresh pasture ad lib after each milking. There were no significant differences (P > 0.05)
found in milk yield and fat corrected milk yield between treatment groups. Milk fat percentage was
significantly higher (P < 0.05) in the low starch treatment than in the high starch treatment. Milk fat yield
was significantly higher (P < 0.05) in both the low starch and the medium starch treatment when
compared to the high starch treatment. Milk protein and lactose percentages, as well as milk urea
nitrogen and somatic cell count, did not differ significantly (P > 0.05) between treatments. Live weight
change, as well as body condition score change, was unaffected (P > 0.05) by treatments indicating body
reserves were not used to maintain milk production in the low starch treatment. A rumen metabolism study was also done with ten lactating, cannulated Jersey cows [body weight, 332 ±
56.3 kg; MY, 17.3 ± 1.73kg/d (mean ± SD)] were used. The cows were divided into two groups of five
each, on the basis of lactation number, DIM, and MY. The five cows from each group were randomly
allocated to one of two treatment groups (high starch and low starch concentrate supplementation) and
used in a cross-over design. Cows were fed 6kg concentrate per day and were allocated fresh pasture ad
lib after each milking. The volatile fatty acid (VFA) concentration was significantly higher (P < 0.05) in the
high starch treatment when compared to the low starch treatment. The individual VFA’s, acetic-,
propionic- and butyric acid concentrations were also significantly higher (P < 0.05) in the high starch
treatments when compared to the low starch treatment. The acetic to propionic acid ratio was unaffected
(P > 0.05) by treatment. Rumen ammonia-nitrogen concentration was significantly higher (P < 0.05) in the
high starch treatment. Rumen pH was unaffected (P > 0.05) by supplementation type. The in sacco dry
matter and neutral detergent fibre digestibilities of the kikuyu/ryegrass pasture were unaffected (P > 0.05)
by treatment type.
Results indicated that milk production could be maintained with low starch concentrates which also
improved milk composition. Results further suggested that the rumen environment was relatively
unaffected by low starch concentrate supplementation.

behandelings (hoëstysel-, mediumstysel- of laestysel-kragvoeraanvulling) geallokeer. Koeie is daagliks

[liggaamsmassa, 332 ± 56.3 kg; melkproduksie, 17.3 ± 1.73kg/d (gem ± standard afwyking)].

hoëstyselbehandeling as in die laestyselbehandeling. Die asynsuur- tot propionsuurverhoudings het geen
verskille (P > 0.05) tussen behandelings getoon nie. Rumen-ammoniakstikstof was beduidend hoër (P <

Herbage production and transformations of the fertilizer nitrogen in irrigated tanzania grass.Martha Júnior, Geraldo Bueno 28 July 2003 (has links)
Whilst grazing management is a key component in pastoral systems little effort has been made to determine the adequate grazing intensity for tropical pastures, especially under irrigated conditions. The economical attractiveness of irrigated pasture systems depends on high pasture productivity, meaning that fertilizer nitrogen (N) must be used. However, to make effective changes toward sustainable N management, an understanding of the balance between N inputs and outputs and the efficiency of N cycling and transformation within the grassland system is required. In this context, seven experiments were carried out to evaluate the effect of post-grazing residue or nitrogenous fertilizer levels on herbage production and on fertilizer-N recovery in irrigated Panicum maximum cv. Tanzania pasture. The post-grazing residue for irrigated, N-fertilized Tanzania grass pasture during the winter, considering a 36-day grazing cycle, should be around 1900 to 2100 kg/ha of green dry matter. During the spring/summer seasons the residual stubble mass should be around 1700 to 2700 kg/ha of green dry matter. This management ensured that leaf production and leaf/stem ratio were close to the maximum. A plot-size of 1 m 2 , with a tussock of the grass in its geometric center, was adequate to study 15 N-fertilizer recovery irrespective of grazing intensity and season of the year. Increasing the grazing intensity resulted in decreased tussock mass. The smaller the tussock mass the greater was the reliance on fertilizer-N. The combination of high soil water content, lack of rainfall in the day following fertilization and high temperature determined low urea- 15 N recovery in the soil-plant system (< 35% of applied N) and high ammonia volatilization losses (> 40% of applied N) at fertilizations levels higher than 80 kg N/ha during the summer. The absorption of ammonia volatilized from urea applied to soil varied from 2.5% (120 kg N/ha) to 16.4% (40 kg N/ha) of the volatilized nitrogen. With fertilizations lower than 60 kg N/ha it was observed sub-optimal N nutrition and a trend of higher 13 C discrimination during the summer.

Potential productivity modelling of Brachiaria brizantha (cultivars Marandu and Xaraés).Detomini, Euro Roberto 20 May 2004 (has links)
With the main goal to purpose a model to estimate the total plant biomass (dry matter) potential productivity (forage removed under cutting management intensive system and optimal conditions) of Brachiaria brizantha, cultivars Marandu and Xaraés, as a function of global solar radiation and air temperature, besides the following goals: (i) acquiring a response curve that might show the shoot and root dried biomass potential productivity regarding to the relative development; (ii) characterizing the dry-matter productivity of leaves, stems and senescing structures of shoots; (iii) identifying the optimal time to pasture cutting; and (iv) characterizing the leaf area index temporal variation; field experiments were carried in Crop Production Department experimental area of Universidade de São Paulo (Piracicaba, SP, Brazil) from November 22nd of 2003 to January 26th of 2004. According to results acquired, it is possible to conclude: (i) the purposed model greatly simulates the total plant biomass (dry matter) potential productivity of Brachiaria brizantha, cultivars Marandu and Xaraés, as a function of year season (from cutting date), climatic variables (daily mean values of global solar radiation and air temperature) and local latitude; (ii) from modeling and field experiments taken together; it is possible to obtain a response-curve reporting the biomass potential productivity of shoot and root compartments as a function of relative development from observed shoot and root dry-matter values, leaf area index (LAI), air temperature and global solar radiation; (iii) the empirical equations greatly denote the productivity temporal variation of leaves, stems and senescing structures dry matter; (iv) the optimal time to both forage utilization (cutting) is around 64% of relative development due to after this moment the shoot dry-matter productivity is strongly affected by increases in stems and senescing structures productivities, otherwise leaf productivity rates decrease as much as relative development raises; and (v) the leaf area index temporal variation of both genotypes might be characterized as a exponential-growth response.

Grazing management techniques are improving the quality of the forage consumed by grazing dairy cows. The objective of this study was to determine if forage crude protein (CP) would be sufficient to achieve the nutritional requirements of midlactating dairy cows, making unnecessary the use of protein supplements on the concentrate. It also aimed to characterize the forage CP fractions and to balance the diet protein content to optimize nitrogen use efficiency. In the first trial, 33 Holstein and crossbred (Holstein X Jersey) mid-lactating dairy cows, producing 20 L d-1, were grouped in 11 blocks, according to breed, days in milk and milk yield and were fed three levels of CP on the concentrate supplement for 76 days. Concentrates contained fine ground corn, minerals, vitamins and three CP levels (T1:8.7, T2:13.4 and T3:18.1% DM) that were achieved by replacing corn with soybean meal. Pasture was fertilized with 50 kg N ha-1 after each grazing cycle and averaged 18.5% CP. There was no difference (P>0.05) in milk yield, fat corrected milk, fat, protein and casein content and yield, microbial synthesis and dry matter (DM) intake among treatments. Milk urea nitrogen increased linearly (P<0.01) as the concentrate CP content increased. The N urine excretion was greater (P<0.05) with the soybean meal concentrates (T2 and T3) than with the corn concentrate (T1). In the second trial, 4 rumen cannulated Holstein dry cows were used in a 4x4 Latin square and assigned to the same treatments used in the first study plus a fourth concentrate (T4) with 13,4% CP, containing urea instead of soybean meal as the N source. Dry matter intake was greater for T4 (P<0.05), suggesting a PDR deficiency in the other treatments. However, this hypothesis was not confirmed by ruminal parameters (pH and short chain fatty acids concentration) neither by microbial synthesis, that were not affected by the levels or sources of CP on the concentrate. Ruminal ammonia N content numerically increased as the concentrate CP content increased, but no differences were detected by the statistic analysis. The third study used the same animals and treatments of the second study to evaluate the ruminal kinetics of DM, CP and NDF of Elephant grass by an in situ assay. Protein concentrates (T2, T3 and T4) didnt improve forage ruminal degradation in relation to the energy concentrate (T1), confirming that the forage protein content was sufficient to optimize ruminal fermentation. The inclusion of soybean meal in the concentrate of midlactating dairy cows grazing intensively managed tropical grasses didnt result in productive advantage and increased the N excretion to the environment.

Mombaça, em regiões do Estado de São Paulo / Mathematical models to estimate seasonal pastures production of Panicum maximum cv. MombaçaAraujo, Leandro Coelho de 13 February 2012 (has links)
Two experiments were held simultaneously at Embrapa South-East Cattle Research Center (São Carlos, SP, Brazil, 21°57\'42\"S , 47°50\'28\"W) in 2010-2011 using Panicum maximum cv. Mombaça (Guineagrass). They were differentiated by the presence or absence of irrigation. The randomized complete block design with four replications was used. The treatments were determined by cumulative thermal times during each growth cycles (i.e. 250, 500, 750 and 1000 °C, base temperature = 0 °C). After the last sample time (1000° C) of each cycle, all the plots were uniformly cut up to 0.3 m above the soil surface to begin a new cycle of re-growth and sampling. At every new growth cycle the pasture was fertilized (N-P-K). The variables recorded were the dry matter above-residue, identification of apical meristem at vegetative state or flowering one, calculation of water index (WI) by ratio between real evapotranspitarion to crop evapotranspiration using the direct method (Diviner) or actual evapotranspitarion to potential evapotranspitarion usgin an indirect method (Climatologic). The data sets from the experiments were used to determine the critical values of the main variables that may influence the Guineagrass growth, besides parameterizing and evaluating an agroclimatic degree-day model (DDi); a photo-thermal-units model (PUi); a growth climate index model (GCIi) and one biophysical function model (APSIM-Growth). The results showed that the relative production of Guineagrass decreases linearly with the reduction of the WI. The base temperature was estimated equal to 15.6 ° C and Guineagrass is possibly identified as a short-day plant with a photoperiod critical value close to 11.81 h. All models performed well during the parameterization (R2 from 0.78 to 0.86 and coefficient of variation from 27 to 32.1%), and evaluation (R2 from 0.69 to 0.78; agreement index from 0.88 to 0.93; coefficient of variation from 37.6 to 50.2% and mean bias error from 6 to 470 kg DM ha/cutting). APSIM model was the one that showed a minor error and UF model was the empirical model that provided predicted values closer to the observed.

Relação entre características funcionais e o desempenho de espécies arbóreas nativas em um plantio de restauração / Relationship between functional traits and performance of native tree species in restoration plantingSano, Bruno 09 August 2016 (has links)
In degraded environments, ecological restoration aims to accelerate the process of ecological succession. The most widely used technique for restoration of degraded environments is the planting of seedlings. The selection of species is crucial to the success of the restoration projects, because the species need to establish and grow under the harsh conditions of the environment. Using the approach of functional traits can be a promising tool to assist in the selection of species in restoration projects. In forest environments, there is evidence that functional traits affect the performance of the species, however, there are few studies conducted in areas undergoing restoration. In this study, the relationship between the functional leaf traits and the relative growth rate in height and in diameter at ground level of native tree species planted in a degraded area that is being restored was evaluated. To describe the relationship between the functional traits and the relative growth rate (RGR) of individuals, we used the approach of model selection based on linear mixed effects models (LMEM). The selection of models was performed using Akaike\'s Information Criterion (AIC). The functional traits showed no relationship with the relative growth rate in height and diameter of the individuals of the species, possibly due to differences in environmental conditions between forest environments and degraded environments. Despite not finding a relationship between the functional traits and the RGR, the most plausible model selected indicates that larger individuals at the beginning of monitoring have lower relative growth rates in height, that is, taller individuals have lower rates of growth in height. Considering that no relationship between the morphological functional traits and the relative growth rate in an area undergoing restoration was found, it is reasonable to assume that this approach cannot be applied in all environments. Studies that assess functional traits of the leaf in the different stages of plant development in degraded environments can refine these analyses and help define whether the selection of species for restoration projects can be accomplished using the approach of functional characteristics
