Estudo fitoquímico biomonitorado da espécie Poiretia bahiana (Leguminosae)

No. of bitstreams: 1 Dissertação Floricéia Araújo.pdf: 1292484 bytes, checksum: c676611610eaf0fd75433352b2e0f9dd (MD5) / CNPq / O presente trabalho descreve o estudo fitoquímico das folhas e galhos de espécimens de Poiretia bahiana (Leguminosae) coletados no Morro do Pai Inácio, na Chapada Diamantina, Bahia, em julho de 1997 e em julho de 1999. A espécie Poiretia bahiana é endêmica da Chapada Diamantina e até o presente trabalho, nenhum estudo fitoquímico foi relatado para o gênero. As folhas do espécimen coletado em 1997 foram submetidas à extrações com hexano e a seguir com metanol. Do extrato hexânico foram isoladas seis isoflavonas, duas rotenonas, uma mistura contendo os triterpenos lupeol e β- amirina e uma mistura contendo os esteróides estigmasterol e sitosterol. Do extrato metanólico foram isoladas a 2’,4’,5’-trimetoxi-7-O-β-D-glicopiranosil isoflavona inédita, o aminoácido prolina e o ciclitol 3-O-metilchiroinositol. Das partes aéreas do espécime coletado em 1999 foi obtido o óleo essencial através de hidrodestilação, onde foram identificados onze constituintes, sendo nove monoterpenos e dois sesquiterpenos. O constituinte majoritário do óleo essencial foi identificado como o monoterpeno tujenona, que foi posteriormente isolado através de coluna cromatográfica em sílica. Os fracionamentos dos extratos e os isolamentos das substâncias foram realizados através de técnicas cromatográficas. As identificações das substâncias foram realizadas através de métodos espectrométricos, tais como RMN de 1H e 13C (incluindo DEPT, HOMOCOSY e HETCOR), espectrometrias de massas e de absorção na região do infravermelho. Os extratos obtidos foram submetidos a bioensaios para verificação de suas atividades fungitóxicas, utilizando esporos do fungo fitopatogênico Cladosporium sphaerospermum, sendo que todos mostraram-se ativos. As substâncias 7,3’,4’-trimetoxiisoflavona, 7,2’,4’,5’-tetrametoxiisoflavona, rotenona e 12a-hidroxirotenona, isoladas do extrato hexânico, foram as responsáveis pela atividade fungitóxica deste extrato. / The present work describes the phytochemical study of leaves and stems of Poiretia bahiana (Leguminosae) specimens collected on Morro do Pai Inácio, in Chapada Diamantina, Bahia, on July 1997 and July 1999. Poiretia bahiana is a endemic species from Chapada Diamantina, and to the best of our knowledge no phytochemical study has been reported to this genus. The leaves of the specimens collected in 1997 were extracted with hexane followed by methanol. From the hexane extract were isolated six isoflavones, two rotenones, a mixture of the triterpenes lupeol and β-amirin and a mixture of stigmasterol and sitosterol. From the methanolic extract the new 2’,4’,5’- trimethoxyisoflavone-7-O-β-D-glucopyranoside was isolated, besides of this the proline amino acid and the ciclytol 3-O-metilchiroinositol. From the aerial parts of the specimen collected in 1999, an essential oil was obtained by hydrodistillation, where eleven constituents were identified. Nine of them were monoterpenes and two of them were sesquiterpenes. The major constituent of the essential oil was indentified as the monoterpene thujenone, which was isolated later by chromatography column over silica gel. Chromatographic techniques were used for the fractionation of the extracts and the isolation of the substances. The identification of the substances was accomplished by spectral analysis such as 1H and 13C NMR (including DEPT, HOMOCOSY and HETCOR), mass and IR spectra. The extracts obtained were submitted to the bioassays in order to verify its antifungal activities using spores of the phytopathogenic fungus Cladosporium sphaerospermum; where both were actives. The substances 7,3’,4’- trimethoxyisoflavone, 7,2’,4’,5’-tetramethoxyisoflavone, rotenone e 12a- hydroxyrotenone, isolated from the hexane extract, were responsible for the antifungal activity.
