Resumo:As ativinas são citocinas membros da superfamília do fator de crescimento transformador-β (TGFβ) que podem ser sintetizadas por quase todos os tipos celulares no organismo e assim a maioria dos tecidos têm a capacidade de ser fonte de ativina A na circulação. Desta forma, os objetivos desse estudo foram determinar se as concentrações plasmáticas e peritoneais da ativina A sofrem alterações em bezerros portadores ou não de hérnia umbilical (HU) antes e após a correção cirúrgica e em qual compartimento o seu aumento refletiria melhor e mais rapidamente as alterações inflamatórias, seu papel como indicador da inflamação na cavidade abdominal e sua correlação entre concentrações plasmáticas e peritoneais e ainda estabelecer intervalos de referência para a variável em cada grupo. Foram utilizados vinte bezerros holandeses com (n=10) ou sem HU (n=10), bezerros com HU foram submetidos à herniorrafia. Sangue total e líquido peritoneal foram coletados dos 20 animais para a quantificação da ativina A pelo teste ELISA nos dias 0 (antes da cirurgia) 1, 3, 5, 7 e 15. Dados dos dois grupos foram comparados. As concentrações plasmáticas e peritoneais de ativina A no grupo controle não tiveram alterações ao longo do tempo, porém no grupo experimental houve um aumento da concentração plasmática 15 dias após a herniorrafia em relação ao momento basal. Quando comparados os grupos, as concentrações plasmáticas de ativina A do grupo experimental estavam diminuídas no 3º dia e aumentadas no 15º dia após a cirurgia em relação aos animais do grupo controle, enquanto a concentração peritoneal apresentou-se aumentada 5 dias após a herniorrafia no grupo experimental em relação ao grupo controle / Abstract:Activins are members of the transforming growth factor β (TGF-β) superfamily and can be synthesized by almost all cell types in the body and most tissues has the ability to be a source of activin A in circulation. Thus, the objectives of this study were to determine if plasma and peritoneal concentrations of activin A change in calves with or without umbilical hernia (UH) before and after surgery and in which compartment its increase would reflect better and faster inflammatory changes. Its role as an inflammatory biomarker in the abdominal cavity and the correlation between plasma and peritoneal concentrations, and establish reference intervals for the variable in each group. Twenty Holstein calves were used with (n = 10) or without HU (n = 10), calves with HU underwent herniorrhaphy. Whole blood and peritoneal fluid were collected from 20 animals for quantification of activin A by ELISA on days 0 (before surgery) 1, 3, 5, 7 and 15. Data were compared between the two groups. Plasma and peritoneal concentrations of activin A in the control group did not change over time, but in the experimental group there was an increase in the plasma concentration 15 days after herniorrhaphy compared to baseline.
