An assessment of water quality and endocrine disruption activities in the Eerste/Kuils River catchment system, Western Cape, South AfricaFourie, Shani 04 1900 (has links)
Thesis (MSc)--University of Stellenbosch, 2005. / ENGLISH ABSTRACT: Water quality analysis forms the basis in assessing and monitoring catchments. As
urban development continuously increase, pollution sources increase in either point
source (wastewater treatment works, industrial effluents) and/or non-point source
origin (storm water discharge, domestic pollutants), accumulating pollutants in the
environment. It was only recently discovered that certain pollutants have subtle
disrupting effects on the endocrine system resulting in health related problems
associated with the reproductive system and thyroid system (growth and
development) of animals and potentially humans. Natural water resource
management proves to include limited biological assays measuring endpoints for
cytoxicity, inflammatory activity and endocrine disruption. The broad objective of
this study was therefore to include several bioassays, not normally used in municipal
(City of Cape Town) monitoring programmes, along with water quality data collected
by the City of Cape Town. The Eerste/Kuils River catchment system, Western Cape,
under the auspices of the City of Cape Town was chosen, and although this catchment
does not contribute to drinking water resources, is subjected to a range of
anthropogenic influences (industrial effluents, household wastewater, agricultural
runoff). Within the short time-frame available for this study (six months) two months,
July (following a dry summer and autumn season) and October (following a wet
winter and early spring season) were selected for water quality monitoring. Spatial
variation (with relevance to specific point and non-point contamination) among
sampling sites were also obtained by choosing several (n=10) along the catchment.
Specific aim of the study therefore included: Firstly (Chapter 2), the use of in vitro
bioassays, lactate-dehydrolises assay (LDH) for cytotoxic activity, pro-inflammatory
hormone Interleukin-6 (IL-6) secretion by human blood cells and a specific
Salmonella ELISA for faecal contamination, in conjunction with routine chemical and
biological (mostly microbiological) monitoring activities. The study indicated
significant variation among sites in all microbiological measures as well in IL-6
secretion and Salmonella presence. Between months, variations were also evident in
certain variables. Secondly (Chapter 3), two bioassays using the yolk precursor
protein, vitellogenin (Vtg) as endpoint was implemented in a) an in vitro Xenopus
laevis liver slice assay (five day exposure) and b) an in vivo Zebrafish (Danio rerio)
bioassay (seven day exposure) assessing estrogenic activity in the Eerste/Kuils River catchment. Although estrogen spiked positive control water samples stimulated Vtg
production in vitro as well as in vivo, no dramatic estrogenic activity was measured at
any of the selected sites. Thirdly (Chapter 4), a bioassay using the thyroid controlled
metamorphosis in Xenopus laevis tadpoles to assess effects on the thyroid hormonal
system was implemented. Thyroid stimulatory activity, compared with a negative
control sample, was measured at two sites along the catchment. Although the
practical implementation of the tadpole semi-static exposure protocol (water
replacement) proved to be labour intensive, all the added bioassays proved to be
valuable tools to add valuable information regarding water quality. It is clear that
more research related to anthropogenic influences along the Eerste/Kuils River
catchment system are needed, specifically in monitoring monthly variations to better
understand annual variation in several of the endpoints studied. / AFRIKAANSE OPSOMMING: Waterkwaliteit vorm die basis vir die evaluering en monitering van opvangsgebiede.
Voortdurende stedelike ontwikkeling gee aanleiding tot ‘n toename in die voorkoms
van besoedelstowwe in die natuurlike omgewing deur gelokaliseerde (punt) bronne
(rioolwerke/industriële uitvloeisel) en/of nie gelokaliseerde (nie punt) bronne (vloed
uitlaat/huishoudelike uitvloeisels) van besoedeling. Dit het onlangs aan die lig gekom
dat van hierdie chemise besoedelstowwe subtiel die endokriene sisteem versteur en so
aanleiding gee tot gesondheidsprobleme in terme van die voortplantingsisteem en
tiroied sisteem (groei en ontwikkeling) by diere en moontlik ook die mens. Daar is
beperkte gebruik van biologiese toetse wat inligting verskaf oor sitotoksiteit,
inflammatoriese aktiwiteit en endokriene versteuring. Die doel van hierdie studie was
dus om van hierdie biologiese toetse, wat normaalweg nie deel uitmaak van die
roetine munisipale (Stad van Kaapstad) opvangsgebied monitering nie, gebruik te
maak. Die Eerste-, Kuilsrivier, Wes Kaap, onder beheer van Stad Kaapstad is gekies
en alhoewel die opvangsgebied nie water bydra tot drinkwaterbronne nie, word die
opvangsgebied beïnvloed deur verskeie mensgemaakte bronne van besoedeling
(afloop vanuit omliggende lanbougebiede). Binne die kort tydsraamwerk van die
projek (ses maande) is besluit om twee maande, Julie (volg ‘n droë somer en herfs
seisoen) en Oktober (volg ‘n nat winter en vroeë lente seisoen) vir water kwaliteit
monitering te kies. Ruimtelike variasie langs die loop van die opvangsgebied is
ingesluit deur moniteringspunte (n=10), met in ag name van die potensiële
besoedelingsbronne. Spesifieke doelwitte van die projek sluit in: Eerstens (Hoofstuk
2), om die in vitro biotoetse, laktaat hidrolise (LDH) vir sitotoksisiteit, proinflammatoriese
hormoon Interleuikin-6 (IL-6) vir inflammatoriese aktiwiteit,
vrygestel deur menslike bloedselle en ‘n Salmonella ELISA vir ontlasting besoedeling

voorgekom. Tweedens (Hoofstuk 3), is twee biotoetse wat die
dooiervoorloperproteïen, vitellogeen (Vtg) as eindpunt gebruik geimplimenteer in a)
‘n in vitro Xenopus laevis lewersnit biotoets (vyf dag blootstelling) en b) ‘n in vivo
Zebravis (Danio rerio) biotoets (sewe dag blootstelling) om estrogenisiteit in die Eerste-, Kuilsrivier opvangsgebied te evalueer. Alhoewel, die estrogeen behandelde
positiewe kontrole water monsters Vtg produksie veroorsaak het in beide die in vitro
lewer-kulture en in vivo vistoets, is geen dramatiess estrogeniese aktiwiteit by enige
van die moniteringspunte gevind nie. Derdens (Hoofstuk 4), is ‘n biotoets wat die
tiroïedbeheerde metamorfose in Xenopus laevis paddavisse gebruik om effekte op die
tiroïedsisteem te evalueer. Die differensiële stimulering (versnelling), in vergelyking
met ‘n negatiewe kontrole watermonster, van die tiroied sisteem is by twee
moniteringspunte in die opvangsgebied waargeneem. Alhoewel die praktiese
implementering van die paddavis semi-statiese (water word gereeld vervang) biotoets
arbeidsintensief is, het alle bykomstige biotoetse waardevolle bykomstige inligting
oor water kwaliteit verskaf wat ‘n belangrike bydrae tot ingeligte bestuursbesluite kan
verleen. Dit is duidelik dat meer navorsing int verband met die menslike invloed
langs die Eerste-, Kuilsrivier opvangsgebied nodig is, veral met maandlikse
monitering vir seisoenale veranderinge.

Groundwater purification using functionalised magnetic nanoparticles (electromagnetic separation)Aigbe, Uyiosa Osagie 01 1900 (has links)
Most developing countries are faced with drinking water problems, with conditions becoming more severe due to water pollution. Meeting the growing demands for clean water in most countries, there are difficult challenges as the availability and supply of drinkable water are diminishing. Due to economic and environmental concerns, development of additional physical means for the removal of organic compounds from wastewater using permanent magnets, electromagnetic coils, electrodes and ultrasonic pretreatment is desirable. Improving the adsorption and separation process, magnetic field exposure method has progressively drawn consideration. Magnetic field exposure method has demonstrated its capacity for increasing the adsorptive elimination of contaminants from water as static magnetization is suitable, simple and cost-effective.
The polypyrrole magnetic nanocomposite use for adsorption experiments influenced by exter-nal magnetic field was prepared using the in-situ polymerization method, which was charac-terized using TEM, SEM, EDX, XRD, BET, FTIR, VSM, and ESR spectrophotometers. The magnetic nanocomposite (PPy/Fe3O4) was observed to have an average particle size of 10 nm with the elementary composition of carbon, oxygen, nitrogen, chloride and iron. The magnetic nanocomposite had a crystalline structure of face-centred cubic lattice of Fe3O4, an adsorption-desorption isotherm shape indicating a typical type-IV mesoporous material with a surface area of 28.77 m2/g. Characteristic peaks of Fe3O4 and PPy were also observed using FTIR spectro-photometer. From the VSM and ESR characterization, the synthesized superparamagnetic ma-terial was shown to have a saturation magnetization of 23 emu/g and an effective g-value of 2.25 g which was attributed to Fe3+ spin interaction.
An enhanced removal of Cr(VI), fluoride and congo red dye were observed under the influence of magnetic field, with parameters like pH, adsorbent dosage, the initial concentration of ad-sorbate, magnetic field and magnetic exposure time been varied. The enhanced adsorption of contaminants using magnetic field is attributed to the increase in the magnetic field induced on the particles over a magnetic exposure time, resulting in the rotating particles forming aggre-gates due to the increased magnetic force and torque on the particles from the PSV results. This leads to increase in the chain collision and area of particle interaction with the aqueous solution of hexavalent chromium, fluoride and congo red dye. / Physics / Ph. D. (Physics)

Project water (Grahamstown) : a case study of the development of an environmental education projectAshwell, Alice Nicola January 1992 (has links)
Environmental education is an approach to education which emphasises the interrelatedness of people and their human and non-human environments and seeks to encourage environmental awareness, concern and action. This case study documents the implementation and development of Project WATER, Grahamstown, a practical environmental education project dealing with catchment conservation and water quality monitoring. The Grahamstown project is one of a number of local water quality monitoring initiatives affiliated to GREEN (the Global Rivers Environmental Education Network). Participants in the project included student teachers from the Department of Education at Rhodes University and pupils and teachers from three farm schools in the district and four high schools in the town. Project WATER, Grahamstown developed as an Action Research and Community Problem-Solving project. The study focuses on fragmentalist and holistic approaches to education, people's responses to Project WATER and the choice of action research as the research method.

Land pollution and population density : the case of Kwekwe City residential areas, ZimbabweChatsiwa, Jaison 02 1900 (has links)
In most developing countries, the problem of inefficient municipal solid waste management (MSWM) is endemic. The problem is manifested by heaps of uncollected solid waste found on open areas and by the street sides. This dissertation examines the relationship between land pollution and population density resulting from solid waste generation in the City of Kwekwe in Zimbabwe. Questionnaires and interviews were conducted to 375 randomly selected households in low, medium and high density residential suburbs. The household survey was triangulated with field observations, key informant interviews and secondary data sources. The average household size in high, medium and low density residential areas was 11, 8 and 5 people respectively. The study revealed that the amount and composition of solid waste generated varied according to household size, income and education levels of residents. The higher the population density, the more the solid waste produced. However, waste generated was not carried to the dumpsites for disposal. Also the higher the income, the higher the amount of solid waste produced, although residents with high income were able to ferry their wastes to the dumpsites hence less land pollution. The solid waste generation rate in high density areas was 0.04 kg/capita day, in medium density areas was 0.35 kg/capita/day and in low density areas were 0.84 kg/capita/day. It was also observed that among high income earners, the educated people produced more waste than low income earners. However, the low income earners and the less educated people could not take care of their environment to limit roadside and open space littering. However, lack of public awareness coupled with inefficient and ad hoc waste management system continue make provision of effective solid waste management services illusive. Based on the research findings, it is suggested sound environmental stewardship amongst residents will limit land pollution in the city of Kwekwe. / Environmental Sciences / M.A. (Environmental Management)

Previous issue date: 2007 / Resumo: Esta pesquisa insere-se no âmbito da atual problemática ambiental urbana, mais especificamente, na contaminação atmosférica ou poluição do ar. Esta tem se agravado nas últimas décadas e passa a exigir estudos mais aprofundados, caracterizando-se como fator importante na busca da preservação do meio ambiente e de alternativas de desenvolvimento que possam diminuir o ritmo acelerado desta contaminação. Discute então, como os problemas de escalas globais podem ser analisados desde um estudo de escala local. Mostra que para saber quais são as causas e efeitos dos problemas ambientais, que envolvem os aspectos tanto social, cultural, econômico, como político, é necessária uma pesquisa sócio-espacial. A área do estudo é a cidade de Campinas, estado de São Paulo, Brasil, cuja população é de mais de um milhão de habitantes, decorrente principalmente do avanço tecnológico e industrial. Utilizando pequenas territorialidades, de escalas locais, procurou-se comparar dois bairros da Cidade de Campinas: Cambuí e Real Parque, muito diferentes nas suas condições sócio-espaciais. Nesse sentido, considerou-se importante averiguar como a contaminação do ar afeta a população desses bairros. O grande problema da poluição do ar, é que esta é ainda mais complexa em ambientes urbanos e suas conseqüências afetam sobretudo as relações dos homens entre si com a natureza. A pesquisa revelou que os dois bairros são afetados, independentemente das suas diferenças. Isto fica constatado na comparação das análises e entrevistas realizadas com os moradores, que afirmam estar enfrentando muitos problemas, principalmente com relação à saúde e ao clima. No final, a dissertação propõe a edu-comunicação sócio-ambiental para as políticas públicas como veículo de conhecimento e conscientização dos graves riscos à que estão exposto moradores e cidadãos. Sabe-se que a contaminação atmosférica afeta o mundo todo sem distinção de raça, cor, ou condições sociais, mas os mais afetados são geralmente aqueles que não tem as mínimas condições de fazer frente ao problema. Assim sendo, algumas sugestões de alternativas são propostas a fim de contribuir com a redução dos poluentes, mediante à re-educação dos moradores inseridos nas cidades. Isto, porque as pequenas partes afetam o todo e fazer nossa parte é tarefa de cidadania / Abstract: This research is inserted to the urban context of environmental problems in ours days, more specifically related to the city¿s air contamination or air pollution¿. This problem during the past decades, has been asking for deeper studies, revealing itself as a very important issue for improving urban environment and for a development able to help reduces the accelerated rythm of air pollution, as well. This research tries to discusses how global space¿s scales problems can be analyzed from the local¿s space scale. In order to recognize causes and consequences from environmental problem which involves different aspects such physical, economic, social, political and cultural needs , this work shows the need of a social space research. The study area is the city of Campinas, state o São Paulo -Brazil, which population reaches more than a million peoples, generated from urbanization, industrial and technological development. Using small territorialities of local scales, this work tried to compare two neighborhoods in Campinas, the Cambui and Real Park, very different from social conditions . In this sense, was important to detect how the air contamination is affecting those people. The great problem is that the air contamination is even worst and more complex in urban environment, that affects mostly the relationshisps between men and the nature. The results of this research showed that both neighborhoods has been affected. despite their differences. This is showed by the compared analysis and interviews with housekeepers that says they are fighting mostly against problems related to health and clime. The conclusion of this dissertation brings the ¿edu - comunication¿ as an environmental purpose, a mechanism to help clarify people and citizens about the risks they are exposed. The air contamination affects all the world without discriminating race, color, or social conditions, but generally the most affected are poorest people who doesn¿t have any condition to fight against it. So, some alternatives comes to the end trying to contribute reduces air pollution through urban people¿s re-education. Même si la réduction des apports de nutriments dans les écosystèmes aquatiques est généralement envisagée avec des approches « effects-based », ce n’est pas le cadre qui a été suivi dans cette thèse. En effet, dans une démarche de développement durable, il semble plus pertinent, de définir des mesures de contrôle de la pollution agissant sur les relations de cause à effet. Selon cette logique, la méthodologie proposée par la Directive Cadre sur l’Eau (DCE) pour protéger les écosystèmes aquatiques présente plusieurs faiblesses. Par le fait qu’elle réfère à certains principes de l’économie standard de l’environnement, l’analyse économique de la DCE pose plusieurs problèmes pour agir sur la causalité de la pollution. Néanmoins, la méthodologie proposée par la DCE est pertinente par le fait qu’elle recommande l’utilisation combinée de mesures techniques et d’instruments pour diminuer les dommages environnementaux. Conformément à cette disposition, l’effet de mesures techniques et d’instruments sur le secteur agricole et sur la chaine alimentaire l’englobant a été évalué puisque cette dernière est responsable de la majorité des émissions diffuses et ponctuelles d’azote et de phosphore dans les eaux de surface. La définition de mesures techniques en fonction de l’efficience de l’utilisation des nutriments (Nutrient Use Efficiency, NUE) dans la production agricole a été complétée par l’étude des instruments économiques utilisés dans la Politique Agricole Commune (PAC) pour développer une agriculture multifonctionnelle. Si la thèse concerne de manière générale l’eutrophisation des écosystèmes aquatiques, elle se penche plus particulièrement sur les pressions au sein du bassin de l’Escaut et sur leurs impacts jusqu’à la zone côtière de la mer du Nord. Des mesures techniques agissant sur les causes des émissions diffuses du secteur agricole et sur les émissions ponctuelles des stations d’épuration ont été proposées pour ce bassin. Ces mesures techniques ont été caractérisées prioritairement par leur efficacité environnementale (grâce aux modèles Sénèque-Riverstrahler et MIRO) et en fonction de leurs coûts directs. <p><p>/<p><p>Although reducing the load of nutrients in aquatic ecosystems is usually looked at through "effects-based" approaches, this is not the context which this paper has chosen to follow. In fact, keeping in line with sustainable development, it seems more relevant to define measures to control pollution which act on cause and effect relations. According to this way of thinking, the methodology proposed by the Water Framework Directive (WFD) to protect aquatic ecosystems presents a number of weaknesses. By the fact that it refers to certain standard economic principles for the environment, the WFD’s economic analysis poses several problems towards acting on the cause of the pollution. Nevertheless, the methodology proposed by the WFD is relevant, as it recommends the combined use of technical measures and of instruments to reduce environmental damage. In accordance with this disposition, an assessment has been made of the effect of technical measures and instruments on the agricultural sector and on the food chain encompassing it, since the food chain is responsible for the majority of diffuse and point emissions of nitrogen and phosphorus in surface waters. The definition of technical measures in terms of efficient use of nutrients (Nutrient Use Efficiency, NUE) in agricultural production has been complemented by a study of economic instruments used in the Common Agricultural Politicy (CAP) to develop a multifunctional type of agriculture. Although the paper deals with the eutrophication of aquatic ecosystems in a general way, it concentrates particularly on the increasing pressures in the Scheldt basin and on their impacts as far as the North Sea coastal zone. Technical measures acting on the causes of diffuse emissions in the agricultural sector and on point emissions in the water treatment plants have been proposed for this basin. Estimation of the emissions of gases from a two landfill sites using the LandGEM and Afvalzorg models: Case study of the Weltervenden (Polokwane) and Thohoyandou landfillsNjoku, Prince Obinna 21 September 2018 (has links)
MENVSC / Department of Ecology and Resource Management / Over the years it has been observed that the solid waste sector has been an increasingly major contributor to the amount of Greenhouse gases (GHGs) in the atmosphere. To some extent a great chunk of these GHGs in the atmosphere is from Landfill gas (LFG). This study employs two theoretical models (LandGEM and Afvalzorg models) to estimate the amount of LFG emitted from Weltervenden and Thohoyandou landfill sites located in Limpopo province of South Africa. Furthermore, the study investigated the appropriate technique of the LFG utilisation as a source of electricity and the number of households using electricity. LFGcost model was used to estimate the cost and benefits related to the implementation of a LFG utilisation technology. Also, the possible health and environmental impacts of the landfill emissions on the people living closer to the landfill site were determined.
The LandGEM model’s simulation concludes that CH4 and CO2 peaked in the year 2020 with values of 3.323 × 103 Mg/year and 9.118 × 103 Mg/year, respectively, for the Thohoyandou landfill. Results from the Afvalzorg model indicate that the CH4 peaked in the year 2020 with value of 3.501 × 103 Mg/year. Meanwhile the total emission of CH4 from 2005-2040 by the LandGEM and Afvalzorg models are 66200 Mg/year and 69768 Mg/year, respectively. However, for the Weltervenden landfill, the total LFG peaked in the year 2023 while the CH4 peak at 4061 Mg/year and 3128 Mg/year for LandGEM and Afvalzorg models, respectively.
Furthermore, results from the cost analysis and benefits for the implementation of a LFG utilisation technology in both landfills show that the implementation of such a utilisation technology will be economically feasible considering the sale of t CO2 equivalent in the carbon market. However, without considering the sales of t CO2 equivalent, not all the LFG engines are economically feasible for both landfills.
This study also shows that the residents living closer to the Thohoyandou landfill are at a higher risk of environmental pollution and could suffer negative impacts from the landfill than residents living far from the landfill site. However, the Weltervenden landfill did not have lots of communities living closer to the landfill and therefore it was not included in this study. / NRF

Detection of aeromonas species in relation to the occurrence of estrogens and testosterone in various water resources in Limpopo Province, South Africa and Lusaka, ZambiaManavhela, Murendeni 18 May 2019 (has links)
MSc (Microbiology) / Department of Microbiology / Background: The occurrence of microorganisms and endocrine disrupting chemicals
(EDCs) in water poses a serious concern due to their effects on humans, animals and
environment. In recent years, EDCs have been increasingly reported in rivers that
receive large amounts of wastewater effluents. Of all the EDCs, natural and synthetic
hormones are among those that are recognized for their potential to mimic or interfere
with normal hormonal functions of humans and animals. The present study aimed at
assessing the occurrence of these hormones in relation to the molecular diversity of
Aeromonas and evaluating the resistance of Aeromonas to antibiotics as well as to
assess anti-bacterial activity of two selected traditional medicinal plants.
Methods: Wastewater, water and fish samples were collected from various sources
(rivers, wastewater treatment plants, taps, and dams) for the detection of hormones
and isolation of Aeromonas species. The analysis of hormones from various organs
of the fish and from water samples was conducted, after extraction using enzymelinked
immunosorbent assays (ELISA). Different types of hormones including Estriol,
Estradiol, Ethinylesradiol and Testosterone were detected, and their concentrations
determined. Aeromonas spp were isolated rom the samples using microbiological
methods and Conventional PCR was used for genotyping as well as for detection of
the beta-lactamase genes. Kirby-bauer method was used to determine the
susceptibility profiles of Aeromonas to different antibiotics. Microdilution assay was
used to determine the Anti-bacterial activity of the plant (Annoniceae and Zornia
milneana) extracts against Aeromonas species.
Results: A total of 144 samples were collected from 23 different locations in two
countries: South Africa and Zambia. These included wastewater and treated
wastewater, River water, fish and tap water. 17α-ethinylestradiol (EE2) was detected
in most of the samples (92.7%) with concentrations varying from 0.59 ng/ml to 65
ng/ml. The hormones were also detected from drinking water, with testosterone
detected at high concentrations of up to 140 ng/ml in tap water. Most sewage treatment
plants were not able to remove the EE2 from the wastewater as the concentration of
this hormone in the final effluent was almost always higher than that in the influent.
These homones were also detected in drinking water at high concentrations of up to
53.49 ng/ml in the tap water for EE2 and 1777 ng/ml for E2. The overall detection of
Aeromonas species in the samples was 84.5%. A. caviae was the most prevalent
species accounting for 73.6%, followed by A. veronii with 64.6%. The bacteria were
completely resistant to cefuroxime accounting for 100% resistance. Aeromonas
isolates also showed high resistance to trimethroprim (88.7% for A. hydrophila),
cefazolin (highest 97.8% for A. cavie), and ceftazidime (83.9% for A. sobria). TEM was
the most prevalent beta-lactamase gene with detection rate of 87%. All isolates lacked
the presence of the CTX-M3 gene. Also, wastewater had the highest prevalence of A.
veronni and A. caviae accounting for 87.5% and 82.5% respectively. Multiple antibiotic
resistance was also observed with the Aeromonas isolates being resistant to up to 11
antibiotics. High prevalence of 77.1% of Aeromonas hydrophila was observed in the
presence of ethinylestradiol (EE2). Aeromonas veronii and Aeromonas caviae were
the most predominant species in the presence of total estriol, A. veronii had a
prevalence of 57.1% and A. caviae had a prevalence of 52.8%. Aeromonas hydrophila
and Aeromonas caviae had the lower prevalence in the presence of hormones with
the percentages of 26.1% and 27.8% respectively. The methanol extracts of both
Zornia milneana and Annona species showed good activity against the Aeromonas
spp with the lowest MIC of 0.078 mg/ml. Ethyl acetate extracts were the least effective.
Conclusion: This study has shown high occurrence of steroid hormones in all types
of environmental samples tested. These included tap water, river water, wastewater
and fish both in Zambia and South Africa. Therefore, steroid hormones constitute and
important health problem in the Southern African Sub-Region. The incapacity of the
wastewater treatment plants to remove EE2 is an important problem that needs to be
tackled immediately. The prevalence of Aeromonas species is very high in our
environmental water as well as in drinking water, with the highest prevalence observed
in fish and wastewater. It was also revealed that there is relationship between steroid
hormones and Aeromonas species, with the hormones supporting the growth of
Aeromonas species. The presence of beta-lactamase genes which causes
Aeromonas to be resistant to antibiotics was also noted. Methanol extracts of Zornia
milneana and Annona spp were the most effective against Aeromonas spp and could
serve as primary sources for the isolation of lead compounds. / NRF

Transport of nitrogen oxides and nitric acid pollutants over South Africa and air pollution in Cape TownOjumu, Adefolake Mayokun 24 October 2013 (has links)
The deteriorating air quality in Cape Town (CT) is a threat to the social and economic development of the city. Although previous studies have shown that most of the pollutants are emitted in the city, it is not clear how the transport of pollutants from neighbouring cities may contribute to the pollution. This thesis studies the transport of atmospheric nitrogen oxides (NOx) and nitric acid (HNO3) pollutants over South Africa and examines the role of pollutant transport from the Mpumalanga Highveld on pollution in CT.
The study analysed observation data (2001 - 2008) from the CT air quality network and from regional climate model simulation (2001 - 2004) over South Africa. The model simulations account for the influences of complex topography, atmospheric conditions, and atmospheric chemistry on transport of the pollutants over South Africa. Flux budget analysis was used to examine whether the city is a net source or sink for NOx and HNO3.
The results show that north-easterly flow transports pollutants (NOx and HNO3) at low level (i.e., surface to 850 hPa) from the Mpumalanga Highveld towards CT. In April, a tongue of high concentration of HNO3 extends from the Mpumalanga Highveld to CT, along the southern coast. The flux budget analysis shows that CT can be a net sink for NOx and HNO3 during extreme pollution events. The study infers that, apart from the local emission of the pollutants in CT, the accumulation of pollutants transported from other areas may contribute to pollution in the city. / Environmental Sciences / M. Assessment of polycyclic aromatic hydrocarbon (PAHs) and heavy metals in the vicinity of coal power plants in South AfricaOkedeyi, Olumuyiwa Olakunle 12 November 2013 (has links)
The distribution and potential sources of 15 polycyclic aromatic hydrocarbons (PAHs) in soils and Digitaria eriantha in the vicinity of three South African coal-fired power plants, Matla, Lethabo and Rooiwal were determined by gas chromatography–mass spectrometry. An ultrasonic assisted dispersive liquid-liquid microextraction (UA-DLLME) method was developed for the extraction of polycyclic aromatic hydrocarbon in soil, followed by determination using gas chromatography mass spectrometry. The study showed that an extraction protocol based on acetonitrile as dispersive solvent and C2H2Cl2 as extracting solvent, gave extraction efficiencies comparable to conventional soxhlet extraction for soil samples. The extraction time using ultrasonication and the volume of the extraction solvent was also investigated. Using a certified reference material soil (CRM), the extraction efficiency of UA-DLLME ranged from 64 to 86% in comparison with the Soxhlet result of 73 to 95%. In comparison with the real sample, the CRM result did not show a significant difference at 95% C.I. The UA-DLLME proved to be a convenient, rapid, cost-effective and greener sample preparation approach for the determination of PAHs in soil samples. PAH compound ratios such as phenanthrene/phenanthrene + anthracene (Phen/ Phen + Anth) were used to provide a reliable estimation of emission sources. The total PAH concentration in the soils around three power plants ranged from 9.73 to 61.24 μg g−1, a range above the Agency for Toxic Substances and Disease Registry levels of 1.0 μg g−1 for a significantly contaminated site. Calculated values of the Phen/Phen + Anth ratio were 0.48±0.08, 0.44±0.05, and 0.38+0.04 for Matla, Lethabo and Rooiwal, respectively. The flouranthene/fluoranthene + pyrene (Flan/ Flan + Pyr) levels were found to be 0.49±0.03 for Matla, 0.44±0.05 for Lethabo, and 0.53±0.08 for Rooiwal. Such values indicate a

pyrolytic source of PAHs. Higher molecular weight PAHs (five to six rings) were predominant, suggesting coal combustion sources. The carcinogenic potency B[a]P equivalent concentration (B[a] Peq) at the three power plants ranged from 3.61 to 25.25, indicating a high carcinogenic burden. The highest (B[a] Peq) was found in samples collected around Matla power station. It can, therefore, be concluded that the soils were contaminated with PAHs originating from coal-fired power stations.
Nine metals (Fe, Cu, Mn, Ni, Cd, Pb, Hg, Cr and Zn) were analysed in soil and the Digitaria eriantha plant around three coal power plants (Matla, Lethabo and Rooiwal), using ICP-OES and GFAAS. The total metal concentration in soil ranged from 0.05 ± 0.02 to 1835.70 ± 70 μg g-1, 0.08 ± 0.05 to 1743.90 ± 29 μg g-1 and 0.07 ± 0.04 to 1735.20 ± 91 μg g-1 at Matla, Lethabo and Rooiwal respectively. The total metal concentration in the plant (Digitaria eriantha) ranged from 0.005 ± 0.003 to 534.87 ± 43 μg g-1 at Matla, 0.002 ± 0.001 to 400.49 ± 269 μg g-1 at Lethabo and 0.002 ± 0.001 to 426.91 ± 201 μg g-1 at Rooiwal. The accumulation factor (A) of less than 1 (i.e. 0.003 to 0.37) at power plants indicates a low transfer of metal from soil to plant (excluder). The enrichment factor values obtained (2.4 – 5) indicate that the soils are moderately enriched, with the exception of Pb that had significant enrichment of 20. The Geo-accumulation Index values of metals indicate that the soils are moderately polluted (0.005 – 0.65), except for Pb that showed moderate to strong pollution (1.74 – 2.53). / Chemistry / D.
