Development of Cancer-Genomics-Guided Precision Immunotherapy for Triple-Negative Breast CancerSun, Yifan 05 1900 (has links)
Indiana University-Purdue University Indianapolis (IUPUI) / Triple-negative breast cancer (TNBC), which accounts for 15-20% of all breast cancers, is highly aggressive and metastatic with the poorest overall rates. While surgery, radiation, and chemotherapy remain the main treatment options, TNBC represents an unmet medical need for better treatment strategies. Tremendous efforts have been made to develop effective therapies over the past years. However, TNBC treatment options are still very limited due to the lack of good drug targets and the low response rate of current therapies. In this study, we developed two different strategies to treat TNBC based on its cancer genomic features: 1) heterozygous loss of chromosome 17p (17p loss) and 2) high mutation load.
17p loss is one of the most frequent genomic events in breast cancer including TNBC, rendering cancer cells vulnerable to the inhibition of POLR2A via α-amanitin (POLR2A-specific inhibitor). Here, we developed a new drug T-Ama (α-amanitin-conjugated trastuzumab) targeting HER2-low TNBC with 17p loss by combining the effects of α-amanitin and trastuzumab (HER2+ breast cancer therapy). Our results showed that T-Ama exhibited superior efficacy in treating HER2-low TNBC with 17p loss in vitro and in vivo, and surprisingly induced immunogenic cell death (ICD) which further enhanced T cell infiltration and cytotoxicity levels and delivered greater efficacy in combination with immune checkpoint blockade therapy. Collectively, the therapeutic window created by 17p loss and HER2 expression will make HER2-low TNBC clinically feasible targets of T-Ama. As another genetic feature of TNBC, the higher genomic instability and mutational burden results in more neoantigens presented on MHC-I, along with the higher level of tumor-infiltrating T cells, making TNBC a perfect model for immunotherapy compared to the other breast cancer subtypes. Here, we designed a deconvolution-algorithm-derived library screening to find new therapeutic targets and identified PIK3C2α as a key player that determines MHC-I turnover and reduces the MHC-I-restricted antigen presentation on tumor cells. In preclinical models, inhibition of PIK3C2α profoundly suppressed breast tumor growth, increased tumor-infiltrating CD8+ T cells, and showed high potential enhancing the efficacy of anti-PD-1 therapy, suggesting that PIK3C2α is a potential therapeutic target for TNBC immunotherapy. / 2025-05-22

Influence of the Negative IAPS and Method of Hemispheric Presentation on Performance on the Affective Auditory Verbal Learning TestCosenzo, Keryl Ann 21 May 2002 (has links)
This investigation examined the effects of emotion and lateralized presentation of a list of affective words on the learning of that list. This investigation also attempted to assess the separate influences of emotion and arousal on verbal performance. Experiment I was a 2x2 factorial design: two types of pictures from the International Affective Picture System (IAPS), negative or neutral and two gender conditions, male and female. Experiment I was used to verify that the IAPS results in an emotional response. Heart rate (HR) and skin conductance level (SCL) were measured during IAPS presentation. Emotion was assessed after IAPS presentation. Results showed the negative IAPS condition elicited significantly greater increases in SCL and decreases in HR, less self-reported of pleasantness and more self-reported activation than the neutral IAPS condition. Women had significantly lower SCL and higher HR than men; men and women did not differ in reported emotion. Experiment I verified that the negative IAPS elicit changes in SCL and negative emotion in men and women. Experiment II was a 2 x 2 x 2 mixed factorial design: two types of IAPS (negative or neutral), two AAVL conditions (positive and neutral AAVL words or negative and neutral AAVL words) and two presentation conditions (AAVL presented to the left ear (LE) or right ear (RE)). The measure of performance on the AAVL was the number of correct responses. HR and SCL were measured during presentation of the IAPS and of the AAVL. In the negative IAPS condition, performance on negative AAVL was significantly better than that for the positive AAVL; for presentation of the AAVL to the RE, performance on the negative AAVL was significantly better than that for the positive and neutral AAVLs. IAPS condition or ear of presentation alone did not significantly impact on AVVL performance or on SCL and HR during the recitation of the AAVL. Evidence showed that the effect of emotion on performance is a function of mood congruent processing and possibly the allocation of hemispheric resources. / Ph. D.

Receptor-Mediated Antigen Delivery by Α<Sub>2</Sub>-Macroglobulin: Effect on Cytotoxic T Lymphocyte Immunity and Implications for Vaccine DevelopmentBowers, Edith Villette January 2009 (has links)
<p><p>The receptor-recognized form of α<sub>2</sub>-macroglobulin (α<sub>2</sub>M*) targets antigens (Ag) to professional Ag-presenting cells (APCs) for rapid internalization, processing, and presentation. When employed as an Ag delivery vehicle, α<sub>2</sub>M* amplifies major histocompatibility complex (MHC) class II presentation as demonstrated by increased antibody (Ab) titers. Recent evidence, however, suggests that α<sub>2</sub>M*-encapsulation may also enhance Ag-specific cytotoxic T lymphocyte (CTL) immunity. In these studies, we demonstrate that α<sub>2</sub>M*-delivered Ag (ovalbumin, OVA) enhances the production of specific <italic>in vitro</italic> and <italic>in vivo</italic> CTL responses. <br><p>Murine splenocytes expressing a transgenic T cell receptor (TCR) specific for CTL peptide OVA<sub>257-264</sub> (SIINFEKL) demonstrated up to 25-fold greater IFN-γ and IL-2 secretion when treated <italic>in vitro</italic> with α<sub>2</sub>M*-OVA compared to soluble OVA. The frequency of IFN-γ -producing cells was increased ~15-fold as measured by ELISPOT. Expansion of the OVA-specific CD8<super>+</super> T cells, as assayed by tetramer binding and [<super>3</super>H]thymidine incorporation, and cell-mediated cytotoxicity, as determined by a flow cytometric assay, were also significantly enhanced by α<sub>2</sub>M*-OVA. Furthermore, CTL responses were observed at Ag doses tenfold lower than those required with OVA alone. <br><p>We also observed enhanced humoral and CTL responses by naïve mice following intradermal immunization with α<sub>2</sub>M*-OVA. These α<sub>2</sub>M*-OVA-immunized mice displayed increased protection against a subcutaneously implanted OVA-expressing tumor, as demonstrated by delayed tumor growth and prolonged animal survival. The anti-tumor response observed with α<sub>2</sub>M*-mediated Ag delivery was comparable to that of an accepted vaccine adjuvant (CpG 1826) and appeared superior to a cell-based vaccine technique. <br><p>To further understand the mechanism underlying this enhanced CTL immunity, the subsets of professional APCs capable of cross-presenting α<sub>2</sub>M*-encapsulated Ag were investigated. Although both dendritic cells (DCs) and macrophages appear to stimulate some degree of cross-priming in response to α<sub>2</sub>M*-encapsulated Ag, CD8<super>+</super>CD4<super>-</super> and CD8<super>-</super>CD4<super>+</super> DCs appear to do so with the greatest efficiency. The implications of this finding to the ongoing debate regarding the relative contributions of APC subsets to Ag cross-presentation and the determinants of which cells cross-present with high efficiency are discussed. <br><p>These observations demonstrate that α<sub>2</sub>M*-mediated Ag delivery promotes cross-presentation resulting in enhanced Ag-specific CTL immunity. Considered in the context of previous work, these results support α<sub>2</sub>M* as an effective Ag delivery system that may be particularly useful for vaccines based on weakly immunogenic subunits or requiring dose sparing.</p> / Dissertation

Presentation became an important communication phenomenon of today. However, its quick onset caused that presenting has been considered as an ability without theoretical fundamentals. This diploma thesis makes an effort to convince the readers about the opposite by connecting practical presentation procedures with theoretical knowledge based on literature concerned with psychology and social communication. The emphasis is put especially on application of the theory of perception, attention and memory. In the application part of this theses all stages of preparing and delivering a presentation are analyzed step by step. Belonging to the empirical part, an evaluation of presenters was carried out. For this very purpose an evaluation sheet was created as a formalized methodical tool. This thesis is usable by any communicator who wants to understand the theoretical phenomena on which presentation is based.
