<p>The aim of this work was to understand the molecular mechanism of translation and the mechanism of translation termination, in particular. Cleavage of peptidyl-tRNA and peptide release terminates translation of mRNA on the ribosome. In prokaryotes, three release factors (RFs) are involved in this process. RF1 and RF2 recognise the three stop codons on mRNA and induce hydrolysis of the ester bond in peptidyl-tRNA. RF3 accelerates the rate of RF1 and RF2 recycling between ribosome in a GTP-dependent manner. We have clarified the mechanism of action of peptide release factor RF3. In the cell, free RF3 is in the GDP conformation. When RF3∙GDP binds to ribosome in complex with RF1 or RF2, these ribosome complexes act as guanine exchange factors for RF3 by inducing rapid dissociation of GDP. If, and only if, the peptide has been removed from tRNA, GDP is quickly replaced by GTP. Binding of GTP to RF3 induces a conformation of the factor with high affinity for the ribosome, which forces RF1 or RF2 to rapidly dissociate. Subsequent hydrolysis of GTP on RF3 induces a factor conformation with low affinity for the ribosome and rapid release of RF3∙GDP. It was further shown how the position of peptidyl-tRNA on the ribosome and the presence or absence of its peptide regulates the binding and GTPase activity of translation factors IF2, EF-G and EF-Tu. The result explains how idling GTPase hydrolysis and negative interference between different translation factors are minimized in living cells. The present biochemical observations, in conjunction with cryo-EM results, lead to new proposals for the role of hybrid sites in translocation of tRNAs, recycling of RF1 and RF2 by RF3 and recycling of post-termination ribosomes back to a new round of initiation.</p>

The Dangers of International Awards: A Lesson from Aung San Suu Kyi’s Nobel Peace Prize

Having spent over 20 years under house arrest fighting for democracy in Myanmar, Aung San Suu Kyi has been a bastion for peace for decades. She has received many international awards, including the Nobel Peace Prize in 1991, which she accepted in person in 2012. The plight of the Rohingya, a Muslim minority in Myanmar, has marred Suu Kyi’s reputation as a bastion of peace, leading to calls for her to lose her Peace Prize. Why is it that Suu Kyi’s image as the future of peace so different from reality? That question is what this research attempts to answer. Through a rhetorical analysis of Suu Kyi’s Nobel lecture and the media coverage that followed it, the impact of the use of tropes becomes evident. Metonymy, synecdoche, and narrative emerge in both the lecture and media coverage. Suu Kyi’s use of tropes heavily influences public perception of her.

La correspondance transatlantique des Prize Papers (1744-1763) : famille, commerce et communications en temps de guerre

Au cours des années comprises entre 1744 et 1763, le monde atlantique connaît deux guerres majeures. La France et ses colonies américaines sont impliquées dans une lutte acharnée contre l’Empire britannique. Les deux puissances rivales misent alors largement sur la guerre de course. Au-delà des conséquences commerciales et militaires évidentes, la guerre de course perturba aussi les communications transatlantiques. En effet, la prise d’un navire signifiait le plus souvent la perte des lettres qu’il transportait ou, dans le meilleur des cas, leur saisie, interrompant du même coup l’acheminement maritime du courrier. Les archives des Prize Papers, conservant des lettres et autres documents saisis par des corsaires britanniques, incarnent bien cette réalité. On y trouve notamment de nombreuses lettres françaises interceptées dans ces circonstances. Considérant que l’intérêt historique de ce fonds d’archives mérite d’être davantage souligné, cette recherche analysera donc certaines de ces correspondances françaises trouvées au sein des Prize Papers. À travers celles-ci, nous tenterons de reconstituer le système de communication transatlantique français et de ses acteurs en temps de guerre. Nous nous pencherons d’abord sur l’histoire de la constitution de ce fonds d’archives bien particulier, avant de présenter les pratiques de communication employées par les correspondants transatlantiques et les façons dont la guerre a pu les affecter. Il sera ensuite question des informations elles-mêmes qui furent véhiculées dans ce contexte si difficile. / Between 1744 and 1763, two major wars shook the Atlantic world. France and her colonies struggled against the British empire. Both belligerents made considerable use of privateers. Beyond its obvious commercial and military consequences, this form of maritime warfare disrupted transatlantic communications. The letters carried by a prize were often lost or, at best, were confiscated, interrupting postal communications. The Prize Papers archives, which contain letters and other documents confiscated by British privateers, embody this reality. The collection includes numerous intercepted French letters. Many of them will be analyzed here, by way of showing the potential of this collection for the study of the French transatlantic communications system and its main actors in wartime. The constitution of this very special archive is presented first. Correspondents’ transatlantic communications practices as they evolved in wartime are analyzed next. Finally, the types of information transmitted in this difficult context are the subject of the last part of the thesis.

Nosso projeto consiste na discussão sobre a literatura e cultura contemporânea na Rússia, especialmente a questão do Pós-modernismo, por um recorte específico: a análise das obras ganhadoras do concurso Russian Booker Prize com enfoque no processo de classificação adotado pelos júris , bem como um apanhado geral da premiação na Rússia. Para tal, analisaremos, em um primeiro momento, os pensadores e críticos mais representativos dessa prosa alternativa ou pós-moderna, sempre com um embasamento histórico. Entender o indivíduo pós-soviético será de fundamental importância para lidar com os temas e personagens dos romances vencedores. Em um segundo momento, nosso trabalho será o de investigar os critérios utilizados pela banca, a fim de tentar encontrar (se houver) algum ponto comum entre algumas das obras ganhadoras. Pensar em tais pontos será de suma importância para compreender os sistemas de valores dos críticos envolvidos nesse processo de premiação, além de colaborar para o enriquecimento de uma nova abordagem analítica da nova literatura russa e de alguns de seus nomes mais prolíficos. / Our project consists in the discussion of contemporary literature and culture in Russia, especially the question of post-modernism, by a specific focus: the analysis of the winning works of the Russian Booker Prize contest focusing on the process of classification adopted by the judges as well as a overview of the awards in Russia. To this end, we will analyze, at first, critical thinkers and most representative of this \"prose alternative\" or post-modern, always with a historical foundation. Understanding the individual post-Soviet is of fundamental importance to deal with the issues and characters of novels winners. In a second step, our work is to investigate the criteria used by banks in order to try to find (if any) a common point among some of the winning works. Think about these points will be extremely important to understand the value systems of the critics involved in the awards process, and contribute to the enrichment of a new analytical approach of the new Russian literature and some of its most prolific names.
