Com isso, pode-se promover a multiplicação rápida e geneticamente confiável, bem como a preservação de espécies ameaçadas de extinção. O objetivo do presente trabalho foi estabelecer um protocolo para a propagação e conservação in vitro e ex situ de Vriesea sucrei, visando à implantação de um banco de germoplasma dessa Bromeliaceae ameaçada de extinção. Sementes de V. sucrei foram inoculadas em meios de cultura e substratos in vitro. Foram testados sete diferentes meios de germinação: MS líquido, MS gelificado, K líquido, K gelificado, Areia lavada, fibra de coco e controle (água destilada). O índice de velocidade de germinação (IVG), porcentagem de germinação e de sobrevivência foram calculados. Também foram realizadas medidas de crescimento inicial como altura e número de folhas das plântulas germinadas. Plântulas provenientes da germinação in vitro foram submetidas aos tratamentos de conservação, que consistiram de uma combinação fatorial (2x3x2x2) das seguintes variáveis fisiológicas: formulação do meio de cultura (MS e K), concentração de macro e micronutrientes do meio de cultura (1, ½ e ¼), estado físico do meio de cultura (gelificado e líquido) e temperatura de incubação (15°C e 25°C). Após 120 dias em cultura, foram real izadas medidas de porcentagem de sobrevivência, incremento em altura, número de folhas, número de raízes, comprimento da maior raiz, teores de clorofila e análise anatômica do limbo. Plântulas de V. sucrei com seis meses de idade, obtidas por meio da germinação in vitro, foram utilizadas como explantes para a indução de brotações laterais. Os tratamentos de indução de brotações laterais consistiram de uma combinação fatorial (2x2x4) de dois meios de cultura (MS e K), dois estados físicos do meio (gelificado e liquido) e quatro combinações de auxinas (AIA e ANA), citocininas (cinetina e BAP), e uma amina (adenina) em diferentes proporções. A eficiência da multiplicação das brotações laterais foi calculada pela porcentagem de plântulas que geraram brotações laterais em cada tratamento e número médio de brotações produzidas por plântula. A germinação in vitro de sementes de V. sucrei nos tratamentos MS líquido, K líquido, areia e controle se mostram igualmente efetivos, apresentando altas taxas de germinação, sobrevivência e IVG. Os tratamentos contendo meio nutritivo (MS líquido e K líquido) seriam os mais indicados para a obtenção de plantas com crescimento inicial mais vigoroso em altura e número de folhas. O crescimento naturalmente lento da espécie V. sucrei favoreceu sua conservação em bancos de germoplasma in vitro. A baixa temperatura de 15°C foi o fator determinante para a redução do crescimento in vitro, sem, contudo, comprometer a sobrevivência das plantas. O tratamento ½ MS liquido a 15°C apresentou alta porcentagem de sobrevivência, plantas com crescimento reduzido, altos teores de clorofila e estrutura anatômica sem anormalidades. Com base nestes resultados, podemos sugerir que esse tratamento seja o mais indicado para a implantação de bancos de germoplasma in vitro de V. sucrei. Quanto ao experimento de indução de brotações laterais, o tratamento MS liquido suplementado com 0,5mg/L de ANA e 2mg/L de BAP promoveu maior porcentagem de plantas com brotações laterais (80%) e maior número de brotações laterais/planta (2,3). No entanto, todos os tratamentos testados geraram um número baixo de brotações laterais/planta.

Differentiating Habits for Pro-Environmental Behaviours

To help protect the environment people need to change current non-environmental behaviours into more sustainable behaviours. By understanding habits for pro-environmental behaviours, people can strive toward building a more sustainable future. The goal of this thesis was to identify different types of repeated pro-environmental behaviours and how to best foster strong habits for pro-environmental behaviours. To achieve this purpose we combined the theoretical framework of habits (Bargh, 1994; Verplanken, 2006) with the theoretical framework of self-determination theory (SDT; Deci & Ryan, 1985; 2000) to address current gaps in the habit literature. The present thesis is comprised of two articles. The first article determined if the proposed indicators of habits (i.e., behaviour frequency, habit strength, and behaviour interference) could identify different patterns of repeated pro-environmental behaviours predicted by habit and self-determination theory using cluster analysis. The three studies provided support for the three proposed types of repeated behaviours: weak habits, repeated behaviours with interference, and strong habits. These results were very robust since the same pattern was found across three studies, three samples, and three different target behaviours. The second article used the groups (i.e., weak habit, repeated behaviour with interference, and strong habit) in exploratory multinomial logistic regression analyses to identify factors related to pro-environmental behaviours which distinguished between the different types of repeated behaviours. Once again, the same three types of repeated behaviours were found across these three studies, with three new samples and two different target behaviours. Three predictors fairly consistently identified differences between the groups: autonomous motivation toward the environment, perceived importance of the environment, and the frequency of other pro-environmental behaviours. The implications of the thesis findings are discussed in relation to self-determination theory and the study of habits.

In this diploma thesis Union for the Mediterranean. From Sarkozy's ambition to European reality I follow an interesting case of one foreign policy. A presidential candidate Nicolas Sarkozy suggested within his pre-election campaign to set up Mediterranean with ambitious goals; when he became president he began to realize this initiative. I argue, that his motivation was not to primarily solve the regional issues or/and integrate the Mediterranean a little bit more, but through this activity to attract media attention. It corresponds to the theory of mediatization which states that politicians hold a permanent campaign; and to attract publicity they adapt to media as much as they can. Instead of following the political logic they follow media logic. Nicolas Sarkozy has usually done so in terms of national/domestic politics, however what I show is the fact that a foreign policy can also be lead by the permanent campaign - or to be more precise that politicians are trying to appeal on the national audience through the permanent campaign, and on public abroad through public diplomacy, it is practically the same thing. This study begins to fill the gap in sociological literature on movements and countermovements by exploring the dynamic environment of two movement organizations. After documenting the climate of public opinion on abortion, it investigates the strategy and tactics employed by a movement to maintain that opinion and a countermovement to reverse that opinion. It relates social movements to their social environments, social change, opposition, and strategy and tactics. It illustrates the efficacy of single-issue groups in the American social and political environment. Finally, this thesis demonstrates the validity of exploratory studies by uncovering elements of social movements and countermovements that had not been previously investigated.

Hartwell, D.D.S., M.S.Chairman and Professor, Department of EndodonticsIt was the aim of this study to investigate the incidence of file breakage and distortion when the Pro Taper, K3 Endo and ProFile systems were used to instrument canals in the severely curved roots of extracted molars.Forty-five roots of extracted mandibular and maxillary molars with curvatures between 40 and 75 degrees were chosen for use in this study. The roots in Group One were instrumented with the Pro File (PF) system and served as the control group. Roots in Group Two were instrumented with the Pro Taper (PT) system and those in Group Three were instrumented with the K3 Endo (K3) system. The three systems were used according to the manufacturers' instructions.The proportion of files distorted was 15.3% for the PF group, 2.4% for the PT group, and 8.3% for the K3 group. There was a statistically significant difference between the PF and PT groups (p = .0079). The percentage of broken files was 1.7% for the PF group, 6.0% for the PT group and 2.1% for the K3 group. No statistically significant differences were found between these three groups (p = .4243). The results of this study showed that these three rotary tapered systems were not significantly different with regard to breakage. There were significantly more distorted files in the PF group when compared to the PT group, with regard to distortion there was no significant difference between the PT and K3 and the PF and K3 groups.

The perception between the pro-Islamic and pro-Kurdish political streams in Turkey, 1980-2011

Official state ideology (Kemalism) whose core principles are Turkish nationalism and secularism excluded Kurds and Islamists from the newly formed social and political structure of modern Turkey. By insisting on a Kemalist modernization project in which assimilation of Kurdish ethnic identity within Turkish nationalism, and a top down imposition of secular policies on public and state affairs, Turkey has had to deal with two profound issues—the Kurdish question, and political Islam. As these social and political rivals of official state ideology present an alternative way of modernization projects, the Kemalist state apparatus has until recently considered their existence and development within Turkish society as a menace to their core existence. That is why the nearly century-long Kurdish question and Islamism (reactionism-gericilik) have occupied the agenda. The striking point of this reality of Turkey is that while Kurdish nationalist and Islamist social and political groups, whether they are legal or illegal, have sought to supersede the official state ideology with their understandings, they have never united or operated joint activities against their, roughly speaking, enemy. The research, because of this focuses on the reasons why these two social and political groups in Turkey did not work together to eliminate their common rival. Regarding pro-Kurdish and pro-Islamic groups, ideological distinctiveness and rigidity led them to consider each other as a part of or as an extension of official state ideology which tries to eliminate its rivals. Despite the fact that they both ideologically and practically confirmed the existence of repression towards Kurds and religious people, the leftist-oriented pro-Kurdish political stream considered religion as a component of denial and assimilationist policy of official state ideology, so that they did not differentiate between Islamist groups and the established state structure, whereas the pro-Islamic political stream refused to co-operate with any member of the leftist-oriented pro-Kurdish stream because of its secular characteristic. The reaction of these groups towards assimilation of Kurdish ethnic origin, and repression over religion, initially embodied itself in several uprisings and revolts in the Kurdish populated eastern areas of Turkey, and religiously sensitive cities, respectively. Thanks to the multi-party system, these groups have found a way in which they can express themselves through political parties, labour unions, associations and foundations so that they have become social movements forcing the Kemalism dominated state to meet their demands. As long as these demands are not met, the interaction between these two social movements and the official state ideology has been hostile. Within this process, the hostile approach to their common enemy reflected in each other’s activities and understandings as they see each other as a part of official state ideology. In the next phase in which their transformation from social movements to a legal political stream competed, the inherited ideological rigidity between pro-Kurdish and pro-Islamic political streams preserved itself. Overall, the research will indicate that since the mid-20th century, ideological barriers between pro-Kurdish and pro-Islamic streams have become the fundamental determinant of how they perceive each other.
