Probiotics may represent an alternative prophylactic disease control, replacing the use of antibiotics. In this respect, this study isolated, tested and identified potential probiotic bacteria from the gut of cobia, Rachycentron canadum, a potential candidate for marine aquaculture. 40 animals were captured, 10 from a private hatchery and 30 from an offshore culture system (PE, Brazil) between November 2010 and July 2011. Fishes from the hatchery had weight of 139.30 ± 31.52 g and length of 27.13 ± 1.46 cm, while those from offshore culture system weighed 456.77 ± 264.46 g and had length of 37.29 ± 6.05 cm. 45 bacterial were isolated and tested in vitro against five known pathogenic species, Aeromonas hydrophila (IOC/FDA 110-36), Pseudomonas aeruginosa (ATCC 15442), Streptococcus agalactiae (ATCC 13813), Vibrio parahaemolyticus (ATCC 17802) and Vibrio vulnificus (ATCC 27562). Fifteen strains (33.33%) had antibacterial activity to at least one pathogen, while eight (17.77%) were inhibited all pathogens tested. There was significant difference (P<0.05) between percentage of potential probiotic obtained from different seasons, rainy and dry. Strains that presented the best results antagonism test in vitro were identified as Lactobacillus plantarum, Bacillus coagulans, Klebsiella spp., Bacillus circulans, Lactococcus lactis subsp. lactis and Bacillus firmus. The largest inhibition zone observed in the antagonism test was produced by B. circulans against V. vulnificus. Some potential probiotic species identified were characterized by others authors, but isolated from the intestine of other fish species. It is suggested in vivo antagonism tests are performed to prove the effectiveness of these bacteria as probiotic to cobia. / O uso indiscriminado de drogas antimicrobianas na aquicultura pode contribuir para o desenvolvimento de bactérias resistentes aos medicamentos, o que torna esses micro-organismos mais difíceis de serem controlados e eliminados. Probióticos podem representar uma alternativa profilática no controle de doenças, em substituição ao uso de antibióticos. Neste contexto, o estudo foi realizado com o objetivo de isolar, testar e identificar bactérias com potencial probiótico do beijupirá, Rachycentron canadum, potencial candidato para a piscicultura marinha. Foram coletados 40 animais, 10 na fase de berçário e 30 na fase de engorda em sistema offshore (PE, Brasil), no período de novembro de 2010 a julho de 2011. Os peixes coletados do berçário apresentaram peso de 139,30 ± 31,52 g e comprimento de 27,13 ± 1,46 cm, enquanto os animais provenientes do sistema offshore apresentaram peso de 456,77 ± 264,46 g e comprimento de 37,29 ± 6,05 cm. Foram obtidos 45 isolados bacterianos testados in vitro frente a cinco espécies patogênicas conhecidas, Aeromonas hydrophila (IOC/FDA 110-36), Pseudomonas aeruginosa (ATCC 15442); Streptococcus agalactiae (ATCC 13813); Vibrio parahaemolyticus (ATCC 17802) e Vibrio vulnificus (ATCC 27562). Quinze isolados (33,33%) apresentaram atividade antibacteriana a pelo menos um patógeno e oito (17,77%) inibiram todos os patógenos testados. Houve diferença significativa (P<0,05) na proporção de isolados potenciais probióticos obtidos nas distintas épocas do ano, chuvosa e seca. Os isolados que apresentaram os melhores resultados no teste de antagonismo in vitro foram identificados como Lactobacillus plantarum, Bacillus coagulans, Klebsiella spp., Bacillus circulans, Lactococcus lactis subsp. lactis e Bacillus firmus. O maior halo de inibição observado no teste de antagonismo foi produzido por B. circulans frente ao V. vulnificus. Algumas espécies potenciais probióticas identificadas já foram caracterizadas por outros autores, porém isoladas de intestino de outras espécies de peixes. Sugere-se a realização de testes de antagonismo in vivo para que seja comprovada a efetividade das bactérias como probióticas para o beijupirá.

When isolated from the host itself is safe to use, being able to adhere and colonize the intestine. The objective was to isolate probiotic bacteria from intestine of juvenile Litopenaeus vannamei and evaluate the effects of probiotic administration in the performance of cultivated animals experimental challenged infection. Intestinal bacteria were from cultivated shrimp in oligohaline water and saltwalter in dry and rainy seasons, and confronted with Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio alginolyticus and Aeromonas hydrophila. Bacteria showed antimicrobial effect have been identified by molecular biology. Twenty-nine isolates showed antagonistic effect at least one of the tested pathogens. The species that most occurred was Bacillus cereus, the producing of the largest zones of inhibition to the V. alginolyticus and V. vulnificus. The bacterium Citrobacter freundii, from oligohaline water showed the best antimicrobial effect before the four pathogens. The probiotic Bacillus cereus was tested in diet for marine shrimp post-larvae against challenge with V. parahaemolyticus, and V. alginolyticus which evaluated the growth performance of animals, colonization capacity of the probiotic bacteria, pathogens count and histopathological lesions. The use of probiotic had no effect on animals survival, but not for treatments which was used probiotic had a lower weight gain. Animals fed dietary supplementation of probiotic had lower pathogens count those fed without the use. Histopathological lesions were observed in organs and tissues of animals. It can be concluded that it was possible to isolate bacteria having probiotic effect of marine shrimp intestine, wherein the strain Bacillus cereus demonstrated high capacity to colonize the host itself, causing a significant reduction of pathogens. / Bactérias com efeito probiótico contribuem para saúde de camarões marinhos em cultivo, em substituição aos antibióticos, exercendo ação antagonista à micro-organismos patogênicos ou por competição por espaço e nutrientes, além de melhorar o apetite levando a um maior crescimento dos animais. Quando isolados do próprio camarão seu uso é seguro, por serem capazes de aderir ou colonizar o intestino destes. Neste sentido, objetivou-se isolar bactérias probióticas do intestino de juvenis de Litopenaeus vannamei e avaliar os efeitos da sua administração sobre o desempenho dos animais cultivados frente infecção experimental. Foram isoladas bactérias do intestino de camarão cultivado em águas oligohalina e salgada, em períodos seco e chuvoso, e confrontadas com Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio alginolyticus e Aeromonas hydrophila, em testes in vitro. As bactérias que produziram efeito antimicrobiano foram identificadas através de biologia molecular.Vinte e nove isolados apresentaram efeito antagônico a pelo menos um dos patógenos testados. A espécie que mais ocorreu foi Bacillus cereus, produzindo os maiores halos de inibição frente ao V. alginolyticus e V. vulnificus. A bactéria Citrobacter freundii, proveniente de água oligohalina apresentou melhor efeito antimicrobiano, perante os quatro patógenos. O probiótico Bacillus cereus foi testado em ração para pós-larvas de L. vannamei, frente a desafio com V. parahaemolyticus, e V. alginolyticus quando avaliou-se o desempenho zootécnico dos animais, a capacidade de colonização das bactérias probióticas, contagem de patógenos e lesões histopatológicas. O uso do probiótico não influenciou nas taxas de sobrevivência dos animais, porém nos tratamentos em que não se utilizou probiótico houve menor ganho de peso. Os animais que receberam ração suplementada de probiótico, tiveram contagem de patógenos inferior aqueles alimentados sem o uso. Não foram observadas lesões histopatológicas nos órgãos e tecidos dos animais. Concluiu-se que é possível isolar bactérias com efeito probiótico do intestino do camarão marinho, sendo que a cepa de Bacillus cereus demonstrou maior capacidade de colonizar o próprio hospedeiro, diminuindo os patógenos.

Os antibióticos têm sido utilizados como aditivos na alimentação animal para aumentar o desempenho e manter a saúde dos animais, como suínos e frangos, destinadas à produção de alimentos de origem animal. No entanto, desde 2006, a Comunidade Europeia proibiu o uso de antibióticos para esse proposito devido ao desenvolvimento de resistência bacteriana aos antibióticos. Como resultado, várias alternativas foram estudadas e propostas para substituir os antibióticos utilizados na alimentação animal. Os óleos essenciais têm recebido considerável atenção devido às suas propriedades antimicrobianas. Portanto, o objetivo do presente trabalho foi avaliar in vitro a atividade antibacteriana dos óleos essenciais contra a microbiota patogênica e probiótica de ocorrência no trato gastrointestinal de suínos e aves, destinadas à produção de alimentos de origem animal. A atividade antibacteriana seletiva, a qual, significou uma alta atividade antibacteriana contra bactérias patogênicas e reduzida ou nenhuma atividade sobre bactérias probióticas, foi avaliada como característica fundamental dos óleos essenciais com alto desempenho. Esta característica foi avaliada nos óleos essenciais usados individualmente e em combinações binarias. Inicialmente, no Capítulo 2, uma triagem de vinte e oito óleos essenciais (OEs) através do método de difusão em disco mostrou que Eucalyptusglobulus, E. exserta, Pimenta pseudocaryophylllus, Orange Oil Phase Essence, e CitrusTerpens (Os dois últimos OEs foram subprodutos do processamento da laranja para obtenção de suco) tiveram uma atividade antibacteriana seletiva sobre a bactéria patogênica Salmonella Enteritidis e a bactéria probiótica Lactobacillus plantarum. Numa fase posterior, esses cinco óleos foram avaliados individualmente e em misturas binárias, contra cinco bactérias patogênicas e três bactérias probióticas. Os melhores resultados foram observados quando os OEs foram avaliados isoladamente e não em misturas. Assim, Orange Oil Phase Essence e Citrus Terpens destacaram-se por ter a melhor atividade antibacteriana seletiva contra essas bactérias. No Capítulo 3, uma análise mais detalhada da atividade antibacteriana dos óleos essenciais foi realizada utilizando Orange Oil Phase Essence e a mistura composta pelos óleos de E. globulus e P. pseudocaryophyllus. Estes dois óleos foram selecionados com base nos resultados do Capitulo 2 e da disponibilidade de OEs em nosso estoque. Ambos, óleo e a mistura foram avaliados sobre a bactéria patogênica mais resistente, E. faecalis, e a bactéria probiótica menos resistente L. rhamnosus, como observado no Capitulo 2. A avaliação da Concentração Inibitória Mínima e Concentração Bactericida Mínima do Orange Oil Phase Essence e da mistura mostrou que não tiveram um efeito antibacteriano seletivo sobre E. faecalis e L. rhamnosus. Finalmente, no Capitulo 4, foi avaliada a atividade antibacteriana individual e combinada dos óleos de E. globulus e P. pseudocaryophyllus sobre E. faecalis e L. rhmanosus. Os resultados mostraram que a combinação destes dois OEs, avaliadas pelo método checkerboard, não potencializou a atividade antibacteriana seletiva dos dois OEs. Portanto, observou-se que o óleo de E. globulus isoladamente apresentou a melhor atividade antibacteriana seletiva contra E. faecalis e L. rhamnosus. Em conclusão, este trabalho permitiu identificar óleos essenciais com perfil antibacteriano seletivo para eles serem possíveis alternativas botânicas aos antibióticos utilizados na alimentação animal. / Antibiotics have been used in animal feed to maintain health and increase performance, as in the case of pigs and poultry intended for food production of animal origin. However, since 2006 the European Community has banned the use of antibiotics for this purpose due to the emergence and increase of antibiotic-resistant bacteria. As a result, several alternatives have been studied and proposed to substitute antibiotics used in animal feed. Essential oils have received considerable attention due to their antimicrobial properties. Therefore, the objective of this work was to evaluate in vitro the antibacterial activity of essential oils against pathogenic and probiotic bacteria that occur in the gastrointestinal tract of swine and poultry, intended for food production of animal origin. The selective antibacterial activity, which means high antibacterial activity on pathogenic bacteria and reduced or no activity on probiotic bacteria, was evaluated as a fundamental feature of the highest-performance essential oils. This feature was evaluated in essential oils used individually and in binary combinations. Initially, in Chapter 2, the screening of twenty-eight essential oils (EOs) by disk diffusion method showed that Eucalyptus globulus, E. exserta, Pimenta pseudocaryophylllus, Orange Oil Phase Essence, and Citrus Terpens (the last two EOs were by-products of orange juice production) had a selective antibacterial activity against the pathogenic Salmonella Enteritidis and probiotic Lactobacillus plantarum. At a later stage those five oils were evaluated, individually and in binary blends, against five pathogenic bacteria and three probiotic bacteria. Better results were observed when the EOs were checked alone and not in blends. Orange Oil Phase Essence and Citrus Terpens stood out for having the two best selective antibacterial activities against those bacteria. In Chapter 3, a more detailed analysis of essential oil antibacterial activities was perfomed using Orange Oil Phase Essence and the blend composed of E. globulus and P. pseudocaryophyllus. These two oils were selected based on the results of Chapter 2 and from the availability of our EO stock. Both oil and blend were checked on the most resistant pathogenic bacterium, E. faecalis, and on the less resistant probiotic bacterium of the Lactobacillus genus, L. rhamnosus, as observed in Chapter 2. The evaluation of Minimal Inhibitory Concentration and Minimal Bactericidal Concentration for Orange Oil Phase Essence and the blend showed that there was not a selective antibacterial effect against E. faecalis and L. rhamnosus. Finally, in Chapter 4, the individual and combined antibacterial activities of E. globulus and P. pseudocaryophyllus essential oils on E. faecalis and L. rhmanosus were evaluated. The results showed that the combination of two EOs evaluated by checkerboard method did not potentiate the selective antibacterial activity of the two EOs. Therefore, it was observed that the E. globulus essential oil alone had the best selective antibacterial activity against E. faecalis and L. rhamnosus. In conclusion, this work enabled the identification of essential oils with selective antibacterial profile that can become possible botanical alternatives to antibiotics used in animal feed.

In etlichen Studien konnte die gesundheitsfördernde Wirkung von probiotischen Bakterien nachgewiesen werden. Heutzutage werden diese zahlreich in der Nahrungsmittel- und Kosmetikindustrie verwendet, um das Immunsystem des Menschen positiv zu beeinflussen. Probiotische Bakterien oder deren Extrakte können zum Beispiel die Produktion von Zytokinen anregen oder herabsetzen und dadurch Entzündungsreaktionen verhindern oder die körpereigene Abwehr steigern. Auch die Produktion des humanen Beta-Defensins-2 (hBD-2) kann durch probiotische Bakterien verändert werden. Dieses wirkt im menschlichen Körper wie ein körpereigenes Antibiotikum und wird von verschiedenen Zellen kontinuierlich gebildet. Gerade in entzündetem Gewebe unterliegt es einer gesteigerten Produktion. Inwieweit sich die hBD-2-Produktion in oralen Keratinozyten der Zelllinie OKF6/hTERT-2 verändert, während diese mit den Extrakten der probiotischen Bakterien Lactobacillus salivarius (L.s.) und Lactobacillus rhamnosus GG (L.GG.) inkubiert wurden, war das Ziel dieser Arbeit. Die Ergebnisse zeigen deutlich eine verringerte hBD-2-Produktion der oralen Keratinozyten nach der Stimulation mit den Bakterienextrakten zu verschiedenen Konzentrationen und Zeiten. Ein günstiger Therapieansatz durch die Gabe von probiotischen Bakterienextrakten bei entzündlichen Erkrankungen der Mundschleimhaut, wäre durch eine gesteigerte hBD-2-Produktion denkbar. Dieses bekämpft nämlich pathogene Erreger und unterstützt somit die Immunabwehr. Mit den Bakterienextrakten von L.s. und L.GG. ließ sich die Produktion von hBD-2 in den oralen Keratinozyten nicht steigern. Einen anti-inflammatorischen Effekt der Extrakte auf die Zellen ist dennoch nicht auszuschließen, da die hBD-2-Konzentration auch indirekt als Indikator einer Entzündung angesehen werden kann und hier mit zunehmender Bakterienextraktkonzentration sinkt. Jedoch sind andere Ursachen für den Konzentrationsabfall ebenso denkbar.

Charakterizace adsorbčních vlastností probiotických bakterií / Characterization of adsorption properties of probiotic bacteria

The aim of the diploma thesis is to characterize the adsorption properties of probiotic bacteria. Characterization was performed using of designed and optimized spectrophotometric method of determination of adhesion of probiotic bacteria to mucin as the main viscoelastic component of mucus. In order to provide a more detailed description of the adhesion interactions involved in the adhesion of probiotic bacteria to mucin, the viability of selected probiotic bacterial strains Lactobacillus rhamnosus CCM 1825, Lactobacillus plantarum CCM 7039, Lactobacillus delbrueckii subsp. bulgaricus CCM 7190, Lactobacillus acidophilus CCM 4833, Lactobacillus casei CCM 4798, Bifidobacterium breve CCM 7825, Bacillus coagulans CCM 2658 and a potential probiotic strain of Lactobacillus zeae CCM 7069. Moreover, bacterial adhesion to carbohydrates was also determined for all the bacteria tested. The last proposed and optimized technique was dynamic and electrophoretic light scattering characterizing surface properties such as -potential, average size distribution and isoelectric point. From this method, information was obtained on aggregation of bacterial cells and on electrostatic interactions. The combination of these methods was used as a complex tool to characterize adhesion of the tested bacterial cultures as a very specific, sensitive and key parameter of a successful probiotic microorganism that is influence by the multiple effects.

Využití různých technik enkapsulace k řízenému uvolňování aktivních látek v potravinářských a kosmetických přípravcích / Use of some encapsulation techniques to controlled release of active substances in food and cosmetics products.

The presented doctoral thesis is focused on preparation, characterization and application of organic micro- and nanoparticles as transport systems for active components and some their complex natural sources. Active component were packed into liposomes and polysaccharide particles. As active components were used caffeine, some drugs – clotrimazole and ibuprofen, further antioxidants and vitamins. Antimicrobial herbs and spices extract, antimicrobial peptides lysozyme, nisin and other antimicrobial ingredients were encapsulated too. Encapsulation of selected hydrolytic enzymes was tested, too. Particles were also used for encapsulation of probiotic strains Bifidobacterium breve and Lactobacillus acidophilus and prebiotic components. These prebiotics were co-encapsulated into capsules with probiotic cells. Natural extracts were encapsulated e.g. extracts of guarana, ginseng, goji, green barley, propolis, black, green and white tea, coffee, fruit and vegetable extracts. The efficiency of encapsulation was determined by HPLC/PDA and by spectrophotometry. Long-term stability of particles and amount of released component in model/real foods, in model cosmetic conditions and in a model physiological environment were monitored too. Size of prepared liposomes and polysaccharide particles was determined by dynamic light scattering and by light microscopy and electron microscopy, respectively. Stability of the particles was measured using a zeta potential. Also, analytical centrifugation was used to measurement of sedimentation velocity and stability of the prepared particles. The antimicrobial activity were tested using two Gram-positive (Bacillus subtilis, Micrococcus luteus), two Gram-negative (Escherichia coli, Serratia marcescens) bacteria and one fungal strains (Candida glabrata). For determining the antimicrobial properties of active component and prepared particles two the most widely used methods were used - agar diffusion method and broth dilution method. The viability of probiotic strains were performed using flow cytometry and fluorescence microscopy. Encapsulation of active component was successful in all types of particles. Liposome showed a very good long-term stability mainly in water conditions with neutral pH and polysaccharide particles were stable in acidic conditions. Prepared particles showed a very good stability in model stomach environment, while in model intestines environments particles were disintegrated and active component were released. Prepared particles with encapsulated caffeine as well as other tested antioxidants and vitamins could be used to modern types of energy drinks, food supplements and also for some cosmetics applications. Encapsulated antimicrobial components could be used for food application as well as for cosmetics and pharmaceutical application like antimicrobial wound formulation. Encapsulated enzymes can be used for controlled release of proteases in wound healing, as delivery systems in digestive tract and as a part of pharmaceutical preparative and food supplements for enzyme therapy. The study revealed that encapsulation of probiotics and also co-encapsulation of probiotics with prebiotics exhibited longer stability of particles and survival bacterial cells. So, prepared particles are suitable for use to food product with beneficial effects on the human body.
