La séparation du CO2 d'un mélange de gaz par cristallisation d'hydrates de gaz est un procédé qui pourrait à terme présenter une alternative intéressante aux techniques conventionnelles de capture du CO2. L'objectif de cette thèse était d'évaluer le potentiel de ce procédé "hydrates" pour séparer le CO2 d'un mélange CO2-CH4 riche en CO2. Nous avons étudié en particulier la sélectivité de la séparation vis-à-vis du CO2 et la cinétique de cristallisation des hydrates, ainsi que l'effet d'additifs thermodynamiques et cinétiques (et de certaines de leurs combinaisons) sur ces deux paramètres pour différentes conditions opératoires (pression, température, concentrations). Les expériences de formation/décomposition d’hydrates ont été réalisées en mode "batch" dans un réacteur haute pression faisant partie d'un pilote expérimental conçu et construit entièrement pendant cette thèse. Un modèle semi-empirique a été également développé pour estimer le taux de conversion de l’eau en hydrate et la composition des différentes phases en présence (hydrates, liquide et vapeur) à l'équilibre. Les résultats obtenus montrent que l'association du sodium dodécyl sulfate (SDS), utilisé en tant que promoteur cinétique, avec du tétrahydrofurane (THF), utilisé en tant que promoteur thermodynamique, permet d'obtenir des résultats intéressants en terme de quantité d'hydrates formés et de cinétique de formation. La sélectivité de la séparation vis-à-vis du CO2 reste cependant trop faible (en moyenne quatre molécules de CO2 piégées dans la structure de l'hydrate pour une de CH4) pour envisager d’utiliser ce procédé "hydrates" à plus grande échelle afin de séparer le CO2 de ce type de mélange de gaz. / The separation of CO2 from a gas mixture by crystallization of gas hydrates is a process that could eventually provide an attractive alternative to the conventional techniques used for CO2 capture. The aim of this thesis was to evaluate the potential of this "hydrate" process to separate CO2 from a CO2-CH4 gas mixture, rich in CO2. We have studied in particular the selectivity of the separation toward CO2 and the hydrate crystallization kinetics. The effects of thermodynamic and kinetic additives (and some additive combinations) on these two parameters for different operating conditions (pressure, temperature, concentrations) were evaluated. Hydrate formation and dissociation experiments were performed in "batch mode” in a high pressure reactor, and with an experimental pilot rig designed and built entirely during this thesis. A semi-empirical model was also developed to estimate the water to hydrate conversion and the composition of the different phases (hydrates, liquid and vapor) at equilibrium. The results show that the combination of sodium dodecyl sulfate (SDS) used as a kinetic promoter, with tetrahydrofuran (THF) used as a thermodynamic promoter, provides interesting results in terms of both the amount of hydrates formed and the hydrate formation kinetics. The selectivity of the separation toward CO2 remains too low (an average of four CO2 molecules trapped in the hydrate structure for one of CH4) to consider using this "hydrate" process on a larger scale to separate CO2 from such a gas mixture.

Com isso, o crescimento, produtividade e exportação de nutrientes da cultura do trigo podem ser maiores, porém a resposta a esta inoculacao pode variar conforme a afinidade da bacteria com os cultivares de trigo. Dessa forma, objetivou-se avaliar o acúmulo de matéria seca e extração de nutrientes em estádios fenológicos, exportação de nutrientes pelos grãos, componentes de produção e produtividade de grãos de cultivares de trigo inoculados ou não com Azospirillum brasilense. O experimento foi desenvolvido em Selvíria - MS, em um Latossolo Vermelho Distroférrico de textura argilosa, em sistema plantio direto. O delineamento experimental foi em blocos ao acaso com quatro repetições, com os tratamentos dispostos em esquema fatorial 3 x 2, sendo três cultivares de trigo (CD 116, IPR CATUARA TM e IAC 385), com ou sem inoculação de sementes por Azospirillum brasilense (300 ml ha-1 do produto, com estirpes Abv5 e Abv6 (garantia de 2x108 UFC mL-1). Procedeu-se também a análise de regressão em esquema de parcela subdividida com quatro repetições, em que as parcelas foram constituídas pelos seis tratamentos descritos acima, e as subparcelas no tempo, por sete épocas de coletas de plantas (antes do afilhamento, afilhamento e antes da adubação nitrogenada de cobertura, folha bandeira, emborrachamento, florescimento, grão pastoso e maturação fisiológica), para o acúmulo de matéria seca e extração de nutrientes. A inoculação com Azospirillum brasilense propiciou maior índice de clorofila foliar (ICF) e acúmulo de matéria seca de raízes, independentemente do cultivar de trigo. Os cultivares de trigo extraíram quantidades totais de nutrientes semelhantes, porém a extração por tonelada de grãos produzida foi diferenciada para K, Mg, S, B, Cu, Mn e Zn. A extração de nutrientes total ou por tonelada de grãos produzida em ordem decrescente foi de N>K>P>S>Ca>Mg>Fe>Mn>Zn≥B>Cu, com ou sem a inoculação com Azospirillum brasilense. As maiores exportações totais de K, Ca, Mg, S e Mn foram obtidas sem a inoculação com Azospirillum brasilense, entretanto, esta bactéria diazotrófica proporcionou maiores exportações de N, P, K, Ca e Mg por tonelada de grãos produzida e de Zn total. As maiores exportações relativas (acima de 50% do que é extraído) foram de Zn, Cu, P, N e B e de Mg apenas com a inoculação. O cultivar CD 116 foi o mais produtivo e que propiciou as maiores exportações totais de nutrientes, apesar da menor absorção de K, Mg, S, Cu, Mn e Zn por grãos produzido em relação aos demais cultivares. A inoculação com Azospirillum brasilense interfere de forma diferenciada nos cultivares de trigo, sendo que o cultivar CD 116 proporcionou a maior produtividade de grãos quando inoculado. / Wheat seeds inoculated with Azospirillum brasilense bacteria can provide biological nitrogen fixation (BNF), but has been observed more pronounced effect of this inoculation in the initial plant growth (phytohormonal effect), thereby, with the further development of root system, the nutrients and water uptake may be greater. Thus, the growth, productivity and nutrients removal from the wheat crop may be higher, but the response to this inoculation may vary according to the affinity of the bacterium with the wheat cultivars. The aim of this study was to evaluate the dry matter accumulation the accumulation of dry matter and nutrients uptake at different growth stages, nutrients export by grains, yield components and grains yield of wheat cultivars inoculated or not with Azospirillum brasilense. The experiment was conducted in Selvíria - MS, in a distroferric Oxisol clayey in no-till system. The experimental design was a randomized block with four replications, with the treatments arranged in a factorial 3 x 2, with three wheat cultivars (CD 116, IPR CATUARA TM and IAC 385) with or without seed inoculation by Azospirillum brasilense (300 ml ha-1 product with strains Abv5 Abv6 (2x108 guarantee UFC mL-1). Also proceeded regression analysis in a split plot scheme with four replications, where the plots were the six treatments described above, and the subplots, seven plants collections seasons (before of tillering, tillering and before nitrogen topdressing, leaf flag, booting, flowering, doughy grain and physiological maturity), for the accumulation of dry matter, extraction and export of nutrients. Azospirillum brasilense inoculation provided greater leaf chlorophyll index (LCI) and accumulation of roots dry matter, regardless of the wheat cultivar. The wheat cultivars uptake similar total amounts of nutrients, but the uptake per ton of grain produced was differentiated for K, Mg, S, B, Cu, Mn and Zn. Total nutrient extraction or per ton of grain produced in descending order was N>K>P>S>Ca>Mg>Fe>Mn>Zn>B>Cu, with or without inoculation with Azospirillum brasilense. The highest total removal of K, Ca, Mg, S and Mn were obtained without inoculation with Azospirillum brasilense; however, this diazotrophic bacterium provided higher removal of N, P, K, Ca and Mg per ton of grain produced and Zn total. The highest relative removal (above 50% that is uptake) was the Zn, Cu, P, N and B and Mg only with inoculation. The CD 116 cultivar was the most productive and provided the highest total nutrients removal, despite the lower uptake of K, Mg, S, Cu, Mn and Zn by grains produced in relation to the other cultivars. The inoculation with Azospirillum brasilense interferes differently in wheat cultivars, and cultivar CD 116 obtained the highest grain yield when inoculated.

Extratos vegetais como promotores do crescimento de leitões recém-desmamados / Herbal extracts as growth promoters of weanling pigs

Apesar da comprovada capacidade de melhorar o desempenho de suínos, o uso de antimicrobianos como promotores do crescimento vem sendo progressivamente restringido em diversos países. As novas regulamentações têm forçado a procura por alternativas ao uso de antibióticos e quimioterápicos como promotores do crescimento de suínos. Os extratos vegetais constituem uma dessas alternativas pesquisadas. Sendo assim, o objetivo do trabalho foi avaliar os efeitos de antimicrobianos e extratos vegetais sobre a digestibilidade aparente dos nutrientes, desempenho, morfometria dos órgãos, histologia do epitélio intestinal, microbiologia intestinal e frequência de diarréia de leitões recém-desmamados. Foram realizados três experimentos em blocos casualizados, nos quais foram testados, durante 35 dias, cinco tratamentos: controle - ração basal; antimicrobiano - basal com bacitracina de zinco, olaquindox e colistina (50 ppm de cada); extrato vegetal A, B e C - basal com 700, 1200 e 2100 ppm de extrato vegetal, respectivamente. O extrato vegetal foi constituído de quantidades iguais de óleo essencial de cravo, tomilho, orégano, eugenol e carvacrol. Foram utilizados 40 leitões em cada experimento, com idades iniciais e finais de, respectivamente, 21 e 56 dias. Foram alocados dois leitões (um macho castrado e uma fêmea) em cada baia (unidade experimental). No ensaio de digestibilidade (Experimento I) utilizou-se o método da coleta parcial de fezes, tendo o óxido crômico (Cr2O3) como marcador. Ao final do Experimento I, um animal de cada unidade experimental foi abatido para análise de morfometria dos órgãos e coleta de amostras do epitélio intestinal para análise histológica e microbiológica. Nos Experimentos I, II e III, foram medidas a frequência de diarréia e as variáveis de desempenho. A digestibilidade da matéria seca foi maior (P<0,05) para o tratamento com extratos vegetais em relação ao tratamento controle e com antimicrobianos. Durante os períodos de 1 a 14 e 1 a 35 dias de experimentação, o tratamento antimicrobiano apresentou melhores resultados (P<0,05) de peso vivo, consumo diário de ração e ganho diário de peso que os demais tratamentos. Dentre os níveis de extratos vegetais, o maior nível de inclusão foi o que proporcionou melhores resultados de desempenho. Na análise de morfometria dos órgãos, foram encontradas diferenças (P<0,05) entre os tratamentos apenas para o peso relativo do trato gastrintestinal total e do intestino delgado vazio, sendo que os melhores (menores) resultados foram obtidos para os animais suplementados com antimicrobianos. Para a histologia do epitélio intestinal, novamente os melhores resultados foram obtidos para o tratamento com antimicrobianos, tendo apresentado maior altura de vilosidade (P<0,05) e menor relação altura de vilosidade e profundidade de cripta do íleo que os tratamentos com extratos vegetais. Não foram encontradas diferenças significativas (P>0,05) para os resultados de microbiologia intestinal. A frequência de diarréia dos animais que receberam antimicrobianos foi inferior (P<0,05) àquela do tratamento controle ou extratos vegetais no período de 1 a 35 dias de experimentação, indicando eficácia dos antimicrobianos em controlar a diarréia. Os extratos vegetais apresentaram resultados intermediários entre o tratamento controle e com antimicrobianos para essa variável. / The use of antimicrobials as growth promoters of swine has been gradually restricted in many countries. The new regulations have forced the search for alternatives to the antibiotic use as growth promoters for swine. The herbal extracts are one of these alternatives. The purpose of this work was to evaluate the antimicrobial agents and herbal extracts as growth promoters, based on intestinal morphology, histology, microbiology, digestibility, fecal score, and on performance of weanling pigs. Three randomized complete block design experiments were carried out to compare five treatments: control - basal diet; antimicrobial - basal diet plus Zn bacitracin, olaquindox, and colistin (50 ppm of each); herbal extract A, B and C - basal diet plus 700 ppm, 1200 ppm and 2100 ppm of herbal extract, respectively. The herbal extract consisted of a mixture of equal amounts of essential oils of thyme, clove, oregano, eugenol and carvacrol. In each Experiment, forty 21-d-weaned pigs were allotted to 20 suspended pens, with two pigs (a castrated male and a female) per pen (experimental unit) and four replications per treatment. Digestibility assay (Experiment I) was conducted using chromic oxide (Cr2O3) as fecal marker. On 35th day of experimental period, an animal of each experimental unit was slaughtered for morphological analysis and samples of the intestine epithelium were collected for histological and microbiological analysis. Performance data and fecal score of pigs were collected from Experiments I, II and III. The herbal extracts increased (P<.05) the dry matter apparent digestibility of the diet compared to control and antimicrobial treatments. The antimicrobial agents improved (P<.05) body weight, feed intake and average daily gain of weanling pigs during 1 to 14 and 1 to 35 days of experimental period compared to control and herbal extract. Among all levels of herbal extract, higher levels showed better performance results. The antimicrobial treatment improved the relative weights of the intestinal tract and empty small intestine (P<.05) and improved ileum villus height and the ratio of ileum villus height:crypt depth of animals. There was no statistical difference (P>.05) in microbiological analysis. During 1 to 35 days of experimental period, incidence of diarrhoea of pigs fed antimicrobials was statisticaly lower (P<.05) than those fed control or herbal extract diet, suggesting the efficacy of antimicrobials in diarrhoea control. Pigs fed herbal extract showed intermediary results between antimicrobial and control diets for this variable.

Detecció de l'administració de 17(beta)-nandrolona en animals

17β-nandrolone (17βN) is illegally used in sport and in livestock as a growth promoter.17βN and some of their metabolites have been described as endogenous compounds in some animals, such as adult boars and entire male horses. The aim of this thesis was to identify markers of exogenous administration of 17βN in boars and horses. A method to quantify and identify metabolites of 17βN in the different metabolic fractions has been optimized and validated in boar and horse urine. The method has been applied to urine samples collected after administration of 17βN preparations and to urine samples from non-treated animals to identify endogenous compounds. The following metabolites were identified after the administration of 17βN: 17βN, NorA, NorE, NorEpia, BAB and ABB in boars and 17βN, 17αN, NorEpiA, ABA and ABB in horses. The endogenous compounds detected were: 17βN in boars and horses, and ABA and 17αN only in entire male horses. A chromatographic method by LC/MS/MS has been evaluated to quantify and 17βN sulfate in horse urine and it has been applied to analyze samples from non-treated and treated horses. Concentrations above 30 ng/ml of 17βN sulfate would be evidences of the exogenous administration of 17βN in entire male horses. Preliminary results indicate the capability of GC/C/IRMS analysis to differentiate the endogenous or exogenous origin of 17βN and their metabolites in boars and horses. / La 17β-nandrolona (17βN) s’usa il•legalment en l’esport i com a promotor del creixement en el bestiar. Tant la 17βN com alguns dels seus metabòlits s’han descrit com a compostos endògens en alguns animals, com els porcs adults i els cavalls mascles. L’objectiu de la tesi va ser identificar els marcadors de l’administració exògena de 17βN a porcs i cavalls. Es va optimitzar i validar un mètode basat en la CG/EM per quantificar i identificar els metabòlits de la 17βN en orina de porcs i cavalls en les diferents fraccions metabòliques i es va aplicar a l’anàlisis de mostres procedents d’estudis d’excreció de 17βN i de mostres procedents d’animals no tractats per identificar els compostos endògens. Els metabolits identificats després l’aministració de 17βN van ser: 17βN, NorA, NorE, NorEpiA, BAB i ABB en porcs i 17βN, 17αN, NorEpiA, ABA i ABB en cavalls. Els compostos endògens detectats van ser: 17βN per porcs i cavalls mascles i ABA i 17αN únicament pels cavalls. S’ha avaluat un mètode per CL/EM/EM per quantificar 17βN sulfat en orina de cavall que ha estat aplicat a l’anàlisi de mostres procedents de cavalls no tractats i tractats amb 17βN. Concentracions superiors a 30 ng/ml de 17βN sulfat en orina, seria indicació d’una administració exògena de 17βN. Resultats preliminars obtinguts pel mètode de GC/C/IRMS permeten indicar que mitjançant aquesta tècnica és possible diferenciar l’origen endogen o exogen de 17βN i els seus metabòlits en porcs i en cavalls.

In particular, the choice of promoters available needs to be broadened to include promoters which are active in conditions other than those conducive to induction of cellulase expression. Using proteomics, the HEX1 protein was identified as an abundant protein of the cell envelope of T. reesei when grown on a range of carbon sources, suggesting that a strong constitutive promoter drives the expression of this physiologically important protein. This thesis is an exploration into the hex1 gene promoter and the role of hex1 in the maintenance of mycelium integrity in T. reesei with consideration for the application of this gene in the further development of filamentous fungi as protein expression systems. -- The single copy hex1 gene and flanking regions were isolated from T. reesei and another biotechnologically important fungus, Ophiostoma floccosum. The fluorescent reporter protein DsRed1-E5 was expressed under the T. reesei hex1 promoter and promoter activity was monitored by fluorescence CLSM and RNA analysis. During the rapid growth phase of a culture, the hex1 promoter was active in a range of carbon sources and three transcipt types with alternative tsp and splicing sites were discovered for the hex1 gene. The distribution of fluorescence throughout the mycelium suggested spatial regulation of the hex1 promoter as well as temporal regulation. The promoter was continually active in the absence of a functional hex1 gene product suggesting that the hex1 promoter is regulated in part, by negative feedback from the endogenous gene product. Interruption of the hex1 gene produced hyphae that leaked excessive volumes of cytoplasm when physically damaged which may be advantageous for the externalisation of selected protein products. Cloning, characterisation and sequencing of promoters of Helicobacter pylori 4187E

Published information on the structure and regulation of H. pylori promoters is limited. The work presented in this thesis describes the cloning and characterisation of promoter regions from a clinical isolate of H. pylori, and the development of an alternative, non-radioactive method for verifying the location of transcriptional start sites of bacterial promoters. H. pylori 4187E promoters were randomly cloned into the promoter-trap vector pKK232-8 in Escherichia coli DH5α using two sets of restriction enzymes. Vector pKK232-8 contains a promoterless chloramphenicol acetyltransferase (CAT) gene. Seventy-four promoter-containing clones were isolated from selective media based on their resistance to chloramphenicol. The strength of each promoter was analysed qualitatively, using chloramphenicol minimum inhibitory concentrations, and quantitatively, using CAT assays following exposure of the clones to pH 4 and pH 7. Selected promoter fragments were subcloned into the GFP reporter vector pFPV25, containing a promoterless gfp gene. The subclones were exposed to buffered LB broth at pH 4, 5, 6, 7 and 8, for varying lengths of time, to study acid-induced regulation of gene expression. Subclones were examined qualitatively, using visual examination of GFP fluorescence and fluorescence microscopy, and quantitatively, using flow cytometry following acid shock. DNA sequences were determined for 61 of the 74 H. pylori promoters, and sequence alignments with the published H. pylori strains (26695 and J99) were performed. The transcriptional start site of 27 H. pylori promoter fragments was experimentally mapped using a fluorescence-based primer extension protocol developed by our group. Potential -35 and -10 sequences were identified for each promoter, and a new consensus sequence for H. pylori promoters was proposed based upon these results. This study has considerably expanded knowledge of H. pylori promoter sequences and transcriptional start sites based on those which also function in E. coli. It has also revealed several H. pylori promoters which appear to respond to acid stress

Orientador: Marcelo Carvalho Minhoto Teixeira Filho / Resumo: Inocular sementes de trigo com a bactéria Azospirillum brasilense pode propiciar a fixação biológica de nitrogênio (FBN), porém tem sido observado efeito mais pronunciado desta inoculação no crescimento inicial de plantas, assim, com o sistema radicular mais desenvolvido, a absorção de nutrientes e água pode ser maior. Com isso, o crescimento, produtividade e exportação de nutrientes da cultura do trigo podem ser maiores, porém a resposta a esta inoculacao pode variar conforme a afinidade da bacteria com os cultivares de trigo. Dessa forma, objetivou-se avaliar o acúmulo de matéria seca e extração de nutrientes em estádios fenológicos, exportação de nutrientes pelos grãos, componentes de produção e produtividade de grãos de cultivares de trigo inoculados ou não com Azospirillum brasilense. O experimento foi desenvolvido em Selvíria - MS, em um Latossolo Vermelho Distroférrico de textura argilosa, em sistema plantio direto. O delineamento experimental foi em blocos ao acaso com quatro repetições, com os tratamentos dispostos em esquema fatorial 3 x 2, sendo três cultivares de trigo (CD 116, IPR CATUARA TM e IAC 385), com ou sem inoculação de sementes por Azospirillum brasilense (300 ml ha-1 do produto, com estirpes Abv5 e Abv6 (garantia de 2x108 UFC mL-1).
