Studying low frequency vibrational modes using ultrafast techniques

In this thesis, I report on the investigation of the low frequency vibrational modes in a number of different systems using ultrafast spectroscopic techniques. These consist of biological systems, including the enzyme, morphinone reductase (MR) and the related biomolecules, riboflavin (Rb) and flavin mononucleotide (FMN), as well as non-biological systems, including the semiconductor gallium nitride (GaN) and gold nanoparticles (Au NPs). The term low frequency refers to terahertz (THz) frequencies, where vibrational modes exist at the molecular level, with molecular rotations, lattice vibrations and inter- and intra-molecular vibrations occurring in the THz spectral range. These vibrational modes occur on sub-picosecond timescales and therefore ultrafast techniques utilising femtosecond laser pulses provide a means of studying these modes, and are employed throughout this thesis. The two ultrafast techniques of transient absorption (TA) spectroscopy and terahertz time-domain spectroscopy (THz-TDS) were used. Firstly, a high-repetition rate transient absorption (HRRTA) spectrometer was commissioned to perform pump-probe measurements with an ultraviolet pump and broadband visible probe. The performance of the HRRTA spectrometer was benchmarked using Au NPs and used to investigate the existence of a promoting vibration in MR contributing to the catalysis process, predicted to occur at THz frequencies. Weak oscillations were detected in the charge-transfer absorption band of MR bound to the non-reactive cofactor 1,4,5,6-tetra-hydro-nicotinamide adenine dinucleotide (NADH4), with a frequency of approximately 1.5 THz and provide evidence of the first direct observation of a promoting vibration in an enzyme. To complement the TA measurements, THz-TDS was also used to obtain direct measurements of the absorption at THz frequencies. Due to the challenge of studying water-based biological samples, an initial investigation was performed on a wurtzite GaN wafer, which exhibited optical phonon modes in the THz frequency range that were found to determine the dielectric response of the semi-insulating semiconductor wafer. Use of a non-polar m-plane wafer allowed the anisotropic nature to be observed and values of 9.22 ± 0.02 and 10.32 ± 0.03 for the static dielectric constants were obtained for the THz electric field polarised both perpendicular and parallel to the c-axis of the wurtzite GaN wafer, respectively. Finally, biological studies using THz-TDS were performed with measurements on Rb pellets and films revealing vibrational modes in the THz region. The sharp absorption features were not observed in FMN, despite a small difference in molecular content from Rb, and dehydration was required to reveal small amplitude absorption features. Final measurements on MR and MR-NADH4 films were carried out and evidence of absorption features in the THz frequency range were observed, however further work is required to determine the precise origin of these features.

Cooperative Games: Promoting Prosocial Behaviors in Children

Children who develop more prosocial behaviors tend to be more competent socially than those children who develop fewer prosocial behaviors. Group games are especially effective in the facilitation of prosocial behaviors. This study compared the number of prosocial or positive behaviors and negative behaviors displayed during cooperatively and competitively structured game treatments using the Observational Checklistand the Teacher Checklist. We controlled for possible differences in teacher nuturance through the Caregiver Interaction Scale. Participants included 20 boys and 19 girls (mean age = 4 years 7.3 months) enrolled in one of two classes at Utah State University s Adele and Dale Young Child Development Lab. There were no statistically significant effects of treatment found according to The Teacher Checklist; however, statistically significant differences in positive and negative behaviors were found on The Observational Checklist across treatment conditions. Specifically, after cooperative games, positive behaviors were higher than expected while negative behaviors were lower than expected . During competitive games , positive behaviors were lower than expected and negative behaviors were higher than expected. When the two factors on The Teacher Checklist, Aggression and Immaturity, were analyzed, no statistically significant relationships were found.

<p>The aim of this study was to investigate how a workplace should be formed to promote health according to the employees. The study was made in IKEA handla hemma in Älmhult, Sweden. The aim of the study was to find out why people experience health at their workplace environment, from a salutogenetic point of view. The study is based on 12 strategically selected interviews and where analysed according to Grounded Theory. This resulted in a model where the core category was “The prevailing spirit of the organisation” and the five main categories was “That the organisation creates favourable conditions to promote health”, “That there is a god atmosphere amongst the colleagues and the bosses”, “The personal point of view about health”, “The relationship to goals and responsibility” and “The personal point of view about their surroundings”. The result shows that it’s important to look upon health in its entirety. Health isn’t stronger than its weakest point. The findings should work as a guide in the on going work with the employees at IKEA handla hemma. For the future it would be interesting to do a study in a workplace with worse health and then compare them with each other.</p>

The empirical research reported in this thesis is along-term case study carried out at Volvo Car Corporation. Inthe licentiate thesis that preceded this doctoral thesis, itwas suggested that assembly work should be based on modules.The further topics researched in this thesis includeexamination of a modularisation process, examination of thecommunication interface between the assembly organisation andthe product development organisation, the effects ofmodularisation on the assembly process, and an investigation ofthe development of two assembly efficient products. The product of the research is a model representing theorganisational prerequisites for developing competence in thecreation of assembly efficient products. The key elements inthe model are a transparent organisational structure, clearintention in the organisation, and accommodation ofcross-functional exchange. Modularisation is a means ofcreating a transparent organisational structure that makes itpossible for members of the organisation to understand thecontext in which they work, which is a basic requirement fordeveloping competence. A structure for cross-functionalcooperation should be put in place to facilitatecross-functional exchange and learning. To succeed indeveloping assembly efficient products, an organisation mustalso clear signal that this is its intention. Visions, goalsand strategies must express this ambition. HUR SAMTAL OM SEXUALITET FRÄMJAS BLAND SJUKSKÖTERSKOR : En litteraturöversikt / HOW CONVERSATIONS ABOUT SEXUALITY IS PROMOTED AMONG NURSES : A literature review

Bakgrund: Patienter som upplever sexuell ohälsa känner ofta ett behov av information och vill prata om sina problem med vårdpersonal. Sjuksköterskor är ansvariga att hantera sexuellt relaterade frågor och problem, men trots att det förväntas av dem att initiera dessa samtal, visar forskning att det sällan sker. Syfte: Att sammanställa och belysa forskning som beskriver hur samtal om sexualitet med patienter främjas bland sjuksköterskor. Metod: Litteraturöversikt. Resultat: I analysen framkom sju teman som beskriver hur samtal om sexualitet främjas bland sjuksköterskor, nämligen genom; Att ha kunskap, Att vara bekväm och ansvarstagande, Att ha en god patientrelation, Motiverande patientsituationer, Att ha en god arbetsmiljö, Hög jobbposition och utbildning samt Att ha erfarenhet. Diskussion: Resultatet visar ett antal olika faktorer som främjar samtal om sexualitet mellan sjuksköterska och patient, men endast ett fåtal med tillräcklig reliabilitet för att kunna utgöra en generaliserbar slutsats. Faktorer som ålder, erfarenhet, jobbposition och civilstånd visar osäkra resultat med låg kongruens. Kunskap, utbildning och att känna sig bekväm med ämnet är de huvudsakliga faktorer som främjar sjuksköterskans förmåga att samtala om sexualitet med patienter. / Background: A persons sexual health may be affected by a number of diseases and different treatments, for example cancer and chemotherapy. Patients who experience sexual health problems often feel a need for information and want to talk about their illness with a health professional.. Nurses have a responsibility to deal with patients’ sexual issues and problems, however, even though expectations lies with them to initiate the conversation, studies reveal it rarely happens. Method: Litterature review. Objective: To compile and highlight research that describes how conversations about sexuality with patients is promoted among nurses. Results: The analysis revealed seven themes that describe how conversations about sexuality is promoted among nurses, namely by: Having knowledge, Being comfortable and responsible, Having a good patient relationship, Motivational patient situations, Having a good working environment, High job position and education and Having experience. Discussion: The results show a number of different factors that promotes nurse-patient conversations about sexuality, although, only a few with sufficient reliability to be a generalisable conclusion. Factors such as, age, experience, job position and marital status show uncertain results with low congruence. Knowledge, education and to feel comfortable with the topic are the main factors underpinning nurses’ ability to discuss sexuality with patients.

Sequence analysis of the genome of the plant growth-promoting bacterium Pseudomonas putida UW4

The plant growth-promoting bacterium (PGPB) Pseudomonas putida UW4, previously isolated from the rhizosphere of common reeds growing on the campus of University of Waterloo, promotes plant growth in the presence of different environmental stresses, such as flooding, high concentration of salt, cold, heavy metals, drought and phytopathogens. The known mechanisms used by P. putida UW4 to promote plant growth include 1-aminocyclopropane-1-carboxylate (ACC) deaminase, indole-3-acetic acid (IAA) synthesis and siderophore production. In this work, the genome sequence of UW4 was obtained by pyrosequencing and the gaps between the contigs were closed by directed PCR. The P. putida UW4 genome contains a single circular chromosome that is 6,183,388 bp with a 60.05% G+C content. The bacterial genome contains 5,431 predicted protein-coding sequences that occupy 87.4% of the genome. Nineteen genomic islands were predicted and thirty one complete putative insertion sequences were identified. Genome analyses were conducted in order to better characterize the general features of the UW4 genome. Genes potentially involved in plant growth promotion such as IAA biosynthesis, trehalose production, siderophore production, and acetoin synthesis were identified, which will facilitate a better understanding of the mechanisms of plant-microbe interactions. Moreover, genes that contribute to the environmental fitness of UW4 were also determined including genes responsible for heavy metal resistance such as nickel, copper, cadmium, zinc, molybdate, cobalt, arsenate, and chromate. Central metabolic pathways helped elucidate the physiological roles of diverse metabolites of UW4. Unexpectedly, whole-genome comparison with other completely sequenced Pseudomonas sp. revealed that UW4 is more similar to the fluorescens group rather than to the putida group. More surprisingly, a putative type III secretion system (T3SS) was found in the UW4 genome, and T3SS was thought to be essential for bacterial pathogenesis. Although putative T3SS was observed in other non-pathogenic Pseudomonas spp. previously, this is the first report indicating that a T3SS in a Pseudomonas sp. is highly similar to the one from Salmonella spp.

Plant Growth-Promoting Bacterial Endophytes that contain ACC Deaminase: Isolation, Characterization, and Use

Bacteria that provide benefit to plants are considered to be plant growth-promoting bacteria (PGPB) and can facilitate plant growth by a number of different mechanisms. Plant growth-promoting bacteria that are able to utilize the plant compound 1-aminocyclopropane-1-carboxylate (ACC) as a sole source of nitrogen, as a consequence of possessing the enzyme ACC deaminase, can protect host plants from a number of environmental stresses. In addition to ACC deaminase, PGPB may utilize other mechanisms to facilitate plant growth including IAA synthesis, siderophore production, phosphate solubilization activity, ammonia production, and antibiotic production. Plant growth-promoting bacterial endophytes employ similar plant growth promotion mechanisms to those used by rhizospheric PGPB. In fact, bacterial endophytes are PGPB that go one step further and colonize the inside of the plant tissues and provide more efficient and prompted protection to their hosts compared to those that bind exclusively to the plant’s rhizosphere. Therefore, it is likely that endophytic plant growth-promoting bacteria will be superior to similar non-endophytic bacterial strains in promoting plant growth under a wide range of environmental conditions. In the work reported here, new bacterial endophytes were isolated and characterized. Among twenty-five ACC deaminase positive strains, two best strains were selected and ACC deaminase deficient mutants were constructed. The ability of two newly isolated 1-aminocyclopropane-1-carboxylate (ACC) deaminase-containing plant growth-promoting bacterial endophytes Pseudomonas fluorescens YsS6, Pseudomonas migulae 8R6 and their ACC deaminase deficient mutants was shown to 1) delay the senescence of mini carnation cut flowers and 2) to facilitate tomato plant growth under salinity stress. In the mini carnation flower senescence evaluation, the only difference between wild-type and mutant bacterial endophytes was ACC deaminase activity, our results demonstrate that this enzyme is directly responsible for a significant delay in flower senescence. Despite containing ACC deaminase activity, the rhizosphere-binding PGPB Pseudomonas putida UW4 was not taken up by the cut flowers and therefore had no effect on prolonging flower shelf life. In evaluating the effect of bacterial endophytes under salt stress, tomato plants treated with either of the wild-type strains of the two selected bacterial endophytes demonstrated early flowering and fruiting and had significantly greater numbers of flowers, buds, and fruits than either the corresponding ACC deaminase mutant strain-treated plants or the control plants. Although both bacterial endophytes P. fluorescens YsS6 and P. migulae 8R6 showed significant plant growth-promotion capabilities, P. migulae 8R6 demonstrated better plant growth facilitation under salt stress than did P. fluorescens YsS6. P. migulae 8R6 treated tomato plants demonstrated the least sodium uptake, the highest chlorophyll content, and highest fresh and dry biomass. The results of the work presented here suggest that ACC deaminase containing selected bacterial endophytes could be employed as environmentally friendly adjuncts to agricultural and horticultural practice.

Health-promoting behaviors in Thai persons with chronic renal failure

The purpose of this cross-sectional, descriptive, correlational study was to describe the relationships among demographic factors (age, gender, education, income), perceived severity of illness, perceived barriers to action, perceived self-efficacy, and interpersonal influences (social support) and health-promoting behaviors (HPB) and to identify predictors of HPB. A nonprobability sample of 110 participants with Chronic Renal Failure (CRF) was recruited from the outpatient clinic of Burirum hospital and Surin hospital located in north-eastern Thailand. All participants were individually interviewed by the principal investigator in a private area within an outpatient clinic. Six variables were significantly correlated with the HPB. They were age, education, perceived severity of illness, perceived barriers to action, perceived self-efficacy and social support. According to the results from the analyses of demographic data and HPB, participants who were younger and had higher educational levels practiced more HPB. From the analyses of perceived severity of illness, perceived barriers to action, perceived self-efficacy, and social support and HPB, the results showed that participants with lower levels of perceived severity of illness and lower levels of perceived barriers to action reported better HPB. In contrast, the participants with higher levels of perceived self-efficacy and social support reported better HPB. By using a stepwise multiple regression analysis, two predictors were identified from 8 predictor variables, and those two accounted for 78.2% (p < 0.01) of the variance in HPB. Two variables that contributed significantly to the variance in the HPB were perceived self-efficacy ([beta] =.769, p < 0.01), and social support ([beta] = .162, p < 0.01); whereas age, gender, income, educational level, perceived severity of illness, and perceived barriers did not contribute to the variance in the HPB. The study found that participants who experienced better perceived self-efficacy and social support reported better HPB. In contrast age, gender, income, educational level, perceived severity of illness, and perceived barriers did not enter as predictors in this stepwise regression equation. / text
